背景技术

然而,采用红豆杉种子育苗,因种子在育苗前需要经过长达两年的休眠及层积二道关,种苗生产周期长;同时,种子育苗可能会导致红豆杉发生劣变,从而影响红豆杉原有优良性状的保持。而采用红豆杉枝条扦插育苗,可以缩短种苗生产周期;同时,能够保持母株的优良性状,避免发生劣变,能够较快较多地育成良种苗木。扦插枝条的生根状况是影响红豆杉幼苗是否能存活的主要因素。用于扦插枝条生根的植物生长调节剂作为一种新兴的生物技术,正向高效、低毒、生物活性高等方向发展。而传统的单一生根剂使那些难以生根的植物在枝条扦插或苗木移栽时不能得到较为满意的生根效果。
发明内容
本发明的目的是针对上述现状,提供一种能有效提高生根率的用于红豆杉枝条扦插生根的生根剂配方及其制备方法。
一种用于红豆杉枝条扦插生根的生根剂配方,其包括:按各组分质量计算,吲哚丁酸200~300mg、萘乙酸200~300mg、茉莉酸甲酯20~30mg、冠菌素0.1~0.5mg、苯丙氨酸10~20mg、水杨酸10~20mg、维生素B250~100mg、维生素B65~10mg、维生素B1250~100mg、维生素C 1~5mg、维生素H 1~5mg、蔗糖2~5g。
一种用于红豆杉枝条扦插生根的生根剂制备方法,其包括以下步骤:
按各组分质量计算,提供吲哚丁酸200~300mg、萘乙酸200~300mg、茉莉酸甲酯20~30mg、冠菌素0.1~0.5mg、苯丙氨酸10~20mg、水杨酸10~20mg、维生素B250~100mg、维生素B65~10mg、维生素B1250~100mg、维生素C 1~5mg、维生素H 1~5mg、蔗糖2~5g;
将吲哚丁酸、萘乙酸、茉莉酸甲酯,冠菌素用乙醇溶解,然后加入200~500mL蒸馏水,再加上其他原料混合均匀;
将混合溶液和1~1.5%吐温80的水溶液以1∶9的比例混合均匀,使溶液总体积达到2~5L,将混合溶液在25~30℃下进行超声处理,超声功率为100~150W,超声时间为60~90min,当溶液呈现白色乳光,且清澈无沉淀,即得红豆杉枝条生根剂。
本发明采用的技术方案,其有益效果在于:(1)本生根剂能有效提高扦插枝条中的叶绿素含量,保证扦插枝条有足够的光合色素进行光合作用,植物生长所需的营养物质的合成也随之增加,直接引起可溶性糖含量的增加,并使蛋白质的合成增多,为生长恢复时期的插条提供了生长的物质基础;(2)本生根剂还能提高扦插枝条的过氧化物酶(POD)、多酚氧化酶(PPO)、吲哚乙酸氧化酶(IAAO)活性,有利于不定根的表达,促进根的伸长生长;同时本生根剂能有效提高扦插枝条的超氧化物歧化酶(SOD)活性,提高插条的防御能力,利于提高其成活率;(3)采用纳米混悬技术,有效提高生根剂的饱和溶解度和溶出速率,提高其在红豆杉体内的利用度。

各原料的介绍如下:
吲哚丁酸是一种内源生长素,主要用于插条生根,能促进体内过氧化物酶、多酚氧化酶、吲哚乙酸氧化酶的等酶的活性表现,从而促进细胞的脱分化,产生愈伤组织,有利于新根生成和维管束系统的分化,促进插条不定根的形成。广泛应用于树木、花卉的扦插生根。
萘乙酸是一种植物生长调节剂,能够促进插条、圈枝生根,提高成活率,加速植株生长发育,促进分蘖,增强抗性。萘乙酸促发的根数量少但较粗壮,与其他促进剂混合使用能使受体更多更好的生根。
茉莉酸甲酯是一种在植物体内普遍存在的植物生长物质,具有诱导植物防御基因的表达,诱导多种次生代谢产物的合成,抑制光合作用,促进呼吸作用、气孔关闭、叶片衰老以及果实成熟等广泛的生理功能。
水杨酸是一种具有生理调节功能的新型植物激素,不仅直接参与植物的生长发育,还可以通过影响其它内源植物激素的水平来调节植物的生长发育。
冠菌素是假单胞菌产生的一种致病毒素,可以提高植物的抗病性、抗旱性,提高植物产生乙烯的量,对植物具有极大生理活性调节功能。
苯丙氨酸是植物一种必需氨基酸,它可以通过参与蛋白质的合成和作为碳源来影响细胞的生长。在植物细胞的次生代谢中,苯丙氨酸又是一个很重要的代谢中间体,它是合成一些生物碱、木质素、黄酮、异黄酮及香豆素等次生物质的重要前体。
蔗糖作为一种重要的碳源,在根的形成和生长方面,不仅作为碳水化合物提供营养,而且像激素一样可以诱导根的形成和促进根的生长。
维生素B2、维生素B6和维生素B12均有利于促进根的伸长。维生素C在扦插中也有促进生根作用。维生素H可以促进生根必需的生穗基部呼吸作用,使插穗内部的养分转化为可给状态之外,还有消毒灭菌、抑制不良微生物繁殖、促进生根的作用。

S1:按各组分质量计算,提供吲哚丁酸200~300mg、萘乙酸200~300mg、茉莉酸甲酯20~30mg、冠菌素0.1~0.5mg、苯丙氨酸10~20mg、水杨酸10~20mg、维生素B250~100mg、维生素B65~10mg、维生素B1250~100mg、维生素C 1~5mg、维生素H 1~5mg、蔗糖2~5g;
S2:将吲哚丁酸、萘乙酸、茉莉酸甲酯,冠菌素用乙醇溶解,然后加入200~500mL蒸馏水,再加上其他原料混合均匀;
S3:将混合溶液和1~1.5%吐温80的水溶液以1∶9的比例混合均匀,使溶液总体积达到2~5L,将混合溶液在25~30℃下进行超声处理,超声功率为100~150W,超声时间为60~90min,当溶液呈现白色乳光,且清澈无沉淀,即得红豆杉枝条生根剂。
试验例:
枝条的选择:选用红豆杉当年生半木质化的枝条,剪成长度约6-8cm的扦插枝条,扦插枝条靠下部2cm处,减去脚叶,将扦插枝条扎把捆住,1捆50株,待用;
苗床基质处理:苗床基质采用沙子+草炭土+珍珠盐,采用高效低毒广谱杀虫剂及杀菌剂对苗床进行消毒,备用;
浸泡枝条:将红豆杉扦插枝条,共80捆,在红豆杉枝条生根剂中浸泡5分钟;
扦插:将浸泡好的枝条扦插在处理好的苗床上,扦插深度为2cm;
苗床管理:保持苗床内温度不超过25℃,湿度保持在70%,按时喷水及叶面肥,补充水分及营养,定期做杀菌处理。
3个月后,观察扦插枝条的生根率,经统计,红豆杉扦插枝条的生根率在85%以上,部分扦插枝条的根部已木质化。10个月后,对其幼苗进行移栽,移栽的成活率在95%以上。
本发明采用的技术方案,其有益效果在于:(1)本生根剂能有效提高扦插枝条中的叶绿素含量,保证扦插枝条有足够的光合色素进行光合作用,植物生长所需的营养物质的合成也随之增加,直接引起可溶性糖含量的增加,并使蛋白质的合成增多,为生长恢复时期的插条提供了生长的物质基础;(2)本生根剂还能提高扦插枝条的过氧化物酶(POD)、多酚氧化酶(PPO)、吲哚乙酸氧化酶(IAAO)活性,有利于不定根的表达,促进根的伸长生长;同时本生根剂能有效提高扦插枝条的超氧化物歧化酶(SOD)活性,提高插条的防御能力,利于提高其成活率;(3)采用纳米混悬技术,有效提高生根剂的饱和溶解度和溶出速率,提高其在红豆杉体内的利用度。

Claims (8)

1.一种用于红豆杉枝条扦插生根的生根剂配方,其包括:按各组分质量计算,吲哚丁酸200~300mg、萘乙酸200~300mg、茉莉酸甲酯20~30mg、冠菌素0.1~0.5mg、苯丙氨酸10~20mg、水杨酸10~20mg、维生素B2 50~100mg、维生素B6 5~10mg、维生素B12 50~100mg、维生素C 1~5mg、维生素H 1~5mg、蔗糖2~5g。
2.如权利要求1所述的用于红豆杉枝条扦插生根的生根剂配方,其特征在于:按各组分质量计算,吲哚丁酸200mg、萘乙酸200mg、茉莉酸甲酯20mg、冠菌素0.1mg、苯丙氨酸10mg、水杨酸10mg、维生素B2 50mg、维生素B6 5mg、维生素B12 50mg、维生素C 1mg、维生素H 1mg、蔗糖2g。
3.如权利要求1所述的用于红豆杉枝条扦插生根的生根剂配方,其特征在于:按各组分质量计算,吲哚丁酸300mg、萘乙酸300mg、茉莉酸甲酯30mg、冠菌素0.5mg、苯丙氨酸20mg、水杨酸20mg、维生素B2 100mg、维生素B6 10mg、维生素B12 100mg、维生素C 5mg、维生素H 5mg、蔗糖5g。
4.如权利要求1所述的用于红豆杉枝条扦插生根的生根剂配方,其特征在于:按各组分质量计算,吲哚丁酸250mg、萘乙酸250mg、茉莉酸甲酯25mg、冠菌素0.3mg、苯丙氨酸15mg、水杨酸15mg、维生素B2 75mg、维生素B6 7.5mg、维生素B12 75mg、维生素C 3mg、维生素H 3mg、蔗糖3.5g。
5.一种用于红豆杉枝条扦插生根的生根剂制备方法,其包括以下步骤:
按各组分质量计算,提供吲哚丁酸200~300mg、萘乙酸200~300mg、茉莉酸甲酯20~30mg、冠菌素0.1~0.5mg、苯丙氨酸10~20mg、水杨酸10~20mg、维生素B2 50~100mg、维生素B6 5~10mg、维生素B12 50~100mg、维生素C 1~5mg、维生素H 1~5mg、蔗糖2~5g;
将吲哚丁酸、萘乙酸、茉莉酸甲酯,冠菌素用乙醇溶解,然后加入200~500mL蒸馏水,再加上其他原料混合均匀;
将混合溶液和1~1.5%吐温80的水溶液以1∶9的比例混合均匀,使溶液总体积达到2~5L,将混合溶液在25~30℃下进行超声处理,超声功率为100~150W,超声时间为60~90min,当溶液呈现白色乳光,且清澈无沉淀,即得红豆杉枝条生根剂。
6.如权利要求5所述的用于红豆杉枝条扦插生根的生根剂制备方法,其特征在于:按各组分质量计算,吲哚丁酸200mg、萘乙酸200mg、茉莉酸甲酯20mg、冠菌素0.1mg、苯丙氨酸10mg、水杨酸10mg、维生素B2 50mg、维生素B6 5mg、维生素B12 50mg、维生素C 1mg、维生素H1mg、蔗糖2g。
7.如权利要求5所述的用于红豆杉枝条扦插生根的生根剂制备方法,其特征在于:按各组分质量计算,吲哚丁酸300mg、萘乙酸300mg、茉莉酸甲酯30mg、冠菌素0.5mg、苯丙氨酸20mg、水杨酸20mg、维生素B2 100mg、维生素B6 10mg、维生素B12 100mg、维生素C 5mg、维生素H 5mg、蔗糖5g。
8.如权利要求5所述的用于红豆杉枝条扦插生根的生根剂制备方法,其特征在于:按各组分质量计算,吲哚丁酸250mg、萘乙酸250mg、茉莉酸甲酯25mg、冠菌素0.3mg、苯丙氨酸15mg、水杨酸15mg、维生素B2 75mg、维生素B6 7.5mg、维生素B12 75mg、维生素C 3mg、维生素H 3mg、蔗糖3.5g。
