技术领域
本发明属于水产养殖技术领域,尤其涉及一种治疗泰国斗鱼打印病的环保制剂;本发明还涉及一种上述治疗泰国斗鱼打印病的环保制剂的制备方法。
背景技术
随着人们的生活水平的提高,物质生活极大的丰富,人们对休闲娱乐生活的要求越来越明显,在家里或庭院中养些色彩艳丽的观赏鱼是最好的选择,因为饲养观赏鱼几乎不占用空间;经济费用可以依人而定;条件相对简单,有水即可。养殖观赏鱼不仅可以修身养性,陶冶情操,水中所释放出来的负离子和水蒸气还可过滤和湿润空气,给人营造出良好的生活环境。
泰国斗鱼,原产于泰国,又被称为五彩搏鱼。和其它斗鱼品种一样,环境的适应能力很强,外形艳丽夸张,属于比较容易饲养的观赏鱼之一。由于泰国斗鱼雄鱼的好斗性,在很早以前,就有很多的地方将它视为娱乐的一种工具。泰国斗鱼以好斗闻名,两雄相遇必定来场决斗,相斗时张大腮盖和鱼鳍,用身体互相冲击挑衅,然后用嘴互相撕咬。因此在饲养中,不能把2尾以上的成年雄鱼放养1缸。
打印病又称腐皮病,其病原体为点状产气单胞菌的点状亚种,属革兰氏阴性菌。斗鱼患打印病后的主要症状为病鱼通常在肛门附近的两侧或尾柄部位(极少数在身体前部)、皮肤、肌肉开始发炎,出现红斑,有时似脓疱状,随着病情发展,该部位的鳞片脱落,肌肉逐渐腐烂,形成边缘充血发红、呈圆形或椭圆形病灶,好象打上一个红色印记,故称之为打印病。病鱼身体瘦弱,游泳迟钝;发病严重时,可陆续出现死亡。当水质不清洁或鱼体受损伤时,常会感染此病。没有明显的流行季节。目前,针对斗鱼的打印病尚无专门的药物治疗,通常采用的手段是用2克/立方米水体呋喃唑酮药浴鱼体30分钟对病鱼进行消毒,然而其治疗效果有限,毒性较大,对环境容易造成污染,而且操作较为繁琐,操作过程极易造成斗鱼的鳞片脱落,在杀菌去病的同时对斗鱼也造成了较大的损伤。
发明内容
本发明的目的之一是提供一种治疗泰国斗鱼打印病的环保制剂,其原料组分均为环保的天然原料,对环境无污染,对泰国斗鱼打印病治愈率高,使用方便、施药过程中对斗鱼的伤害小、斗鱼的健康恢复快。
为了解决上述技术问题,本发明采用以下技术方案,一种治疗泰国斗鱼打印病的环保制剂,由下列质量份数的各组分组成:茶水27-35份、白薇20-26份、马齿苋19-25份、土荆芥19-25份、皂荚19-25份、牡丹皮17-23份、茵陈草17-23份、鱼腥草15-23份、夏枯草16-22份、五倍子15-21份、雷丸14-22份、槟榔14-20份、蜈蚣12-20份、紫云英13-19份、荠菜13-18份、茜草12-19份、胡粉11-19份、生姜10-19份、大蒜10-17份、蝎子9-17份、车前草9-17份、竹盐8-16份、白醋8-15份。
优选地,所述治疗泰国斗鱼打印病的环保制剂,由下列质量份数的各组分组成:茶水30-32份、白薇22-24份、马齿苋21-23份、土荆芥21-23份、皂荚21-23份、牡丹皮19-21份、茵陈草19-21份、鱼腥草18-20份、夏枯草18-20份、五倍子18-19份、雷丸17-19份、槟榔16-18份、蜈蚣15-17份、紫云英15-17份、荠菜15-16份、茜草14-15份、胡粉14-15份、生姜13-16份、大蒜13-15份、蝎子12-14份、车前草12-14份、竹盐11-13份、白醋11-12份。
优选地,所述治疗泰国斗鱼打印病的环保制剂,由下列质量份数的各组分组成:茶水31份、白薇23份、马齿苋22份、土荆芥22份、皂荚22份、牡丹皮20份、茵陈草20份、鱼腥草19份、夏枯草19份、五倍子18份、雷丸18份、槟榔17份、蜈蚣16份、紫云英16份、荠菜16份、茜草15份、胡粉15份、生姜14份、大蒜14份、蝎子13份、车前草13份、竹盐12份、白醋11份。
本发明的另一目的是提供一种上述治疗泰国斗鱼打印病的环保制剂的制备方法,该治疗泰国斗鱼打印病的环保制剂的制备方法包括以下步骤:
(1)按配比称取白薇、马齿苋、土荆芥和皂荚,将白薇、马齿苋、土荆芥和皂荚的总重量记作1份,称取8份的纯化水,70-80℃下水浴浸泡3-5h,得浸泡液;
(2)将步骤(1)得到的浸泡液加热至沸腾后煮沸2h,分离取出煎出液,继续向药材内加入4份的纯化水,加热至沸腾后继续煮沸2h,合并两次煎出液即为提取液;
(3)按上述配比分别称取牡丹皮、茵陈草、鱼腥草、夏枯草、五倍子、雷丸、槟榔、蜈蚣、紫云英、荠菜、茜草、胡粉、生姜、大蒜、蝎子和车前草,装入球磨机研磨2h,过80目筛,得研磨粉;
(4)将步骤(2)得到的提取液及步骤(3)得到的研磨粉放置于超临界萃取仪中的萃取釜中,利用二氧化碳作为介质进行萃取,萃取釜压力为40Mpa,萃取温度为45℃,分离器压力12Mpa,分离器温度为70℃,萃取2小时,得到萃取液;
(5)按照配比称取茶水、竹盐和白醋,与步骤(4)所得的萃取液混合均匀,即得目标物。
所述茶水的制备方法为:将绿茶与开水以1:20的重量比冲泡30分钟后,过滤提取所得的滤液即为茶水。
本发明在治疗泰国斗鱼打印病时的使用方法如下:每吨水倒入60-90ml的本发明的制剂,每12小时换水一次,换水后按相同比例加入本发明的制剂,连续使用直至病鱼治愈。
本发明在预防泰国斗鱼打印病时的使用方法如下:每吨水倒入30-40ml的本发明的制剂,24小时后换水一次,换水后按相同比例加入本发明的制剂,连续使用3天后,停1天;如此循环6-8个周期。

泡制茶水所用的绿茶中所含的茶多酚有较强的收敛作用,对病原菌、病毒有明显的抑制和杀灭作用,对消炎止泻有明显效果。白薇别名薇草、知微老、老瓜瓢根、山烟根子、百荡草、白马薇、白前、老君须,萝藦科、鹅绒藤属直立多年生草本,根须状,有香气,有除虚烦、清热散肿、生肌止痛之效,可治产后虚烦呕逆,小便淋沥,肾炎,尿路感染,水肿,支气管炎和风湿性腰腿痛等。马齿苋为马齿苋科一年生草本植物,全草供药用,有清热利湿、解毒消肿、消炎、止渴、利尿作用。土荆芥别名红泽蓝、天仙草、臭草、钩虫草、虱子草等,土荆芥可祛风除湿,杀虫,止痒,能用于蛔虫病,钩虫病,蛲虫病,外用治皮肤湿疹,瘙痒,并杀蛆虫。皂荚具有杀虫、解毒的作用:皂荚中所含有的皂苷素是三萜烯类和低聚糖,有消炎、抗溃疡、抗病变效果,还有抗癌和提高艾滋病免疫力等功效。牡丹皮为毛茛科植物牡丹干燥根皮,味苦、辛,性微寒,具有清热凉血、活血化淤、退虚热等功效,能用于治疗跌扑伤痛,痈肿疮毒。茵陈草又名:蛇蒿草、龙蒿草、白蒿草、他力干,为菊科多年生草本植物,茵陈以果穗精油含量较多,主要成分有B一旅烯、茵陈炔酮、茵陈原色酮、茵陈烯酮等,性微寒、味苦辛,具有清湿热、退黄疽、解毒疗疮的作用,长期饮用,保肝利胆,有直接阻碍肝胆系统肿瘤细胞增殖的功效。鱼腥草具有清热解毒,消痈排脓,利尿通淋的作用。大青叶主治热毒发斑、丹毒、咽喉肿痛、口舌生疮、疮痈肿毒等症。夏枯草的煎剂、水浸出液、乙醇-水浸出液及乙醇浸出液均可明显降低实验动物血压,茎、叶、穗及全草均有降压作用,但穗的作用较明显;夏枯草的水煎醇沉液小鼠腹腔注射,有明显的抗炎作用;夏枯草的煎剂在体外对痢疾杆菌、伤寒杆菌、霍乱弧菌、大肠杆菌、变形杆菌、葡萄球菌及人型结核杆菌均有一定的抑制作用。五倍子性寒,味酸、涩,具有敛肺降火、涩肠止泻、敛汗止血、收湿敛疮等功效,主治肺虚久咳、久泻久痢、便血痔血、痈肿疮毒、皮肤湿烂等症。雷丸,中药名,为白蘑科真菌雷丸的干燥菌核,可以杀虫消积消肿瘤,用于治疗肿瘤,绦虫病,钩虫病,蛔虫病,虫积腹痛,小儿疳积。槟榔为棕榈科槟榔属的一个种,槟榔具有“杀虫,破积,降气行滞,行水化湿”的功效,曾被用来治疗绦虫、钩虫、蛔虫、绕虫、姜片虫等寄生虫感染。蜈蚣为陆生节肢动物,能退炎治疮,用于炎肿疮疡,可快速消肿、止痛。紫云英主治破血、止血生肌,可利养五脏,明耳目,祛热风,令人轻健,久食不厌,补人,能止热疟,活血平胃。荠菜具有和脾、利水、止血、明目的功效,常用于治疗产后出血、痢疾、水肿、肠炎、胃溃疡、感冒发热、目赤肿疼等症。茜草味苦,性寒,归肝经,有凉血止血,化瘀,通经的作用,主治血热吐衄,崩漏下血,血瘀经闭,跌打损伤,风湿痹痛,由于茜草有活血通经的作用,临床上常用来治疗风湿痹痛。胡粉为原生质毒,能破坏癌细胞的生活能力,能直接杀灭寄生虫,有制止出血与粘液分泌的作用。生姜具有杀菌抑菌作用,且能增强消化能力。大蒜解毒杀虫,消肿止痛,止泻止痢,治肺,驱虫,此外还有温脾暖胃,治痈疽肿毒,白秃癣疮,痢疾泄泻,肺痨顿咳,蛔虫蛲虫,饮食积滞,脘腹冷痛,水肿胀满。蝎子可以消炎杀菌,能治中风、口眼歪斜、半身不遂后遗症,可以用于治疗皮肤湿疹。车前草又名车轮菜,为车前科多年生草本,不仅可以药用,而且可以食用,其能清热利尿,凉血,解毒,主治热结膀胱,小便不利,淋浊带下,暑湿泻痢,衄血,尿血,肝热目赤,咽喉肿痛,痈肿疮毒。竹盐是将日晒盐装入三年生的青竹中,两端以天然黄土封口,以松树为燃料,经1000℃~1500℃高温煅烧后提炼出来的物质,具有抗菌、消炎、中和毒素的作用。白醋有散瘀、解毒、消肿的功用。

此外,本发明使用方便,使用时,只需按照施药比例向水族箱中泼洒即可,无需将患病的斗鱼捞出上药,因此也减少了打捞斗鱼过程中和给斗鱼上药过程中给斗鱼造成的损伤。

实施例1
本发明第一个具体实施方式中饲料成分及其制备方法如下:
一种治疗泰国斗鱼打印病的环保制剂,由下列质量份数的各组分组成:茶水32份、白薇24份、马齿苋23份、土荆芥23份、皂荚23份、牡丹皮21份、茵陈草21份、鱼腥草20份、夏枯草20份、五倍子19份、雷丸19份、槟榔18份、蜈蚣17份、紫云英17份、荠菜16份、茜草15份、胡粉15份、生姜16份、大蒜15份、蝎子14份、车前草14份、竹盐13份、白醋12份。
上述治疗泰国斗鱼打印病的环保制剂的制备方法包括以下步骤:
(1)按配比称取白薇、马齿苋、土荆芥和皂荚,将白薇、马齿苋、土荆芥和皂荚的总重量记作1份,称取8份的纯化水,70-80℃下水浴浸泡3-5h,得浸泡液;
(2)将步骤(1)得到的浸泡液加热至沸腾后煮沸2h,分离取出煎出液,继续向药材内加入4份的纯化水,加热至沸腾后继续煮沸2h,合并两次煎出液即为提取液;
(3)按上述配比分别称取牡丹皮、茵陈草、鱼腥草、夏枯草、五倍子、雷丸、槟榔、蜈蚣、紫云英、荠菜、茜草、胡粉、生姜、大蒜、蝎子和车前草,装入球磨机研磨2h,过80目筛,得研磨粉;
(4)将步骤(2)得到的提取液及步骤(3)得到的研磨粉放置于超临界萃取仪中的萃取釜中,利用二氧化碳作为介质进行萃取,萃取釜压力为40Mpa,萃取温度为45℃,分离器压力12Mpa,分离器温度为70℃,萃取2小时,得到萃取液;
(5)按照配比称取茶水、竹盐和白醋,与步骤(4)所得的萃取液混合均匀,即得目标物。
所述茶水的制备方法为:将绿茶与开水以1:20的重量比冲泡30分钟后,过滤提取所得的滤液即为茶水。

实验结果
治疗效果实验
为了验证本发明的治疗效果,于2015年6月-7月对患有打印病的泰国斗鱼进行了治疗实验。实验过程中,选取60个水族箱、患有打印病的斗鱼60只,将患有打印病的泰国斗鱼随机分到60个水族箱,每10个水族箱为一个处理组,对照组和实验组分别设置3个平行。实验组的处理如下:每吨水倒入60-90ml的本发明实施例6的制剂,每12小时换水一次,换水后按相同比例加入本发明实施例6的制剂,连续使用直至打印病治愈。对照组的处理方式为:将病鱼捞出,用2克/立方米水体呋喃唑酮药浴鱼体30分钟对病鱼进行消毒,每日一次,直至治愈。
实验结果表明,本发明的治疗泰国斗鱼打印病的环保制剂对泰国斗鱼打印病的治愈效果好,治愈率高,使患病的斗鱼的成活率达到90%以上,用药2-3天后病情有显著地抑制,肌肉腐烂现象得到抑制,斗鱼的摄食频率逐渐提高。此外,本发明的环保制剂起来较为安全,对斗鱼损伤小,使用本发明的治疗泰国斗鱼打印病的环保制剂后,斗鱼健康恢复快,用药5-10天后,斗鱼打印病的症状消除,斗鱼恢复正常活动,游资矫健,摄食完全正常。实验结果见表1:

由表1可见,本发明的环保制剂对泰国斗鱼打印病的治疗效果好,治愈率高,治愈周期短,对患病的斗鱼的治愈率达到90%以上,此外本发明使用起来方便安全,而且对斗鱼损伤小,使用本发明的治疗泰国斗鱼打印病的环保制剂后,斗鱼健康恢复快。
预防效果实验
选取60个水族箱、健康泰国斗鱼60只,将健康泰国斗鱼随机分到60个水族箱,每个水族箱1只泰国斗鱼,在每个水族箱中再分别加入1只患有打印病的泰国斗鱼,每10个水族箱为一个处理组,对照组和实验组分别设3个平行。以正常饲喂组为空白对照组,以水族箱中添加本发明实施例6的环保制剂的处理组为实验组。实验组处理如下:每吨水倒入30-40ml的本发明实施例6的制剂,24小时后换水一次,换水后按相同比例加入本发明实施例6的制剂,连续使用3天后,停1天;如此循环6-8个周期。实验结果见表2:

由表2可见,本发明的治疗泰国斗鱼打印病的环保制剂能有效降低泰国斗鱼打印病的发病概率,能提高斗鱼的免疫力,对打印病的发生具有很好的预防作用。
抑菌试验检查
采用呋喃唑酮做对照组,本发明实施例6的治疗泰国斗鱼打印病的环保制剂为实验组进行抑菌检查。
培养基的制备:称取牛肉浸膏5g溶于水800mL中,加入蛋白胨10g溶解,加入酪蛋白34g溶解,再加入NaCl5g溶解,调至pH7.6~7.8,稀释至1000mL。加入琼脂粉18g,高压灭菌15min,冷却至40℃~50℃时,倒板,凝固,放置无细菌污染的4℃冰箱备用。
菌种为:点状产气单胞菌。
对照组抑菌液配制:将1份呋喃唑酮分别溶解在10ml无菌去离子水中,将药液浓度分别进行稀释,与原液比为1:2,1:4,1:8,1:16,1:32。
实验组抑菌液配制:将本发明实施例6的治疗泰国斗鱼打印病的环保制剂溶解在10ml无菌去离子水中,将药液浓度分别进行稀释,与原液比为1:2,1:4,1:8,1:16,1:32。对点状产气单胞菌的抑菌效果见表3:

3.根据权利要求1所述的治疗泰国斗鱼打印病的环保制剂,其特征在于:所述治疗泰国斗鱼打印病的环保制剂,由下列质量份数的各组分组成:茶水31份、白薇23份、马齿苋22份、土荆芥22份、皂荚22份、牡丹皮20份、茵陈草20份、鱼腥草19份、夏枯草19份、五倍子18份、雷丸18份、槟榔17份、蜈蚣16份、紫云英16份、荠菜16份、茜草15份、胡粉15份、生姜14份、大蒜14份、蝎子13份、车前草13份、竹盐12份、白醋11份。
4.一种权利要求1-3中任一项所述的治疗泰国斗鱼打印病的环保制剂的制备方法,其特征在于:包括以下步骤:
(1)按配比称取白薇、马齿苋、土荆芥和皂荚,将白薇、马齿苋、土荆芥和皂荚的总重量记作1份,称取8份的纯化水,70-80℃下水浴浸泡3-5h,得浸泡液;
(2)将步骤(1)得到的浸泡液加热至沸腾后煮沸2h,分离取出煎出液,继续向药材内加入4份的纯化水,加热至沸腾后继续煮沸2h,合并两次煎出液即为提取液;
(3)按上述配比分别称取牡丹皮、茵陈草、鱼腥草、夏枯草、五倍子、雷丸、槟榔、蜈蚣、紫云英、荠菜、茜草、胡粉、生姜、大蒜、蝎子和车前草,装入球磨机研磨2h,过80目筛,得研磨粉;
(4)将步骤(2)得到的提取液及步骤(3)得到的研磨粉放置于超临界萃取仪中的萃取釜中,利用二氧化碳作为介质进行萃取,萃取釜压力为40Mpa,萃取温度为45℃,分离器压力12Mpa,分离器温度为70℃,萃取2小时,得到萃取液;
(5)按照配比称取茶水、竹盐和白醋,与步骤(4)所得的萃取液混合均匀,即得目标物。
5.根据权利要求4所述的治疗泰国斗鱼打印病的环保制剂的制备方法,其特征在于:所述茶水的制备方法为:将绿茶与开水以1:20的重量比冲泡30分钟后,过滤提取所得的滤液即为茶水。
6.一种权利要求1-3中任一项所述的治疗泰国斗鱼打印病的环保制剂在治疗泰国斗鱼打印病时的使用方法,其特征在于:每吨水倒入60-90ml的本发明的制剂,每12小时换水一次,换水后按相同比例加入本发明的制剂,连续使用直至病鱼治愈。
7.一种权利要求1-3中任一项所述的治疗泰国斗鱼打印病的环保制剂在预防泰国斗鱼打印病时的使用方法,其特征在于:每吨水倒入30-40ml的本发明的制剂,24小时后换水一次,换水后按相同比例加入本发明的制剂,连续使用3天后,停1天;如此循环6-8个周期。
