发明背景
CD4为在大多数胸腺细胞和成熟的T细胞亚群(CD4+T细胞)的表面表达的非多态性跨膜糖蛋白(参见Germain,Curr.Biol.,7:R640-R644(1997))。它由四个免疫球蛋白样细胞外结构域、跨膜区段和与src-家族酪氨酸激酶Lck非共价缔合的胞质尾区组成。作为免疫系统的重要组分,CD4起到帮助CD4+T细胞上的T细胞受体(TCR)与抗原呈递细胞(APC)上的II类主要组织相容性复合物(MHCII)更强缔合的共受体的作用。该缔合足以触发T-细胞信号转导,从而导致CD4+T细胞的活化。
CD4也经由其激酶连接的胞质尾区通过驱动信号级联来直接促进信号转导。人CD4-鼠MHCII的晶体结构显示仅CD4的第一细胞外结构域(D1)与MHCII接触(参见Wang etal.,Proc.Natl.Acad.Sci.USA,98:10799-10804(2001))。然而,突变分析表明除了D1之外,其它结构域也影响与MHCII的结合(参见Moebius等,Proc.Natl.Acad.Sci.USA,90:8259-8263(1993))。而且,CD4的寡聚化对于与MHCII的稳定相互作用以及有效的T-细胞活化是必需的(参见Sakihama et al.,Proc.Natl.Acad.Sci.USA,92:6444-6448(1995))。
CD4还是HIV-1的主要受体(参见Dalgleish et al.,Nature,312:763-767(1984))。HIV-1进入通过病毒包膜糖蛋白(Env)gp120结合于细胞受体CD4来引发。这一相互作用在共受体(CCR5或CXCR4)参与之后引起gp120以及随后gp41的广泛构象重排。Envs的结构重排以及Envs与细胞受体及共受体的相互作用将病毒膜带向靶细胞膜,并最终引起膜融合和病毒进入。因为CD4在HIV-1感染中起着重要作用,所以含有所有四个(T4)(参见Deenet al.,Nature,331:82-84(1988))或前两个胞外结构域(D1D2)(参见Traunecker et al.,Nature,331:84-86(1988))的重组可溶性表达的CD4(sCD4)为HIV-1进入的有效抑制剂并被用于单独的结晶化(参见Wu et al.,Nature,387:527-530(1997)和Ryu et al.,Nature,348:419-429(1990))或与gp120一起的结晶化(参见Kwong et al.,Nature,393:648-659(1998))。
在Sharma et al.(Biochemistry,44:16192-16202(2005))中,通过使用突变策略生成D1并将其从大肠杆菌的超声处理上清液中纯化。然而,纯化的蛋白质仅在低pH(4.0)下为稳定的,部分错误地折叠,并且具有的亲和力比D1D2或T4所具有的亲和力低数倍。
在Chen et al.(J.Virol.,85:9395-9405(2011))中,鉴别了两种稳定的单体D1突变体,mD1.1和mD1.2,所述突变体比D1D2显著更可溶的并且与Env gp120的结合强于(50倍)D1D2,能比D1D2更有效地中和一组来自不同进化枝的HIV-1原代分离物,诱导gp120的构象变化以及使HIV-1对于通过CD4诱导的抗体中和敏感。
然而,仍存在对以下新的D1的需要:其在生理条件下为正确折叠的、高度可溶的并且稳定的;同时不仅保存了结合活性和特异性,而且保存了其它功能,如诱导HIV-1gp120的构象变化。
发明概述
本发明提供了包含单结构域CD4的多肽,如SEQ ID NO:1或SEQ ID NO:2以及包含单结构域CD4和一种或多种融合伴侣的融合蛋白,其中所述一种或多种融合伴侣任选地经由连接子连接于单结构域CD4。另外,提供了包含多价融合蛋白的构建体。还提供了编码单结构域CD4和融合蛋白的核酸以及包含单结构域CD4、融合蛋白、构建体或核酸的组合物或细胞。
本发明还提供了抑制细胞或宿主中HIV感染的方法,其包括向细胞或宿主施用多肽、融合蛋白或构建体以使得HIV感染被抑制。

发明详述
本发明提供了具有全长CD4的结合活性和特异性并维持其它功能,如诱导HIV-1gp120的构象变化的单结构域CD4。由于分子尺寸减小,单结构域CD4具有极好的生物学性质,包括改善的结合动力学,在大肠杆菌中的可溶性表达,较高的溶解度、稳定性和特异性,使在动物中的免疫原性最小化以及更好地渗透入组织,如HIV-1主要在其中复制和扩展的浓密填充的淋巴样环境(例如脾、淋巴结和肠)。
本发明的单结构域CD4包含SEQ ID NO:1或SEQ ID NO:2的氨基酸序列,基本上由或由SEQ ID NO:1或SEQ ID NO:2的氨基酸序列组成。本发明还提供包含具有至多20(例如0、1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19或20)个添加、缺失、取代或插入的SEQ ID NO:1或SEQ ID NO:2的变体的多肽。优选地,本发明提供包含具有至多10个添加、缺失、取代或插入的SEQ ID NO:1或SEQ ID NO:2的多肽,其中所述多肽不包含SEQ ID NO:4。
本发明的多肽优选为重组或合成多肽,其并非天然存在的。
本发明的多肽可单独提供,或作为包含所述多肽和一种或多种融合伴侣的融合蛋白的一部分来提供。融合伴侣可为基本上不抑制多肽结合其靶标的能力的任何合适的部分。理想地,与不存在融合伴侣时多肽的稳定性或效力相比,融合伴侣增强多肽的稳定性和/或效力。例如,融合伴侣可为天然存在的蛋白质或其片段,与不存在融合蛋白时的多肽相比,其通过体内内源机制抵抗降解或消除从而增加融合蛋白的半衰期。

在一个实施方案中,融合伴侣为免疫球蛋白Fc区或其部分(例如CH2或CH3区),尤其是人免疫球蛋白的Fc区,如人IgG1Fc区。在本发明中使用的Fc区或其部分的实例包括但不限于SEQ ID NO:7和SEQ ID NO:8的氨基酸序列。
在另一实施方案中,融合伴侣为抗体或抗体片段(例如Fab、scFv、dAb等)。优选地,抗体为单结构域抗体(a.k.a.“结构域抗体(“dAb”)或“工程化抗体结构域”(“eAd”)),其为由来自重链或轻链的单个单体可变抗体结构域组成的抗体片段。例如,eAd可包含SEQ IDNO:12、SEQ ID NO:13、SEQ ID NO:14、SEQ ID NO:15或SEQ ID NO:16,在本文也分别被称为m36、m36.1、m36.2、m36.4或m36.5抗体。

融合蛋白包含一种或多种融合伴侣(例如两种、三种、四种、五种或更多种融合伴侣)。例如,融合蛋白可包含本发明的多肽、eAd(例如分别为SEQ ID NO:12-16的m36、m36.1、m36.2、m36.4或m36.5抗体)以及稳定性增强的融合伴侣,如免疫球蛋白Fc区(例如人IgG1Fc)或其部分(例如CH3)。
本发明的多肽和一种或多种融合伴侣可经由连接子(即柔性分子连接,如柔性多肽链)连接。连接子可为任何长度的任何合适的连接子,但优选长度为至少约15(例如至少约20、至少约25、至少约30、至少约35、至少约40、至少约45、至少约50或它们的范围)个氨基酸。在一个实施方案中,连接子为天然存在于宿主的免疫球蛋白分子中的氨基酸序列,这样使得连接子的存在不会产生针对哺乳动物的连接子序列的免疫应答。合适的连接子的实例包括但不限于包含一一个或多个(例如两个、三个、四个、五个、六个、七个、八个、九个或十个)G4S基序的连接子,如SEQ ID NO:9-11的连接子。

可选地,可将以上式(II)的两个融合蛋白以及A-(任选的连接子)-C(式III)的两个融合蛋白组装在单个构建体中,如图2所示(4Dm2m)。在一个实施方案中,式(III)的融合蛋白包含SEQ ID NO:21以及式(II)的融合蛋白包含SEQ ID NO:20。

在一个实施方案中,组装融合蛋白(例如自组装)以形成图2中所示的其中一个构建体,其中A表示抗体或抗体片段(例如m36.4eAd),B表示本发明的多肽(例如mD1.22),C表示免疫球蛋白轻链恒定区(例如人IgG1κ轻链恒定区),D表示免疫球蛋白重链恒定区(例如人IgG1重链恒定区),并且E表示Fc区(例如来自人IgG1的Fc区)。
多肽和融合蛋白可为聚乙二醇化的,或被偶联至类似结构、功能和目的的聚合物以赋予增强的稳定性和半衰期。相对于非聚乙二醇化的(例如抗体)多肽,聚乙二醇化可提供增加的半衰期和对降解的抗性而无活性的丧失(例如结合亲和力)。因为聚乙二醇化对于一些靶标可能不是有利的,特别是那些空间受阻的表位,所以应使多肽或融合蛋白最低程度地聚乙二醇化以便不会负面影响尺寸受限的抗原的可及性。多肽或融合蛋白可通过本领域已知的任何合适的手段偶联至PEG或PEG样聚合物。合适的PEG或PEG样部分可为合成的或天然存在的,并且包括但不限于直链或支链聚亚烷基聚合物、聚亚烯基聚合物或聚氧亚烷基聚合物、或分枝或不分枝的多糖,诸如同多糖或杂多糖。合成聚合物的优选实例包括直链或支链聚(乙二醇)(PEG)、聚(丙二醇)或聚(乙烯醇)及其衍生物或取代形式。用于连接至结构域抗体的取代的聚合物也包括取代的PEG,包括甲氧基(聚乙二醇)。除了或代替PEG之外,可使用的天然存在的聚合物部分包括例如,乳糖、直链淀粉、右旋糖酐或糖原及其衍生物。
多肽或融合蛋白可被多聚化为例如,异源-或同源二聚体、异源-或同源三聚体、异源-或同源四聚体、或更高阶异源-或同源多聚体。多聚化可增加抗原结合的强度,其中结合的强度与多个结合位点的结合亲和力的总和相关。具体地说,可将半胱氨酸残基引入多肽或融合蛋白的氨基酸序列,从而允许在多聚化形式中形成链间二硫键。同源二聚的或异源二聚的(或多聚化)融合蛋白可包括相同或不同融合伴侣(例如eAd)的组合,使得相同构建体可在一次靶向多于一个表位。此类表位可最近地定位于靶标(例如在HIV靶标上),使得一个表位的结合促进多聚化融合蛋白与第二个或更多个表位的结合。多聚化融合蛋白靶向的表位也可位于远端。

也可将诸如荧光化合物的可检测试剂添加至多肽或融合蛋白(或含有融合蛋白的构建体)。示例性荧光可检测试剂包括荧光素、异硫氰酸荧光素、罗丹明、5-二甲胺-l-萘磺酰氯、藻红蛋白等。多肽或融合蛋白也可用诸如以下的可检测酶衍生化:碱性磷酸酶、辣根过氧化物酶、葡萄糖氧化酶等。当多肽或融合蛋白构建体用可检测酶衍生化时,通过添加酶利用其产生可检测的反应产物的另外试剂来检测多肽或融合蛋白构建体。多肽或融合蛋白构建体也可用生物素衍生化,并通过间接测量亲和素或链霉亲和素结合来检测。
本发明还提供编码多肽或融合蛋白的氨基酸序列的核酸。该核酸可包含DNA或RNA,可为单链或双链的。此外,核酸可包含核苷酸类似物或衍生物(例如肌苷或硫代磷酸酯核苷酸等)。
可将核酸提供为构建体的一部分,所述构建体包含核酸以及能够使核酸递送至细胞和/或能够使核酸在细胞中表达的元件。此类元件包括例如,表达载体、启动子以及转录和/或翻译序列。合适的载体、启动子、转录/翻译序列和其它元件以及制备此类核酸和构建体的方法为本领域已知的(例如Sambrook et al.,同上;和Ausubel et al.,同上)。

可将多肽、融合蛋白、缀合物、核酸、载体或细胞施用于有需要的任何宿主(例如哺乳动物,优选为人)。由于将多肽、融合蛋白、缀合物、核酸、载体或细胞施用于哺乳动物的结果,哺乳动物的病毒感染(例如HIV感染)被抑制。本发明的方法可预防性或治疗性抑制任何类型的HIV的感染,但优选地抑制HIV-1和/或HIV-2感染。可将本发明的方法用于抑制任何HIV群组(例如群组M和/或O)和亚型(例如进化枝A、B、C、D、E、EA、F、和/或G)的感染。

多肽、融合蛋白、缀合物、构建体、核酸、载体、细胞或其组合物的功效可以以熟练的从业者熟知的不同方式评估。例如,本领域普通技术人员通过观察到多肽或融合蛋白降低病毒载量或延迟或防止病毒载量进一步增加,会理解本发明的多肽或融合蛋白在治疗或抑制对象中的病毒(例如HIV)感染中是有效的。病毒载量可通过本领域已知的方法来测量,例如,使用PCR测定以检测病毒(例如HIV)核酸的存在或抗体测定以检测来自对象或患者的样品(例如血液或另一体液)中病毒(例如HIV)蛋白的存在,或通过测量患者循环中抗病毒(例如抗-HIV)抗体的水平。多肽或融合蛋白治疗的功效也可通过测量HIV-感染的对象中CD4+T细胞的数目来确定。延迟或抑制HIV-阳性对象或患者中CD4+T细胞的起始或进一步降低或引起HIV-阳性对象中CD4+T数目增加的治疗可被认为是有效的。

将多肽、融合蛋白、缀合物、构建体、核酸、载体、细胞或其组合物施用于宿主的合适方法为本领域已知的。该宿主可为任何合适的宿主,如哺乳动物(例如啮齿类动物(如小鼠、大鼠、仓鼠或荷兰猪)、兔、猫、犬、猪、羊、牛、马、灵长类或人)。
施用可为局部的(包括眼科的、阴道的、直肠的、鼻内的、经皮的等)、口服、通过吸入或肠胃外(包括通过静脉滴注或皮下注射、腔内注射、腹腔注射或肌内注射)。局部的鼻内施用是指通过一个或两个鼻孔将组合物递送至鼻和鼻道,并且可包括通过喷雾机制或微滴机制或通过核酸、载体或融合蛋白的雾化来递送。通过吸入施用组合物可以经由通过喷雾或微滴机制的递送通过鼻或口来实现。也可经由插管法直接递送至呼吸系统(例如肺)的任何区域。
用于局部施用的制剂包括软膏、洗剂、乳膏、凝胶、滴剂、栓剂、喷雾、液体和粉剂。常规药物载体、水性、粉状、或油性基质、增稠剂等可以是必需或期望的。

一些组合物可以潜在地作为药学上可接受的酸加成盐或碱加成盐被施用,所述药学上可接受的酸加成盐或碱加成盐通过与无机酸,如盐酸、氢溴酸、高氯酸、硝酸、硫氰酸、硫酸和磷酸以及与有机酸,如甲酸、乙酸、丙酸、乙醇酸、乳酸、丙酮酸、草酸、丙二酸、琥珀酸、马来酸和富马酸反应,或者通过与无机碱,如氢氧化钠、氢氧化铵、氢氧化钾以及有机碱,如单-、二-、三烷基和芳基胺以及取代的乙醇胺反应而形成。
多肽、融合蛋白、缀合物、构建体、核酸、载体或细胞可与药学上可接受的载体一起施用并且可通过本领域熟知的多种机制(例如裸DNA的摄取、脂质体融合、经由基因枪的DNA的肌内注射、胞吞等)体内和/或离体递送至哺乳动物细胞。
另外,本发明设想了益生菌治疗。可将含有本发明的核酸或载体并表达融合蛋白或缀合物的活宿主细胞直接用作将融合蛋白递送至体内期望部位的递送媒介物。将融合蛋白或缀合物直接递送至期望部位,诸如例如递送至选定的体腔的优选宿主细胞可包括细菌。更具体地,此类宿主细胞可包括乳杆菌、肠球菌或已知通常定居于体腔的其它常见细菌如大肠杆菌正常菌株的合适工程化菌株。更具体地,此类宿主细胞可包括一种或多种选定的乳杆菌的非病原菌株,如Andreu et al.,J.Infect.Dis.,171(5),1237-43(1995)所述的那些,尤其是与上皮细胞(例如阴道上皮细胞)具有高的粘附性能并且使用本发明的核酸或载体适于转化的那些菌株。
如果使用离体方法,根据本领域已知的标准方案,可取出细胞或组织并将其维持在体外。可经由诸如以下的任何基因转移机制将组合物导入细胞:磷酸钙介导的基因递送、电穿孔、微注射或蛋白脂质体。然后,根据针对细胞或组织类型的标准方法,可注入转导的细胞(例如与药学上可接受的载体一起)或将转导的细胞同伦(homotopically)移植回哺乳动物。将不同细胞移植或注入哺乳动物的标准方法是已知的。
治疗病毒感染(例如HIV感染)所需的组合物的精确量因哺乳动物的不同而不同,取决于哺乳动物的物种、年龄、性别、体重和一般状况,病毒的性质,病毒感染的存在性和程度,使用的具体融合蛋白、核酸、载体或细胞,施用途径以及其它药物是否包括在方案内。因此,规定每种组合物的精确量是不可能的。然而,考虑到本文给出的教导,本领域普通技术人员仅利用常规实验就能够确定合适的量。可凭经验测定施用本发明的核酸分子、载体、细胞和融合蛋白的有效剂量和方案并且进行此种测定在本领域技术人员范围内。施用组合物的剂量范围为足够大以产生期望效果的那些;然而,剂量不应大到引起显著的不良副作用,诸如不良的交叉反应、过敏反应等。给药剂量可以变化且可以以每日一次或多次(例如两次或多次、三次或多次、四次或多次或五次或多次)的剂量来施用,持续一天或多天。组合物可在病毒(例如HIV)感染前或在确定病毒(例如HIV)感染时立即施用并且持续施用直至检测不到病毒。
将多肽、融合蛋白、缀合物、构建体、核酸、载体、细胞或其组合物以预防性或治疗性抑制HIV感染的有效量施用于宿主(例如哺乳动物,诸如人)。多肽、融合蛋白、缀合物、构建体、核酸、载体、细胞或其组合物作为HIV感染抑制剂的功效可以通过本领域已知的体内或体外参数来测定。
可将任何合适剂量的多肽融合蛋白、缀合物、核酸、载体、细胞或其组合物施用于宿主。适当剂量取决于诸如以下的因素而变化:宿主的年龄、体重、身高、性别、一般医疗状况、先前的医疗史和病毒(例如HIV)感染进展,并且可由临床医生来确定。例如,可以以每天约1μg/kg至高达100mg/kg体重(例如5μg/kg、10μg/kg、50μg/kg、100μg/kg、200μg/kg、300μg/kg、400μg/kg、500μg/kg、600μg/kg、700μg/kg、800μg/kg、900μg/kg、1mg/kg、2mg/kg、5mg/kg、10mg/kg、20mg/kg、30mg/kg、40mg/kg、50mg/kg、60mg/kg、70mg/kg、80mg/kg、90mg/kg及其范围)或更多的剂量将多肽、融合蛋白或缀合物施用于宿主(例如哺乳动物,如人)。可提供几种剂量(例如1、2、3、4、5、6或更多种)(例如在数周或数月的时间内)。
当载体为病毒载体时,合适的剂量可包括约1 x 105至约1 x 1012(例如1 x 106、1x 107、1 x 108、1 x 109、1 x 1010、1 x 1011及它们的范围)个噬斑形成单位(pfu),尽管可将更低或更高剂量施用于宿主。例如,可施用约2 x 108个pfu(例如在约0.5mL的体积中)。

实施例1

将结构引导的腔填充和文库筛选用于鉴定具有改善的可溶性表达、稳定性和特异性的D1突变体。筛选产生两种突变体:mD1.22(SEQ ID NO:1)和mD1.23(SEQ ID NO:2)(参见图1)。

如图3A和3B所显示,两种突变体针对测试的gp140为交叉反应的。
还通过利用单循环方法的Biacore X100装置(GE Healthcare)上的表面等离子体共振(SPR)分析遗传上多样化的HIV-1Envs的结合。简言之,将纯化的HIV-1gp140在乙酸钠(pH 5.0)中稀释并经由标准的胺偶联方法直接固定在CM5传感芯片上。将参考池用N-羟基琥珀酰亚胺-1-乙基-3-(3-二甲基氨基丙基)碳二亚胺和乙醇胺注射而未注射gp140。将D1突变体和D1D2用电泳缓冲液HSB-EP(100mM HEPES(ph 7.4),1.5M NaCl,30mM EDTA,0.5%表面活性剂20)稀释。在500、100、20、4和0.8nM浓度下测试所有分析物。由用BiocoreX100评价软件2.0的单价结合模型拟合的传感图(sensograms)计算动力学常数。
在四种HIV-1Env gp140的SPR分析中,mD1.22显示出的亲和力与mD1.2的亲和力相当或高于mD1.2亲和力(参见表8)。
实施例3
该实施例显示D1突变体维持全长CD4的功能活性。
CD4诱导gp120的构象变化,从而导致CD4-可诱导的(CD4i)表位的暴露。为确定D1突变体是否诱导此类构象变化,在不存在或存在mD1.2(SEQ ID NO:3)、mD1.22(SEQ ID NO:1)或mD1.23(SEQ ID NO:2)的情况下测试基于CD4i抗体的融合蛋白,m36h1Fc(参见Chen etal.,Proc.Natl.Acad.Sci.USA,105:17121-17126(2008))与gp140Con-s的结合。将两个结构域的人CD4(D1D2;SEQ ID NO:5)用作阳性对照,并且将Fab b12用作阴性对照,所述Fabb12为靶向gp120上CD4-结合位点的HIV-1广泛中和抗体(参见Roben et al.,J.Virol.,68:4821-4828(1994))。
相对于对照,在存在D1突变体的情况下,m36h1Fc与gp140Con-s的结合增加(参见图4)。另外,m36h1Fc与gp140Con-s的结合在分别存在D1D2和mD1.2的情况下增加或与在分别存在D1D2和mD1.2的情况下所观察到的结合相当(参见图4)。
实施例4
该实施例显示D1突变体的其它表征。
通过圆二色性(CD)光谱法测定mD1.2(SEQ ID NO:3)、mD1.22(SEQ ID NO:1)和mD1.23(SEQ ID NO:2)的二级结构和热稳定性。将纯化的蛋白质以0.33mg/mL的终浓度溶解于PBS并将CD光谱记录于AVIV Model 202CD光谱仪(Aviv Biomedical)。在25℃下使用针对天然结构测量的0.1-cm光程长比色杯记录波长谱。通过记录在25-90℃的温度范围,加热速率为1℃/min的CD信号来测量216nm处的热稳定性。用外部探针传感器记录温度并通过校准计算微量比色池内部的温度;它比通过外部传感器测量的温度低约2-3℃(对于20至80℃的温度,范围为1.9至3.8℃)。在加热之后,在90℃记录波长谱。
如通过图5A所证明,mD1.22和mD1.23主要由β-链二级结构组成。mD1.22的热稳定性高于mD1.2和mD1.23(参见图5B)。
与mD1.2类似,mD1.22和mD1.23在pH 7.4的PBS中为均一单体的,如通过尺寸排阻色谱法所测定的。D1D2也为单体,但其未洗脱为单峰(参见图6A)。mD1.22在高浓度(10mg/mL)下不倾向于聚集,如通过动态光散射所显示的(参见图6B)。
实施例5
该实施例显示包含D1突变体的Fc融合蛋白的表征。
使用常规方法制备D1D2、D1D2m(具有A55V突变;SEQ ID NO:6)、mD1.2和mD1.22的Fc-融合蛋白(分别为D1D2Fc、D1D2mFc、mD1.2Fc和mD1.22Fc)。
评估m36h1Fc(阴性对照)、D1D2Fc、D1D2mFc、mD1.2Fc和mD1.22Fc与表达MHC的细胞-人血细胞系BJAB(B细胞)和SUP-T1(T细胞)的结合。如图7和图8B所证实的,相对于其它融合蛋白,mD1.22或D1D2m的Fc融合物显示出与表达MHC的细胞的结合降低。
为确定A55V突变是否导致构象变化,评估与CD4D1-特异性抗体(D9)的相互作用。如图9所显示,在mD1.22Fc和D1D2mFc中存在的A55V突变大幅降低了与D9抗体的结合。这表明该突变诱导D1的构象变化。然而,如上所述,存在A55V突变导致与HIV-1gp140的结合增加(参见图8A)。
实施例6
该实施例显示包含mD1.22的双特异性多价融合蛋白的表征。
制备如图2中例示的mD1.22(SEQ ID NO:1)和m36.4的双特异性多价融合蛋白(2Dm2m、4Dm2m和6Dm2m)。m36.4(SEQ ID NO:15)为靶向HIV-1gp120的CD4i表位的工程化的单一人抗体结构域。
在每个双特异性多价融合蛋白中,A对应于m36.4,B对应于mD1.22,C对应于轻链恒定区,D对应于重链恒定区,并且E对应于Fc区。具体地说,2Dm2m包含含有SEQ ID NO:18的两个融合蛋白以及含有SEQ ID NO:19的两个融合蛋白。4Dm2m包含含有SEQ ID NO:20的两个融合蛋白以及含有SEQ ID NO:21的两个融合蛋白。6Dm2m包含含有SEQ ID NO:22的两个融合蛋白以及含有SEQ ID NO:23的两个融合蛋白。
类似于实施例5中所述的结果,mD1.22-m36.4多价融合蛋白与表达MHC的BJAB细胞的结合比D1D2Fc与表达MHC的BJAB细胞的结合低得多(参见图10)。
为测定D1突变体中和HIV-1的效力和广度,包括具有来自代表进化枝A-E的HIV-1分离物的Env的假型病毒,并使用CCR5(R5)或CXCR4(X4)或两者(R5X4)作为共受体。假病毒来源于293T细胞,并使用HOS-CD4-CCR5(对于所有R5和双嗜性病毒)或HOS-CD4-CXCR4细胞系以一式两份进行中和测定,如Chen et al.,Proc.Natl.Acad.Sci.USA,105:17121-17126(2008))所述。在感染后48小时测量发光并通过下式来计算中和百分比:(1-含有抑制剂的孔的平均RLU/平均RLU或仅含病毒的孔)x 100。中和的IC50和IC90分别被指定为观察到50%和90%中和时的抑制剂浓度。
如表2和表3所显示,mD1.22和含有mD1.22的融合蛋白中和来自几种不同进化枝的HIV-1原代分离物。明显地,4Dm2m和6Dm2m比广泛中和单克隆抗体VRC01更有效(参见表3)。
另外,mD1.22和含有mD1.22的融合蛋白抑制HIV-1包膜糖蛋白介导的细胞融合(参见表4和5)。如表5所显示,4Dm2m和6Dm2m在抑制HIV-1包膜糖蛋白介导的细胞-细胞融合中比VRC01更有效。
在基于TZM-bl细胞的测定中(参见Brown et al.,Virology,375:529-538(2008)),其中使用TZM-bl细胞和随机选自A至D、AE和AG(总计n=12)的各个进化枝的HIV-1分离物,当比较其IC50和IC90的几何平均值时,4Dm2m和6Dm2m比b12和CD4-Ig有效得多,并且比VRC01的效力大约10倍(参见表6)。
在使用一组来源于主要在中国流行的41HIV-1分离物的具有Env的假病毒的另一基于TZM-bl细胞的测定中(参见Yao et al.,J.Biol.Chem.,287:6788-6796(2012);和Shang et al.,J.Biol.Chem.,286:14531-14541(2011)),基于针对上述组的中和活性,将4Dm2m和6Dm2m与T20(恩夫韦地)、C34和CD4-Ig进行比较,所述T20为FDA-批准的来源于HIV-1Env gp41C-末端七肽重复(CHR)的肽抑制剂,所述C34为另一有效的CHR肽抑制剂。4Dm2m和6Dm2m中和所有病毒,IC50几何平均值为大约0.20nM,该值比T20、C34和CD4-Ig的值(11、3.5和42nM)分别低约50倍(P=0.002,学生配对t检验)、16倍(P<0.001,学生配对t检验)和200倍(P<0.001,学生配对t检验)(参见表7)。当比较IC90时,也观察到4Dm2m和6Dm2m的优越效力。
也已经显示4Dm2m和6Dm2m在低浓度时不会增强HIV-1感染性,这与D1D2相反(参见图11)。
实施例7
该实施例显示双特异性多价融合蛋白的溶解性、稳定性和聚集倾向。
为评价进一步开发成药物的潜能,测试了几种药物相关的性质,包括溶解度、稳定性和聚集倾向。将PBS中的4Dm2m和6Dm2m在高速离心之后分别浓缩至25.0和25.9mg/ml,而无可见沉淀。将它们在4℃储存2周,未观察到另外的沉淀,这表明蛋白质具有高溶解度。
为测试4Dm2m和6Dm2m的可溶性聚集体是否在延长孵育期间形成,使用了动态光散射(DLS)。将在PBS中浓缩至10mg/ml的蛋白质储存在80℃并在测量之前于冰上缓慢解冻。结果显示:4Dm2m和m909(呈IgG1形式的对照人抗体(参见Feng et al.,ArthritisRes.Ther.13:R59(2011))的颗粒主要为小的(平均直径分别为16.3和12.7nm)。4Dm2m的少量聚集体在4℃和37℃孵育的第一天内消失,而m909聚集体在37℃孵育之后继续存在。相比之下,约30%至50%的CD4-Ig颗粒为大的,平均直径为大约100nm并且在孵育的全部时段内聚集体持续存在。在冻融循环之后,6Dm2m部分聚集;虽然孵育之后聚集被削弱,但是其产生了相对宽的粒径分布。
这些结果表明:如同典型的IgG1,4Dm2m具有低的聚集倾向,其低于CD4-Ig的聚集倾向。
然后,在与PBS(参见图12A)或人血清(参见图12B)于37℃下孵育15天期间评估蛋白质针对降解的稳定性。在PBS中孵育10天未引起功能性4Dm2m的量的显著下降,然而分别为约20%和30%的6Dm2m和CD4-Ig丧失与gp140Con-s的结合活性(参见图12A)。此后,4Dm2m开始降解,6Dm2m继续降解,而CD4-Ig看起来为稳定的。最终,约90%的4Dm2m保留功能性,略微超过保留功能性的6Dm2m和CD4-Ig的百分比(80%)。
关于人血清,在前5天孵育期间,所有蛋白质水平以类似速率下降(参见图12B)。然而,在接下来5天的孵育期间,4Dm2m和6Dm2m的降解变得比CD4-Ig的降解缓慢。在测量期结束时,保留结合活性的CD4-Ig的水平略微高于功能性4Dm2m和6Dm2m的水平,但量的差异并非统计学上显著的。
本文所述的实施例显示了新的D1突变体的鉴定。具体地说,mD1.22具有改善的可溶性表达和稳定性,与人血B和T细胞系没有显示出可测量的相互作用,并保存了与HIV-1gp120的结合和交叉反应性。mD1.22-m36.4融合蛋白已经显示为有效的HIV-1中和剂并且多价mD1.22-m36.4融合蛋白不倾向于增强HIV-1感染性。因此,这些新鉴定的突变体可用于HIV-1的预防和治疗,以及也可作为研究HIV-1进入的机制以及免疫应答中CD4的生物学功能的有价值的工具。

Claims (19)

其中A为抗体或抗体片段,B为由SEQ ID NO:1的氨基酸序列组成的多肽,C为免疫球蛋白轻链恒定区,D为免疫球蛋白重链恒定区,并且E为Fc区或其部分,其中所述式(III)的融合蛋白包含SEQ ID NO:21,并且所述式(II)的融合蛋白包含SEQ ID NO:20。
2.构建体,其包含:
式(I)的两个融合蛋白,其中式(I)为:A-(任选的连接子)-C-(任选的连接子)-B,以及
式(II)的两个融合蛋白,其中式(II)为:B-(任选的连接子)-D-(任选的连接子)-E-(任选的连接子)-B,
