Abstract
本发明公开了红麻(Hibiscus?cannabinus)植物提取物以及利用其作为皮肤美白、保湿及抗氧化活性的化妆品添加剂及其在化妆品中的组合应用。所述红麻提取物为红麻根部的乙醇提取物,该提取物具有良好的DPPH自由基清除能力,是良好的天然抗氧化剂。在人体护肤方面,红麻提取物具备优秀的抗皱、美白、防晒等皮肤化妆品应用潜力。本发明有效利用中国传统的粗纤维植物材料——红麻,得到了具有很高皮肤化妆品活性的红麻提取物,这些提取物可被用于制作具良好的抗氧化剂,是良好的天然植物化妆品原料。
Description
技术领域
本发明属于化妆品和医药技术领域,具体涉及从一种中国传统粗纤维材料来源植物红麻中提取天然活性物质--红麻提取物,以其为活性成分的化妆品或药物,以及红麻提取物在制备保湿、嫩肤、抗皱、防晒、美白等多种皮肤化妆品中的应用,还有在制备治疗或预防皮肤病的药物中的应用
技术背景
红麻(Hibiscuscannabinus),又名洋麻、槿麻、刺红花,为锦葵科木槿属植物,一年生草本韧皮纤维作物,花淡黄或乳白色,柱头紫或红色,叶分掌状裂叶型和阔卵叶型。在我国,红麻常见于广东、广西、浙江、河南、山东以及安徽等地,大多数为人工种植。
我国对红麻的使用拥有悠久的历史,一直以来,我国都是传统的红麻种植大国,主要将红麻用于制作麻绳和麻布。随着多种合成纤维对市场的冲击,红麻的利用也日渐减少,传统的利用方式正逐渐消失。如今,对于红麻的使用,大多数利用方式都局限于提取红麻纤维,用于复合材料的增强纤维或直接用于建筑材料和包装材料。虽然,人类对红麻的使用历史悠久,但目前还未见有红麻或者红麻提取物在皮肤化妆品或者药物中活性的报道。
发明内容
本发明目的在于提供红麻提取物,以其为活性成分的化妆品,它们的制备方法,及其在制备化妆品中的应用。本发明明确了红麻提取物具有良好的护肤效果和安全性,使其成为很好的天然护肤化妆品原材料。
为了实现本发明的上述目的,本发明提供了如下的技术方案:
红麻提取物,由下述方法制备而得:取红麻根部,研成粉末,用红麻的15-30倍量的85%-95%乙醇,冷浸18-28h,收集滤液,重复2-5次,滤液减压蒸馏,浓缩滤液,低温干燥,得粗提物,将粗提物经大孔树脂吸附脱色,85%-95%乙醇洗脱后,浓缩蒸干,低温冷冻干燥,得红麻提取物。
本发明同时提供了红麻提取物的制备方法:取红麻根,研成粉末,用红麻的15-30倍量的85%-95%乙醇,冷浸18-28h,收集滤液,重复2-5次,滤液减压蒸馏,浓缩滤液,低温干燥,得粗提物,将粗提物经大孔树脂吸附脱色,85%-95%乙醇洗脱后,浓缩蒸干,低温冷冻干燥。
本发明还提供了一种化妆品,由上述的红麻提取物和化妆品常用载体组成,所述的载体选自一种或多种含水体系,甘油、C1-4醇、脂肪醇、多元醇、脂肪醚、脂肪酯、乙二醇、丙二醇、植物油、矿物油、脂质体、层状类脂材料、硅油、水,或者它们的组合。
以及,所述的红麻提取物在制备化妆品中的应用。
所述的红麻提取物在制备保湿或嫩肤或抗皱或防晒或美白化妆品中的应用。
所述的红麻提取物在制备麻醉剂、抗真菌剂、抗炎剂、防腐剂、螯合剂、着色剂、脱色剂、软化剂、乳化剂、脱落剂、成膜剂、芳香剂、湿润剂、驱虫剂、润滑剂、增湿剂、药剂、光稳定剂、防腐剂、皮肤保护剂、皮肤渗透增强剂、防晒剂、稳定剂、表面活性剂、增稠剂、粘度调节剂、维生素中的应用。

本发明还提供了一种预防或治疗皮肤病的药物组合物,由所述的红麻提取物和药物常用载体组成。
本发明所述的红麻提取物在制备预防或治疗皮肤病的药物中的应用。
与现有技术相比,本发明具备如下的优益性:本发明通过对红麻植物进行浓缩提取得到目标提取物,对目标提取物进行了细胞毒性实验和皮肤化妆品活性实验,证明了目标提取物的安全性,明确了目标提取物优良的化妆品活性和药物活性。本发明提高了红麻的利用功能和附加值,又拓展了天然护肤品植物原料的来源。本发明通过实验提取红麻根部的活性提取物,并进行了细胞毒性及皮肤化妆品活性实验,明确了红麻提取物不仅拥有良好的安全性还表现出良好的抗氧化活性,使之成为了很好的天然护肤品植物原材料,突破了红麻传统利用方式的局限,拓宽了红麻的利用途径,对于提高红麻的资源利用价值具有重大意义。

具体实施方式:
以下结合附图,用本发明的实施例对本发明进行更详细叙述,但并不限制本发明的范围。
实施例1:
红麻提取物的制备:
称取红麻(Hibiscuscannabinus)根部14g,将其研成粉末状,用85%乙醇25倍量(421-422mL)冷浸24h,然后用布氏漏斗收集滤液,重复3次。将滤液用旋转蒸发仪减压蒸馏,浓缩滤液,浓缩滤液低温干燥,得粗提物。将粗提物经大孔树脂(D101)吸附脱色,95%乙醇洗脱后,将洗脱液浓缩蒸干,低温冷冻干燥,得红麻根部提取物0.3g,提取率为2.14%。
或者,称取红麻(Hibiscuscannabinus)根部样品14g,将样品研成粉末状,用95%乙醇20倍量冷浸20h,然后用布氏漏斗收集滤液,重复4次。将滤液用旋转蒸发仪减压蒸馏,浓缩滤液,浓缩滤液低温干燥,得粗提物。将粗提物经大孔树脂(D101)吸附脱色,90%乙醇洗脱后,将洗脱液浓缩蒸干,低温冷冻干燥,得红麻提取物0.4g,提取率为2.85%。
实施例2:
红麻提取物的细胞毒性实验:通过MTS比色法,确定样品的CC50(50%cytotoxicconcentration),即对50%的细胞产生毒性时的药物浓度,从而确定以下活性实验的安全样品浓度。
实验方法:96孔细胞培养板上,将B16细胞与不同浓度的待测药物溶液混合,设定3个重复孔,同时设置不含药物的空白对照,37℃,5%CO2培养24h,采用MTS比色法检测细胞毒性,酶标仪测定OD值,测定波长490nm。计算得到CC50值。
细胞存活率(%)=实验孔OD490nm/空白孔OD490nm×100%
实验结果:红麻根部提取物的细胞存活率为79.714%,属于正常水平,可明显看出红麻提取物对细胞没有毒害作用,若作为皮肤化妆品的天然原料的安全性是有保证的。
实施例3:
DPPH自由基清除实验:将待测药物与DPPH(终浓度为100μM)混合反应,设定3个重复孔,同时设置不含药物的空白对照和Trolox阳性对照,30℃,1h,酶标仪测定OD值,检测波长为515nm。计算得到抗氧化率。
抗氧化率(%)=(1-实验孔OD515nm/空白孔OD515nm)×100%
实验结果:在实验中,红麻根部提取物表现出显著的DPPH自由基清除能力,并且根据浓度梯度,呈现良好的量效关系(见图1),可被用作具备抗皱或防晒潜力化妆品活性的良好天然植被来源。
本发明提供具有红麻提取物成分或红麻提取物混合物的局部组合物,它在药学上或化妆品上可接受的载体。这些组合物能抑制皮肤的酪氨酸酶活性,B16细胞黑色素生成抑制活性,弹性蛋白活性,TNF-α分泌抑制活性,促胶原蛋白分泌活性,抑制皮肤的氧化。这些组合物能用于细纹和/皱纹,皮肤红疹,干性、敏感肌肤,皮肤瘙痒,肤色不均或皮肤黑斑,皮肤发炎和其它与此相关的皮肤不适症。这些组合物包含有从红麻中得到的提取物或从该植物中得到的活性成分。这些组合物能进一步抑制皮肤的脂肪氧化酶,COX(环氧化酶)活性,MMP-1和MMP-3活性。这些组合物中红麻植物提取物的含量是组合物总重的0.001wt%至20.0wt%。这些组合物可以是护肤液、护肤霜或乳剂等。红麻的提取物可以是水溶性提取物,该提取物可以用醇(例如甲醇、乙醇、丙醇,丁醇等)提取。
本发明中含有红麻提取物的组合物可用于局部皮肤护理或护发产品。这些组合物可以做成乳化剂(例如水包油、油包水、硅油包水,水包硅油,水包油包水,油包水包油,油包水包硅油等),霜剂,护肤液,液体(例如水剂或水-醇溶液),无水基质(例如唇膏或粉剂等),啫喱,油膏,奶液,软膏,喷雾剂,固体剂,眼霜等。
本发明中的局部护肤组合物包含有红麻的提取物或从红麻提取物中获得的组分或是成分。局部施用这些组合物可减轻皮肤红斑的外观(如皮肤红斑、过敏性皮肤、皮肤红肿、发炎等),还可减轻由于红斑引起的疼痛。红斑一般可由皮肤日光灼伤、皮肤的电疗、皮肤烧伤、接触性过敏、全身过敏、皮肤毒性、锻炼、虫咬、细菌感染、病毒感染、真菌感染、寄生原虫感染、按摩和吹风引起的皮炎等。可治疗干性皮肤、敏感皮肤或皮肤搔痒或减小肤色不均的外观。可减小细纹或皱纹的外观。这些提取物可以是水溶性的或非水溶性的提取物,它们可用醇(甲醇、乙醇、丙醇或丁醇等)、甘醇、油、水等来提取。
本发明中的局部护肤组合物包含有红麻的提取物或从红麻提取物中获得的组分或成分。局部施用这些组合物可治疗或预防皮肤病。皮肤病包括干性皮肤、皮肤搔痒、皮肤发炎、红斑、皮肤过敏、瘙痒症、蜘蛛血管、雀斑、老年斑、老年性紫癜、角化症、黑斑病、疙瘩、细纹或皱纹、结节、皮肤太阳损伤、皮炎(脂溢性皮炎、钱币状皮炎、接触性皮炎、异位性皮炎、剥脱性皮炎、口周皮炎和淤滞性皮炎等)、牛皮癣、毛囊炎、红斑痤疮、痤疮、脓胞病、丹毒、红癣、湿疹、日光灼伤、皮肤烧伤、开放性伤口、感染性皮肤病等。皮肤病还可由暴露在紫外光下、年龄、辐射、长期暴露在太阳下、环境污染、空气污染、风、冷、热、化学品、病理、吸烟或缺乏营养等引起。本方法通过局部施用本发明的组合物来增加皮肤角质层的转化速度,增加成纤维细胞中胶原蛋白的合成,增强细胞抗氧化防御机理(如外层抗氧化的增强能改善、补充或防止细胞抗氧化剂如皮肤细胞——角质形成细胞、黑色素细胞、朗格汉斯细胞中减小或防止皮肤、细胞、蛋白和脂质氧化性损伤的催化酶和谷胱甘肽的损失),抑制黑色素细胞中黑色素的产生,减少或阻止皮肤氧化性损伤(包括减少脂质过氧化酶的数量和/或皮肤中蛋白的氧化)。
本发明中的组合物能减小细胞中内部氧化和/或外部氧化性损伤的数量,增加细胞中胶原蛋白的合成,减小细胞中感染细胞因子的产生。这些细胞包括人表皮角质形成细胞,人真皮成纤维细胞,或人黑色素细胞,或它们的任意组合(如人角质形成细胞和人成纤维细胞的组合或人角质化细胞和人黑色素细胞的组合)。
本发明中的组合物能治疗色素沉着过度,本方法也可包含确定一个人是否需要治疗皮肤色素过度沉着。通过施用本发明中的组合物还可用于减少皮肤外观中的老年斑、皮肤色素减退、雀斑,减少或防止皮肤的细纹或皱纹,增加皮肤的紧固性。
本发明中含红麻提取物的组合物具有抗氧化、抑制酪氨酸酶、抑制弹性酶、抑制TNF-α酶、抑制脂氧化酶、抑制COX-1和/或COX-2酶和抑制MMP-1和/或MMP-2的活性的能力。这些组合物可作为局部皮肤组合物,包括皮肤学上可接受的载体。这些组合物可治疗或预防与皮肤氧化、皮肤细胞中酪氨酸酶、弹性酶、TNF-α酶、脂氧化酶、COX-1和/或COX-2酶和MMP-1和/或MMP-2的活性相关的皮肤病。
实施例4:
按实施例1制得红麻提取物,按提取物与赋形剂重量比1:1的比例加入赋形剂,制粒压片。
实施例5:
按实施例1制得红麻活性提取物,按提取物与赋形剂重量比1:2的比例加入赋形剂,制粒压片。
实施例6:
按实施例1制得红麻提取物,按常规胶囊制剂方法制成胶囊。
实施例7:
先按实施例1制得红麻提取物,再按下述方法制成片剂
片剂:红麻提取物100mg
淀粉适量
玉米浆适量
硬脂酸镁适量
实施例8:
胶囊剂:红麻提取物100mg
淀粉适量
硬脂酸镁适量
制备方法:将红红麻提取物与助剂混合,过筛,在合适的容器中均匀混合,把得到的混合物装入硬明胶胶囊。
实施例9:
制备方法:搅拌下于适当体积的重蒸馏水中每次加入一种成分,直至完全深解,然后再加入另一种成分。加水至2ml后,将该溶液在无菌过滤器上过滤,装入瓶中并按照适当的剂量分隔。
实施例10:
滴丸:红麻提取物1g
聚乙二醇60009g
制法:红麻提取物6000熔融液的制备:按上述处方量称取红麻提取物,加入适量无水乙醇,微热溶解后,加入处方量的聚乙二醇熔融液中(60℃水浴保温),搅拌混合均匀,直至乙醇挥尽为止,静置于60℃水浴中保温30分钟,待气泡除尽,然后将除尽气泡的上述混匀熔融液转入贮液筒内,在保温80-85℃的条件下,控制滴速,一滴滴地滴入冷凝液中,等冷凝完全,倾去冷凝液,收集滴丸,沥净和用滤纸除去丸上的冷凝液,放置硅胶干燥器中或自然干燥即可。
实施例11:
片剂:实施例1所得红麻提取物10mg,乳糖180mg,淀粉55mg,硬脂酸镁5mg;
制备方法:将提取物、乳糖和淀粉混合,用水均匀湿润,把湿润后的混合物过筛并干燥,再过筛,加入硬脂酸镁,然后将混合物压片,每片重250mg,化合物含量为10mg。
实施例12:
安瓿剂:实施例1所得红麻提取物2mg,氯化钠10mg;
制备方法:将红麻提取物和氯化钠溶解于适量的注射用水中,过滤所得溶液,在无菌条件下装入安瓿瓶中。
实施例13:
胶囊剂:实施例1所得红麻提取物10mg,乳糖187mg,硬脂酸镁3mg;
制备方法:将红麻提取物与助剂混合,过筛,均匀混合,把得到的混合物装入硬明胶胶囊,每个胶囊重200mg,活性成分含量为10mg。
实施例14:
含有红麻提取物的美白霜配方(W%):
按常规制作化妆品的方法制得本发明上述配方的化妆品。
实施例15:
含有红麻提取物的乳剂配方(W%):
按常规制作化妆品的方法制得本发明上述配方的化妆品。
实施例16:
配方:
A油相w(质量百分比)
蜂蜡2%,硬脂酸5%,十八醇3%,羊毛脂2%,司盘803%,土温803%,石蜡油3%。
B水相w(质量百分比)
甘油5%,本发明红麻提取物5%,三乙醇胺0.3%,蒸馏水75%。
C香精适量,尼泊金丙酯0.2%
方法:
将油相置于恒温水浴锅内搅拌加热至90℃完全熔化成均相后,缓慢加入水相,继续搅拌30min;缓慢降温至45℃时加入香精、尼泊金丙酯,继续搅拌;待温度降至35℃左右时,停止搅拌,静置,冷却,得到乳膏状产品。

1.红麻提取物,由下述方法制备而得:取红麻根,研成粉末,用红麻的15-30倍量的85%-95%乙醇,冷浸18-28h,收集滤液,重复2-5次,滤液减压蒸馏,浓缩滤液,低温干燥,得粗提物,将粗提物经大孔树脂吸附脱色,85%-95%乙醇洗脱后,浓缩蒸干,低温冷冻干燥,得红麻提取物。
2.红麻提取物的制备方法,其特征在于该方法包括下述步骤:取红麻根,研成粉末,用红麻的15-30倍量的85%-95%乙醇,冷浸18-28h,收集滤液,重复2-5次,滤液减压蒸馏,浓缩滤液,低温干燥,得粗提物,将粗提物经大孔树脂吸附脱色,85%-95%乙醇洗脱后,浓缩蒸干,低温冷冻干燥,得红麻提取物。
3.化妆品,由权利要求1所述的红麻提取物和化妆品常用载体组成,所述的载体选自一种或多种含水体系,甘油、C1-4醇、脂肪醇、多元醇、脂肪醚、脂肪酯、乙二醇、丙二醇、植物油、矿物油、脂质体、层状类脂材料、硅油、水,或者它们的组合。
4.权利要求1所述的红麻提取物在制备化妆品中的应用。
5.权利要求1所述的红麻提取物在制备保湿或嫩肤或抗皱或防晒或美白化妆品中的应用。
6.权利要求1所述的红麻提取物在制备麻醉剂、抗真菌剂、抗炎剂、防腐剂、螯合剂、着色剂、脱色剂、软化剂、乳化剂、脱落剂、成膜剂、芳香剂、湿润剂、驱虫剂、润滑剂、增湿剂、药剂、光稳定剂、防腐剂、皮肤保护剂、皮肤渗透增强剂、防晒剂、稳定剂、表面活性剂、增稠剂、粘度调节剂、维生素中的应用。
7.权利要求1所述的红麻提取物在制备抗氧化剂中的应用。
8.预防或治疗皮肤病的药物组合物,由权利要求1所述的红麻提取物和药物常用载体组成。
9.权利要求1所述的红麻提取物在制备预防或治疗皮肤病的药物中的应用。
