背景技术
肥胖症是一种由某些特定的生理遗传因素参与的能量代谢平衡发生紊乱的疾病状态。随着全球工业化、都市化进展以及社会经济的迅猛发展和生活方式的剧烈改变,近二三十年肥胖症的发病率急剧增长,已成为“全球蔓延的慢性非传染性疾病”,严重威胁人类的健康。研究表明,美国成年男性肥胖患者在年龄大于20岁人群中所占的比例高达35.5%,成年女性高达35.8%。本中心全国10万人大型流行病学调查资料显示,年龄大于18岁的自然人群经抽样调查,超重人群比例达到30%,而肥胖人群比例则达到了5.72%,由此可见国内肥胖症的发病形势也日益严峻。事实上,国内儿童、青少年肥胖已初见端倪。由于肥胖症是2型糖尿病、心血管疾病以及某些肿瘤如大肠癌,乳腺癌等疾病的重要危险因素,肥胖相关疾病患者的死亡率明显增加,该疾病已成为严重影响全球公共卫生的社会经济问题。为此世界卫生组织(WHO)认为肥胖是21世纪最严重的公共健康问题,2013年美国医学会已经正式将肥胖定义为一种疾病。因此开展肥胖症发病机制相关研究对于筛查肥胖易感人群、治疗肥胖症、预防肥胖症在儿童和青少年群体的大规模爆发极为重要。
脂肪组织来源于中胚层的间充质干细胞(MSC)。间充质干细胞可以分化为骨骼,肌肉,脂肪和软骨。在调节间充质干细胞向脂肪分化发育的信号通路和因子中,Wnt信号通路发挥重要作用。经典Wnt信号通路通过调节β-catenin核转位传递信号,参与细胞命运决定、增殖、分化以及肿瘤生成等多个生理病理过程。在缺乏Wnt配体刺激时,β-catenin在胞浆内与Axin、APC和GSK3等组成的降解复合体结合并被磷酸化,经蛋白酶体而进入泛素化蛋白降解途经;而当Wnt配体(如Wnt1、Wnt10b或Wnt3a等)刺激存在时,其与细胞表面辅助受体包括Fzd(Frizzled)-LRP5或Fzd-LRP6结合,促发β-catenin降解复合体解离,于是胞浆β-catenin量增加,进而转位至细胞核,作为TCF/LEF共激活因子促进下游基因转录。多项研究均已表明Wnt/β-catenin信号通路是白色脂肪分化发育的负性调控因素。目前已发现Wnt/β-catenin/TCF/LEF下游靶基因CyclinD1、WISP2、COUP-TFII以及Id2等均可通过抑制脂肪分化转录因子PPARγ和C/EBPα的表达,发挥抑制白色脂肪分化的作用。
临床病例研究以及转基因动物模型研究的结果表明,Wnt信号通路上的分子异常能够导致肥胖及代谢综合征的发生。比如,在肥胖家系可检测出Wnt10b基因突变,LRP6基因突变可导致高血脂、高血压以及糖尿病的发生,LRP5基因突变可以引起人体脂肪的分布改变,TCF7L2的多个基因多态性位点在多个人群被证实与糖尿病发生存在显著关联,Ap2-Wnt10b转基因小鼠抵抗高脂饮食诱导或遗传异常导致的肥胖。这些证据均表明,经典Wnt/β-catenin信号通路可抑制白色脂肪的分化,参与体重调节或影响能量代谢。在对棕色脂肪发育调节方面,仅有零星研究提示,早期激活该通路可抑制棕色脂肪分化,而后期激活则可促进棕色脂肪向白色脂肪转化,但具体证据仍十分有限。随着白、棕色脂肪成为肥胖抵抗的干预靶点,阐明经典Wnt信号通路在脂肪发育及能量代谢方面的参与作用具有非常重要的科学意义和临床意义。
遗憾的是,在脂肪组织特异性敲除β-catenin的工作至今未见报道;而这对于认识经典的Wnt通路在肥胖发生与体重调节中的作用至关重要。
发明内容
发明人利用β-cateninflox/flox和adiponectin-cre小鼠进行交配,首次建立脂肪组织特异性β-catenin敲除小鼠(β-cateninflox/flox;AdipoQ-cre),从而从体内水平探究β-catenin在脂肪发育过程中的作用。这对于从机体整体水平认识WNT通路,尤其是经典WNT通路在脂肪发育与体重调节中的作用至关重要。
具体而言,本发明一方面提供了一种成熟脂肪组织β-catenin敲除的小鼠模型的构建方法,其特征在于,包括以下步骤:
分别选用β-cateninflox/flox小鼠与Adiponectin-Cre小鼠繁殖,得到F1代:β-cateninflox/+;AdipoQ-Cre;
将F1代进行繁殖,得到F2代β-cateninflox/flox和β-cateninflox/flox;AdipoQ-Cre的敲除小鼠。
在某些具体实施方式中,所述β-cateninflox/flox小鼠为SPF级;
在某些具体实施方式中,所述β-cateninflox/flox小鼠具有C57BL/6J背景。
在本发明的另一方面,提供了一种通过权利要求1所述方法构建的成熟脂肪组织β-catenin敲除的小鼠模型在研究β-catenin在脂肪发育过程中的作用的应用。
通过上述技术方案,本发明提供了一种构建的小鼠模型的方法,其在探讨经典wnt信号通路核心分子β-catenin在体重调节和肥胖发生中的作用具有较为重要的意义。

具体实施方式
小鼠繁殖
采用SPF级β-cateninflox/flox小鼠(C57BL/6J背景)与Adiponectin-Cre小鼠繁殖,获得F1代(β-cateninflox/+;Ap2-Cre或者β-cateninflox/+;AdipoQ-Cre)之后,再用双杂合的F1进行繁殖,即可获得F2代对照小鼠β-cateninflox/flox;AdipoQ-Cre的敲除小鼠。
小鼠在以下条件分笼喂养:标准商业小鼠饲料(4.5%脂肪,4%纤维素,21%蛋白质、1.404kcal/g)、高脂饲料购自ResearchDiet(60%脂肪,20%碳水化合物,20%蛋白质),室温20-22℃,自由摄食和饮水,稳定的人工光照周期(6:00到18:00光照,18:00到6:00黑暗)。8周龄雄鼠和雌鼠按1:2比例合笼交配。
一周幼鼠基因型鉴定,三周幼鼠离乳、标记、分笼。没有特殊说明的,所有试验均采用雄性小鼠。实验所用动物均由上海交通大学医学院动物中心伦理委员会提供监管。
如图1所示,发明人建立了Adiponectin启动子驱动的cre重组酶介导的成熟脂肪细胞β-catenin敲除的小鼠模型。在获得的F2子代小鼠中,通过基因组DNAPCR的方法初步筛出敲除小鼠。
基因型鉴定

PCR反应体系见下:
96孔PCR反应板中加入上述混合液后离心数秒,每孔加石蜡油一滴。PCR反应在EppendorfMastercycleGradientPCR热循环仪上进行。
PCR扩增条件为:94℃3min;94℃30s,51.7℃60s,72℃60s35cycles,72℃2min,Hold14℃。
取PCR产物5μl在1.5%琼脂糖凝胶上电泳,溴化乙锭染色。用DL2000标记鉴定。
鼠尾组织DNA抽提
1.取2周龄小鼠,用剪刀取0.5cm长鼠尾,移入1.5ml离心管并标记。
2.加350μl裂解液,50μl蛋白酶K储存液56℃烤箱溶解过夜。
3.次日样本于室温12000rpm离心10分钟。
4.上清倒入1.5ml已标记离心管,加800uL无水乙醇,盖后振摇,可见絮状沉淀。
5.12000rpm4℃离心10分钟。
6.弃上清,倒立吸水纸约3-5分钟,待乙醇挥发。7.加100ulTE液吹打溶解后-20℃冻存备用。
琼脂糖凝胶电泳法鉴定PCR产物
1.将洗净的水平电泳凝胶槽放入夹板。
2.1XTAE电泳缓冲液在微波炉上将琼脂糖颗粒完全溶解,琼脂糖的浓度为2%。
3.溶化的琼脂糖溶液冷却至60℃时,加入溴化乙锭,充分摇匀。
4.在胶模上放置好梳子后,将温热凝胶倒入胶模中,避免产生气泡。
5.待凝胶凝固后,放入电泳槽,加入1XTAE电泳缓冲液,使液面高出凝胶表面5mm。
6.将PCR产物与6Xloadingbuffer混合,用微量加样器将样品依次加入加样孔内。
7.电压140V电泳,45min。电泳完成后切断电源,取出凝胶,在凝胶成像系统中对凝胶进行观察和摄影记录。
对小鼠一般情况观察
观察动物的一般表现、行为、进食及死亡情况。3周小鼠分笼,6周开始称重到实验终止,依次每周定时称量体重一次。发明人对小鼠进行高脂饮食喂养,探究β-cateninflox/flox;AdipoQ-cre小鼠对于高脂诱导的肥胖的反应性。
如图2所示,体重增长曲线显示,高脂饮食喂养12周后ABKO小鼠体重即明显低于对照小鼠,而高脂喂养到8个月时,ABKO小鼠平均体重比对照小鼠体重减轻了近15%,约8克。
鼠糖代谢指标观察
IPGTT试验
腹腔注射葡萄糖耐量试验(Intra-peritonealglucosetolerancetest,IPGTT):正常饮食小鼠饥饿16小时后,2g/kg腹腔注射葡萄糖,0、30、60、120分钟后尾静脉采血,测定血糖;高脂饮食小鼠葡萄糖的剂量减半为1g/kg体重,其余操作不变。
如图3所示,相应于体重的下降,ABKO小鼠葡萄糖耐量也明显改善。
血清胰岛素ELISA测定
1.实验前将所有试剂复温至室温。
2.10X洗液稀释10倍备用。
3.用洗涤液将反应板洗涤3次,每孔加300μl洗涤液,弃去洗涤液,在滤纸上轻轻地拍干。
4.标准品、空白对照及NSB孔加入10μlMatrix液。
5.标准孔加入胰岛素标准品。
6.质控孔分别加入10μlQC1和QC2。
7.检测孔加入10μl待测样品。
8.所有的孔加入80μl检测抗体,用封口膜封好,室温摇床500转孵育2小时。
9.甩去孔内液体,轻轻拍干。
10.每孔加300μl洗涤液洗涤3次,避免产生气泡。
11.每孔加入100μl酶反应液,用封口膜封好,室温500转每分钟孵育30分钟。
12.倒去孔内液体,轻轻拍干。
13.每孔加300μl洗涤液洗涤6次,避免产生大气泡。
14.每孔加入100μl底物工作液,用封口膜封好,室温500转每分钟孵育15分钟反应将根据胰岛素浓度的不同表现为深浅不同的蓝色。
15.每孔加入100μl终止液,轻轻晃动检测板使液体充分混匀。蓝色将变成黄色在5分钟内检测450nm和590nm的吸光度。检测前确保孔中没有气泡。
16.所有OD值都应减除空白值,计算出校正OD值;
17.以标准品之校正OD。
如图4所示,相应于体重的下降,ABKO小鼠血胰岛素水平也显著降低。
成熟脂肪组织β-catenin敲除的小鼠模型的应用
本发明人还通过成熟脂肪组织β-catenin敲除的小鼠模型对β-catenin在脂肪发育过程中的作用进行了研究,发现β-catenin调控SAA3表达,可能是脂肪组织增生、肥胖发生的分子机制。
为了验证和寻找这种可能的因子,发明人对ABKO小鼠和正常小鼠这两组小鼠的皮下脂肪组织mRNA进行了芯片筛查。在皮下脂肪组织的芯片中,发现SAA3是表达量下降最显著的基因,并且SAA3也是可以被分泌的。文献显示重组SAA显著促进3T3-L1细胞的增殖。通过实时定量PCR,证实敲除小鼠的皮下脂肪组织SAA3的表达显著下降。
进一步发明人在体外诱导分化成熟的原代脂肪细胞和3T3-L1细胞中加入WNT3a作用16小时,可以显著上调SAA3的表达,提示SAA3有可能是WNT信号通路在脂肪组织的的下游。发明人构建了SAA3启动子序列的报告基因,在转入过表达质粒β-catenin之后,SAA3启动子的活性显著上调,这说明β-catenin转录调控SAA3的表达。β-catenin通常作为转录共激活因子与下游转录因子TCF4/LEF1共同调控下游靶基因的转录。
通过软件分析预测,发明人在SAA3的启动子区域发现了四个TCF4结合区域,发明人对以上四个位点进行了突变,发现所有位点突变之后,SAA3的启动子活性在β-catenin过表达条件下显著下调,从而证明β-catenin对SAA3的直接调控是TCF4依赖的。发明人正在进行染色质免疫共沉淀实验。SAA3的启动子序列存在TCF4结合的种子序列,发明人预期在过表达β-catenin之后,TCF4的结合显著增多,而在过表达种子序列突变之后的体系中,这种结合显著降低。
在以上实验数据的基础上,发明人提出了这样一个模式图:高脂饮食诱导的肥胖过程中,成熟脂肪细胞β-catenin可以上调SAA3,引起SAA3分泌增加,进而前脂肪细胞增殖并分化成熟脂肪细胞以应对巨大的能量压力。而成熟脂肪细胞中β-catenin失活之后,SAA3的表达则下调,前脂肪细胞的增殖减少,终末的成熟脂肪细胞数目相应下降。
Claims (4)
1.一种成熟脂肪组织β-catenin敲除的小鼠模型的构建方法,其特征在于,包括以下步骤:
分别选用β-cateninflox/flox小鼠与Adiponectin-Cre小鼠繁殖,得到F1代:β-cateninflox/+;AdipoQ-Cre;
将F1代进行繁殖,得到F2代β-cateninflox/flox和β-cateninflox/flox;AdipoQ-Cre的敲除小鼠。
2.如权利要求1所述的方法,其特征在于,所述β-cateninflox/flox小鼠为SPF级。
3.如权利要求1所述的方法,其特征在于,所述β-cateninflox/flox小鼠具有C57BL/6J背景。
4.一种通过权利要求1所述成熟脂肪组织β-catenin敲除的小鼠模型在研究β-catenin在脂肪发育过程中的作用的应用。
