本发明涉及一种纺锤形和侧扁形鱼类快速摘取矢耳石的方法,包括:用不同方法取出鱼的矢耳石;将得到的耳石于蒸馏水中去除外包膜和黏液,吸干表面水分后保存。本发明操作简单,耗时短,能保证取出的鱼类耳石完整,具有良好的应用前景。

Description

一种纺锤形和侧扁形鱼类快速摘取矢耳石的方法
技术领域
本发明属于矢耳石摘取领域,特别涉及一种纺锤形和侧扁形鱼类快速摘取矢耳石的方法。
背景技术
耳石是鱼类左右内耳膜迷路内成对的结石,起平衡和听觉作用。鱼类孵化时耳石就开始沉积出现,随后伴随着生长、发育,耳石不断围绕核心部沉积、增厚、扩大至最边缘。许多鱼的耳石每天都增厚一层,通过计算鱼耳石的“环层”数和特点,便能得知鱼的日龄甚至年龄。鱼类从水环境中吸收的元素可以在耳石中沉积并长期保存栖息水环境中相应元素特征,对这些元素特征进行深入的研究,可以恢复、重现和掌握大量水生动物实际生活过的环境以及生理学等方面的信息。
基于上述原理,近些年来在国际上飞速发展的研究鱼类耳石微结构、耳石元素微化学以及耳石形态等研究,有着非常独特而重要的鱼类资源学和保护生物学的价值。矢耳石一般为耳石研究的首选对象。
耳石的研究涉及到从鱼体的头部摘取耳石,并且在耳石微结构和形态学的研究方面需要大量的耳石标本进行图像采集和统计判别。但目前耳石的摘取方法均采用解剖剪刀沿着鱼体方向将头盖骨剪开,将头部其余部分剥离后寻找耳石,由于剥离时头部有机组织和杂质较多,易造成混淆,整个耳石摘取过程比较复杂、费时,并可能导致耳石流入鱼体的头部内部后丢失。
发明内容
本发明所要解决的技术问题是提供一种纺锤形和侧扁形鱼类快速摘取矢耳石的方法,该方法操作简单,耗时短,能保证取出的鱼类耳石完整,具有良好的应用前景。
本发明的一种纺锤形鱼类快速摘取矢耳石的方法,包括:
(1)翻开纺锤形鱼类的鱼体鳃部;
(2)用镊子将鱼内耳的球状囊戳穿,并取出鱼的矢耳石;
(3)将得到的矢耳石于蒸馏水中去除耳石外包膜和黏液,吸干表面水分后保存。
所述步骤(3)中用带有编号的耳石槽或离心管保存。钙质薄的耳石,以耳石槽或离心管保存;钙质厚的耳石也可用有编号的牛皮纸袋保存。
本发明的一种侧扁形鱼类快速摘取矢耳石的方法,包括:
(1)将侧扁形鱼类的鱼背部在上、腹部朝下,在鱼眼斜上方的背部处用解剖刀下切;
(2)掰开切口处,从鱼内耳的球状囊中取出鱼的矢耳石;
(3)将得到的矢耳石于蒸馏水中去除耳石外包膜和黏液,吸干表面水分后保存。
所述步骤(1)中用解剖刀下切至鳃盖下缘。
所述步骤(3)中用带有编号的耳石槽或离心管保存。钙质薄的耳石,以耳石槽或离心管保存;钙质厚的耳石也可用有编号的牛皮纸袋保存。
有益效果
本发明操作简单,耗时短,能保证取出的鱼类耳石完整,具有良好的应用前景。

实施例1
(1)用镊子掀开小黄鱼(纺锤形鱼类)的鳃盖(图1中的1),轻轻拨开鱼鳃,出现透明的球状囊(图1中的2)。
(2)用尖头镊子将球状囊戳穿,取出小黄鱼的一对矢耳石(图1中的3)。
(3)将分离出的耳石于蒸馏水中去除耳石外包膜和黏液,用吸水纸吸干表面水分后放入有耳石编号的牛皮纸袋(长7cm×宽5cm)中保存。
实施例2
(1)切头盖骨:将银鲳(侧扁形鱼类)背部在上、腹部朝下,从银鲳的眼外缘至鳃盖缘之间约2/3处,沿与眼外缘平行方向的切割线(图2中的1),在鱼的背部用解剖刀切入,切至鳃盖的下缘。
(2)掰开切口处,出现“心”型外框(图2中的2),即为鱼内耳的球状囊纵切面,用尖头镊子轻轻取出银鲳的一对矢耳石(图2中的3)。
(3)将分离出的耳石于蒸馏水中去除耳石外包膜和黏液,用吸水纸吸干表面水分后,放入有耳石编号的1.5ml离心管中保存。

Claims (2)

1.一种快速摘取侧扁形鱼类矢耳石的方法,包括步骤:
(1)将侧扁形鱼类的鱼背部在上、腹部朝下,从鱼眼外缘至鳃盖缘之间约2/3处,沿与眼外缘平行方向的切割线,在鱼眼斜上方的背部处用解剖刀下切至鳃盖下缘;
(2)掰开切口处,出现“心”型外框,即为鱼内耳的球状囊纵切面,从鱼内耳的球状囊中取出鱼的矢耳石;
(3)将得到的矢耳石于蒸馏水中去除耳石外包膜和黏液,吸干表面水分后保存。
2.根据权利要求1所述的一种快速摘取侧扁形鱼类矢耳石的方法,其特征在于:所述步骤(3)中得到的耳石用带有编号的耳石槽或离心管保存。
