技术领域
本发明涉及一种富硒地肤子粉及其制备方法、应用。
背景技术
硒是一种人和动物所必需的微量无素,具有清除自由基、抗氧化、延缓衰老、抗癌,增强免疫等生物学特性,被称之为“抗癌之王”。同时,也是人体重要的长寿微量元素。 硒总体分为无机硒和有机硒两类,无机硒主要有硒酸盐、亚硒酸盐等,对于人和动物来说,毒性风险大,吸收效果差,使用不当易造成中毒。有机硒主要是指与生物有机硒分子结合形态的硒,对人和动物来说,毒性风险小,吸收率高,更适合补硒的要求。由于人体内不能合成而必需由外界供应,为达到人体补硒的目的,出现了一些富含硒含量的水果,如 中国专利95109479.3“一种提高水果硒含量的方法”、《 安微农业科学》公开的“高硒西瓜无公害栽培技术”等,这些技术实施较为复杂,富硒水果应用范围较小。
发明内容
本发明的目的是为了克服现有技术的不足而提供一种工艺合理、操作方便、产量高、应用范围广的富硒地肤子粉及其制备方法、应用。
为实现上述目的,本发明所采用的技术方案是:
一种富硒地肤子粉,由下列方法制得:
a、当年秋天采集地肤子种子,播种于平地上;
b、第二年自然生长,间苗后培育,在开花结果时期向其喷洒无机硒复合液,相隔5-10天进行第二次喷雾;
c、喷后经过20天后收割;
d、然后切断,晾干,粉碎,100目以上过筛制得富硒地肤子粉。
所述的无机硒复合液是由下列原料配制而成:二氧化硒或亚硒酸钠130克、小苏打200克、柠檬酸150克、谷氨酸钠150克、水2500克。
所述的制得富硒地肤子粉中含有70~160mg/kg的有机硒。
所述的无机硒复合液喷雾时为阳光照射下的午后时间14-16时。
一种富硒地肤子粉的制备方法,包括下列步骤:
a、当年秋天采集地肤子种子,播种于平地上;
b、第二年自然生长,间苗后培育,在开花结果时期向其喷洒无机硒复合液,相隔5-10天进行第二次喷雾;
c、喷后经过20天后收割;
d、然后切断,晾干,粉碎,100目以上过筛制得富硒地肤子粉。
所述的一种富硒地肤子粉可作为食品、保健品或饮料的添加剂的应用。
本发明中的地肤子是食用植物,野生自长的地肤子在东北地区自然界广泛存在,且生长快。到秋天采集,往平地一扔,第二年即可自长,间苗后培育,到秋天九月份开花结果时期往叶柱上喷酒无机硒复合液后,收割、切断、凉干、粉碎、制粉,就完成了整个生产过程,整个工艺合理、操作方便,原料易得。特别是地肤子为食用植物,生命力和耐药性强,产量高,比灵芝多产16倍以上,中等土壤每垧可产2500斤以上。通过实施叶肥法,可节约无机硒复合液的药原料,植株富集有机硒含量高,有机硒易为人体代谢和吸收。用富含有机硒的地肤子作为食品、保健品的添加剂,或直接泡成水溶混悬液,添加少量蜂蜜作为饮料直接饮用,可以持补充人体缺乏的微量元素硒,有效的预防因为缺硒导致的各种疾病。
本发明的地肤子粉经延边大学分析测试中心检测,测试项目及含量mg/kg:
Cr 170.01,Mn 126.4 , Fe 32.73,Cu 8.21,Zn 91.81,
Se 46.14,Mo 80.31,K 7.53%,Ca 2.00%,Mg 1.00%。
本发明的地肤子水溶液经国家化学和光谱研究分析中心分析测试,测试项目及含量ng/ml:

为了检验本发明地肤子有机硒产品的安全性和效能,特进行了急性毒性试验和对老龄鼠的短期喂养试验:
1材料和方法
1.1材料:
供试品: 1.地肤子有机硒(由金银赫、金哲浩等研制,延吉市喜康科技研究所提供). 2.有机硒提纯品(由延边大学农学院生物技术实验教学中心提纯).实验动物: 小白鼠(由延大实验动物中心提供).试剂: 包括联苯三酚,谷胱甘肽等(国产分析纯).仪器: 紫外分光光度计,电子天平,酸度计,转棒疲劳仪,组织匀浆器。
1.2方法
1.2.1急性毒性实验: 1. 一日内地肤子有机硒掺加在饲料中最大浓度
· (3g*313.1mg/1000g)=0.939 g
· 0.939/30/1000=0.313g/Kg
· 2. 把有机硒提纯浓缩物悬浮于1%的羧甲基纤维素钠溶液中,一次灌胃最大浓度.
· 小鼠观察,没有出现可见的毒性反应,观察到给药7日为止,计算存活率.
1.2.2短期喂养及抗疲劳实验:
将28只年龄在十个月以上的试验小白鼠根据雌雄不同随机分为6组(R、S、T、U、V、W),R、S、T为雌鼠组,其中又分为对照组(R)、低硒组(S)(0.2%)和高硒组(T)(1%),U、V、W为雄鼠组,其中又分为对照组(U)、低硒组(V)(0.2%)和高硒组(W)(1%),试验鼠自由采食每组对应的有机硒饲料。实验结束前利用转棒疲劳仪测定小鼠的抗疲劳能力. 实验结束时测量体重,脏器指数,抗氧化指标:过氧化物歧化酶(SOD);丙二醛(MDA);谷胱甘肽(GSH)。
1.2.3组织匀浆的制备
小鼠实验结束时剖检,取主要脏器:肝脏,大脑,肾脏;称取所需的少量脏器,用生理盐水溶液研磨,离心,制备10%的组织匀浆,用于抗氧化指标的测定.
SOD的测定及比活力的计算: 取PH=8.3的磷酸氢钾于比色杯中,再向其加入样品溶液,再向其加入联苯三酚,λ=340nm下测分光光度值,一共测六次,然后做一分钟内的差值,得到四组差值后,算其平均值(A325)。

1.2.4 MDA和GSH的测定
0.12ml样品,0.12ml8.1%SDS,0.6mlDW,0.9ml0.2M的乙酸,0.9ml1%的TBA,混匀后90度水浴15分钟,迅速冷却后再离心,取上清液,λ=532nm下测分光光度值。
0.1ml的样品稀释液,0.5ml的双蒸水(DW),0.3ml的四氧嘧啶,0.3ml0.24M的磷酸钠缓冲液,0.3ml0.1M的NaOH,混匀后,避光静止10分钟后,再加入0.3ml1M的NaOH,混匀后λ=305nm 的紫外可见分光光度计下测分光光度值。
2 结果
2.1.急性毒性实验结果
小鼠观察,没有出现可见的毒性反应,观察到给药7日为止100%存活.
2.2.抗疲劳实验结果

2.3.1动物剖检,观察,没有肉眼可见的异常变化。取主要脏器称重,并计算脏器指数;制取主要脏器的组织匀浆,测定抗氧化指标。结果如下表。

从以上实验得出:本发明的地肤子有机硒及提取浓缩物在本实验条件下对于小鼠没有出现急性毒性,是低毒安全的.饲喂有机硒对于小鼠有抗疲劳作用.有机硒能增强老龄小鼠的抗氧化能力,并且最适的浓度是0.2%。
具体实施方式

第二年自然生长,间苗后培育,在开花结果时期向其喷洒无机硒复合液,相隔5-10天进行第二次喷雾,无机硒复合液喷雾时为阳光照射下的午后时间14-16时,优选15时;所述的无机硒复合液是由下列原料配制而成:二氧化硒或亚硒酸钠130克、小苏打200克、柠檬酸150克、谷氨酸钠150克、水2500克;
喷后经过20天后收割;

当年秋天采集地肤子种子,播种于平地上;
第二年自然生长,间苗后培育,在开花结果时期向其喷洒无机硒复合液,相隔5-10天进行第二次喷雾,无机硒复合液喷雾时为阳光照射下的午后时间14-16时,优选15时;所述的无机硒复合液是由下列原料配制而成:二氧化硒或亚硒酸钠130克、小苏打200克、柠檬酸150克、谷氨酸钠150克、水2500克;
喷后经过20天后收割;
然后切断,晾干,粉碎,100目以上过筛制得富硒地肤子粉。
所述的一种富硒地肤子粉可作为食品、保健品或饮料的添加剂的应用。
用富含有机硒的地肤子作为食品、保健品的添加剂,或直接泡成水溶混悬液,添加少量蜂蜜作为饮料直接饮用,可以持补充人体缺乏的微量元素硒,有效的预防因为缺硒导致的各种疾病。

Claims (6)
1.一种富硒地肤子粉,由下列方法制得:
a、当年秋天采集地肤子种子,播种于平地上;
b、第二年自然生长,间苗后培育,在开花结果时期向其喷洒无机硒复合液,相隔5-10天进行第二次喷雾;
c、喷后经过20天后收割;
d、然后切断,晾干,粉碎,100目以上过筛制得富硒地肤子粉。
2.根据权利要求1所述的一种富硒地肤子粉,其特征在于所述的无机硒复合液是由下列原料配制而成:二氧化硒或亚硒酸钠130克、小苏打200克、柠檬酸150克、谷氨酸钠150克、水2500克。
3.根据权利要求1所述的一种富硒地肤子粉,其特征在于所述的制得富硒地肤子粉中含有70~160mg/kg的有机硒。
4.根据权利要求1所述的一种富硒地肤子粉,其特征在于所述的无机硒复合液喷雾时为阳光照射下的午后时间14-16时。
5.一种富硒地肤子粉的制备方法,包括下列步骤:
a、当年秋天采集地肤子种子,播种于平地上;
b、第二年自然生长,间苗后培育,在开花结果时期向其喷洒无机硒复合液,相隔5-10天进行第二次喷雾;
c、喷后经过20天后收割;
d、然后切断,晾干,粉碎,100目以上过筛制得富硒地肤子粉。
6.权利要求1所述的一种富硒地肤子粉作为食品、保健品或饮料的添加剂的应用。
