本发明公开了一种红色裸甲藻培养基,所述红色裸甲藻培养基包括以下质量浓度的组分:NaNO380‑120mg/L、NaH2PO48‑12mg/L、柠檬酸铁铵2.5‑3.5mg/L、Na2EDTA10‑12mg/L、Na2SiO3·9H2O20‑30mg/L、VB10.005‑0.007mg/L和VB120.04‑0.06mg/L;还有植物生长调节因子,植物生长调节因子包括以下质量浓度的组分:二苯基脲磺酸钙0.05‑0.1mg/L和抗坏血酸0.1‑1mg/L。本发明所述的培养基在培养红色裸甲藻时具有生长速率快、细胞密度大的优点,适于规模化批量生产。

一种红色裸甲藻培养基
技术领域
本发明涉及一种藻类培养基,尤其涉及一种红色裸甲藻培养基。
背景技术
红色裸甲藻,裸甲藻科的1属,鞭毛藻类,腰鞭毛虫的一个属,绝大多数为海产,多分布在热带和温带海域。能够产生贝类毒素,具有一定的科研和药用价值。但是红色裸甲藻在人工培养室,生长较慢,生物量低,不利于毒素的生产和提取。
发明内容
本发明所要解决的技术问题是提供一种利于提高红色裸甲藻生长速率、增大细胞密度的红色裸甲藻培养基。
为解决上述技术问题,本发明的技术方案是:所述红色裸甲藻培养基包括以下质量浓度的组分:NaNO3 80-120mg/L、NaH2PO48-12mg/L、柠檬酸铁铵2.5-3.5mg/L、Na2EDTA10-12mg/L、Na2SiO3·9H2O20-30mg/L、VB10.005-0.007mg/L和VB12 0.04-0.06mg/L。
作为优选的技术方案,所述红色裸甲藻培养基包括以下质量浓度的组分:NaNO360-120mg/L、NaH2PO4 8-12mg/L、柠檬酸铁铵2.5-3.5mg/L、Na2EDTA 10-12mg/L、Na2SiO3·9H2O 20-30mg/L、VB1 0.005-0.007mg/L、VB12 0.04-0.06mg/L和植物生长调节因子,植物生长调节因子包括以下质量浓度的组分:二苯基脲磺酸钙0.05-0.1mg/L和抗坏血酸0.1-1mg/L。
本发明的优点是:本发明提供的红色裸甲藻培养基利于藻细胞快速生长,培养出的红色裸甲藻细胞密度大,特别是由于添加了植物生长调节因子二苯基脲磺酸钙和抗坏血酸,配方配伍合理,利于促进藻细胞快速增殖,使得藻细胞浓度增大,适于大批量规模化培养生产。
具体实施方式
下面结合具体实施例,对本发明提供的红色裸甲藻培养基进行详细的说明。
实施例一:
(1)用自然海水配置如下配方的红色裸甲藻培养基:NaNO3 80-120mg/L、NaH2PO48-12mg/L、柠檬酸铁铵2.5-3.5mg/L、Na2EDTA10-12mg/L、Na2SiO3·9H2O20-30mg/L、VB10.005-0.007mg/L和VB12 0.04-0.06mg/L。
(2)取对数生长期的红色裸甲藻接种到该培养基中进行培养,每天光照10小时,光照强度约为50μmol/(m2·s),培箱温度控制在25摄氏度,每隔一段时间记录培养基中红色裸甲藻的细胞浓度,详见表1。
实施例二:
(1)用自然海水配置如下配方的红色裸甲藻培养基:NaNO3 80-120mg/L、NaH2PO48mg/L、柠檬酸铁铵2.5mg/L、Na2EDTA10mg/L、Na2SiO3·9H2O20mg/L、VB10.005mg/L、VB12 0.04mg/L、二苯基脲磺酸钙0.1mg/L和抗坏血酸1mg/L。
(2)取对数生长期的红色裸甲藻接种到该培养基中进行培养,每天光照10小时,光照强度约为50μmol/(m2·s),培箱温度控制在25摄氏度,每隔一段时间记录培养基中红色裸甲藻的细胞浓度,详见表1。
对比实施例
使用海水f/2培养基作为对照培养基,取对数生长期的红色裸甲藻接种到该培养基中进行培养,每天光照10小时,光照强度约为50μmol/(m2·s),培箱温度控制在25摄氏度,每隔一段时间记录培养基中红色裸甲藻的细胞浓度。
对比实施例与实施例一、实施例二进行对比,对比的结果如表1所示:

从表1可以看出,使用本发明所提供的培养基比使用f/2培养基可以更快速地使红色裸甲藻增殖,红色裸甲藻能达到的最高浓度也高于对照培养基f/2约35%,由此说明本发明提供的红色裸甲藻培养基具有明显的培养优势,更利于藻细胞的快速生长。
