Abstract

本发明涉及一种产黄青霉菌菌种选育用培养基,该培养基分为种子培养基和发酵培养基两部分,其主要成分有葡萄糖浆、玉米浆、甘油、碳酸钙和微量元素等。该培养基成本低,发酵效价高,表现稳定,为产黄青霉菌的育种验证提供了一个良好的培养体系。降低了育种工作的成本,增加了育种工作的效率。

本发明属于产黄青霉菌菌种选育用验证培养基,为验证工艺降低了成本,提高了效率。
背景技术
在青霉素发酵生产过程中,选育优良的种子一直是提高产量和质量的有力保证。在通过各种诱变育种、基因构建改造选育或者纯化筛选育种的过程中,一个稳定的验证区分所获得的新菌株的平台就成了迫切需要解决的问题。
在这个问题中,培养基的配方是成功的关键之一,努力寻找更廉价、更有效的培养基配方一直是努力地方向。
发明内容
本发明的目的就是为产黄青霉菌的菌种选育寻找一个成本更低、效率更高的验证培养基的配方,从而为菌种选育的验证打下基础。
本发明的技术方案是:利用工业生产中更加廉价的葡萄糖浆、玉米浆替代传统工艺培养基配方中的蔗糖和乳糖,经过多次摸索验证,获得一个组方,比传统工艺成本更低,但是效果更好。将同一株产黄青霉菌分别接种到本发明的培养基中和传统工艺的培养基中,经过发酵培养,测发酵液的效价进行对比,发现本发明的培养基效价更好,结束发酵时菌丝体状态更年轻。
具体地,本法的技术方案如下: 一种产黄青霉菌菌种选育用培养基,分为种子培养基和发酵培养基两部分。
其中种子培养基含有:葡萄糖浆7-8%,玉米浆9~10%,甘油2%,碳酸钙0.5%,pH5.8。
发酵培养基含有:葡萄糖浆15~17%,玉米浆25~30%,碳酸钙1%,硫酸钠0.5,磷酸二氢钾0.3,硫酸镁0.005%, 硫酸铜0.002%,磷酸二氢钾0.03,硫酸亚铁0.0015,pH5.9。
本发明的优点:
本发明所述的产黄青霉菌菌种选育用培养基,适用于产黄青霉菌工业发酵生产育种的验证用更加廉价的葡萄糖浆和玉米浆作为替代物改良后的培养基,成本降低了,效果更好了。
具体实施方式:
实施例1:
将同一株产黄青霉菌菌株分别接种到:传统工艺种子培养基(蔗糖2%,玉米浆6.7%,碳酸钙0.5%,pH5.8);本发明种子培养基(葡萄糖浆7%,玉米浆9%,甘油2%,碳酸钙0.5%,pH5.8),在同一个摇床上,控温25℃,摇床180转/分钟,培养58小时,然后分别移种到装有30ml传统培养基(乳糖13,玉米浆3,甘 油1.5,碳酸钙1,硫酸钠0.5,磷酸二氢钾0.3,硫酸镁0.005%, 硫酸铜0.002%,磷酸二氢钾0.03,硫酸亚铁0.0015,pH5.9)和本发明的培养基(葡萄糖浆15%,玉米浆25%,碳酸钙1%,硫酸钠0.5,磷酸二氢钾0.3,硫酸镁0.005%, 硫酸铜0.002%,磷酸二氢钾0.03,硫酸亚铁0.0015,pH5.9)的250ml摇瓶中。在同一摇床上,控温26℃,摇床190转/分钟,培养6天。测两种发酵液青霉素的效价分别为传统工艺:3.0×104IU/ml,本发明:3.1×104IU/ml。
实施例2:
将同一株产黄青霉菌菌株分别接种到:传统工艺种子培养基(蔗糖2%,玉米浆6.7%,碳酸钙0.5%,pH5.8);本发明种子培养基(葡萄糖浆8%,玉米浆10%,甘油2%,碳酸钙0.5%,pH5.8),在同一个摇床上,控温25℃,摇床180转/分钟,培养58小时,然后分别移种到装有30ml传统培养基(乳糖13,玉米浆3,甘 油1.5,碳酸钙1,硫酸钠0.5,磷酸二氢钾0.3,硫酸镁0.005%, 硫酸铜0.002%,磷酸二氢钾0.03,硫酸亚铁0.0015,pH5.9)和本发明的培养基(葡萄糖浆17%,玉米浆30%,碳酸钙1%,硫酸钠0.5,磷酸二氢钾0.3,硫酸镁0.005%, 硫酸铜0.002%,磷酸二氢钾0.03,硫酸亚铁0.0015,pH5.9)的250ml摇瓶中。在同一摇床上,控温26℃,摇床190转/分钟,培养6天。测两种发酵液青霉素的效价分别为传统工艺:2.9×104IU/ml,本发明:3.2×104IU/ml。

2. 如权利要求1所述的一种产黄青霉菌菌种选育用培养基,其特征在于,种子培养基含有:葡萄糖浆7-8%,玉米浆9~10%,甘油2%,碳酸钙0.5%,pH5.8。
3. 如权利要求1所述的一种产黄青霉菌菌种选育用培养基,其特征在于,发酵培养基含有:葡萄糖浆15~17%,玉米浆25~30%,碳酸钙1%,硫酸钠0.5,磷酸二氢钾0.3,硫酸镁0.005%, 硫酸铜0.002%,磷酸二氢钾0.03,硫酸亚铁0.0015,pH5.9。
4. 如权利要求1~3所述的一种产黄青霉菌菌种选育用培养基,适用于产黄青霉菌工业发酵生产育种的验证。
