Abstract
本发明公开了一种具有发酵产碱功能的微生物菌及其应用的有机肥料,本菌株在不同的发酵条件下均可产碱,可以针对过度耕作土壤的土壤酸化问题,芽孢杆菌能够在逆境中存活和生长,能够为土壤提供各种酶类和增加土壤微生物多样性,筛选能够产碱的芽孢杆菌应用在酸性土壤,同时能够优化微调酸性土壤的作物根际微生态;菌株HNPY23来源于甘蔗根际土壤中,能够明显促进油麦菜、生菜等作物对酸性土壤的耐受性,提高土壤尿酶活性,为油麦菜、生菜等作物在酸性土壤中生长提供了一种优良的液体菌肥;菌株HNPY23制备的有机肥能够明显的提高油麦菜、生菜等作物的叶片组织的叶绿素含量、叶片Vc含量、以及产量,具有增加质量增加产量的良好效果。
Description
技术领域
本发明涉及微生物菌及有机肥料领域,具体涉及一种产碱能够微调酸性土壤的根际微环境,促进油麦菜、生菜等作物在酸性土壤中的生长的微生物有机肥料及应用。
背景技术
芽孢杆菌广泛地存在于自然界,具有繁殖快速、代谢快、四小时增殖10万倍,标准菌四小时仅可繁殖6倍。该属细菌的重要特性是能够产生对不利条件具有特殊抵抗力的芽孢,无湿状态可耐低温-60℃、耐高温+280℃,耐强酸、耐强碱、抗菌消毒、耐高氧(嗜氧繁殖)、耐低氧(厌氧繁殖)。尤其是该属的许多菌能水解淀粉,分解蛋白质、果胶、藻酸盐等工业上用于提取淀粉酶、蛋白酶、果胶酶、葡聚糖酶、纤维素酶等。它们也存在于土壤、植物组织内、水和空气中,与自然界物质转化、土壤肥力、环境卫生均密切相关。芽孢杆菌体积比一般病源菌分子大四倍数,占据空间优势,抑制有害菌的生长繁殖。同时它们对有机质分解力强,增殖的同时,会释出高活性的分解酵素,将难分解的大分子物质分解成可利用的小分子物质,产生丰富的代谢生成物,合成多种有机酸、酶、生理活性等物质,及其它多种容易被利用的养份,促进作物生长。随着农药、化肥等的长期的、持续施用,造成耕作土壤的生物多样性退化及土壤酸化,芽孢杆菌能够在逆境中存活和生长,为土壤提供各种酶类和增加土壤微生物多样性。筛选能够产碱的芽孢杆菌应用在酸性土壤,微调酸性土壤的作物 根际微生态,促进作物生长具有重要的意义。
发明内容
针对上述问题,本发明旨在提供一种具有产碱能力、生长速度快、对环境适应性强和耐逆性强的微生物菌;通过菌株产碱改善作物根际微生态及分解和降解土壤有机物为小分子,供作物利用,从而能够显著提高蔬菜及作物产量和品质的菌株具有发酵产碱功能的微生物有机肥料及应用。
为实现该技术目的,本发明的方案是:一种具有发酵产碱功能的生物菌,包括菌株HNPY23,为枯草芽孢杆菌(Bacillus subtilis),保藏号为CGMCC 10069;
菌株HNPY23的形态及生理生化特性如下:
枯草芽孢杆菌为革兰氏阳性菌,细胞呈杆状,能够产生芽孢;
LB琼脂平板上生长速度较快,菌落圆形凸起(37℃,生长48小时),奶油色不透明,菌落直径3-6毫米。细胞杆状,大小3.5-4.5x1.8μm,具周生鞭毛。最适生长温度是37℃及最佳生长pH值6-8;
兼性厌氧,革兰氏阳性,氧化酶阴性,过氧化氢酶阳性。菌株在改性LB液体培养基上发酵产碱,使发酵液的pH值升高到pH8以上;能以吐温80,D-阿拉伯糖,L-阿拉伯糖,D-半乳糖,L-果糖,D-纤维二糖,D-棉子糖,L-鼠李糖,木糖,D-木糖,D-松二糖,D-核糖,甘露醇,己六醇,半乳糖醇,侧金盏花醇,葵二酸,羟基苯乙酸,琥珀酸钠,L-苹果酸,L-谷氨酸,L-赖氨酸,L-脯氨酸,L-组氨酸为碳源的培养基上生长;pH生长范围较广,可耐pH3-pH10;对NaCl敏感, 在低于1%NaCl浓度下可生长;对100μg/mL的链霉素、四环素、红霉素有耐受性,而对卡拉霉素、氯霉素、新霉素、氨苄青霉素、庆大霉素的耐受性差、低浓度(50μg/ml)的这几种抗生素就能抑制其生长;
吲哚反应呈阳性,且能分泌生长素IAA,能液化明胶。
用菌株HNPY23生产的微生物有机肥料,所述菌株HNPY23含量为1×108cfu/g以上的菌株,全氮含量质量比为3~7%,总氮磷钾养分质量比为5~11%、有机质含量质量比不低于40%。
本发明涉及的产酸功能的微生物菌及其制备有机肥具有如下有益效果:
1.菌株HNPY23为一种发酵产碱功能的枯草芽孢杆菌,在不同的发酵条件下均可产碱,可以针对过度耕作土壤的生物多样性退化及土壤酸化问题,芽孢杆菌能够在逆境中存活和生长,能够为土壤提供各种酶类和增加土壤微生物多样性,筛选能够产碱的芽孢杆菌应用在酸性土壤,同时能够优化微调酸性土壤的作物根际微生态。
2.菌株HNPY23来源于甘蔗根际土壤中,能够明显促进油麦菜、生菜等作物对酸性土壤的耐受性,提高土壤尿酶活性,为油麦菜、生菜等作物在酸性土壤中生长提供了一种优良的液体菌肥。
3.菌株HNPY23制备的有机肥能够明显的提高油麦菜、生菜等作物的叶片组织的叶绿素含量、叶片Vc含量、以及产量,具有增加质量增加产量的良好效果。
具体实施方式
下面结合具体实施例对本发明做进一步详细说明。
本发明通过采用80℃处理杀死非芽胞杆菌,用牛肉膏蛋白胨培养基(1L),葡萄糖20g,蛋白胨15g,氯化钠5g,牛肉膏0.5g,琼脂20g,在厌氧和好氧条件下培养,对生长于广东省甘蔗土壤中的芽孢杆菌进行分离、纯化得到的。具有在改性LB培养基上产碱的能力,生长速度快,能够迅速分解土壤中的有机质为作物提高营养,可作为接种剂促进油麦菜、生菜、玉米等作物的生长。
一、菌株HNPY23的形态及生理生化特性如下:
菌株HNPY23,为枯草芽孢杆菌(Bacillus subtilis),保藏号为CGMCC 10069,保藏日期为2014年11月27日,保藏单位:中国微生物菌种保藏管理委员会普通微生物中心,地址:北京市朝阳区北辰西路1号院3号中国科学院微生物研究所。
(一)、菌株HNPY23的形态及生理生化特性
枯草芽孢杆菌为革兰氏阳性菌,细胞呈杆状,能够产生芽孢;
LB琼脂平板上生长速度较快,菌落圆形凸起(37℃,生长48小时),奶油色不透明,菌落直径3-6毫米。细胞杆状,大小3.5-4.5x1.8μm,具周生鞭毛。最适生长温度是37℃及最佳生长pH值6-8;

(二)、本发明解淀粉芽孢杆菌HNPY23的分子分类地位的确定
采用细菌16S rDNA基因通用引物25f和1492r进行扩增,将PCR产物直接进行序列测定,将获得的DNA序列输入GenBank进行比对,初步确定本发明的枯草芽孢杆菌(Bacillus subtilis)在分类学中的菌株种属的位置。结果发现本发明的枯草芽孢杆菌属于芽孢杆菌属(Bacillus),与枯草芽孢杆菌(Bacillus subtilis)模式菌株DSM10相似性为98%,但二者的序列不完全一致,有2%的差异,表明二者是不同的菌株。
结合上述的生理生化特性、16S rDNA序列分析,本发明的枯草芽孢杆菌(Bacillus subtilis)应归属于芽孢杆菌属(Bacillus)。
(三)、本发明菌种的分离纯化方法
3.1富集:称取甘蔗根际土壤20g于含50mL无菌水的三角瓶中,充分振荡打散,制成悬浊液,与富集营养液按1:2比例装人三角瓶中,静置于培养箱中,30±2℃,培养5天,小心取出上清液于另一灭菌的烧杯中,加热到80℃杀死非芽孢菌。
取此第一代培养瓶中的加热处理菌悬液500μL至250mL富集营养液进行第二代培养,培养方法与第一代相同。
3.2分离纯化:吸取第二代菌体培养液1ml于9ml无菌水试管制成10-1倍稀释菌液,再取此浓度菌悬液如法稀释至10-2、10-3倍, 直至稀释到10-6倍。分别吸取10-4、10-5、10-6三个梯度的菌悬液100μL,依次采用平板涂布培养法和倾注厌氧培养法,30±2℃下培养,观察不同培养方法下菌体的生长情况,挑取长势较好并且具有不同特征的菌落用平板划线法进行反复传代培养,直至菌落的形状、颜色、质地、透明度一致。最后通过简单染色和镜检,进一步观察其形态,以长度均一、宽度一致、染色情况统一为菌株纯化标准。
3.3保存:将分离纯化后的菌株在分离培养基平板及斜面上培养2-5d后,收集平板上的菌体。部分保存于15%灭菌甘油(-20℃)和(-80℃)。
富集培养液(1L):KH2PO4 0.5g,K2HPO4 0.6g,MgSO4·7H2O 0.2g,NaCl 0.2g,酵母膏1g,蛋白胨10.0g,1L水),PH为7.0±0.2。
其中分离培养基(1L):酵母膏3g,蛋白胨3g,琼脂粉17g,pH 7.0±0.2。
二、生物有机肥料制备:
将菌株HNPY23接种到蛋白膏体培养基进行发酵生产,发酵生产的条件为室温下,发酵5-8天,溶液pH值为7.6-8.3,发酵之间中后期形成芽孢,使发酵液中含菌或芽孢量≥1×108cfu/ml,其中所用蛋白膏体培养基的配方为:1L含有酵母粉2g,糖蜜15g,蛋白胨5g,KH2PO4 0.01g,氯化钠1g,豆饼10g,K2HPO4 0.01g。完成发酵 后加热至80℃杀死非芽孢菌。
将菌株HNPY23发酵液加入到猪粪堆肥和菜粕原料中,调节含水量55-65%和pH7.5-8.5,进行发酵,待发酵温度上升至50℃时立即开始翻堆,以后每天翻堆,使堆温始终维持在35-50℃(共5-8天),固体发酵开始后,堆物的pH会不断上升,每次翻堆时用含酸液体调节含水量和pH。
最后得到菌株HNPY23含量为1×108cfu/ml以上,全氮含量质量比为3~7%,总氮磷钾养分质量比为5~11%、有机质含量质量比不低于40%的微生物有机肥料。
三、盆栽实验:
(一)生菜盆栽实验
1.1发酵产碱:将菌株HNPY23接种到发酵液中,1L发酵液含有酵母粉2g,糖蜜15g,蛋白胨5g,KH2PO4 0.01g,氯化钠1g,豆饼10g,K2HPO4 0.01g,室温下发酵5-7天,溶液pH值为7.7-8.2之间。
1.2施用:土壤混合均匀一致,原初始pH值为5.0,盆栽试验做4个处理,5个重复,将培养基1发酵产物如下处理。I1:化肥;I2:无菌培养液;I3:发酵液;CK:只浇清水不施肥。以上四个处理中种植大小、长势相近的生菜苗,每盆1颗苗,其他日常管理一致。
1.3结果:
其中,I3组的土壤尿酶活性为0.556±0.18mg/g,Vc含量 0.255±0.009mg/g。

从上述数据可以看出,用含有菌株HNPY23的肥料种植的生菜产量高,Vc含量和土壤尿酶活性的含量明显高于I1化肥组、I2等量空白营养液组和CK只浇清水不施肥组,其中叶绿素相对于CK只浇清水不施肥组有明显提升。
(二)油麦菜盆栽实验
2.1发酵产碱:将菌株HNPY23接种到发酵液中,1L发酵液发酵液中,含有氯化钠0.5g,酵母浸膏5g,KH2PO4 0.02g,麸皮12g,蛋白胨6g,K2HPO4 0.02g,花生饼15g,室温下发酵5-8天,溶液pH值为7.6-8.2之间。
2.2施用:土壤混合均匀一致,原初始pH值为5.0,盆栽试验做4个处理,5个重复,将培养基1发酵产物如下处理。I1:化肥;I2:无菌培养液;I3:发酵液;CK:只浇清水不施肥。以上四个处理中种植大小、长势相近的油麦菜苗,每盆3颗苗,其他日常管理一致。

2.一种运用权利要求1所述微生物菌的微生物有机肥料,其特征在于:所述菌株HNPY23含量为1×108cfu/g以上的菌株,全氮含量质量比为3~7%,总氮磷钾养分质量比为5~11%、有机质含量质量比不低于40%。
3.权利要求2所述的微生物有机肥料在油麦菜和生菜栽培中的应用。
