Abstract

本发明提供一种通过添加磷酸二甘油酯在喷雾干燥过程中保护乳酸菌的方法,其主要是通过添加抗热保护剂磷酸二甘油酯提高在喷雾干燥处理过程的乳酸菌的活菌存活量。具有使用方便、效果明显、无污染、对环境友好等特点。本发明方法用于乳酸菌的喷雾干燥过程,获得的乳酸菌干粉活菌数可达5.4*1010CFU/g以上。

技术领域
本发明涉及一种喷雾干燥过程中保护乳酸菌的方法,尤其是一种通过添加磷酸二甘油酯在喷雾干燥过程中保护乳酸菌的方法。
背景技术
乳酸菌是一类可发酵碳水化合物,主要生成乳酸的细菌总称。随着乳酸菌对人体健康有益作用机理研究的不断深入以及人们保健意识的增强,乳酸菌得到越来越多的关注和研究开发,目前已被广泛应用于营养保健、食品、医药、饲料添加剂等行业中。
乳酸菌对营养及环境条件要求较高,且对外界抵抗力差,难以制备和长期保存,这严重限制了其在实际生产生活中的应用。目前乳酸菌多以冻干粉的形式供应市场,但乳酸菌冻干成本较高,因此也有喷雾干燥乳酸菌的工艺的应用,但是乳酸菌不耐高温,喷雾干燥过程中容易造成大量的乳酸菌死亡,因此加入保护剂可以降低喷雾干燥过程对于微生物细胞造成的损伤,保护细胞的生物活性,有效提高活菌率。目前国内广泛采用的乳酸菌保护剂有脱脂乳粉、海藻糖、乳糖、麦芽糊精、菊粉、吐温等;选用的菌种有双歧杆菌、德式乳杆菌等。而实际上,冻干保护剂的作用会随菌种的变化以及保护剂的组成和配比表现出一定的差异性,因此,开发性质独特、作用高效的乳酸菌保护剂是目前急需解决的问题。
磷酸二甘油酯(diglycerol phosphate, DP)是嗜热菌体内蓄积的一种溶质,主要发现存在于嗜热菌和超嗜热古菌中。其主要作用为具有高温下保护蛋白、核酸、细胞膜、酶等生物大分子的功能,从而达到协助嗜热菌抵御外界高温的侵害。
发明内容
本发明的目的是提供一种无毒、安全、使用方便的喷雾干燥保护方法。
所述的乳酸菌为乳酸杆菌或乳酸球菌中的一种或以上。
所述磷酸二甘油酯的含量为0.01wt%~5wt%。
与现有技术相比,本发明的有益效果如下:
利用磷酸二甘油酯处理过的乳酸菌喷雾干燥提高其活性的机制为:(1)待喷雾干燥的乳酸菌通过加入磷酸二甘油酯处理后,能够提高其对干燥、高温的防护能力,有效减少菌在喷雾干燥过程中的活性损失,并且在恢复过程中加快复苏。(2)磷酸二甘油酯成分是能够进入乳酸菌细胞中保留的一种相容性物质,因此在持续的喷雾干燥过程中保护细胞。
具体实施方式
实施例 1
将50升保加利亚乳杆菌经液体发酵26小时,将50g磷酸二甘油酯添加到发酵液中,混合均匀后进行喷雾干燥,设置对照组(不添加保护剂)。喷雾干燥条件为:进风口温度为150℃,出风口温度为65℃,喷头压力为140KPa。
实施例2
将100升乳酸链球菌经液体发酵24小时,将200g磷酸二甘油酯添加到发酵液中,混合均匀后进行喷雾干燥,设置对照组(不添加保护剂)。喷雾干燥条件为:进风口温度为170℃,出风口温度为70℃,喷头压力为130KPa。
实施例3
将300升双歧杆菌经液体发酵26小时,将30g磷酸二甘油酯添加到发酵液中,混合均匀后进行喷雾干燥,设置对照组(不添加保护剂)。喷雾干燥条件为:进风口温度为180℃,出风口温度为75℃,喷头压力为110KPa。
喷雾干燥得到乳酸菌干粉,通过平板计数法检测活菌数。本发明通过添加磷酸二甘油酯在喷雾干燥过程中保护乳酸菌的方法对三种乳酸菌的保护效果如下表:

1.磷酸二甘油酯用于喷雾干燥乳酸菌,其特征在于:所述的乳酸菌为乳酸杆菌或乳酸球菌中的一种或以上。
2.根据权利要求1所属的磷酸二甘油酯用于喷雾干燥乳酸菌,其特征在于:乳酸菌喷雾干燥之前搅拌均匀加入磷酸二甘油酯。
3.根据权利要求2所述的磷酸二甘油酯用于喷雾干燥乳酸菌,其特征在于:所述磷酸二甘油酯的用量为0.01wt%~5wt%。
