本发明公开了一种抗SM-RNP抗体试剂盒,所述试剂盒包括用于表达抗SM-RNP抗体的细胞载体、阴性对照血清、阳性对照血清、酶底物、浓缩样本稀释液,所述阳性对照血清为病人的阳性对照血清筛选后的对照血清。本发明通过对所采集的阳性对照血清进行筛选,取分布比较密集的数值区域的阳性对照血清作为抗SM-RNP抗体试剂盒中的阳性对照血清,克服了现有试剂盒在检测人血清中抗SM-RNP抗体的含量时,因每个批次的阳性对照抗体不稳定而导致结果不稳定的问题。

一种抗SM-RNP抗体试剂盒
技术领域
本发明涉及试剂盒领域,具体地,涉及一种抗SM-RNP抗体试剂盒。
背景技术
系统性红斑狼疮简称SLE是一种以大量自身抗体形成为特征,导致多系统、多器官损害的慢性系统性自身免疫疾病。系统性红斑狼疮是一种常见的风湿性疾病,经调查显示,该病的发病率为千分之一左右,好发于育龄女性,男女比列约为1:7~10,非生育年龄女性优势不明显。主要病因可能与遗传、环境因素和雌激素有关系,已有研究报道年龄是一大危险因素,尤其是青年女性发病最为常见。发病机制主要与免疫调节异常、大量的自身抗体以及免疫复合物的形成有关系,目前,具体的发病机制尚不清楚。
SLE血清检测是SLE普查和辅助诊断的重要方法。目前,医院实验室主要采用酶联免疫吸附实验及ELISA法对患者的血清进行初步的诊断,但是目前的诊断方法都是需要阳性对照液,而阳性对照液的来源一般都是来自病人的血清。目前,在检测人血清中抗SM-RNP抗体的检测试剂盒中,阳性对照抗体多数都来自病人的阳性对照血清,这样,每个批次的试剂盒,阳性对照或许不同,导致每个批次中阳性对照抗体不稳定的问题。
发明内容
本发明所要解决的技术问题是提供一种抗SM-RNP抗体试剂盒,以克服现有试剂盒在检测人血清中抗SM-RNP抗体的含量时,因每个批次的阳性对照抗体不稳定而导致结果不稳定的问题。
本发明解决上述问题所采用的技术方案是:一种抗SM-RNP抗体试剂盒,其特征在于,所述试剂盒包括用于表达抗SM-RNP抗体的细胞载体、阴性对照血清、阳性对照血清、酶底物、浓缩样本稀释液,所述阳性对照血清为病人的阳性对照血清筛选后的对照血清。
现有的抗SM-RNP抗体试剂盒中的阳性对照血清来自病人的阳性对照血清,这样,每个批次的试剂盒,阳性对照或许不同,导致每个批次中阳性对照抗体不稳定的问题,本发明通过对所采集的阳性对照血清进行筛选,取分布比较密集的数值区域的阳性对照血清作为抗SM-RNP抗体试剂盒中的阳性对照血清,克服了现有试剂盒在检测人血清中抗SM-RNP抗体的含量时,因每个批次的阳性对照抗体不稳定而导致结果不稳定的问题。
进一步地,细胞载体为哺乳动物细胞。哺乳动物细胞与人体细胞环境具有较高的相似度,且哺乳动物细胞比价容易获得,能够降低实验成本。
一种抗SM-RNP抗体试剂盒的制备方法,包括以下步骤:
首先,细胞载体的制备:将表达有抗SM-RNP抗体的细胞载体活化后,在4℃培育5小时,用PBS缓冲液洗涤、离心、弃上清液,加入一定浓度的血清蛋白封闭液,培育30分钟,完成细胞载体的制备;
其次,其它组分的制备:阴性对照血清是健康人的血清,阳性对照血清为已经确诊的抗SM-RNP抗体阳性血清,浓缩样本稀释液中含有PBS、哺乳动物血清蛋白。
综上,本发明的有益效果是:
1、本发明通过对所采集的阳性对照血清进行筛选,取分布比较密集的数值区域的阳性对照血清作为抗SM-RNP抗体试剂盒中的阳性对照血清,克服了现有试剂盒在检测人血清中抗SM-RNP抗体的含量时,因每个批次的阳性对照抗体不稳定而导致结果不稳定的问题。
2、本发明通过将载体设置为哺乳动物细胞,哺乳动物细胞与人体细胞环境具有较高的相似度,且哺乳动物细胞比价容易获得,能够降低实验成本。

下面结合实施例,对发明作进一步地详细说明,但本发明的实施方式不限于此。
实施例:
一种抗SM-RNP抗体试剂盒,试剂盒包括用于表达抗SM-RNP抗体的细胞载体、阴性对照血清、阳性对照血清、酶底物、浓缩样本稀释液,所述阳性对照血清为病人的阳性对照血清筛选后的对照血清。细胞载体为哺乳动物细胞。
一种抗SM-RNP抗体试剂盒的制备方法,包括以下步骤:
首先,细胞载体的制备:将表达有抗SM-RNP抗体的细胞载体活化后,在4℃培育5小时,用PBS缓冲液洗涤、离心、弃上清液,加入一定浓度的血清蛋白封闭液,培育30分钟,完成细胞载体的制备;
其次,其它组分的制备:阴性对照血清是健康人的血清,阳性对照血清为已经确诊的抗SM-RNP抗体阳性血清,浓缩样本稀释液中含有PBS、哺乳动物血清蛋白。
如上所述,可较好的实现本发明。

1.一种抗SM-RNP抗体试剂盒,其特征在于,所述试剂盒包括用于表达抗SM-RNP抗体的细胞载体、阴性对照血清、阳性对照血清、酶底物、浓缩样本稀释液,所述阳性对照血清为病人的阳性对照血清筛选后的对照血清。
2.根据权利要求1所述的一种抗SM-RNP抗体试剂盒,其特征在于,所述细胞载体为哺乳动物细胞。
3.一种抗SM-RNP抗体试剂盒的制备方法,其特征在于,包括以下步骤:
首先,细胞载体的制备:将表达有抗SM-RNP抗体的细胞载体活化后,在4℃培育5小时,用PBS缓冲液洗涤、离心、弃上清液,加入一定浓度的血清蛋白封闭液,培育30分钟,完成细胞载体的制备;
