背景技术
痤疮,俗称“青春痘”,是毛囊皮脂腺发炎的疾病,85%以上的成人在成长过程中皆有发生痤疮的记录。依症状的不同,痤疮可分为非发炎型痤疮如粉刺,及发炎型痤疮如脓包或毛囊发炎。其中因皮脂过度分泌造成细菌过度生长,所引发的发炎型痤疮最为难以治愈。
目前对于细菌引起的发炎型痤疮常见的治疗方法有外用药剂及内服药剂。外用药剂主要以外用抗生素药膏或药水(Cleocin或Fucidin),可以抑制痤疮杆菌的生长,减轻发炎反应。而内服药剂如口服抗生素是痤疮治疗的第一线用药,常见的有四环霉素、红霉素或磺铵类,主要作用在减少游离性脂肪酸的生成,有抗发炎效果,并非用来杀菌。而利用抗生素来治疗细菌所引起的发炎型痤疮已经造成细菌的抗药性问题越来越严重。
因此,利用天然成分来抑制引起发炎型痤疮的细菌,而减少抗生素的使用或服用,是非常重要的。
发明内容
鉴于上述,本发明的目的在于提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该到手香萃取物是以体积百分比浓度为50%(含)以上的醇类溶液通过抽提方式萃取到手香植株而得。
一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该微生物为细菌。又该细菌可为丙酸菌属(Propionibacterium sp.)属、葡萄球菌属(Staphylococcus sp.)或假单胞菌属(Pseudomonas sp.)。
一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该细菌为丙酸菌属(Propionibacterium sp.)属或葡萄球菌属(Staphylococcussp.)。
另一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该细菌为丙酸痤疮杆菌(Propionibacterium acnes)、金黄色葡萄球菌(Staphylococcus aureus)或绿脓杆菌(Pseudomonas aeruginosa)。
一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该到手香萃取物为以体积百分比浓度为50%(含)以上的甲醇或乙溶液通过抽提方式萃取到手香植株而得。
一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该到手香萃取物为以体积百分比浓度为50%~95%的醇类溶液通过抽提方式萃取到手香植株而得,又该50%~95%的醇类溶液可为甲醇或乙醇溶液。另一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该到手香萃取物为以体积百分比浓度为70%~95%的醇类溶液通过抽提方式萃取到手香植株而得,又该70%~95%的醇类溶液可为甲醇或乙醇溶液。
一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该到手香萃取物为以体积百分比浓度为50%~95%的醇类溶液通过抽提方式萃取到手香植株而得的到手香萃取物,以及以体积百分比浓度为70%~95%的醇类溶液通过抽提方式萃取到手香植株而得的手香萃取物的组合。又该50%~95%的醇类溶液可为甲醇或乙醇溶液,以及该70%~95%的醇类溶液可为甲醇或乙醇溶液。

一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该组合物尚包含一适当赋形剂而制成外用药剂形式或化妆品形式或清洁剂形式。
另一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该组合物可用以治疗或预防痤疮。又该到手香萃取物可与其他抗菌剂、消炎药、清洁或护肤的组成份、或其组合制成外用药剂形式或化妆品形式或清洁剂形式。
一方面,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该组合物还包含积雪草萃取物及/或水杨酸。
本发明亦提供一种治疗或预防痤疮的组合物,其是呈外用剂型,其组合物包含如上述定义的到手香萃取物及积雪草萃取物。
根据本发明具体的实施例,该组合物更进一步包含医药上可接受的载剂。
根据本发明具体的实施例,其中含有到手香萃取物的具有抗微生物功效的组合物可用在抑制或预防由微生物感染所导致的痤疮。
因此,本发明提供一种到手香萃取物在制备具有抗微生物功效组合物中的用途,其中该到手香萃取物为以体积百分比浓度为50%(含)以上的醇类溶液通过抽提方式萃取到手香植株而得。具体而言,其中该组合物用在抑制或预防由微生物感染所导致的痤疮。
本发明的该等及其它方面,可通过以下的较佳具体实施例的描述以及图式,得以更为明晰;即便其中可能会有变化或修饰,但不背离本发明所揭露的新颖观念的精神及范畴。
具体实施方式
除非另有定义,所有本文所用的技术性及科学性术语,对于属于本发明领域的具有通常知识者而言,皆具有与其所习知者相同意义。
除非文中有清楚指明者,在本文中所使用的单数形式“一”、“一种”、及“该”的涵义均为包括“至少一种”的复数形式。因此,例如,当提及“一成分”时,包括复数个该等成分及本领域技术人员所知的同等物。
本发明组成物可调制成药品的型式。可依任何常规技术配合常用的赋形剂或填充剂。亦可依一般医药上常用的技术或方法,含有医疗有效量的本发明组成物,佐以医药上可接受的载剂调制成药品。
本文所使用的“医疗有效量”一词是指可有效治疗或预防痤疮的组合物浓度,其可依照施予的方式及治疗的情况,包括年龄、体重、症状、治疗效果、施用方式及治疗时间调整。
本文所使用的“载剂”或“医药上可接受的载剂”一词是指用在医药的稀释剂、赋形剂或类似物,包括制药业一般技术人员所熟知者。
以上述发明说明以及下列实施例说明本发明,但并非用以限制本发明的范围。
实施例1:50%~95%乙醇溶液抽提的到手香萃取物
取需要量的到手香干燥药材粉末,加入体积百分比浓度为95%的乙醇溶液萃取;将过滤所得药渣再以体积百分比浓度为95%的乙醇溶液进行第二次萃取,经过滤将两次过滤液合并,进行减压浓缩至剩下原滤液体积的约1/20,得萃取浓缩液准备进行管柱层析分离。
将所得萃取浓缩液进行管柱层析,层析树脂为HP-20。将所得萃取浓缩液加入逆渗透(RO)水,混和均匀后导入管柱中进行层析,接着以逆渗透水导入管柱抽提,收集抽提液。接着以4倍管柱体积的逆渗透水与体积百分比浓度为95%的乙醇(体积比1:1)的溶液导入管柱中抽提,收集抽提液,再进行减压浓缩至少量,冷冻干燥得一到手香萃取物,作为体积百分比浓度为0%~50%的乙醇溶液抽提对照组。
接着以4倍管柱体积的体积百分比浓度为95%的乙醇溶液导入管柱抽提,收集抽提液,再进行减压浓缩至干燥得到手香进一步的萃取物,编号为F3,即体积百分比浓度为50%~95%的乙醇溶液抽提萃取物。再以乙酸乙酯(Ethyl Acetate,EA)导入管柱抽提,收集抽提液进行减压浓缩至干燥得到手香乙酸乙酯抽提萃取物。
实施例2:70%~95%乙醇溶液抽提的到手香萃取物
依制备实施例1的步骤进行萃取得萃取浓缩液,准备进行管柱层析。将所得萃取浓缩液进行管柱层析,层析树脂为HP-20。将所得萃取浓缩液加入逆渗透(RO)水,混和均匀后导入管柱中进行层析,接着以逆渗透水导入管柱抽提,收集抽提液。接着以逆渗透水与体积百分比浓度为95%的乙醇(体积比3:7)的溶液导入管柱中抽提,收集抽提液再进行减压浓缩至少量,冷冻干燥得一到手香萃取物。接着以体积百分比浓度为95%的乙醇溶液导入管柱抽提,收集抽提液,再进行减压浓缩至干燥得一到手香进一步萃取物,编号为F4,即体积百分比浓度为70%~95%的乙醇溶液抽提萃取物。再以乙酸乙酯(Ethyl Acetate,EA)导入管柱抽提,收集抽提液进行减压浓缩至干燥为到手香乙酸乙酯抽提萃取物。
实施例3:离体抗菌实验
微生物感染在痤疮发病中扮演重要角色,其中丙酸痤疮杆菌(Propionibacteriumacnes)及金黄色葡萄球菌(Staphylococcus aureus)的影响较为严重且最为多见。本试验使用丙酸痤疮杆菌(Propionibacterium acnes)及金黄色葡萄球菌(Staphylococcusaureus),评估本发明到手香萃取物及其制备的组合物的抑菌能力。
本实验样品为到手香萃取物及其制备的组合物-痘康组合成分。其中到手香萃取物由F3(50%~95%乙醇溶液抽提萃取物)及F4(70%~95%乙醇溶液抽提萃取物)所组成。痘康组合成分除了含有F3及F4,另包含积雪草萃取物、水杨酸及其他赋形成分。将1%的实验样品添加到含待测菌株丙酸痤疮杆菌(Propionibacterium acnes)及金黄色葡萄球菌(Staphylococcus aureus)(2-8×105菌落形成单位/毫升)的测试孔中,在受控的温度及培养基条件下生长1至2天后,利用分光光度计(波长600nm)进行混浊度测定,以检查微生物生长情形。(+)表示抑制生长,(-)表示混浊/不影响微生物生长。所得结果如下表1。

由实验结果可知本发明到手香萃取物及痘康组合成分对于痤疮致病菌丙酸痤疮杆菌(Propionibacterium acnes),及金黄色葡萄球菌(Staphylococcus aureus)具有完全抑制生长的效果。
Claims (26)
1.一种到手香萃取物在制备具有抗微生物功效的组合物中的用途,其中该到手香萃取物是以体积百分比浓度为50%(含)以上的醇类溶液通过抽提方式萃取到手香植株而得。
2.如权利要求1所述的用途,其中该微生物是细菌。
3.如权利要求2所述的用途,其中该细菌是丙酸菌属(Propionibacterium sp.)属、葡萄球菌属(Staphylococcus sp.)或假单胞菌属(Pseudomonas sp.)。
4.如权利要求2所述的用途,其中该细菌为丙酸菌属(Propionibacterium sp.)属或葡萄球菌属(Staphylococcus sp.)。
5.如权利要求3所述的用途,其中该细菌是丙酸痤疮杆菌(Propionibacterium acnes)。
6.如权利要求3所述的用途,其中该细菌是金黄色葡萄球菌(Staphylococcus aureus)。
7.如权利要求3所述的用途,其中该细菌是绿脓杆菌(Pseudomonasaeruginosa)。
8.如权利要求1所述的用途,其中该到手香萃取物是以体积百分比浓度为50%-95%的醇类溶液通过抽提方式萃取到手香植株而得。
9.如权利要求1所述的用途,其中该到手香萃取物是以体积百分比浓度为50%-95%的乙醇溶液通过抽提方式萃取到手香植株而得。
10.如权利要求1所述的用途,其中该到手香萃取物是以体积百分比浓度为70%-95%的醇类溶液通过抽提方式萃取到手香植株而得。
11.如权利要求1所述的用途,其中该到手香萃取物是以体积百分比浓度为70%-95%的乙醇溶液通过抽提方式萃取到手香植株而得。
12.如权利要求1所述的用途,其中该到手香萃取物是以体积百分比浓度为50%-95%的醇类溶液通过抽提方式萃取到手香植株而得的到手香萃取物,以及以体积百分比浓度为70%-95%的醇类溶液通过抽提方式萃取到手香植株而得的到手香萃取物的组合。
13.如权利要求1所述的用途,其中该到手香萃取物是以体积百分比浓度为50%-95%的乙醇溶液通过抽提方式萃取到手香植株而得的到手香萃取物,以及以体积百分比浓度为70%-95%的乙醇溶液通过抽提方式萃取到手香植株而得的到手香萃取物的组合。
14.如权利要求1、8、10及12中任一项所述的用途,其中该醇类溶液是甲醇或乙醇的溶液。
15.如权利要求1、8、10及12中任一项所述的用途,其中该醇类溶液是乙醇的溶液。
16.如权利要求1所述的用途,其中该到手香萃取物是以醇类溶液抽提后再以乙酸乙酯(Ethyl Acetate)抽提而得。
17.如权利要求9所述的用途,其中该到手香萃取物是以醇类溶液抽提后再以乙酸乙酯(Ethyl Acetate)抽提而得。
18.如权利要求11所述的用途,其中该到手香萃取物是以醇类溶液抽提后再以乙酸乙酯(Ethyl Acetate)抽提而得。
19.如权利要求14所述的用途,其中该到手香萃取物是以甲醇或乙醇溶液抽提后再以乙酸乙酯(Ethyl Acetate)抽提而得。
20.如权利要求15所述的用途,其中该到手香萃取物是以乙醇溶液抽提后再以乙酸乙酯(Ethyl Acetate)抽提而得。
21.如权利要求1-13及16-20中任一项所述的用途,其中该具抗微生物功效组合物还包含一适当赋形剂而制成外用药剂形式或化妆品形式或清洁剂形式。
22.如权利要求1-13及16-20中任一项所述的用途,其中该具抗微生物功效组合物可用以治疗或预防痤疮。
23.如权利要求22所述的用途,其中该到手香萃取物可与其他抗菌剂、消炎药、清洁或护肤的组成份、或其组合制成外用药剂形式或化妆品形式或清洁剂形式。
24.如权利要求22所述的用途,其中该组合物还包含积雪草萃取物。
25.如权利要求22或24所述的用途,其中该组合物还包含水杨酸。
26.一种治疗或预防痤疮的组合物,其是呈外用剂型,其组合物包含如权利要求1-13及16-20中任一项定义的到手香萃取物及积雪草萃取物。
