本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該到手香萃取物為以50%(含)以上濃度的醇類溶液沖提方式萃取到手香植株而得。其中該組合物用於抑制或預防由微生物所造成之痤瘡。
Description
本發明有關於一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該到手香萃取物為以50%(含)以上濃度的醇類溶液沖提方式萃取到手香植株而得。具體而言,該具抗微生物功效之組合物可用於抑制或預防由微生物所造成之痤瘡。
痤瘡,俗稱「青春痘」,是毛囊皮脂腺發炎的疾病,85%以上的成人在成長過程中皆有發生痤瘡的記錄。依症狀的不同,痤瘡可分為非發炎型痤瘡如粉刺,及發炎型痤瘡如膿包或毛囊發炎。其中因皮脂過度分泌造成細菌過度生長,所引發的發炎型痤瘡最為難以治癒。
目前對於細菌引起的發炎型痤瘡常見的治療方法有外用藥劑及內服藥劑。外用藥劑主要以外用抗生素藥膏或藥水(Cleocin或Fucidin),可以抑制痤瘡桿菌的生長,減輕發炎反應。而內服藥劑如口服抗生素是痤瘡治療的第一線用藥,常見的有四環黴素、紅黴素或磺銨類,主要作用在減少游離性脂肪酸的生成,有抗發炎效果,並非用來殺菌。而利用抗生素來治療細菌所引起的發炎型痤瘡已經造成細菌的抗藥性問題越來越嚴重。
因此,利用天然成分來抑制引起發炎型痤瘡的細菌,而減少抗生素的使用或服用,是非常重要的。
鑑於上述,本發明之目的為提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該到手香萃取物為以50%(含)以上濃度的醇類溶液沖提方式萃取到手香植株而得。
一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該微生物為細菌。又該細菌可為丙酸菌屬(Propionibacterium sp.)屬、葡萄球菌屬(Staphylococcus sp.)或假單胞菌屬(Pseudomonas sp.)。
一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該細菌為丙酸菌屬(Propionibacterium sp.)屬或葡萄球菌屬(Staphylococcus sp.)。
另一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該細菌為丙酸痤瘡桿菌(Propionibacterium acnes)、金黄色葡萄球菌(Staphylococcus aureus)或綠膿桿菌(Pseudomonas aeruginosa)。
一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該到手香萃取物為以50%(含)以上濃度的甲醇或乙溶液沖提方式萃取到手香植株而得。
一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該到手香萃取物為以50%~95%醇類溶液沖提萃取到手香植株而得,又該50%~95%醇類溶液可為甲醇或乙醇溶液。另一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該到手香萃取物為以70%~95%醇類溶液沖提萃取到手香植株而得,又該

一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該組合物尚包含一適當賦形劑而製成外用藥劑形式或化粧品形式或清潔劑形式。
另一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該組合物可用以治療或預防痤瘡。又該到手香萃取物可與其他抗菌劑、消炎藥、清潔或護膚之組成份、或其組合製成外用藥劑形式或化粧品形式或清潔劑形式。
一方面,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該組合物尚包含積雪草萃取物及/或水楊酸。
本發明亦提供一種治療或預防痤瘡之組合物,其係呈外用劑型,其組合物包含如上述定義之到手香萃取物及積雪草萃取物。
根據本發明具體之實施例,該組合物更進一步包含醫藥上可接受之載劑。
根據本發明具體之實施例,其中含有到手香萃取物之具抗微生物功效組合物可用於抑制或預防由微生物所造成之痤瘡。
因此,本發明提供一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該到手香萃取物為以50%(含)以上濃度的醇類溶液沖提方式萃取到手香植株而得。具體而言,其中該組合物用於抑制或預防由微生物所造成之痤瘡。
本發明之該等及其它方面,可藉由以下之較佳具體實施例之描述以及圖式,得以更為明晰;即便其中可能會有變化或修飾,但不背離
本發明所揭露之新穎觀念的精神及範疇。
除非另有定義,所有本文所用之技術性及科學性術語,對於屬於本發明領域之具有通常知識者而言,皆具有與其所習知者相同意義。
除非文中有清楚指明者,於本文中所使用之單數形式「一」、「一種」、及「該」之涵義均為包括「至少一種」的複數形式。因此,例如,當提及「一成分」時,包括複數個該等成分及對該領域具有通常知識者所知之同等物。
本發明組成物可調製成藥品之型式。可依任何習知技術配合常用的賦形劑或填充劑。亦可依一般醫藥上常用之技術或方法,含有醫療有效量之本發明組成物,佐以醫藥上可接受之載劑調製成藥品。
本文所使用的「醫療有效量」乙詞係指可有效治療或預防痤瘡之組合物濃度,其可依照施予的方式及治療的情況,包括年齡、體重、症狀、治療效果、施用方式及治療時間調整。
本文所使用的「載劑」或「醫藥上可接受之載劑」乙詞是指用於醫藥之稀釋劑、賦形劑或類似物,包括製藥業一般知識者所熟知者。
以上述發明說明以及下列實施例說明本發明,但並非用以限制本發明之範圍。
實施例1、50%~95%乙醇溶液沖提之到手香萃取物
取需要量之到手香乾燥藥材粉末,加入95%乙醇溶液萃取;
將過濾所得藥渣再以95%乙醇溶液進行第二次萃取,經過濾將兩次過濾液合併,進行減壓濃縮至剩下原濾液體積之約1/20,得萃取濃縮液準備進行管柱層析分離。
將所得萃取濃縮液進行管柱層析,層析樹脂為HP-20。將所得萃取濃縮液加入逆滲透(RO)水,混和均勻後導入管柱中進行層析,續以逆滲透水導入管柱沖提,收集沖出液。續以4倍管柱體積逆滲透水與95%乙醇(體積比1:1)之溶液導入管柱中沖提,收集沖出液,再進行減壓濃縮至少量,冷凍乾燥得一到手香萃取物,作為0%~50%乙醇溶液沖提對照組。
續以4倍管柱體積95%乙醇溶液導入管柱沖提,收集沖出液,再進行減壓濃縮至乾燥得到手香進一步之萃取物,編號為F3,即50%~95%乙醇溶液沖提萃取物。再以乙酸乙酯(Ethyl Acetate,EA)導入管柱沖提,收集沖出液進行減壓濃縮至乾燥得到手香乙酸乙酯沖提萃取物。
實施例2、70%~95%乙醇溶液沖提之到手香萃取物
依製備實例1之步驟進行萃取得萃取濃縮液,準備進行管柱層析。將所得萃取濃縮液進行管柱層析,層析樹脂為HP-20。將所得萃取濃縮液加入逆滲透(RO)水,混和均勻後導入管柱中進行層析,續以逆滲透水導入管柱沖提,收集沖出液。續以逆滲透水與95%乙醇(體積比3:7)之溶液導入管柱中沖提,收集沖出液再進行減壓濃縮至少量,冷凍乾燥得一到手香萃取物。續以95%乙醇溶液導入管柱沖提,收集沖出液,再進行減壓濃縮至乾燥得一到手香進一步萃取物,編號為F4,即70%~95%乙醇溶液沖提萃取物。再以乙酸乙酯(Ethyl Acetate,EA)導入管柱沖提,收集沖出液進行減壓濃縮至乾燥為到手香乙酸乙酯沖提萃取物。
實施例3、離體抗菌實驗
微生物感染在痤瘡發病中扮演重要角色,其中丙酸痤瘡桿菌(Propionibacterium acnes)及金黄色葡萄球菌(Staphylococcus aureus)的影響較為嚴重且最為多見。本試驗使用丙酸痤瘡桿菌(Propionibacterium acnes)及金黄色葡萄球菌(Staphylococcus aureus),評估本發明到手香萃取物及其製備之組合物之抑菌能力。
本實驗樣品為到手香萃取物及其製備之組合物-痘康組合成分。其中到手香萃取物由F3(50%~95%乙醇溶液沖提萃取物)及F4(70%~95%乙醇溶液沖提萃取物)所組成。痘康組合成分除了含有F3及F4,另包含積雪草萃取物、水楊酸及其他賦形成分。將1%之實驗樣品添加到含待測菌株丙酸痤瘡桿菌(Propionibacterium acnes)及金黄色葡萄球菌(Staphylococcus aureus)(2-8×105菌落形成單位/毫升)之測試孔中,在受控之溫度及培養基條件下生長1至2天後,利用分光光度計(波長600nm)進行混濁度測定,以檢查微生物生長情形。(+)表示抑制生長,(-)表示混濁/不影響微生物生長。所得結果如下表1。
由實驗結果可知本發明到手香萃取物及痘康組合成分對於痤瘡致病菌丙酸痤瘡桿菌(Propionibacterium acnes),及金黄色葡萄球菌(Staphylococcus aureus)具有完全抑制生長的效果。
Claims (19)
- 一種到手香萃取物用於製備具抗微生物功效組合物之用途,其中該到手香萃取物為以50%~95%醇類溶液沖提萃取到手香植株而得之到手香萃取物、以70%~95%醇類溶液沖提萃取到手香植株而得之到手香萃取物、或以50%~95%醇類溶液沖提萃取到手香植株而得之到手香萃取物以及以70%~95%醇類溶液沖提萃取到手香植株而得之到手香萃取物之組合。
- 如請求項1之用途,其中該微生物為細菌。
- 如請求項2之用途,其中該細菌為丙酸菌屬(Propionibacterium sp.)屬、葡萄球菌屬(Staphylococcus sp.)或假單胞菌屬(Pseudomonas sp.)。
- 如請求項2之用途,其中該細菌為丙酸菌屬(Propionibacterium sp.)屬或葡萄球菌屬(Staphylococcus sp.)。
- 如請求項3之用途,其中該細菌為丙酸痤瘡桿菌(Propionibacterium acnes)。
- 如請求項3之用途,其中該細菌為金黄色葡萄球菌(Staphylococcus aureus)。
- 如請求項3之用途,其中該細菌為綠膿桿菌(Pseudomonas aeruginosa)。
- 如請求項1之用途,其中該醇類溶液為甲醇或乙醇之溶液。
- 如請求項1之用途,其中該醇類溶液為乙醇溶液。
- 如請求項1之用途,其中該到手香萃取物為以醇類溶液沖提後再以乙酸乙酯(Ethyl Acetate)沖提而得。
- 如請求項8之用途,其中該到手香萃取物為以甲醇或乙醇之溶液沖提後再以乙酸乙酯(Ethyl Acetate)沖提而得。
- 如請求項9之用途,其中該到手香萃取物為以乙醇溶液沖提後再以乙酸乙酯(Ethyl Acetate)沖提而得。
- 如請求項1至12中任一項之用途,其中該組合物尚包含一適當賦形劑,而製成外用藥劑形式或化粧品形式或清潔劑形式。
- 如請求項1至12中任一項之用途,其中該組合物可用以治療或預防痤瘡。
- 如請求項1至12中任一項之用途,其中該組合物尚包含其他抗菌劑、消炎藥、清潔或護膚之組成份、或其組合,而製成外用藥劑形式或化粧品形式或清潔劑形式。
- 如請求項14之用途,其中該組合物尚包含積雪草萃取物。
- 如請求項14之用途,其中該組合物尚包含水楊酸。
- 如請求項16之用途,其中該組合物尚包含水楊酸。
- 一種治療或預防痤瘡之組合物,其係呈外用劑型,其組合物包含如請求項1至12中任一項之到手香萃取物、積雪草萃取物及水楊酸。
