本发明公开一种针对单增李斯特菌inlB毒力基因的快速检测方法,首次针对毒力基因inlB设计特异引物,将反转录技术与SYBR Green Ⅰ嵌合荧光定量PCR技术相结合,建立了一种快速检测单增李斯特菌的方法,具体包括单增李斯特菌RNA的提取及cDNA的合成、SYBR Green Ⅰ嵌合荧光定量PCR检测建立标准曲线、人工污染样本的验证;食物样本RNA的提取及cDNA的合成、普通SYBR Green Ⅰ嵌合荧光定量PCR法检测验证。检测速度快、特异性高、灵敏度高、检出限低的优点,具有广泛的应用前景。
Description
技术领域
本发明属于致病菌基因检测技术领域,具体涉及一种单增李斯特菌检测引物以及利用反转录SYBR GreenⅠ嵌合荧光定量PCR技术检测食物中单增李斯特菌的方法。
背景技术
李斯特氏菌是兼性厌氧的革兰氏阳性杆状菌[1]。其中单核细胞增生李斯特菌(Listeria monocytogenes,以下简称单增李斯特菌)是唯一一种与人类有关系的致病菌,广泛存在于我们日常生活中的食品中。单增李斯特菌是保证肉制品质量安全的必检项目,近年来,我国水生生物资源丰富,水产品的出口量也是逐年增加,水产品中单增李斯特菌的检出率也相对较高。目前在世界的很多国家都发生过因该菌引起的中毒事件。感染该菌后临床表现多样化,例如肠炎、脑膜炎、流产等,其中对于一些免疫力低下者最易感染。该菌的致病率远远超过其他食源性细菌,粪便污染食品后经口传播是其主要传播途径,由该菌引起的疾病称为李斯特菌病。该病发病率不高,该病的临床表现,健康成人个体出现轻微类似流感的症状。主要表现为败血症、脑膜炎和单核细胞增加,死亡率可达20%-30%[2],尤其是婴幼儿、老人、孕妇以及免疫功能缺陷者,发病机会更大[3,4]。其中,在新生儿和免疫力低下的人中因单增李斯特菌病致死的高达70%[5]。
种类多、数量少、难于富集和体外培养是食源性致病菌的特点,也决定它们一直是微生物检测中的难点和空白点。现有的检测技术如微生物培养和生化鉴定、免疫学技术、PCR技术等均存在一些问题:常规的生化鉴定方法操作复杂,具体包括富集培养、选择分离、生理生化和血清学实验。这样的方式需要周期长,准确度受到外界影响因素大;免疫学技术虽灵敏性高,但易污染,经常出现假阳性现象;PCR技术一般要与凝胶电泳联用,而电泳结果不能作为最终结论,还需要进行其他探针杂交实验,同时常规PCR容易造成PCR产物对环境的污染,可能导致假阳性结果的出现,对操作环境要求严格分区。而荧光定量PCR技术的出现实现了PCR从定性到定量的飞跃,它具有特异性强、灵敏度高、重复性好、定量准确、速度快、全封闭反应等优点。反转录PCR技术是一种将cDNA合成与PCR技术结合分析基因表达的快速灵敏的方法,主要用于对表达信息进行检测或定量分析。死的细胞RNA的半衰期要比DNA的短的多,因此可以选择用RNA作为活细胞指示器。于是将反转录技术与荧光定量PCR技术相结合,提高检测灵敏度、准确率以及降低假阳性率,使检测过程更高效。
致病的毒力基因常成簇状,扎堆形成毒力岛。单增李斯特菌就是含有100个大小不一样的毒力岛组成,其中单增李斯特菌毒力岛1(LIPI-1)就是毒力基团中心聚集处,位于此中心位置的毒力基因都发挥自己的功能。单增李斯特菌的内化素岛(LIPI-2)主要参与诱导粘附、侵袭,是一个基因家族系inlA-inlH。其中最重要的是inlA和inlB,inlA和inlB是单增李斯特菌中最为重要的内化素蛋白。几乎所有致病性的单增李斯特菌都含有inlA和inlB。对单增李斯特菌具有鉴别意义[6]。其中,inlB基因属于内化素家族,位于菌体表面,介导细菌侵入细胞并内化至吞噬小体。inlB基因具有受体特异性与肝细胞生长因子受体结合,是肝细胞、成纤维细胞、上皮细胞的介导因素等。缺失inlB基因的单增李斯特菌毒力株的毒力严重下降。
发明内容
本发明首次针对毒力基因inlB设计特异引物,将反转录技术与SYBR GreenⅠ嵌合荧光定量PCR技术相结合,建立了一种快速检测单增李斯特菌的方法。死的细胞RNA的半衰期要比DNA的短的多,因此RNA可以作为活细胞指示器。在RNA水平上检测可以检测出样品中是否存在单增李斯特菌活菌,使本发明所述方法的实验结果更加准确。具体采用的方法是通过提取样品细菌总RNA且合成cDNA为模板,继而进行SYBR GreenⅠ嵌合荧光定量PCR扩增,准确率高、高效率、低污染的完成整个过程,具有广泛的应用前景。整个检测过程可用4.5h完成,其中人工操作时间不足60min,而用传统方法检测则需要一周的时间才能完成检测,步骤繁琐,操作时间长,人工量大。
本发明的技术方案为:基于单增李斯特菌毒力基因inlB设计特异性引物,建立反转录SYBR GreenⅠ嵌合荧光定量PCR检测系统,其中包括:单增李斯特菌RNA的提取及cDNA的合成、SYBR GreenⅠ嵌合荧光定量PCR检测建立标准曲线、人工污染样本的验证;食物样本RNA的提取及cDNA的合成、普通SYBR GreenⅠ嵌合荧光定量PCR法检测验证。
本发明公开了一对从20对设计引物中筛选出来的检测引物:SEQ ID NO.1~2。设计引物过程中,经过NCBI Blast比对后发现单增李斯特菌毒力基因inlB的部分序列与其他革兰氏阳性菌毒力基因序列有重合,因此能够选取的序列片段有限且易与其他革兰氏阳性菌重合,即设计一对特异性强的引物是有难度的,经过多次特异性实验进行筛选,得到了特异性较强的引物:SEQ ID NO.1~2,如下表所示。
本发明公开了试剂盒法经过自行改进后RNA的提取、cDNA的合成以及标准曲线的建立方法。
本发明公开一种将反转录技术与SYBR GreenⅠ嵌合荧光定量PCR技术相结合的快速检测单增李斯特菌的方法,其是先将待检样品的RNA反转录成cDNA作为模板,再进行SYBRGreenⅠ嵌合荧光法检测,步骤包括:
在25μl反应体系中:
反应参数:95℃30s;95℃5s,63.3℃25s;72℃15s;80℃15s,40个循环;60~95℃建立熔解曲线。
上述反应体系中,对加入引物的量及模板量进行了改进,运用控制变量法进行体系优化,最终得到上述体系;上述反应参数,对循环数进行了改进,通常循环数为30个,本发明为了得到更精准的数据将循环数延长到40个,从而达到扩增曲线指数期明显的目的,Ct值≤40判定为阳性。
本发明还公开一种将反转录技术与SYBR GreenⅠ嵌合荧光定量PCR技术相结合的快速检测单增李斯特菌的方法,其中,本发明使用的提取RNA的方法为经过自行改进的试剂盒法。目前提取细菌总RNA的试剂盒非常少,本发明选取了天根培养细胞/细菌总RNA提取试剂盒(DP430),按照未经改进的试剂盒说明书所述方法进行RNA提取,得到的RNA浓度普遍较低,不符合合成cDNA的条件,改进试剂盒后本发明使用的提取RNA的方法,RNA浓度明显提高,且达到了cDNA合成条件,为后续的检测提供基础。
本发明使用的提取RNA的方法为经过自行改进的试剂盒法的操作步骤如下:
①4℃12000rpm离心2min收集细菌,仔细去除所有培养基上清,以后的离心步骤均在室温下进行。
注:如果培养基上清去除不完全,将对细胞壁消化过程产生抑制。
②加入120μL溶菌酶(50mg/mL)吹散,37℃30min。
注:原试剂盒中的步骤为加入含有溶菌酶(3mg/mL)的100μL TE缓冲液,室温孵育5-10min。但由于单增李斯特菌是属于较难破壁的革兰氏阳性菌,需要加入溶菌酶进行破壁,直接加入120μL 50mg/mL溶菌酶,37℃处理30min使其破壁充分。
③加入350μL裂解液RL(340μL RL+10μLβ-巯基乙醇),涡旋振荡混匀,70℃10min,12000rpm离心2min,将上清转移到另一个离心管中。
注:原试剂盒中的步骤为346.5μL RL+3.5μLβ-巯基乙醇配成350μL裂解液,然后直接12000rpm离心2min。本发明中加大了β-巯基乙醇的量,同时加入了70℃10min的步骤,其目的是使菌体裂解更充分。
④加入250μL无水乙醇,混匀。将得到的溶液和沉淀一起转入吸附柱CR3,12000rpm离心1min,倒掉废液,将吸附柱CR3放回收集管中。
⑤向吸附柱CR3加入350μL去蛋白液RW1,12000rpm离心1min,弃废液,将吸附柱放回收集管中。
⑥加入80μL DNase I工作液(10μL DNase I+70μL RDD溶液于新的RNase-Free离心管)置于室温15min。
⑦加入350μL RW1,离心12000rpm 1min,弃废液,将吸附柱放回收集管中。
⑧加入500μL RW,室温放置2min,12000rpm离心1min,倒掉废液。
⑨重复步骤⑧。
⑩12000rpm离心2min,倒掉废液,将吸附柱CR3置于室温放置4min,以彻底晾干吸附材料残留的漂洗液。
加入20-30μL RNase-Free ddH2O,室温放置2min,12000rpm离心2min。
重复3次步骤⑾。
注:原试剂盒中的步骤无重复,重复3次目的是提高RNA回收率。
经过未改进试剂盒法与本发明上述改进试剂盒法提取RNA的多次对比实验后,得到提取RNA平均值如表1。

cDNA的合成条件为RNA的浓度至少要大于200ng/μL,根据表1可明显看出未改进的试剂盒法提取RNA得到的浓度不符合合成cDNA的条件,而用改进后的试剂盒法提取RNA的浓度均符合合成cDNA的条件。
综上的试验结果证明了,本发明检测方法检测速度快、特异性高、灵敏度高、检出限低的优点。
本发明还公开了对于上文所述的技术方案中cDNA的合成方法。所述方法中,将提取样品的RNA反转录成cDNA的步骤包括:
①反应体系为6μL,其中:模板RNA浓度为200~1000ng/μL;Oligo(dT)Primer 1μL;RNase free dH2O;
反应条件:70℃10min;4℃2min;4℃放置
反应条件:42℃60min;70℃15min;4℃2min;4℃放置。
本发明还公开一种对于上文所述的方法在检测食品中单增李斯特菌的应用。并在检测过程中,同时采用国标法和普通SYBR GreenⅠ嵌合荧光定量PCR法来与本发明所述方法进行比较,从而有效验证了本申请所述方法结果的准确性。

上述附图1和2中,其中4株单增李斯特菌扩增出来了,分别为单核细胞增生李斯特菌ATCC 19111、ATCC 19112、ATCC 19115和ATCC 15313,而其他7株细菌没有扩增出来,可说明引物SEQ ID NO.1~2特异性较好,可以用作后续试验。
图3是标准菌株的扩增曲线图,图中,横坐标为循环数,纵坐标为荧光信号强度,由图可以看出,对2μL 10倍梯度稀释液模板cDNA溶液扩增时,均得到一条S型曲线。
图4是标准菌株的标准曲线图,图中,横坐标为数量级,纵坐标为CT值,由图可以看出,标准曲线相关系数R2为0.995,扩增效率为102.3%,灵敏度为5.7CFU/mL。
图5是标准菌株的熔解曲线图,图中,横坐标为温度,纵坐标为荧光量,由图可以看出Tm值均在80℃。
上述附图3~5中,菌液8个稀释度(10-1~10-8)的模板DNA溶液分别代表的菌液浓度为:5.7×107、5.7×106、5.7×105、5.7×104、5.7×103、5.7×102、57、5.7CFU/mL。
图6是人工污染样品的扩增曲线图,图中,横坐标为循环数,纵坐标为荧光信号强度,由图6可以看出,对2μL 10倍梯度稀释液模板cDNA溶液扩增时,均得到一条S型曲线,而对稀释度为10-7的模板DNA溶液(对应菌液浓度为1.32CFU/mL)扩增时,扩增曲线基本与空白对照持平,并在背景阈值线以下,表明在该浓度下不能对inlB基因进行有效检测。
图7是人工污染样品的标准曲线图,图中,横坐标为数量级,纵坐标为CT值,由图可以看出,标准曲线相关系数R2为0.994,扩增效率为102.3%,灵敏度为13.2CFU/mL。
图8是人工污染样品的熔解曲线图,图中,横坐标为温度,纵坐标为荧光量,由图可以看出Tm值均在80℃。
上述附图6~8中,菌液7个稀释度(10-1~10-7)的模板DNA溶液分别代表的菌液浓度为:1.32×106、1.32×105、1.32×104、1.32×103、1.32×102、13.2、1.32CFU/mL。
图9、11是水产品样品检测扩增曲线图,如图所示,标准品(单增李斯特菌)和样品均呈现S型曲线,说明样品中含有单增李斯特菌。
图10、12是水产品样品检测熔解曲线图,如图所示,标准品(单增李斯特菌)和样品Tm值均在80℃(±1℃属正常现象),说明样品中含有单增李斯特菌。

下述非限制性实施例,其对本方法的建立及其应用作进一步的说明,可以使本领域的普通技术人员更全面地理解本发明,但不以任何方式限制本发明。
若无特殊说明,本部分所使用的主要试剂、仪器及其商品来源为常规方法制备或商业途径获得,具体信息如下:
细菌基因组总RNA提取试剂盒(DP430)购自天根生化科技有限公司;Premix Ex TaqTM、Oligo(dT)15Primer、5×M-MLV Buffer、dNTP Mixture、RNaseInhibitor、RTase M-MLV(RNase H-)等均购自宝生物工程(大连)有限公司;20×PBS缓冲溶液、RNase Free dH2O、反转录SYBR GreenⅠ嵌合荧光定量PCR用引物均购自生工生物工程(上海)股份有限公司;李斯特菌显色培养基、单增李斯特菌成套生化鉴定管等均购自青岛海博生物技术有限公司;荧光定量PCR仪(BIO-RAD公司,美国)、高速离心机(Eppendorf公司,德国)、水浴锅(精宏实验设备有限公司,上海)。
本申请实施例中所用标准菌株信息如下:
各菌株均购自中国工业微生物菌种保藏管理中心(CICC),见表2。各菌株使用菌种保存管-80℃保存,活化培养等均按照相关的国家标准(GB)方法进行。其中,以单核细胞增生李斯特菌ATCC 19111作为本发明的标准株。

本申请实施例中所用到的实验方法如下:
一.本发明使用的cDNA合成方法为:
①反应体系6μL,其中:模板RNA;Oligo(dT)Primer 1μL;RNase free dH2O;混匀反应条件:70℃10min;4℃2min;
反应条件:42℃60min;70℃15min;4℃2min;4℃放置。
二.本发明使用的改进试剂盒法为:
待检样品的RNA在天根DP430细菌总RNA提取试剂盒基础上改进后进行提取:
①4℃12000rpm离心2min收集细菌,彻底去除所有培养基上清;
②加入120μL浓度为50mg/mL的溶菌酶,吹散,37℃30min;
③加入350μL裂解液RL,涡旋振荡混匀,70℃10min,12000rpm离心2min,将上清液转移到另一个离心管中;其中,所述的裂解液RL为340μL RL中加入10μLβ-巯基乙醇;
④向步骤③获得的上清液中,加入250μL无水乙醇,混匀;将得到的溶液和沉淀一起转入吸附柱CR3,12000rpm离心1min,倒掉废液,将吸附柱CR3放回收集管中;
⑤向吸附柱CR3加入350μL去蛋白液RW1,12000rpm离心1min,弃废液,将吸附柱CR3放回收集管中;
⑥向步骤⑤的吸附柱CR3中加入80μL DNase I工作液,置于室温15min;其中,所述的80μL DNase I由10μL DNase I中加入70μL RDD溶液配制而成;
⑦向吸附柱CR3中再加入350μL RW1,离心12000rpm 1min,弃废液,将吸附柱放回收集管中;
⑧向吸附柱CR3中加入500μL RW,室温放置2min,12000rpm离心1min,倒掉废液;
⑨重复步骤⑧;
⑩12000rpm离心2min,倒掉废液,将吸附柱CR3置于室温放置4min,以彻底晾干吸附材料残留的漂洗液;
加入20-30μL RNase-Free ddH2O,室温放置2min,12000rpm离心2min;
重复3次步骤⑾。
三.本发明使用的RNA反转录成cDNA的方法:
①反应体系为6μL,其中:模板RNA浓度为200~1000ng/μL;Oligo(dT)Primer 1μL;RNase free dH2O;
反应条件:70℃10min;4℃2min;4℃放置
反应条件:42℃60min;70℃15min;4℃2min;4℃放置。
四.本发明使用的食物样本RNA提取前处理方法:
①将装有6g样品混合物的无菌50mL离心管中,加入15mL无菌1×PBS缓冲溶液浸泡溶解,超声波处理1min;
②将离心管放于恒温摇床中300r/min,振荡5min后,将离心管放于4℃低速离心机中700r/min离心5min;
③离心后将离心管中的上层液体小心转移至另一无菌50mL离心管中,加入15mL无菌1×PBS缓冲溶液重复以上步骤处理沉淀;
④将两次获得的上清液过滤(孔径16~30nm);
⑤在无菌条件下,将所得滤液分装于1.5mL无菌Epp离心管中,放置于高速冷冻离心机中4℃10000r/min离心1min,去除上层液体收集沉淀,收集到的沉淀即为所需的菌体。
实施例1引物设计
本发明利用NCBI与FastPCR软件相结合,针对单增李斯特菌毒力基因inlB基因设计引物,共设计20对引物(引物浓度均为10μM,引物SEQ ID NO.1~40扩增片段大小为200~300bp),引物序列及扩增片段长度如表3所示:

通过PCR方法筛选出一对特异性强的引物,其中引物SEQ ID NO.1~2特异性最强。在此基础上用于后续所有检测试验。
PCR扩增体系为20μl,其中:
PCR扩增条件为:
94℃,40s;94℃,30s,57℃,30s;72℃,1min,32个循环;72℃,5min。
实施例2
特异性实验
提取表1所示11株菌的RNA,继而cDNA合成作为模板,SEQ ID NO.1~2作为引物分别进行SYBR GreenⅠ嵌合荧光定量PCR扩增。
检测结束后的结果判定:若引物SEQ ID NO.1~2能使4种单增李斯特菌扩增出来,而不使其他7种细菌扩增,则判定引物特异性高。
引物特异性验证结果如图1和2,由图示内容可知:4株单增李斯特菌扩增出来了,而其他7株细菌没有扩增出来,可说明引物SEQ ID NO.1~2特异性较好,可以用作后续试验。
实施例3
a、标准菌株待检样品的制备、标准曲线的建立及灵敏度检测:
挑取单个菌落于营养肉汤中培养17~19h,使用改进后的细菌基因组总RNA提取试剂盒提取其RNA,继而按照本发明公开的cDNA合成方法合成cDNA作为扩增模板。标记,直接作为模板或-20℃保存。
取单增李斯特菌标准株(ATCC 19111)cDNA溶液1μL进行十倍梯度稀释,取8个不同稀释梯度的cDNA样品(浓度分别为5.7×107、5.7×106、5.7×105、5.7×104、5.7×103、5.7×102、57、5.7CFU/mL)各2μL按照最佳反应体系及条件进行SYBR GreenⅠ嵌合荧光定量PCR反应,最佳体系和条件:
①反应体系为25μL,其中:
②反应参数:
95℃30s;95℃5s,63.3℃25s;72℃15s;80℃15s,40个循环;60~95℃建立熔解曲线。
③检测结束后的结果判定:扩增曲线指数期明显,且Ct值≤40判定为阳性,表明检测出单增李斯特菌。除此之外,均判定为阴性,表明没有检测出单增李斯特菌。
标准曲线由iQ5检测系统自动生成,确定SYBR GreenⅠ嵌合荧光定量PCR检测灵敏度。
结果分析:单增李斯特菌菌液浓度为5.7×108cfu/mL,图3~5中,菌液8个稀释度(10-1~10-8)的模板DNA溶液分别代表的菌液浓度为:5.7×107、5.7×106、5.7×105、5.7×104、5.7×103、5.7×102、57、5.7CFU/mL。图3是标准菌株的扩增曲线图,图中,横坐标为循环数,纵坐标为荧光信号强度,由图可以看出,对2μL 10倍梯度稀释液模板cDNA溶液扩增时,均得到一条S型曲线;图4是标准菌株的标准曲线图,图中,横坐标为数量级,纵坐标为CT值,由图可以看出,标准曲线相关系数R2为0.995,扩增效率为102.3%,灵敏度为5.7CFU/mL,相对于其他方法,扩增效率和灵敏度有了显著的提高;图5是标准菌株的熔解曲线图,图中,横坐标为温度,纵坐标为荧光量,由图可以看出Tm值均在80℃。
b、人工污染样品待检样品的制备、标准曲线的建立及灵敏度检测:
1.食物样本人工污染的方法:
①将剪碎的6g样品放入单增李斯特菌液中,常温放置30min。
②将样品放入无菌离心管(其他暂时不用的部分放在-80℃冻存,以备后续平行试验),加入15mL 1×PBS,浸泡溶解,超声波处理1min。
③摇床300r/min,5min,700r/min,低速离心5min。
④小心转移上层液体于另一无菌离心管,再用15mL 1×PBS重复以上步骤处理沉淀。
⑤将两次获得的上清液在10000r/min条件下离心10min,收集沉淀即为食物样本人工污染的菌体。
2.人工污染样本标准曲线的方法:
采用改进的试剂盒提取法提取人工污染样品的总RNA,继而按照本发明公开的cDNA合成方法合成cDNA作为扩增模板。标记,直接作为模板或-20℃保存。
取人工污染样品的cDNA溶液1μL进行十倍梯度稀释,取7个不同稀释梯度的cDNA样品(浓度分别为1.32×106、1.32×105、1.32×104、1.32×103、1.32×102、13.2、1.32CFU/mL)各2μL,按照最佳反应体系及条件,进行SYBR GreenⅠ嵌合荧光定量PCR反应,最佳体系和条件:
①反应体系为25μL,其中:
反应参数:
95℃30s;95℃5s,63.3℃25s;72℃15s;80℃15s,40个循环;60~95℃建立熔解曲线。

实施例4
应用于实际样品检测试验
2014年8月至10月的3个月期间,共检测水产品285份样品,其中包括杂色蛤、贻贝、海螺各95份。经过RNA提取前处理后用改进试剂盒法提取样品RNA,继而合成cDNA,最后采用实施例2所述的反应体系和条件进行SYBR GreenⅠ嵌合荧光定量PCR检测。结果如图9~12所示:
图9、11是水产品样品检测扩增曲线图,如图所示,标准品(单增李斯特菌)和样品均呈现S型曲线;图10、12是水产品样品检测熔解曲线图,如图所示,标准品(单增李斯特菌)和样品Tm值均在80℃(±1℃属正常现象),说明样品中含有单增李斯特菌。
图9~12所示,筛选检出的4份阳性样本,其中贻贝占3份、杂色蛤占1份。采用国标法进行检测,检出阳性样本同样是4份,其中贻贝占3份、杂色蛤占1份。同时还运用了普通SYBR GreenⅠ嵌合荧光定量PCR方法进行检测(表5为SYBR GreenⅠ嵌合荧光定量PCR加样明细),检出5份阳性样本,其中贻贝占3份、杂色蛤占2份。(表4)

综上实施例1~4的实验结果可知:
首次针对单增李斯特菌毒力基因inlB设计引物NO.1~40共20对引物,经过引物特异性检测得到SEQ ID NO.1~2的特异性强,可以用于后续试验。
用标准菌株(10-1~10-8)和人工污染样品(10-1~10-7)cDNA分别10倍梯度稀释作为模板,继而进行SYBR GreenⅠ嵌合荧光定量PCR扩增,得到的相关系数(R2)分别为0.995和0.994,扩增效率(E)分别为102.3%和102.3%,灵敏度分别为5.7CFU/mL和33CFU/g。上述结果与同水平检测方法相比,灵敏度显著提高。
筛选检出的4份阳性样本,其中贻贝占3份、杂色蛤占1份;采用国标法进行检测,检出阳性样本同样是4份,其中贻贝占3份、杂色蛤占1份;同时还运用了普通SYBR GreenⅠ嵌合荧光定量PCR方法进行检测,检出5份阳性样本,其中贻贝占3份、杂色蛤占2份。
本发明能够快速、准确的检测食物中的单增李斯特菌,该方法具有特异性强、灵敏度高、耗时时间短、准确率高等优点。
Claims (3)
1.一种将反转录技术与SYBR Green Ⅰ嵌合荧光定量PCR技术相结合的快速检测食品中单增李斯特菌的方法,其特征在于:将待检样品的RNA反转录成cDNA作为模板,再进行SYBRGreen Ⅰ嵌合荧光定量PCR法检测,步骤包括:
反应体系为25µl,其中:
2倍浓度的SYBR® Premix Ex TaqTM 12.5µL
浓度为10µmol/L的引物对SEQ ID NO.1~2 各0.5µL
灭菌双蒸水 9.5µL
浓度为200~1000ng/µL的 cDNA模板 2µL
反应参数:95℃ 30s;95℃ 5s,63.3℃ 25s;72℃ 15s;80℃ 15s,40个循环;60~95℃建立熔解曲线。
其中:上文所述的待检样品的RNA使用经改进后的天根DP430细菌总RNA提取试剂盒提取,步骤包括:
①4℃ 12000rpm 离心2min收集细菌,彻底去除所有培养基上清;
②加入120μL浓度为50mg/mL的溶菌酶,吹散,37℃ 30min;
③加入350µL裂解液RL,涡旋振荡混匀,70℃ 10min,12000rpm离心2min,将上清液转移到另一个离心管中;其中,所述的裂解液RL为340µL RL中加入10µL β-巯基乙醇;
④向步骤③获得的上清液中,加入250µL无水乙醇,混匀;将得到的溶液和沉淀一起转入吸附柱CR3,12000rpm离心1min,倒掉废液,将吸附柱CR3放回收集管中;
⑤向吸附柱CR3加入350µL去蛋白液RW1,12000rpm离心1min,弃废液,将吸附柱CR3放回收集管中;
⑥向步骤⑤的吸附柱CR3中加入80µL DNase I工作液,置于室温15min;其中,所述的80µL DNase I由10µL DNase I中加入70µL RDD溶液配制而成;
⑦向吸附柱CR3中再加入350µL RW1,离心12000rpm 1min,弃废液,将吸附柱放回收集管中;
⑧向吸附柱CR3中加入500µL RW,室温放置2min,12000rpm离心1min,倒掉废液;
⑨重复步骤⑧;
⑩12000rpm离心2min,倒掉废液,将吸附柱CR3置于室温放置4min,以彻底晾干吸附材料残留的漂洗液;
⑪加入20-30µL RNase-Free ddH2O,室温放置2min,12000rpm离心2min;
⑫重复3次步骤⑾。
2.根据权利要求1所述的方法,其特征在于:所述的将待检样品的RNA反转录成cDNA的步骤包括:
①反应体系为6µL,其中:模板RNA浓度为200~1000ng/µL ;Oligo(dT)Primer 1 µL;RNase free dH2O;
反应条件:70℃ 10min;4℃ 2min;4℃ 放置
②以上模板RNA变性溶液 6µL
5×M-MLV Buffer 2µL
dNTP Mixture 0.5µL
RNase Inhibitor 0.25µL
RTase M-MLV(RNase H-) 0.5µL
RNase Free dH2O 0.75µL
反应条件:42℃ 60min;70℃ 15min;4℃ 2min;4℃ 放置。
