本发明提供了一种鲜烟叶生产酒精的方法,具体如下:鲜烟叶,破碎,打浆,加入干烟叶粉20%、果胶酶0.15%、纤维素酶0.1%,混合均匀,50℃,酶解90min,加入盐酸2%,140-145℃水解60min,冷却至80℃,离心,得上清液;加入硫酸铵0.1%、磷酸二氢钾0.1%、硫酸镁0.05%,调pH值至5.0-5.5,85℃杀菌5min,冷却至30℃,接种酒精活性干酵母0.1%、果胶酶0.1%、纤维素酶0.05%,28-32℃发酵60h,在发酵至24h、40h时分别添加干烟叶粉10%、8%,蒸馏,即得酒精。

技术领域
本发明属于化工领域,涉及一种鲜烟叶生产酒精的方法。
背景技术
烟草是我国农业经济的主导产业之一。据统计,全国烟草种植面积达1500多万亩,除去可用于深加工的正常烟叶外,还剩有烟秆、烟梗、烟花、烟杈、底脚叶和部分顶叶等数量巨大的烟草废弃物,其中废弃鲜烟叶每年约有180万吨。废弃鲜烟叶含有碳水化合物4-5%、还原糖2.5-3%,纤维素1.5-2%,还有少量的蛋白质、烟碱及挥发酸、矿物质等,废弃了十分可惜,不但浪费了资源,有时还会造成环境污染。
发明内容
本发明提供了一种鲜烟叶生产酒精的方法。
本发明的目的是按下述方式实现的:鲜烟叶,破碎,打浆,加入干烟叶粉20%、果胶酶0.15%、纤维素酶0.1%,混合均匀,50℃,酶解90min,加入盐酸2%,140-145℃水解60min,冷却至80℃,离心,得上清液;加入硫酸铵0.1%、磷酸二氢钾0.1%、硫酸镁0.05%,调pH值至5.0-5.5,85℃杀菌5min,冷却至30℃,接种酒精活性干酵母0.1%、果胶酶0.1%、纤维素酶0.05%,28-32℃发酵60h,在发酵至24h、40h时分别添加干烟叶粉10%、8%,发酵结束后进行蒸馏,即得酒精。
所述的干烟叶粉是指鲜烟叶经烘干、粉碎、过40目筛制得的干烟叶粉。
经过检测,鲜烟叶成分如下:水分85%、水溶性总糖4.48%、还原糖3.98%、纤维素1.87%、蛋白质0.08%、总氮0.03%;干烟叶粉成分如下:水溶性总糖29.85%、还原糖26.50%、纤维素12.46%、蛋白质0.92%、总氮0.37%。由成分分析可以见,鲜烟叶中含有可发酵糖、蛋白质和氨基酸,但总糖含量偏低。为了提高生产效率和酒精收率,并充分利用烟叶资源,本发明将鲜烟叶经烘干、粉碎制成干烟叶粉,其水溶性总糖接近30%,再加回到配料中,解决了初始总糖偏低的问题;同时为了有效利用烟叶含有的纤维素,采用酶水解加酸水解的双重水解方式,来提高利用率。本发明为大量废弃烟叶的综合利用提供了一条有效途径。
四、具体实施例
实施例1
具体实施方式:称取鲜烟叶1000g,破碎,打浆,加入干烟叶粉200g、果胶酶1.5g、纤维素酶1g,混合均匀,50℃,酶解90min,加入盐酸20mL,140-145℃水解60min,冷却至80℃,离心,得上清液;量取上清液500mL,加入硫酸铵0.5g、磷酸二氢钾0.5g、硫酸镁0.25g,调pH值至5.0-5.5,85℃杀菌5min,冷却至30℃,接种酒精活性干酵母0.5g、果胶酶0.5g、纤维素酶0.25g,28-32℃发酵48h,发酵24h时添加干烟叶粉50g,发酵至40h时再添加干烟叶粉40g,发酵结束后进行蒸馏,即得酒精。
实施例2
具体实施方式:称取鲜烟叶500g,鲜烟叶,破碎,打浆,加入干烟叶粉100g、果胶酶0.75g、纤维素酶0.5g,混合均匀,50℃,酶解90min,加入盐酸10mL,140-145℃水解60min,冷却至80℃,离心,得上清液;量取上清液200mL,加入硫酸铵0.2g、磷酸二氢钾0.2g、硫酸镁0.1g,调pH值至5.0-5.5,85℃杀菌5min,冷却至30℃,接种酒精活性干酵母0.2g、果胶酶0.2g、纤维素酶0.1g,28-32℃发酵60h,在发酵至24h、40h时分别添加干烟叶粉20g、16g,发酵结束后进行蒸馏,即得酒精。

1.本发明提供了一种鲜烟叶生产酒精的方法,其特征如下:鲜烟叶,破碎,打浆,加入干烟叶粉20%、果胶酶0.15%、纤维素酶0.1%,混合均匀,50℃,酶解90min,加入盐酸2%,140-145℃水解60min,冷却至80℃,离心,得上清液;加入硫酸铵0.1%、磷酸二氢钾0.1%、硫酸镁0.05%,调pH值至5.0-5.5,85℃杀菌5min,冷却至30℃,接种酒精活性干酵母0.1%、果胶酶0.1%、纤维素酶0.05%,28-32℃发酵60h,在发酵至24h、40h时分别添加干烟叶粉10%、8%,发酵结束后进行蒸馏,即得酒精。
