Abstract

本发明通过对毛蕊异黄酮进行实验研究,首次发现毛蕊异黄酮可明显对抗刀豆蛋白A(ConA)诱导的T淋巴细胞增殖,具有免疫抑制作用,利于未来制备免疫抑制药物。

毛蕊异黄酮在制备免疫抑制剂方面的用途
技术领域
本发明涉及医药领域,具体涉及毛蕊异黄酮在制备免疫抑制剂方面的用途。 
背景技术
黄芪是是一味健脾益气的中药,具有补气固表、利尿托毒、排脓、敛疮生肌的作用,临床用于气虚乏力,食少便溏,中气下陷,久泻脱肛,便血崩漏,表虚自汗,气虚水肿,痈疽难溃,久溃不敛,血虚痿黄,内热消渴,慢性肾炎蛋白尿和糖尿病等症(参见:国家药典委员会.中华人民共和国药典(一部)[z].北京:化学工业出版社,2005:212-213.)。现代药理学研究认为,黄芪能够调节机体免疫功能并具有抗病毒作用(参见:晏洪波,梁文,涂亚庭.黄芪的免疫调节及抗病毒作用[J].华南国防医学杂志,2008,22(6):69-70)。 
黄芪的主要化学成分包括:黄芪黄酮类化合物、黄芪皂苷类化合物、黄芪多糖类化合物、以及黄芪中的氨基酸和微量元素,其中黄芪黄酮类化合物是黄芪的重要有效成分。毛蕊异黄酮是黄芪异黄酮类中重要的化合物之一,其在免疫抑制方面的用途目前尚无人报道。 
发明内容
本发明的目的是提供毛蕊异黄酮在制备免疫抑制剂方面的用途。 
本发明所研究的毛蕊异黄酮化学结构如下,通过从蒙古黄芪中分离制备得到: 
毛蕊异黄酮: 
本发明通过刀豆蛋白A(Con A)诱导小鼠T淋巴细胞增殖试验,首次发现毛蕊异黄酮对T淋巴细胞增殖具有抑制作用,能用于未来制备免疫抑制剂。  
具体实施方式
下面结合实施例对本发明作进一步的描述: 
实施例:毛蕊异黄酮对刀豆蛋白A(ConA)诱导的小鼠T细胞增殖的影响 
1材料 
1.1动物 
BALB/c小鼠,体重18~20g,雄性;由北京大学实验动物部提供(许可证号:SCXK11-00-0008)。动物饲养于标准环境:温度(23±1)℃,相对湿度(55±5)%,明暗周期12h/12h,标准颗粒饲料喂养。 
1.2试剂 
刀豆蛋白A(ConA),噻唑蓝(MTT)购自美国Sigma公司;胎牛血清(FBS),购自美国GIBCO公司;红细胞裂解液,1640培养液,磷酸盐(PBS)缓冲液均购自北京迈晨生物科技公司;DMSO(二甲基亚砜,批号20101008),购自北京化工厂。毛蕊异黄酮样品用DMSO配制。 
1.3仪器 
Bro-Rad iMark酶标仪(美国伯乐公司),Sigma3K-15离心机(德国Sigma公司),CO2培养箱(美国Thermal Technology LLC公司),超净工作台(北京东联哈尔仪器制造有限公司)等。 
2方法 
小鼠摘眼球放血,无菌取脾脏匀浆过滤,离心5min(4℃,1000r·min-1),弃上清,用红细胞裂解液裂解红细胞后用PBS洗涤2次;用1640培养液(含10%FBS)悬浮脾细胞,细胞计数并使用台盘蓝染色法测定细胞的存活率,存活率=未着色细胞数/细胞总数×100%,存活率>95%者用于试验,细胞调至2×106个/mL。 
以丝裂原刀豆蛋白(ConA,2.5μg·mL-1)刺激T细胞使其增殖。分组:细胞对照组(含等浓度的DMSO),Con A刺激组,毛蕊异黄酮样品组(浓度根据预试验结果定为30μmol·L-1),各设3个复孔,重复3次。培养44h(37℃,5%CO2,饱和湿度)后加入MTT20μL(0.5mg·mL-1),培养4h,离心6min(1500r·min-1),弃上清后各孔均加DMSO100μL轻微振荡10min,酶标仪570nm测吸光度值(A值)。 
抑制率(%)=(Con A组A均值样品组A均值)/Con A组A均值×100% 
3实验结果 
ConA能明显促进小鼠T淋巴细胞增殖(p<0.01),首次发现毛蕊异黄酮能明显抑制ConA诱导的T细胞增殖(p<0.01),抑制率为:61.63%,未来可用于制备免疫抑制方面的药物, 具体结果见下表:

与DMSO对照组相比,##P<0.01,与ConA组相比**P<0.01。
