Abstract
本发明以牛膝、苍术和薏苡仁为主要原料制备一种清热解毒凉茶,其重量百分比组成为牛膝齐墩果酸提取物1%~3%、苍术多糖提取物0.5%~2%、薏苡仁多糖提取物0.5%~2%、甜味剂1%~10%、酸味剂0.5%~1%、水82%~96.5%,经过原料粉碎、提取、调配、均质、灌装和灭菌而成。本发明的清热解毒凉茶具有清心火、解热毒、除湿健脾、增强食欲等功效。

本发明属于饮料加工技术领域,特别涉及一种清热解毒凉茶的制备方法。
背景技术
凉茶是我国南方地区根据当地的气候以及生活方式,在长期预防疾病与保健的过程中,以中草药为基础、以中医养生理论为指导制作出的一类具有清热解毒、生津止渴等功效的饮料总称。凉茶的生产主要包括浸提、配制、灭菌等工艺,首先得到用热水或冷水浸提的可溶性成分的提取液(王萍等,广式凉茶研究进展,食品科技,2010,35:77-80),再将提取液与果汁、蜂蜜等结合,赋予凉茶饮料较好的风味和感官特性。根据凉茶产品的包装方式不同,目前其灭菌工艺主要有高温灭菌和超高温瞬时灭菌等工艺。近年来,随着新技术开发,高压灭菌、膜除菌和辐射灭菌等新技术开始受了人们的关注。
凉茶配方多种多样,如广东传统凉茶的配方是“三花三草一叶”再加上水和白砂糖,具体包括菊花、金银花、鸡蛋花、甘草、仙草、夏枯草和布渣叶。具有清热解毒功效的中成药中,著名的有同仁堂的“二妙丸”,其配料为苍术(炒)、黄柏(炒),用于燥湿清热,湿热下注;吉林紫鑫药业的“三妙丸”,其配料为苍术、牛膝、盐黄柏、薏苡仁,具有清热利湿的功效,用于湿热下注所致的痹病,症见足膝红肿、筋骨疼痛。在卫生部2009年公布的《药食同源品名单》中,以上原料中被列入既是食品又是药品的物品名单的有薏苡仁,被列入保健食品的物品名单的有苍术、川牛膝、怀牛膝,而黄柏未被列入。
牛膝(Achyranthesbidentata)为苋科牛膝属的植物,生长在山坡林下。牛膝是多年生草本,根圆柱形,直径5~10mm,高70~120cm。茎有棱角或四方形,带紫色或绿色,有白色贴生或开展柔毛,或近无毛,分枝对生。花期7~9月,果期9~10月。采收和储藏期南方为11月下旬至12月中旬,北方为10月中旬至11月上旬。牛膝的主要功效为补肝肾、清热去火、强筋骨、活血通经、引火下行、利尿通淋等,主治腰膝酸痛、血滞经闭、痛经、产后血瘀腹痛、热淋、血淋、痈肿恶疮、咽喉肿痛等。齐墩果酸(Oleanolicacid)是牛膝中主要的活性物质之一,又名庆四素,属于五环三萜类化合物。齐墩果酸具有消炎、抗病毒、调节免疫力、抑制血小板凝聚、抗过氧化抗衰老等多种药理作用。齐墩果酸对动物无明显毒副作用,安全无毒,除了可以将其作为天然抗氧化剂应用于食品工业外,还可将齐墩果酸含量高的食品制成保健食品(赵培西等,牛膝中齐墩果酸提取工艺研究,陕西中医,2005,26:1099-1101)。
苍术(Atractylodeslancea)为菊科苍术属的植物,多年生直立草本,生长于海拔50米至1900米的地区,多生在灌丛、野生山坡草地或岩缝隙中,目前尚未有人工引种栽培。苍术是著名的野生中药材,是东北野生道地药材的代表,也是野生药材品种中用途最广、用量最大的品种之一。苍术主治湿困脾胃、倦怠嗜卧、胞痞腹胀、食欲不振、痰饮、湿肿、表证夹湿、头身重痛等(陈炎明等,苍术化学成分和药理活性研究进展,上海中医药大学学报,2006,20:95-98)。
薏仁(SemenCoicis)又名薏苡仁、苡米、苡仁、土玉米、薏米、起实、薏珠子、草珠珠、回回米、米仁、六谷子(周爱芬等,薏苡药效物质基础研究进展,亚太传统医药,2012,8:188-190)。薏仁是常用的中药,性味甘淡微寒,有利水消肿、健脾去湿、舒筋除痹、清热排脓等功效,为常用的利水渗湿药。薏仁又是一种美容食品,常食可以保持人体皮肤光泽细腻,消除粉刺、雀斑、老年斑、妊娠斑、蝴蝶斑,对脱屑、痤疮、皲裂、皮肤粗糙等都有良好疗效(陆雅丽等,薏苡仁综合开发利用,中国食物与营养,2013,19:64-66)。薏米因含有多种维生素和矿物质,有促进新陈代谢和减少胃肠负担的作用,可作为病中或病后体弱患者的补益食品;薏米能增强肾功能,并有清热利尿作用,因此对浮肿病人也有疗效;除此之外薏米中含有丰富的维生素B,对防治脚气病十分有益(曾海龙,薏苡仁多糖提取纯化及流变学特性和抗氧化研究,南昌大学硕士学位论文,2011)。
本发明以著名的清热利湿良药“二妙丸”、“三妙丸”和“四妙丸”的配方作为参考,采用牛膝、苍术、薏苡仁为原料,甜味剂和酸味剂为辅料,经过原料粉碎、提取、调配、均质、灌装、灭菌,制备一种凉茶保健饮料。
发明内容
本发明的目的是提供一种清热解毒凉茶的制备方法,以苍术、牛膝、薏苡仁为主要原料,分别经粉碎、提取得到牛膝齐墩果酸、苍术多糖、薏苡仁多糖提取物,以甜味剂和酸味剂为辅料,再经过调配、均质、灌装、灭菌而成。该凉茶的重量百分比组成为牛膝齐墩果酸提取物1%~3%、苍术多糖提取物0.5%~2%、薏苡仁多糖提取物0.5%~2%、甜味剂1%~10%、酸味剂0.5%~1%、水82%~96.5%,其中甜味剂选自蔗糖、葡萄糖、果糖、果葡糖浆中的一种或多种,酸味剂选自柠檬酸、苹果酸、乳酸、酒石酸、抗坏血酸的一种或多种。具体制备过程如下:
1)将牛膝清洗干净,于60~80℃恒温干燥4~6小时,取出后粉碎,过10~40目筛,得牛膝粉;将牛膝粉按照料液比1∶20~1∶40加入60%~80%(v/v)的乙醇,超声波处理10~30分钟后抽滤;滤渣按料液比1∶15用60%~80%(v/v)的乙醇重复提取两次,合并滤液,于60℃减压浓缩,浓缩物于60℃干燥至恒重,得牛膝齐墩果酸提取物,备用;
2)将苍术和薏苡仁清洗干净,于60~80℃恒温干燥4~6小时,取出后分别粉碎,过10~40目筛,分别得苍术粉和薏苡仁粉;分别将苍术粉和薏苡仁粉按照料液比1∶20~1∶40加入水,用盐酸调节pH5~6,超声波处理10~30分钟后抽滤;滤渣按料液比1∶15用水重复提取两次,合并滤液于80℃减压浓缩至原液的1/5,加入4倍量的无水乙醇,4℃放置6小时后离心,沉淀依次用无水乙醇、丙酮、乙醚洗涤,于60℃干燥至恒重,分别得苍术和薏苡仁多糖提取物,备用;
3)将以上提取物以及由蔗糖、葡萄糖、果糖、果葡糖浆组成的一种或多种甜味剂,由柠檬酸、苹果酸、乳酸、酒石酸、抗坏血酸组成的一种或多种酸味剂和水按牛膝齐墩果酸提取物1%~3%、苍术多糖提取物0.5%~2%、薏苡仁多糖提取物0.5%~2%、甜味剂1%~10%、酸味剂0.5%~1%、水82%~96.5%的比例放于调配缸中,升温到60~90℃搅拌30分钟,使之充分溶解;
4)将调配好的凉茶用高压均质机20~40MP均质两次,均质后进行灌装,装入三片罐中;
5)将灌装好的凉茶于120℃灭菌40分钟。
本发明的有益效果:
1)本发明将传统的中药提取物配以糖和酸,不仅提升其药用和营养价值,而且口感更加温和,符合人们的口感需求。
2)本发明具有清热去火、生津止渴等多种功效,通过将传统的汤药制成饮料,方便饮用。
3)传统凉茶生产方法是浸煮而成,本专利在原材料浸提中采用超声波辅助浸提,不仅大大减少了浸提时间,而且能有效保护原料中的功能性成分,得率高,效果好,优化了生产工艺。
具体实施方式
下面的实施例将对本发明作进一步的解释,但是本发明并不仅仅局限于这些实施例,这些实施例不以任何方式限制本发明的范围。
实施例1:
1)将5kg牛膝清洗干净,于60℃恒温干燥4小时,取出后粉碎,过10目筛,得牛膝粉;将牛膝粉按照料液比1∶20加入100kg的60%(v/v)乙醇,超声波处理10分钟后抽滤;滤渣按料液比1∶15用60%(v/v)的乙醇重复提取两次,合并滤液,于60℃减压浓缩,浓缩物于60℃干燥至恒重,得牛膝齐墩果酸提取物60g,备用;
2)将1kg苍术和1kg薏苡仁清洗干净,于60℃恒温干燥4小时,取出后分别粉碎,过10目筛,分别得苍术粉和薏苡仁粉;将苍术粉和薏苡仁粉分别按照料液比1∶20加入20kg水,用盐酸调节pH5,超声波处理10分钟后抽滤;滤渣按料液比1∶15用水重复提取两次,合并滤液于80℃减压浓缩至原液的1/5,加入4倍量的无水乙醇,4℃放置6小时后离心,沉淀依次用无水乙醇、丙酮、乙醚洗涤,于60℃干燥至恒重,分别得到73g苍术多糖提取物和38g薏苡仁多糖提取物,备用;
3)按凉茶总量6kg,将以上提取物以及甜味剂、酸味剂和水按牛膝齐墩果酸提取物1%(即60g)、苍术多糖提取物0.5%、薏苡仁多糖提取物0.5%、蔗糖1%、柠檬酸0.5%、水96.5%的重量百分比于调配缸中,升温到60℃搅拌30分钟,使之充分溶解;
4)将调配好的凉茶用高压均质机20MP均质两次,均质后进行灌装,装入三片罐中;
5)将灌装好的凉茶于120℃灭菌40分钟。
实施例2:
1)将30kg牛膝清洗干净,于80℃恒温干燥6小时,取出后粉碎,过40目筛,得牛膝粉;将牛膝粉按照料液比1∶40加入1200kg的80%(v/v)的乙醇,超声波处理30分钟后抽滤;滤渣按料液比1∶15用80%(v/v)的乙醇重复提取两次,合并滤液,于60℃减压浓缩,浓缩物于60℃干燥至恒重,得牛膝齐墩果酸提取物330g,备用;
2)将6kg苍术和6kg薏苡仁清洗干净,于80℃恒温干燥6小时,取出后分别粉碎,过40目筛,分别得苍术粉和薏苡仁粉;分别将苍术粉和薏苡仁粉按照料液比1∶40加入240kg水,用盐酸调节pH6,超声波处理30分钟后抽滤;滤渣按料液比1∶15用水重复提取两次,合并滤液于80℃减压浓缩至原液的1/5,加入4倍量的无水乙醇,4℃放置6小时后离心,沉淀依次用无水乙醇、丙酮、乙醚洗涤,于60℃干燥至恒重,分别得到420g苍术多糖提取物和210g薏苡仁多糖提取物,备用;
3)按凉茶总量10kg,将以上提取物以及甜味剂、酸味剂和水按牛膝齐墩果酸提取物3%(即300g)、苍术多糖提取物2%、薏苡仁多糖提取物2%、果葡糖浆10%、抗坏血酸0.5%、酒石酸0.5%、水82%的重量百分比于调配缸中,升温到90℃搅拌30分钟,使之充分溶解;
4)将调配好的凉茶用高压均质机40MP均质两次,均质后进行灌装,装入三片罐中;
5)将灌装好的凉茶于120℃灭菌40分钟。
实施例3:
1)将200kg牛膝清洗干净,于70℃恒温干燥5小时,取出后粉碎,过20目筛,得牛膝粉;将牛膝粉按照料液比1∶30加入6000kg的70%(v/v)的乙醇,超声波处理20分钟后抽滤;滤渣按料液比1∶15用70%(v/v)的乙醇重复提取两次,合并滤液,于60℃减压浓缩,浓缩物于60℃干燥至恒重,得牛膝齐墩果酸提取物2.5kg,备用;
2)将30kg苍术和30kg薏苡仁清洗干净,于70℃恒温干燥5小时,取出后分别粉碎,过20目筛,分别得苍术粉和薏苡仁粉;分别将苍术粉和薏苡仁粉按照料液比1∶30加入900kg水,用盐酸调节pH5.5,超声波处理20分钟后抽滤;滤渣按料液比1∶15用水重复提取两次,合并滤液于80℃减压浓缩至原液的1/5,加入4倍量的无水乙醇,4℃放置6小时后离心,沉淀依次用无水乙醇、丙酮、乙醚洗涤,于60℃干燥至恒重,分别得到2.1kg苍术多糖提取物和1.1kg薏苡仁多糖提取物,备用;
3)按凉茶总量100kg,将以上提取物以及甜味剂、酸味剂和水按牛膝齐墩果酸提取物2%(即2kg)、苍术多糖提取物1%、薏苡仁多糖提取物1%、葡萄糖4%、果糖4%、苹果酸0.5%、乳酸0.5%、水85%的重量百分比于调配缸中,升温到70℃搅拌30分钟,使之充分溶解;
4)将调配好的凉茶用高压均质机30MP均质两次,均质后进行灌装,装入三片罐中;
5)将灌装好的凉茶于120℃灭菌40分钟。
Claims (1)
1.一种清热解毒凉茶的制备方法,其步骤如下:
1)将牛膝清洗干净,于60~80℃恒温干燥4~6小时,取出后粉碎,过10~40目筛,得牛膝粉;将牛膝粉按照料液比1∶20~1∶40加入60%~80%(v/v)的乙醇,超声波处理10~30分钟后抽滤;滤渣按料液比1∶15用60%~80%(v/v)的乙醇重复提取两次,合并滤液,于60℃减压浓缩,浓缩物于60℃干燥至恒重,得牛膝齐墩果酸提取物,备用;
2)将苍术和薏苡仁清洗干净,于60~80℃恒温干燥4~6小时,取出后分别粉碎,过10~40目筛,分别得苍术粉和薏苡仁粉;分别将苍术粉和薏苡仁粉按照料液比1∶20~1∶40加入水,用盐酸调节pH5~6,超声波处理10~30分钟后抽滤;滤渣按料液比1∶15用水重复提取两次,合并滤液于80℃减压浓缩至原液的1/5,加入4倍量的无水乙醇,4℃放置6小时后离心,沉淀依次用无水乙醇、丙酮、乙醚洗涤,于60℃干燥至恒重,分别得苍术和薏苡仁多糖提取物,备用;
