技术领域
 本发明涉及一种分析方法,特别是河豚素的分析方法。
背景技术
河豚毒素一种存在于河豚、蝾螈、斑足蟾等动物中的海洋毒素,分子式C11H17N3O8,它是一种化学结构独特、毒性强烈并有广泛药理作用的一种天然毒素,是无色棱柱状晶体,对热不稳定,难溶于水,可溶于弱酸的水溶液,在碱性溶液中易分解,在低的pH值溶液中也不稳定。河豚毒素是一种能麻痹神经的剧毒,通常只需氰化钾五百分之一就可置人于死地的剧毒,中毒的人会因神经麻痹而窒息而死,其中,毒素直接进入血液中毒死亡速度最快。河豚毒素虽为剧毒,但是其高活性和高特异性的生物特征有着潜在的、巨大的医药开发价值,在临床应用方面有广阔的前景,如果利用妥当,“变毒为宝”,既减少河豚毒素处理不当对环境造成的污染,又可以创造经济利润。从稀有昂贵的河豚鱼卵巢、肝脏等内脏器官中提取河豚毒素,技术要求精湛,寻找到先进、合理的提取工艺,可以申请专利,形成技术壁垒,具有很高的商业开发价值。
专利201010227994.2涉及一种用于戒毒、镇痛的河豚毒素制剂的制备方法。专利200880005935.1涉及河豚毒素的生物合成方法。专利01142658.6公开了一种提取河豚鱼毒素的方法。还有不少专利涉及河豚素的制备和应用方面,但未见对其用高效液相色谱分析的报道。
发明内容
本发明的目的是提供河豚素的高效液相色谱分析方法。
本发明为实现上述发明目的采用的技术方案如下:采用高效液相色谱分析仪,以反相C18为填料,以磷酸二氢钠、磷酸氢二钠、庚烷磺酸钠的水溶液为流动相进行分析洗脱,系统温度为25~40℃。本发明具有以下技术效果:本发明采用高效液相色谱仪,分析河豚素,分析条件简单,分离度高,重现性好。
具体实施方式:
实施例1
采用高效液相色谱分析仪,以反相C18为填料,填料粒径5um,色谱柱尺寸为Φ4.6×250mm,以磷酸二氢钠、磷酸氢二钠、庚烷磺酸钠的水溶液为流动相进行分析洗脱,流动相的配置方法为:0.195gNaH2PO4+3.276gNa2HPO4+0.6g庚烷磺酸钠+300ml水,流速0.8ml/min,系统温度为25℃。
采用的紫外光度检测器的检测波长为210nm,得到的样品峰对称度好,分离度达2.3。
实施例2
采用高效液相色谱分析仪,以反相C18为填料,填料粒径5um,色谱柱尺寸为Φ4.6×250mm,以磷酸二氢钠、磷酸氢二钠、庚烷磺酸钠的水溶液为流动相进行分析洗脱,流动相的配置方法为:0.195gNaH2PO4+3.276gNa2HPO4+0.6g庚烷磺酸钠+300ml水,流速0.8ml/min,系统温度为30℃。
采用的紫外光度检测器的检测波长为210nm,得到的样品峰对称度好,分离度达2.2。
 
实施例3
采用高效液相色谱分析仪,以反相C18为填料,填料粒径5um,色谱柱尺寸为Φ4.6×250mm,以磷酸二氢钠、磷酸氢二钠、庚烷磺酸钠的水溶液为流动相进行分析洗脱,流动相的配置方法为:0.195gNaH2PO4+3.276gNa2HPO4+0.6g庚烷磺酸钠+300ml水,流速0.8ml/min,系统温度为28℃。
采用的紫外光度检测器的检测波长为210nm,得到的样品峰对称度好,分离度达2.3。

1.一种河豚素的分析方法,其特征在于:采用高效液相色谱分析仪,以反相C18为填料,以磷酸二氢钠、磷酸氢二钠、庚烷磺酸钠的水溶液为流动相进行分析洗脱,系统温度为25~40℃。
2.根据权利要求1所述的方法,其特征是:流动相的洗脱方式是等度洗脱。
3.根据权利要求1所述的方法,其特征在于所述的系统的最适宜的操作温度为30℃。
4.根据权利要求1所述的方法,其特征在于所述的填料的粒径是5um。
