技术领域
本发明涉及心脏肥大的治疗和预防。更具体地,本发明涉及用于通过抑制离子通道TRPV1来预防或治疗哺乳动物(包括人类)的心脏肥大、心脏重塑(cardiac remodeling)、纤维化、高血压和心力衰竭的方法和组合物。
背景技术
心肌肥大是心脏对长期增加的工作负荷的基本应答,其可以源自诸如高血压或心脏瓣膜病变等病症。心肌肥大的进展代表了心力衰竭的发展和随后的心源性死亡的主要危险因素。
本发明的焦点在于与肥大、细胞凋亡纤维化和心力衰竭做斗争,聚焦于TRPV1 (瞬时型感受器电位阳离子通道亚家族V成员1)的调节,所述TRPV1是复杂的且重要的感受器/通道。TRPV1通常被归类为伤害感受器。公开的数据表明,TRPV1的开放可能性由内源性大麻素N-花生四烯酰基乙醇胺控制,它的内源性配体和调控N-花生四烯酰基乙醇胺水平的通路也会影响TRPV1活化。TRPV1 (瞬时型感受器电位阳离子通道亚家族V成员1)的肥大性调节的病原学不明。关于如何调节TRPV1和TRPV1所存在的几种细胞和组织类型的身份,仅存在一般性理解。
已经在作为疼痛感受器的周围感觉神经元中研究了TRPV1;但是,TRPV1在众多组织和细胞类型中表达,包括心血管系统的那些。TRPV1表达在肥大心脏中被增量调节,并且通道被定位以接收肥大心脏的刺激信号。TRVP1是6个跨膜四聚体非选择性阳离子通道,通常与参与伤害感受的周围感觉神经元有关。TRPV1的外源性活化剂包括大于43℃的温度和辣椒辣素。在内源方面,内源性大麻素、N-花生四烯酰基乙醇胺和N-花生四烯酰基-多巴胺(N-arachidonoyl-dopamine)、低pH、以及蛋白激酶C (PKC)和环AMP依赖性蛋白激酶(PKA)的磷酸化会活化和增强TRPV1。TRPV1活化在周围感觉神经元中的伤害性参与,已经激励了TRPV1作为抑制靶标的大量研究。所以,已经制备了许多有效的TRPV1拮抗剂,并证实在炎症性疼痛和痛觉过敏的控制中是有效的镇痛药。
除了周围感觉神经元以外,TRPV1也存在于其它可应激的和不可应激的组织中,包括心脏和循环系统的那些组织。例如,心肌细胞、心脏血管、血管周围神经、肺动脉平滑肌细胞、和冠状动脉内皮细胞、骨骼肌、肥大细胞、和树突细胞表达TRPV1。
尽管尚未在心脏肥大的背景下研究TRPV1抑制,TRPV1活化已经涉入对抗心肌缺血再灌注损伤的保护。另外,所述通道的内源性配体N-花生四烯酰基乙醇胺已经涉入多种心脏疾病诸如心脏毒性和高血压。
发明内容
本发明提供了用于治疗哺乳动物受试者的心脏肥大的方法,所述方法包括:给所述受试者施用抗肥大有效量的离子通道TRPV1抑制剂。在某些实施方案中,本发明提供了治疗方法,其中所述受试者的心脏肥大的症状包括心脏重塑、心脏纤维化、细胞凋亡、高血压或心力衰竭。
本发明另外提供了哺乳动物受试者的心脏肥大的预防性处理方法,所述方法包括:给所述受试者施用抗肥大有效量的离子通道TRPV1抑制剂。
本发明也提供了可用于本发明的方法中的药物组合物,其包含药学有效量的离子通道TRPV1抑制剂或这类抑制剂的混合物。
本发明也提供了可用于本发明的方法中的药物组合物,其包含药学有效量的离子通道TRPV1抑制剂N-(4-叔丁基-苯基)-4-(3-氯吡啶-2-基)-四氢吡嗪-1(2H)-甲酰胺或该抑制剂与其它离子通道TRPV1抑制剂的混合物。

具体实施方式
心脏肥大经典地被视作在负荷增加情况下的适应性和补偿性应答,其会增加心肌细胞的工作输出量,并从而维持心脏功能。在小鼠中,通常通过使用横主动脉狭窄(TAC)诱导急性压力负荷过度来建立心脏肥大模型。由主动脉狭窄造成的阻力增加最初会损害左心室(LV)功能;随后在TAC以后2周形成的左心室肥大开始恢复心脏收缩功能。向心性左心室肥大在TAC后第2-11周期间持续,可能使左心室质量与对照组相比倍增。左心室功能的下降伴有左心室扩张和心肌纤维化,并且大约一半的TAC处理过的小鼠在第11周时或之前发展成肺充血。因而,TAC是在实验场所中快速产生心脏肥大的有效刺激。TAC模型会为鉴别心脏病的重要治疗靶标和探究分子或药理学抑制剂的作用提供巨大实用性。
本发明表明,就使用靶向TRPV1的治疗剂的针对心脏肥大、纤维化和心力衰竭的保护疗法而言,TRPV1功能是一种新靶标,本发明具有通过开拓现有药物的新用途来改变心脏肥大和心力衰竭的临床治疗范例的潜力。
如下面在实施例中所示,小鼠的TRPV1功能的缺失会改变心脏对TAC诱导的压力负荷过度的应答。响应于压力负荷过度,TRPV1会促进心脏肥大、纤维化、细胞凋亡和收缩功能的缺失。本发明的方法意外地提供了以前已知被用作抗痛觉过敏剂的TRPV1拮抗剂来作为抗肥大剂。
如在实施例中所示,Trpvl的敲除会显著抑制与模型化的压力负荷过度性心脏肥大有关的心室扩大、细胞凋亡、组织重塑和纤维化。该表型会反映抗肥大治疗的一些最希望的作用。通过使用横主动脉狭窄建模,缺乏功能性TRPV1的压力负荷过度性心脏肥大小鼠与野生型小鼠相比具有改善的心脏功能、和减少的肥大性纤维化和细胞凋亡标志物。TRPV1在心脏肥大的进展中起作用,并代表下述目的的治疗靶标:治疗心脏肥大和继发的疾病状态,包括心律失常、肾功能障碍和心力衰竭;治疗和减轻导致心脏肥大和心力衰竭的症状,诸如高血压、心脏瓣膜疾病、心肌无力(心肌病)、心搏异常、贫血、甲状腺障碍、用药过度、肌营养不良和法布里病、主动脉瓣狭窄、导致中毒性心肌病的化疗剂副作用、肥胖、糖尿病、抽烟、病毒性心肌炎(心肌感染)、肌肉渗入(infiltrations of
the muscle)诸如淀粉样变性、HIV心肌病(由人免疫缺陷病毒造成)、结缔组织疾病诸如系统性红斑狼疮、诸如酒精和可卡因等药物的滥用、以及与心律失常或药物(诸如化学治疗剂)有关的副作用。

试验模型
小鼠模型的建立
横主动脉狭窄 (TAC) 。如Rockman所述,执行横主动脉狭窄,通过主动脉的收缩造成左心室肥大(Rockman等人, 1994; Rockman等人,
1991)。用Nair将胸部左侧脱毛,并得到基线2-D超声心动图。然后用氯胺酮和赛拉嗪的混合物深度麻醉小鼠。使用6-0结扎丝线(围绕血管)和26G钝针头绑扎在头臂动脉和左颈动脉之间的横主动脉,此后抽出针头。除了主动脉的收缩以外,假手术组相同。
检查成功的绑扎
多普勒回声心动描记术。在TAC后1周执行多普勒回声心动描记术,以测量收缩水平。用异氟烷气体轻度麻醉小鼠,并剃毛。使用Visualsonics Vevo 770系统进行多普勒回声心动描记术。在胸骨旁短轴切面中,将脉冲多普勒样品体积放入横主动脉中,刚好在绑扎部位的近端和远端。使用Vevo
770软件跟踪峰速度,并使用简化的伯努利方程计算压力梯度。
跟踪在模型化疾病进展过程中心脏尺寸的结构变化
经胸超声心动描记术。使用基线和TAC后经胸超声心动描记术来评估小鼠心脏尺寸和功能的变化。简而言之,在习服2天和脱毛以后,使用30-Mhz传感器(Vevo 770,
VisualSonics)执行未麻醉的经胸超声心动描记术。记录左心室的高质量二维图像和M-模式图像。通过美国超声心动描记术学会采用的前缘至前缘约定,测量左心室舒张末期(LVIDd)和收缩末期(LVIDs)内部尺寸。将左心室射血分数(%EF)计算为(LV Vol; d-LV Vol;s/LV Vol; d x 100) (Visualsonics Inc.)。
测试治疗后的细胞肥大、纤维化和细胞凋亡的程度
通过蛋白质印迹(WB)分析、提取的RNA的实时PCR (RT)、或组织学和免疫组织学分析(H),评估肥大、纤维化、组织重塑、炎症和细胞凋亡的标志物。
肥大标志物可以包括 : ANP (WB 、 RT)、BNP (RT)、ACTA1 (RT)、α-MHC (RT)、β-MHC (RT)、MLC2A
(RT)和麦胚凝集素(H),以产生心肌细胞横截面积。
组织重塑标志物可以包括 : 胰凝乳蛋白酶CMA1 (WB 、 RT)、MMP-2 (RT)、MMP-9
(RT)、TGF-β(RT)、胶原III (RT 、 H)、纤维蛋白原(RT 、 H)、成纤维细胞增殖CD29 (H)。
细胞凋亡标志物可以包括 : 裂解的天冬氨酸特异性半胱氨酸蛋白酶-3 (WB)。
免疫学的、炎症性的和渗入标志物可以包括 : IL-6 (RT)、TNF-α(RT)、NOS3 (RT)、CD68
(巨噬细胞) (WB 、 RT 、 H)、组氨酸脱羧酶(WB)和Fc R1α(WB) (肥大细胞) (WB 、 RT 、 H)、CD4+/CD8+ T-细胞标志物 (WB 、 H)、NK细胞CD161 (RT 、 WB 、 H)。
用于组织学的组织制备 . TAC后8周,通过CO2窒息将小鼠安乐死,并收集心脏用于组织学和分子分析。就组织学而言,用磷酸盐缓冲盐水和10% 福尔马林原位灌注心脏,立即收集,并在10%福尔马林中在4℃固定过夜。然后以矢形取向切割组织,包埋在石蜡中,固定在载玻片上,并保存备用。对石蜡包埋的切片染色,用于下述目的:
胶原 : 通过分析天狼星红染色的切片,确定胶原容积分数。将切成5 μm厚的切片脱石蜡,用Weigert氏苏木精染色,然后用天狼星红(0.1%在苦味酸中的天狼星红)染色。随后对切片进行洗涤和脱水,然后进行图像分析。
心肌细胞横截面积:将心脏切片脱石蜡,并进行透化处理,然后用50 μg/mL浓度的与Alexa488缀合的麦胚凝集素(WGA-Alexa488, Invitrogen, W11261)染色,以鉴别肌纤维膜和测量心肌细胞横截面积(下述)。
图像收集和分析 . 在落射荧光显微镜(Axioscope,
Zeiss)上收集荧光图像和明视野图像。使用ImageJ软件(NIH)定量纤维化和心肌细胞横截面积。为了定量纤维化,将胶原纤维突出显示,并计数染成红色的像素,以确定在每个视野中代表胶原纤维的像素的百分比。从该计算中排除血管周围组织。以每个心脏5副图像,获取得自每只动物的3个心脏切片的图像。取每只动物的图像的平均值,并在Prism
GraphPad中绘图。以不知情方式随机选择得自WGA染色的切片的心肌细胞,然后跟踪以确定单个肌细胞(n=100)的横截面积。
以单盲方式捕获和分析所有图像,但是WGA染色除外,其以双盲方式进行分析。
RT-PCR. 为了提取RNA,从小鼠收集心脏,并用Trizol(Molecular
Research Center, TR 118)从匀浆化的心脏分离总RNA,并用RNA分离试剂盒(Mo Bio
Laboratories, Inc, 15000-250)进一步纯化。使用cDNA合成试剂盒(Qiagen, 205113),从1ug总RNA合成单链cDNA。在Opticon装置(MJ Research,
Waltham, MA)中用QuantiTect SYBR Green (Qiagen, 204245)通过RT-PCR一式三份地定量胰凝乳蛋白酶(CMA1)、心房钠尿肽(ANP)、TGF-、胶原III、基质金属蛋白酶(MMP) 2和9、以及亲环蛋白(CPN)

Max 340)在562nm测量吸光度,并使用基于牛血清白蛋白(BSA)蛋白标准品的标准曲线确定浓度。将浓度标准化至30 μg,并通过十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分离样品。将蛋白样品转移至聚偏氟乙烯(PVDF,
Millipore, IPFL00010)膜在1.4安培保持3.5小时。用针对裂解的天冬氨酸特异性半胱氨酸蛋白酶-3
(Cell Signaling, 9661S)、CMA1 (Gene Tex,

在本文的方法和组合物中可以使用任何合适的TRPV1抑制剂或抑制剂组合。例如,可以用TRVP1选择性抑制剂和非选择性TRPV1抑制剂治疗受试者。
TRPV1抑制剂包括、但不限于:

本发明提供了药物组合物,其含有TRPV1抑制剂或TRPV1抑制剂的混合物。抑制剂可以是下述形式:与常见的药学上采用的载体、稀释剂等相组合的药学上可接受的盐或药学上可接受的溶剂化物。
药物组合物可以是通常采用的形式,例如片剂、胶囊剂、散剂、糖浆剂、溶液、混悬液等,可以在适合的固体或液体载体或稀释剂中或者在适合的无菌介质中含有矫味剂、甜味剂等,以形成可注射的溶液或混悬液。这类组合物通常含有1-25重量%、优选1-15重量%的活性化合物,组合物的其余部分是药学上可接受的载体、稀释剂、赋形剂或溶剂。
适合的药学上可接受的载体包括固体填充剂或稀释剂和无菌的水性或有机溶液。活性化合物在这类药物组合物中的含量将足以提供在上文所述范围内的所需剂量。因而,就口服给药而言,所述化合物可以与适合的固体或液体载体或稀释剂组合,以形成胶囊剂、片剂、散剂、糖浆剂、溶液、混悬液等。如果需要的话,药物组合物可以含有其它组分,例如矫味剂、甜味剂、赋形剂等。可以使用在芝麻油或花生油、含水丙二醇等中的药学上可接受的溶液,以及所述化合物的水溶性药学上可接受的酸加成盐或者碱金属或碱土金属盐的水溶液。以这种方式制备的可注射溶液然后可以通过静脉内、腹膜内、皮下或肌内途径给药,在人类中优选肌内给药。
通过在动物模型中的试验证实本发明的药物组合物是有效的。本发明的药物组合物因而可有效地用于治疗哺乳动物受试者的心脏肥大,包括心脏重塑、心脏纤维化、细胞凋亡、高血压或心力衰竭。也可以施用所述组合物用于哺乳动物受试者的心脏肥大的预防性处理。
一般而言,有效治疗患者的特定病症的剂量可以由医师在治疗期间容易地确定和调节,以减轻该病症或疾病的症状或适应症。一般而言,就为了获得有效结果的给药而言,在约0.01-1000
mg/kg体重的范围内的活性化合物(抑制剂)的每日剂量是适当的。每日剂量可以在单次剂量中或者分成若干剂量施用。在有些情况下,依赖于个体反应,可能需要上调或下调最初处方的每日剂量。典型的药物制剂通常每剂含有约0.2mg至约500mg的式I活性化合物和/或其药学活性盐或溶剂化物。
术语“治疗有效量”、“药学有效量”或“有效量”表示,当给需要这类治疗的哺乳动物单独地或者与其它治疗联合地施用式I化合物或式I化合物的混合物时,足以实现如下定义的治疗的量。本文使用的术语“哺乳动物”意图包括所有哺乳动物,特别是人类。这类哺乳动物在本文中也被称为需要治疗的受试者或患者。治疗有效量将随所治疗的受试者与疾病情况、受试者的体重与年龄、疾病的严重程度、所要采用的给药方案、给药的时机、给药方式等而变化,所有这些都可以由本领域普通技术人员容易地确定。
术语“治疗”是指对哺乳动物疾病的任何处理,包括:
a) 预防所述疾病或病症,也就是说,使所述疾病的临床症状不进展;
b) 抑制所述疾病,也就是说,减慢或阻止临床症状的进展;和/或
c) 消除所述疾病,也就是说,造成临床症状的消退。
下面给出的实施例详细解释了本发明,所述实施例仅仅通过例证来提供,因此不应当解释为限制本发明的范围。

Trpv1
敲除会抑制压力负荷过度性心脏肥大
在8周龄雄性TRPV1敲除的小鼠(Caterina,
2000)和野生型对照组中,通过横主动脉狭窄(TAC)建立压力负荷过度性心脏肥大模型。假对照小鼠接受除了主动脉狭窄以外的相同操作。通过多普勒回波分析,评估在TAC绑扎部位近端和远端的基线压力。在Trpv1-/- 动物和对照动物之间没有显著差异。使用具有30MHz传感器的高分辨率Vevo
770™Echo系统(Visual

Trpv1

从提取的心脏裂解物和切片分析了多种肥大标志物。总体上,在用压力负荷过度性心脏肥大模型化的Trpv1-/- 小鼠中的主要肥大性指示物如胶原(图2)、心房钠尿肽和TGFβ(图3A、3B)表现出表达的显著下降达6周。在图2中,显示了TRPV1缺陷型和C57BU6 WT对照中的天狼星红染色的定量,其在Image J中进行,不包括血管周围组织。对每个心脏进行一式三份的染色。使用Image
J来分析得自每个心脏(x3)的5个图像,并确定每个视野中的像素计数为总像素数目的百分比,用于计算染成红色的胶原/纤维:总组织面积比率。取每只动物的图像的平均值,并在Prism GraphPad中绘图;在绑扎的TRPV1和绑扎的C57BU6之间,p<0.001。在图3A中显示了在C5781/6 (n=7)和TRPV1-/-(n=4)中的TGF-βRNA表达(p<0.0328)。在图3B中显示了在C5781/6 (n=15)和TRPV1(n=14)中的ANP表达(p<0.0431)。用Trizol (Molecular Research Center)从匀浆化的心脏中分离出总RNA,并用RNA分离试剂盒(Mo Bio Laboratories, Inc)进一步纯化。使用cDNA合成试剂盒(Qiagen),从1 μg总RNA合成单链eDNA。使用SYBR qreen方法,通过RT-PCR定量mRNA水平。

细胞外基质重塑
心脏组织组成在心室肥大发展过程中变化,并导致心肌的结构重塑。这些变化之一与胶原的合成和降解之间的平衡的破坏有关,所述破坏导致胶原I型和III型纤维在心肌内的过度堆积。随着胶原和其它细胞外基质组分在间质间隙中的堆积,心肌僵硬度增加,并发生心脏舒张和心脏收缩功能障碍。先前的数据表明,在压力负荷过度性心脏肥大情况下,在Trpvr1-/- 小鼠中存在比对照小鼠更少的间质胶原沉积(Buckley 2011)。通过胶原蛋白测定(SircolTM, Biocolor, Northern Island)和RealTime-PCR,得到了类似的结果。也观察到负责降解胶原的酶——基质金属蛋白酶(MMP)的变化(图4A、4B)。
在图4A中显示了用MMP2通过RT-PCR测得的假手术的小鼠相对于TAC处理过的小鼠(p<0.05)的RNA表达变化:C5781/6 (n=15)和Trpv1-/- (n=14),在图4B中显示了用MMP13进行RT-PCR测得的结果。通常,在TAC后6周,在Trpv1-/- 小鼠中观察到比对照小鼠更多的Mmp和 Timp转录抑制。但是,MMP13表现出增量调节。MMP13靶向胶原I型、II型和III型,且可以用于保护组织免于纤维化。在TAC后8周,在Trpv1-/- 小鼠中比在对照小鼠中表达更少的肥大细胞胰凝乳蛋白酶(CMA 1) 信使和蛋白(图7D)。CMA1是一种属于肽酶家族S1的糜蛋白酶丝氨酸蛋白酶。据称其在肥大细胞中表达,但是似乎在其它组织和细胞类型中表达。它在细胞外基质的降解和血管活性肽的产生中发挥功能。在心脏和血管中,该蛋白,而不是血管紧张素转化酶(ACE),主要负责将肾素-血管紧张素系统中的血管紧张素I转化成血管活性肽血管紧张素II。该系统控制血压,且参与高血压、心脏肥大和心力衰竭的发病机制。

TRPV1
在心脏肥大进展中的涉入
将缺乏功能性TRPV1的小鼠和具有野生型TRPV1的对照小鼠用于压力负荷过度性心脏肥大建模。使用未麻醉的经胸超声心动描记术,随着时间经过测量和对比心脏尺寸和功能,并在8周后收获心脏,用于分子、生化和组织学分析。在缺乏功能性TRPV1的小鼠中更好地保留了心脏尺寸和功能。这些小鼠中的细胞肥大、肥大标志物、纤维化和细胞凋亡也显著减少,从而指示TRPV1在心脏肥大进展中的涉入。
压力负荷过度模型
为了试验TRPV1在与心脏肥大和心力衰竭有关的重塑中的涉入,使10周龄雄性B6.129X1-Trpv1tm1Jul /J小鼠(TRPV1 KO)(Caterina, 2000)和年龄/性别匹配的C57BL/6J (WT) 对照小鼠遭受由横主动脉狭窄(TAC)引起的急性压力负荷过度。得自两种品系的假手术对照小鼠接受除了实际的主动脉狭窄以外的相同外科手术。通过多普勒回声心动描记术在TAC绑扎部位的远端紧邻处评估,TRPV1 KO TAC小鼠和WT

TRPV1

通过经胸超声心动图分析,分析在TAC以后8周的舒张末期左心室内径(LVIDd)。在WT TAC小鼠中,LVIDd在2周时开始增加,并在大约6周时达到平稳态。TRPV1 KO TAC小鼠在6周之前没有表现出LVIDd的变化。图5C显示了在WT (n = 6)和TR
PV1 KO小鼠(n = 8)中0-8周的舒张末期左心室内径(LVIDd)分析。TAC
WT对照小鼠在2周时开始增加它们的内径,而在TAC

TRPV1

通过用荧光地标记的麦胚凝集素(WGA)对质膜染色,检查细胞肥大的程度。通过成像和计算机辅助的心肌细胞横截面积测量,对比细胞大小。该对比会反映样品之间的细胞肥大程度(Shiojima,
2005)。数据表明,与TRPV1 KO TAC小鼠相比,WT TAC的心肌细胞横截面积的显著增加(图6A)。在TAC处理后8周,在TAC WT小鼠和TAC TR PV1 KO小鼠之间,心肌细胞横截面积测量结果是显著不同的(p =

WT小鼠中的心房钠尿肽(ANP)和TGFβ转录物的表达水平显著大于TRPV1 KO小鼠(p =

用TRPV1拮抗剂BCTC治疗的小鼠在压力负荷过度性心脏肥大过程中保留心脏质量、结构和功能
下述试验表明,在暴露于TAC的WT小鼠中使用渗透泵连续施用TRPV1拮抗剂BCTC的治疗,证实了得自先前实施例的试验的发现。
渗透泵安装 . 通过插入渗透泵(不需要重复注射),可以实现药物的长期(多达42天)输注。在外科手术之前10分钟,将小鼠在低水平的麻醉(通过腹膜内(IP)注射氯胺酮/赛拉嗪麻醉剂(50mg/10mg.Kg))下放置。将肩胛骨之间的小区域剃毛,并用Povidine拭子消毒。在该区域切一个小切口,并用在皮肤下面钝性切割,以允许在小鼠不可触及的肩胛骨之间的皮肤下面放置Alzet渗透泵(2006型),所述渗透泵预先装载了选择的药物。用缝线缝几针来封闭切口。将小鼠放入常规笼子中在温暖的垫子上直到完全有意识,此后再将小鼠返回常规圈养室。
使小鼠遭受通过TAC引起的压力负荷过度诱发的心脏肥大,同时施用4mg/kg
BCTC (在20%重量/体积的2-羟丙基-β-环糊精/PBS中),其在整个实验期间使用渗透泵(Alzet, Model
2006)以0.15ul/小时的速率连续泵入。通过泵的功能,将试验限制在约42天(最大值),这样在TAC后36天停止实验,因为泵在TAC之前安装以允许在TAC外科手术之前恢复。在TAC后36天的心脏重量分析揭示,与药物治疗的小鼠相比,在媒介物治疗的WT小鼠中的心脏重量/体重比率明显更大(p = 0.035) (图8A)。在研究期间每9天对小鼠进行的超声心动图评估表明,媒介物和BCTC治疗的假手术小鼠(媒介物假手术(n=2)和BCTC假手术小鼠(n=2))没有表现出左心室内径(LVID,d)的差异(图8B)。但是,BCTC治疗的TAC小鼠(n=8)在0-36天具有与媒介物治疗的TAC小鼠(n=7)相比显著更小的LVIDd (图8C),这指示,TAC媒介物对照小鼠在9天时开始增加它们的内径,而在TAC BCTC治疗的小鼠中,所述直径在试验期间得到维持。在18天以后,LVIDd显著不同(p<0.001)。这种免于左心室扩张的保护反映为通过射血分数(%EF,
