本发明涉及一种HBV持续性感染和纤维化小鼠模型的建立,本发明使用重组病毒AAV2/8‐HBV1.2尾静脉注射C57BL/6小鼠,重组病毒在小鼠肝脏中形成持续性感染,并造成肝脏的纤维化。该方法可以介导HBV的基因组在小鼠肝脏中复制、转录和表达,血清中能检测到HBV的病毒颗粒,模型小鼠不产生急性炎症反应,但在肝脏组织中产生脂肪变性和纤维化的生成,重组病毒的包装信号是AAV2的末端重复序列,包装病毒的血清型可以是对小鼠肝脏嗜性强的AAV6,7,8,9,被包装的HBV基因型可以是A、B、C、D、E、F、G亚型,大小为1.2倍的基因组。

技术领域:
本发明属于生物技术发明领域。本发明涉及用重组AAV病毒介导HBV1.2倍基因组建立HBV持续性感染和肝脏纤维化小鼠模型,用于医学实验。
背景技术:
乙型肝炎病毒:乙型肝炎病毒属于嗜肝DNA病毒科,正嗜肝DNA病毒属。完整的病毒粒子为直径42-49nm的球型,由外膜和呈二十面体对称的核衣壳组成,外膜主要成份为脂质双层和蛋白质,核衣壳的主要成份为HBc蛋白。在电镜下观察,HBV可呈现三种不同的形态结构,包括Dane颗粒、小球形颗粒和管形颗粒。完整的球形HBV颗粒为Dane颗粒,含有双层衣壳且感染性很强,直径为42nm。乙型肝炎患者血清中含量最多的是直径为22nm,呈球型或者棒状的小球形颗粒,小球性颗粒不含HBV DNA和多聚酶,主要成份为包膜蛋白。当小球形颗粒成串排列就会形成直径为50-70nm的管型颗粒。小球形颗粒和管形颗粒均不是完整的HBV颗粒,没有传染性,由过剩的HBV衣壳蛋白组成。
慢性乙型肝炎:乙型肝炎是世界上的一种高发传染病,全球感染HBV的患者人数已高达3.5亿。临床上,5%-10%的HBV感染将发展为慢性肝炎(chronic hepatitis,CH),而慢性肝炎又是肝硬化和肝细胞癌(Hepatocellular carcinoma,HCC)发生的高危因素(几率比正常人高100倍),尽管存在有效的HBV疫苗进行预防,但是每年仍有一百万人死于乙型肝炎病毒感染,治疗种类少疗效有限。
HBV的动物模型:乙型肝炎病毒宿主范围窄,并且没有适合的动物模型进行抗病毒药物的筛选与评估,这些都严重制约了慢性乙型肝炎致病机理的研究与新型抗病毒药物的研发。因此建立一种新型的乙型肝炎病毒慢性感染小鼠模型是研究肝脏纤维化、肝细胞癌病理机制及新型药物研发的关键。到目前为止,大猩猩、树鼩、转基因小鼠、高压水注小鼠以及病毒载体介导的转染小鼠是研究的最为广泛的模型。
(1)大猩猩:大猩猩在研究病毒的传播和自然感染史中发挥着重要的作用,可用于HBV致病机制,安全疫苗评价,抗HBV药物评估等方面的研究。人基因组和大猩猩基因组虽然有98%的同源性,但这两个物种仍具有明显的区别,对疾病的形式和结果影响不同。且受限于价格昂贵、伦理学、及其处于濒临灭绝的边缘等原因,使其做为HBV感染的动物模型受到了严重的限制。
(2)树鼩:一种松鼠样的哺乳动物,进化上属于灵长类。有研究表明树鼩能被HBV和HCV感染。体外实验表明,树鼩原代肝细胞能够感染HBV,具有较好的应用价值,但是感染只造成温和而短暂的复制,相关的研究报道也非常有限。因此必须做更多的工作以确定其是否比大猩猩更有优势,是否更适合作为HBV的感染模型。
(3)土拨鼠肝炎病毒和鸭乙型肝炎病毒:也可用于抗病毒药物的评估。在病毒复制及病毒生活周期的研究中,鸭模型系统发挥了重要作用,土拨鼠模型系统也在病毒感染、慢性化以及肝细胞癌发生机制的研究中做出了重要贡献。但是,鸭和土拨鼠尚未建立纯系动物,缺乏标准化,且与人的亲缘关系较远,因此它们的应用和推广也受到一定的限制,都不是十分理想的小动物模型。
(4)转基因小鼠:自1985年第一只能分泌HBsAg的转基因小鼠诞生以来,转基因小鼠越来越受到广大研究者的关注。1995年,Guidotti等构建的HBV转基因小鼠中HBV的复制水平可达到乙肝慢性感染病人中HBV的复制水平。由于转基因小鼠将HBV基因组整合到小鼠染色体基因组之中,使其成为自身的组成部分,因此不能被免疫系统识别,转基因小鼠免疫耐受转基因产物,并不会产生肝脏病变。但是在转基因小鼠体内注入针对HBsAg的细胞毒性T淋巴细胞(CTL),则可以建立急性肝炎小鼠模型,利用这种模型研究者获得了大量与HBV相关的免疫学信息。转基因小鼠的缺点是小鼠的肝细胞没有HBV的受体,转基因小鼠缺乏HBV进入肝细胞的过程,不适合HBV入侵和扩散机制的研究。HBV转基因小鼠体内从未检测到cccDNA,与人HBV感染也显著不同。
(5)HBV-DNA转染小鼠模型:①HBV-DNA高压注射小鼠模型:Yang等为了增加HBV DNA转染效率,将大量质粒通过尾静脉快速注入小鼠体内,注射可复制性HBV克隆后的第一天即可以检测到HBV的复制,血液中HBV复制水平可达 107GE/mL。但是一周后,小鼠产生了特异性抗病毒抗体和CD8+T细胞,导致HBV被清除。这种小鼠模型持续时间短,只能用于研究急性HBV感染,不能用于HBV慢性感染的研究。②重组腺病毒载体转染小鼠模型:将携带HBV1.3倍基因组的腺病毒载体尾静脉注射小鼠,小鼠肝脏内可成功的检测到HBV复制,并在血清中也能检测到病毒的分泌。注射腺病毒后,小鼠体内也出现了针对HBV的T细胞特异性免疫应答,产生了中和性抗体。虽然腺病毒载体转染效率很高,但是随之而来的针对病毒抗原的免疫反应会摧毁转染细胞,细胞毒性强[,严重的制约了腺病毒载体的应用。
(6)人源化肝脏小鼠:一种新型的人鼠嵌合肝脏模型,基因型为uPA+/+-SCID,是尿激酶纤溶酶原激活物(uPA)的转基因小鼠与重症联合免疫缺陷(SCID)小鼠杂交的子代纯合子,出生后移植入人的原代肝细胞,人的肝细胞可整合到小鼠的肝实质中,HBV和HCV都能感染成功移植的小鼠。Dandri研究表明,虽然该模型人的肝细胞所占比例不到15%,但嵌合小鼠对HBV易感,注射HBV慢性携带者的阳性血清可建立HBV的感染,病毒在小鼠体内能复制,感染小鼠的血清中能检测到病毒基因组DNA(4.5-10×108GE/mL),免疫组织化学法在肝组织切片中能检测到HBcAg的表达。另外Meuleman也报道HBV感染这种嵌合小鼠后,可观察到病毒高水平复制长达八十天,同时嵌合小鼠中人的肝细胞内可以观察到明显的细胞病变效应和严重的肝组织损伤,与免疫抑制的慢性HBV感染病人所表现的生理变化非常相似,但是,高活性的原代人肝细胞是建立人鼠嵌合肝模型的关键,而这对于多数实验室来说来源有限,且移植难度大。另外,人源化肝脏小鼠缺乏完整的免疫系统,很难观察到乙肝发展过程中免疫系统和病毒抗原的相互作用。
(7)乙型肝炎病毒慢性感染小鼠模型:小鼠是医学研究领域最为常用的实验动物之一,具有遗传背景清晰、饲养管理方便,生产繁殖快等优点,已广泛的应用于药物筛选实验、毒性实验和安全评价等领域。因此,如果能够利用小鼠建立HBV感染模型将是一条很好的策略,可是小鼠的肝脏不具有HBV的受体,HBV不能直接感染小鼠肝脏,这就需要我们利用一种载体将HBV介导进入小鼠肝脏。
由于HBV的种属、器官和组织的特异性,到目前为止没有一个好的小动物模型用于HBV的致病机制、抗病毒药物的筛查和评估。
腺相关病毒(Adeno-associated virus,AAV):是无致病性人类细小病毒科(parvoviridae)成员之一,病毒基因组为单链DNA,长约为5kb,病毒衣壳为二十面立体对称,直径为25nm,包含两个开放读码框架,左边开放读码框架编码复制蛋白,右边开放读码框架编码结构蛋白。AAV是一种依赖细小病毒(dependovirus),它的感染需要腺病毒等辅助病毒(或者某些单纯疱疹病毒)的共感染,在缺乏辅助病毒时,AAV通过整合到宿主的染色体基因组建立潜伏感染状态。病毒的衣壳蛋白由VP1、VP2和VP3三个亚单位构成,分子量分别为87、73和62kDa。VP3蛋白是衣壳蛋白的主要构成部分,占80-90%,VP2被认为是非必需的[89],VP1亚单位包含一个磷酸化酶(phospholipase A2,PLA2)结构域,被认为与病毒的感染性相关,重组病毒AAV载体的生产只需要基因组两端145bp的末端重复序列和反式提供其他的所需病毒因子。自第一个AAV感染性克隆建立以来,重组AAV(rAAV2)载体被广泛应用到基因治疗和临床前/临床试验,尝试用于多种疾病的治疗。
目前共发现12种AAV基因型和100多种变异体。不同AAV血清型对于脏器及组织的嗜性不同如表1所示。宾夕法尼亚大学Gao等发现不同血清型AAV的主要区别在于衣壳蛋白的差异,并且这种差异性决定各种血清型AAV对于不同组织和细胞的感染效率不同。

肝脏是最早用于基因治疗的器官,多数肝脏疾病的发生是由于单基因或多基因缺陷所致,因此如果可以通过载体将正常的基因导入,实现肝脏疾病的基因水平治疗,不失为一种良好的治疗策略。但是研究发现,肝细胞是一种很难转导 的细胞,所以病毒载体的出现使解决这一难题成为可能。AAV病毒具有无治病性,低免疫原性,能感染分裂细胞和非分裂细胞,可介导外源基因长期表达等优点,被广大研究者用于肝病的治疗。治疗先天性血友病B的因子IX、抑制肿瘤转移的血管生成抑制素、有抗病毒作用的干扰素α和γ、白细胞介素10和12等都作为与肝病有关的目的基因,被用来进行肝病治疗的研究。
在治疗先天性B型血友病I/II期临床试验中,将剂量为1.8×1012vg/kg的rAAV多点肌肉内注射是安全的,但只能产生不到1%的凝血因子IX。即使通过肝靶向性给药的方法来增加hFIX在人血清的水平,在一段时间过后血清中hFIX水平后会降低至基底水平。这可能是由于rAAV衣壳蛋白诱导产生中和抗体所致,即使仅产生1/10的中和抗体都能抑制高剂量rAAV2载体编码目基因,导致药物治疗失败。还有临床研究还发现如果注射2×1012vg/kg高剂量rAAV,将会导致肝脏血清转氨酶增加,对肝脏造成损伤。因此需要不断优化AAV载体,使其更适合于基因治疗的应用。
由于AAV载体具有可介导外源基因长期表达、免疫原性低等特点,因此利用AAV来介导HBV在小鼠肝脏细胞中表达,建立一种慢性感染小鼠模型不失为一个很好的方法。
发明内容:
本发明经过实验发现,使用靶向小鼠肝脏AAV8病毒载体介导HBV1.2倍基因组到小鼠的肝脏建立了HBV持续性感染和纤维化模型,可用于本领域的相关研究。
为此,本发明提供一种HBV持续性感染和纤维化小鼠模型,其特征在于,利用重组病毒AAV2/8-HBV1.2建立HBV持续性感染和纤维化的小鼠模型。
其中,重组病毒AAV2/8‐HBV1.2的包装信号是AAV2的末端重复序列。
其中,重组病毒AAV2/8‐HBV1.2的包装病毒是AAV血清型6,7,8,9。
其中,重组病毒AAV2/8‐HBV1.2被包装的序列是大于或等于HBV1.2倍的基因组。
其中,重组病毒AAV2/8‐HBV1.2的被包装的是HBV基因型D或其他基因型A、B、C、E、F、G。
所建立的小鼠模型是HBV持续性感染模型以及HBV的肝脏纤维化模型。
本发明还包括,建立本发明HBV持续性感染和纤维化小鼠模型的方法,该方法包括如下步骤:
使用重组病毒AAV2/8‐HBV1.2作为载体,其中,AAV2的末端145bp的重复序列,含有病毒复制和包装信号,包装衣壳蛋白是AAV8的衣壳蛋白,被包装的序列是HBV1.2倍的基因组;该重组病毒可按照实施例1方法构建,其结构列在附图1中。其中,重组病毒AAV2/8‐HBV1.2的制备可以采用以下方法:将1.2倍基因组的HBV片段克隆到去除Rep和Cap基因的含有腺相关病毒2型TR结构的pSSV9骨架质粒中,获得质粒载体pAAV-HBV1.2。
上述重组病毒通过三质粒共转染法在293T细胞中包装后,通过氯化铯密度梯度离心纯化后,重组病毒使用荧光定量PCR法检定,然后通过尾静脉注射到小鼠体内(重组病毒2.0x1011vg),即构建成HBV持续性感染和纤维化小鼠模型。其中小鼠优选C57BL/6小鼠。
以上模型建立中所述的三质粒共转染法(Xiao X,JV,1998),293T细胞,氯化铯密度梯度离心,C57BL/6小鼠等均属于现有技术,在现有文献中有所报道。
本发明的模型小鼠可以产生HBV的持续性感染和纤维化,主要特征有,注射后两周血清中可以检测到病毒血症的发生,也可以检测到HbsAg和HbeAg,肝脏组织可以检测到HBV的复制、转录和表达,所有这些特征持续超过6个月,模型小鼠不产生急性炎症反应,但存在肝脏组织的慢性病理变化,包括少量淋巴细胞浸润、脂肪变性、胶原蛋白的累积以及肝脏显微结构的改变,纤维化分子标示显著升高。该模型小鼠没有形成HBV抗原的血清学转化。该模型是典型的HBV持续性感染和纤维化模型,同时也是免疫耐受模型。可以用于研究HBV的复制、清除和致病机制,也可以用于HBV引起的慢性感染和纤维化的药物筛选和评估。
发明详述:
乙型肝炎病毒宿主范围窄,并且没有适合的动物模型进行慢性乙型肝炎致病机理研究和抗病毒药物的评估,严重制约了相关研究。因此,建立一种新型的乙型肝炎病毒慢性感染小鼠模型是研究慢性乙型肝炎的病理机制及新型药物研发的关键。正如前面背景中所述,利用高压注射或者腺病毒可将HBV基因组转移到小鼠肝脏,但是这些方法建立的小鼠模型,HBV基因组在短期内会被清除;HBV转基因小鼠也是一种有潜力的小鼠模型,但HBV基因组整合到宿主的染色体基因 组中,表达的抗原被宿主识别为自身的组成部分,而且不能被清除,限制其在抗病毒药物评价上的应用。为了能克服这些限制,本课题构建一种能有效模拟人慢性乙型肝炎病毒感染的小鼠模型。
为了克服现有小鼠模型的缺陷,我们寻求建立一种小鼠模型,可以有效地模拟人类慢性乙型肝炎。由于小鼠没有HBV的受体,不能被HBV感染,我们利用AAV介导HBV1.2倍的基因组传输到小鼠的肝脏。1.2倍的HBV基因组被证明在小鼠的肝脏细胞和人HepG2细胞株中可以有效复制(Dong Q,Liu Z,J Immunol.2000;Mabit H,Dubanchet S,J Gen Virol.1994)。重组AAV载体由于其低免疫原性、有效转导分裂和非分裂细胞、无致病性并能介导外源基因在体内长期表达,AAV6,8,9,具有高效的肝脏组织嗜性,因此我们使用AAV8介导HBV的基因转移到小鼠的肝脏。首先我们构建AAV的重组质粒AAVHBV1.2,该质粒包含HBV的1.2倍基因组(基因型D,血清型ayw),两端为AAV2的末端重复序列,然后将该质粒与AAV8的辅助质粒pXR8和腺病毒的辅助质粒pXX680共同转染293T细胞(Xiao X,JV,1998),重组病毒产生后,通过细胞裂解,氯化铯超速离心纯化,Q-PCR检定滴度。
由于AAV8体外没有合适的感染细胞株,为了验证pSSV9-1.2HBV是否可以体外表达HBsAg与HBeAg,将重组质粒pSSV9-1.2HBV转染Huh7.5.1细胞,连续7天收获培养上清液,测定培养上清液中HBsAg和HBeAg分泌情况。为了研究AAV8-1.2HBV是否能介导HBV靶向肝细胞转移,尾静脉注射AAV8-1.2HBV到C57/BL6小鼠体内,注射2天和6个月处死模型小鼠,测定各组织中的HBV基因组DNA的分布和变化。由于本发明是将AAV8-1.2HBV病毒尾静脉注射C57BL/6来建模,因此不能确定小鼠血液和肝组织中的HBV是以重组病毒(AAV8-1.2HBV)的形式存在,还是以HBV复制中间体或HBV的病毒颗粒形式存在,为了观察HBV基因组的存在形式,本课题以pSSV9-1.2HBV质粒为模板,设计2对效率相等的引物来定量HBV基因组的拷贝数。第一对引物(TR-primer)的上游引物位于AAV2-TR区,下游引物位于HBV的X区,只能扩增AAV重组病毒基因组;第二对引物(NO-TR-primer)上下游引物均位于HBV的S区,可用于扩增包括AAV载体、HBV复制中间体、已包装的HBV病毒在内的全部HBV基因组。同时用这两对引物检测血清中HBV的基因组,后者得到的数值减前者就是血清中存在的HBV的病毒 颗粒,可以证明是否存在HBV的病毒血症。在肝脏组织中的检测就可以阐明HBV的基因组存在形式。用ELISA的方法测定AAV8-1.2HBV感染小鼠血清中HBsAg和HBeAg的分泌情况。我们进一步用免疫组织化学检测AAV8-1.2HBV感染小鼠模型肝组织中HBsAg和HBcAg的表达,并用Image pro plus软件对免疫组化结果进行定量分析。
为了检测小鼠模型是否引起急性炎症反应,我们检测血清中ALT和AST的浓度变化。为了检测模型小鼠是否引起慢性炎症和其他病理变化,我们对模型小鼠肝组织进行了HE染色和Masson染色,以检测炎症细胞浸润、脂肪变性和胶原蛋白累积引起的肝脏纤维化。为了定量研究模型小鼠肝脏的纤维化,我们用ELISA和RT-qPCR的方法研究了一些在模型小鼠肝脏和血清中纤维化的分子标示的变化,包括蛋白水平和mRNA水平。转化生长因子β(TGF-β)是肝组织纤维化的关键调节因子,TGF-β家族包括TGF-β1,TGF-β2和TGF-β3,其中TGF-β1在促进肝星状细胞向成纤维细胞转化中起着重要的作用,肝组织纤维化过程中,TGF-β1在肝星状细胞和内皮细胞内会显著增加,TGF-β1的增加,同时又会伴随金属蛋白酶组织抑制因子(TIMP-1)的增加,因此TGF-β1和TIMP-1常被用作检测肝组织纤维化的指标。
在本发明中我们通过AAV介导HBV基因组到小鼠的肝脏建立的小鼠模型,典型的特征是HBV的持续性感染并伴随肝脏的纤维化。AAV病毒载体在小鼠肝脏中持续超过6个月,并伴随HBV的复制、转绿和表达。在小鼠的肝脏中可检测到病毒的转录和复制中间体,但cccDNA未检测到,HBV的病毒血症在注射后的1个月开始检测到,载量达到109HBV-DNA copies/ml,然后缓慢下降,在6个月时降低65%左右。肝脏中HBV的基因组超过AAV载体20%以上,证明有HBV的复制,同时也表明HBV的基因组在小鼠肝脏中以染色体外基因组的形式存在,以AAV-HBV的基因组形式为主要存在方式,作为模板转录HBV的RNA前基因组。HBsAg开始在血清中检测到,2个月后达到峰值,此后开始缓慢下降,HBeAg表达更快,1个月就达到峰值,此后没有明显下降,注射后6个月,大概25%的肝脏细胞被转导。模型小鼠不产生明显的急性炎症反应,但有少量的淋巴细胞浸润,同时也产生了明显的脂肪变性和胶原蛋白的累积,纤维化的分子标示检测表明模型小鼠肝脏形成了明显的纤维化。在整个实验过程中小鼠都没有产生anti-HBs 和anti-HBc等中和抗体,提示该小鼠模型是免疫耐受模型。
总之,本发明介绍了一种在免疫功能完善小鼠体内建立的一种HBV持续性感染并伴随肝脏纤维化的模型,和人类HBV引起的慢性乙型肝炎非常相似,小鼠没有形成针对HBV的特异性血清学转换。这种HBV持续性感染和肝脏纤维化的小鼠模型可以用于研究HBV慢性感染过程中形成纤维化的机制,同时也能用于相关的药物筛选和评估。

将1.2倍基因组的HBV片段克隆到去除Rep和Cap基因的含有腺相关病毒2型TR结构的pSSV9骨架质粒中,获得质粒载体pAAV-HBV1.2(图1)。简要操作过程如下,利用限制性内切酶HindⅢ和SacⅠ双酶切质粒plitmus-1.2HBV获得用于插入的目的片段1.2HBV,利用限制性内切酶XbaⅠ和HindⅢ双酶切质粒pSSV9获得线性化质粒载体pSSV9(rep-/cap-),用DNA Polymerase I,Large(Klenow)Fragment将胶回收试剂盒纯化的1.2HBV目的片段和线性化质粒载体pSSV9(rep-/cap-)补平,将补平的目的片段1.2HBV(平末端)与线性化质粒pSSV9(rep-/cap-)(平末端)利用T4DNA连接酶进行连接,转化大肠杆菌,获得重组质粒。用三质粒共转染技术制备AAV8-1.2HBV,转染试剂为PEI,所用质粒为pSSV9-1.2HBV,pXR8,pXX680(Xiao,X,JV,1998),使用氯化铯密度梯度离心纯化,使用Q-PCR法检测制备病毒的滴度。Q-PCR的引物如下:上游引物q-1.2HBV-f,GCGGGCGTTTTATCATCTTCCTC;下游引物q-1.2HBV-r,GAAAGCCCTACGAACCACTGAAC。
实验方案如下:
利用网站http://www.uri.edu/research/gsc/resources/cndna.html计算质粒pSSV9-1.2HBV拷贝数和所需稀释倍数。经过梯度稀释使质粒pSSV9-1.2HBV的浓度分别为103-109copy/μL,每次试验标准曲线和样本均设置3个平行。qPCR反应体系及程序如下:

实施例2.AAV8-1.2HBV体内有效介导HBV基因组的转移、复制与转录
将AAV8-1.2HBV用PBS稀释为1×1012vg/200μL,即:每只C57BL/6小鼠注射体积为200μL病毒,病毒剂量为2×1011vg/只。将成功注射的小鼠,放回鼠笼继续饲养,饲养条件为25℃,湿度30%-60%,每天给予12h光照,12小时黑暗。到所需时间点取血,或者处死模型小鼠取所需组织。AAV8-1.2HBV小鼠模型建模2day,1month,2month,3month,4month,5month,6month对模型小鼠取血。采集AAV8-1.2HBV小鼠模型组织样本,(1)当模型小鼠到所需时间点时,处 死小鼠,剪开小鼠腹腔,找到下腔静脉,并剪开下腔静脉,然后打开小鼠胸腔,找到小鼠心脏并用装有预冷PBS的注射器从小鼠左心尖刺入进行PBS灌注,此时可见血液从下腔静脉流出,直到流出的液体变为无色停止灌注;(2)灌注结束后,迅速摘取小鼠的肝、肾、肠、肌肉、胰、心等组织,将摘取的组织放置于冰上预冷的PBS中,洗去表面残余血液;(3)将各组织分装于1.5mL离心管内,-80℃冻存,每次取分装的组织进行后续试验。
使用QIAGEN DNA easy试剂盒提取小鼠模型血浆样本和组织样本中DNA,按实施例1的方法定量,使用两套引物分别定量HBV全基因组和AAV重组病毒基因组,使用的引物如下:

使用RNAⅡ提取试剂盒提取模型小鼠肝组织总RNA,逆转录合成cDNA,按实施例1的方法定量mRNA的含量。
如图2a,建模2天后,90%以上重组病毒基因组分布于肝组织,而其它组织分布总量不足10%。同理建模6个月,检测各组织HBV DNA含量,肝组织内HBV DNA含量增加40%,其它组织内HBV DNA显著降低,进一步说明AAV8-1.2HBV可介导HBV基因组向肝组织有效转移。因为6个月的时候肝组织内HBV DNA含量增加40%,我们推测肝组织有HBV DNA的复制。
然后,我们测量尾静脉注射AAV8-1.2HBV建模6个月内小鼠血清中HBV基因组的拷贝数,结果如图2b所示,建模2周,血清中病毒基因组含量达到峰值, 伴随着病毒的清除,血清中病毒基因组含量逐渐降低,2个月以后进入稳定期。2周内,两对引物定量血清中HBV基因组含量没有差别,表明此时HBV基因组是以AAV重组病毒基因组的形式存在,但是2周以后两对引物测量值呈现2-5倍的区别,这说明血清中有HBV病毒的分泌。然后我们又用这两对引物测定了模型小鼠肝组织中HBV基因组的含量,结果如图2c所示,整个试验阶段,模型小鼠肝组织内全部HBV基因组含量要显著高于重组病毒载体(AAV8-1.2HBV)的含量,并且随着时间的推移,呈增加的趋势,这也表明模型小鼠肝组织内有HBV的复制。以上数据证明了注射重组病毒(AAV8-1.2HBV)可以导致病毒血症的发生。
随后我们通过RT-qPCR技术又测定了模型小鼠肝组织中HBV cDNA含量,来观察HBV RNAs的转录情况,结果如图2d所示,建模1个月肝组织内HBV cDNA含量只有3.65×107copy/g,但是3个月的时候HBV cDNA含量呈现200倍的增长,当到达6个月的时候达到1.01×1010copy/g,说明模型小鼠肝组织内有明显的HBVRNAs的转录,AAV载体能有效介导HBV基因组的转录。
综上所述,AAV8-1.2HBV能有效的介导HBV基因组向肝脏转移,并能介导HBV基因组在小鼠体内复制与转录。
实施例3AAV8-1.2HBV能有效介导HBV基因在小鼠体内的表达
虽然AAV8-1.2HBV感染小鼠能检测到病毒复制和病毒血症的发生,但是不足以达到成为一个良好的HBV持续性感染小鼠模型的标准。因此我们又用ELISA的方法测定了AAV8-1.2HBV感染小鼠血清中HBsAg和HBeAg的分泌情况,使用免疫组织化学的方法检测肝组织中HBsAg和HBcAg的表达情况。实验方案可以参照经典的ELISA和免疫组化方案(略)。
我们的结果显示,AAV8-1.2HBV注射C57BL/6小鼠能导致小鼠持续性感染,6个月内模型小鼠血清中均有HBsAg和HBeAg的分泌,模型小鼠肝组织均有HBsAg和HBcAg的表达,且6个月内未检测到HBsAg血清转学转换。
如图3a所示,建模2周即可在血清中检测到HBsAg的表达,2个月达到高峰(4.85×102ng/mL),2个月以后,小鼠血清中HBsAg缓慢降低,直到6个月血清中HBsAg含量均维持在4.0×102ng/mL左右。如图3b所示,血清中HBeAg变化趋势与HBsAg变化趋势相似,只是模型小鼠血清中HBeAg在1个月即达到高峰。虽然在6个月内血清中HBsAg含量相对稳定。
进一步免疫组织化学检测AAV8-1.2HBV感染小鼠模型肝组织中HBsAg和HBcAg的表达,并用Image pro plus软件对免疫组化结果进行定量分析。如图3c所示,1个月HBsAg阳性的肝细胞随机分布,大多数区域未能被染色,只有少量的肝细胞胞质淡染,但是随着时间推移,HBsAg表达逐渐增多,6个月转导效率可达25%,这有可能是模型小鼠肝组织HBsAg表达积累的结果。
同样检测HBcAg在模型小鼠肝组织中表达,结果如图3c所示,1个月就可明显观察到HBcAg分布于肝组织表面,3个月显著增加,随后直到6个月HBcAg表达基本稳定,定量分析显示3个月到6个月模型小鼠HBcAg转导肝细胞百分比没有显著变化(如图3d所示)。同时对照组小鼠免疫组织化学分析未见HBsAg和HBcAg在肝组织有表达。这些结果都证明AAV8-1.2HBV可以有效的介导HBV基因在小鼠体内的高效表达。
实施例4.模型小鼠肝脏损伤的检测。
为检测模型小鼠,是否引起急性炎症反应,我们使用ELISA法检测血清中ALT和AST的浓度变化。结果显示,与正常小鼠相比较,注射AAV8-1.2HBV不会导致模型小鼠血清中ALT增加(图4a),但是血清中AST浓度会在注射1个月和2个月的时候,有略微提升(图4b)。观察AAV8-1.2HBV模型小鼠未见其他的全身毒性症状(未显示数据)。建模1个月和2个月AST的略微升高可能是由HBV基因组的体内表达所致。该数据表明模型小鼠不产生明显的急性炎症反应。
为了进一步研究小鼠肝脏的病理变化,我们对模型小鼠肝组织进行了HE染色。通过对小鼠肝组织HE染色观察,与正常小鼠对比,建模1个月模型小鼠肝细胞没有显著的病理损伤,只有个别炎症灶的出现(见图4c),但是建模6个月后可明显观察到多处出现炎症灶的聚集,具有炎性细胞浸润,而且出现较多的空泡,说明小鼠肝脏中出现了脂肪变性(图4c)。进一步说明了,本课题构建的小鼠模型不会导致急性肝炎,但是会导致模型小鼠肝组织的慢性炎症和脂肪变性。
人乙型肝炎病毒的慢性感染会导致肝组织纤维化,最终发展为肝硬化或者肝细胞癌。HE染色的结果表明了模型小鼠肝细胞产生了脂肪变性,为了验证AAV8-1.2HBV注射小鼠是否与人乙型肝炎病毒慢性感染相似,能诱导肝组织的纤维化。我们对小鼠肝组织进行Masson染色初步观察,并用Image pro plus软件对Masson染色结果进行定量分析。观察Masson染色结果,与对照小鼠相比,建 模1个月小鼠肝组织只有少量胶原纤维增生(图4c),定量结果显示(图4d)1个月模型小鼠肝组织胶原纤维含量与正常小鼠没有显著区别。但是建模3个月和6个月肝组织可观察到大量蓝染胶原纤维的堆积(图4c和d),定量结果显示与对照小鼠相比,3个月和6个月肝组织胶原纤维增加量分别为16.65%和37.23%,统计学分析表明差别显著(P<0.001)。结果均说明肝组织有明显胶原纤维增生。
实施例5模型小鼠肝脏纤维化分子标示显著升高
为了定量研究模型小鼠肝脏的纤维化,我们用ELISA和RT-QPCR的方法研究了一些在模型小鼠肝脏和血清中纤维化的分子指标变化。
ELISA法参照试剂盒说明书,RT-QPCR参照实施例1的方法,不同的是所用引物,见下表:

肝组织纤维化最主要的改变就是细胞外基质量的增加和细胞外基质成分的改变,所以我们用ELISA法直接测量了血清和组织中Ⅰ型胶原纤维和Ⅲ型胶原纤维的变化情况。结果如图5所示,建模6个月内小鼠血清中ColⅠ含量(图5a)和ColⅢ(图5b)含量均高于正常小鼠,模型小鼠血清中ColⅠ含量在建模1个 月以后,浓度保持稳定,ColⅢ含量在建模1个月以后,呈先上升后下降的趋势。说明纤维化初期主要以ColⅠ和ColⅢ为主,而随着纤维化的进程,主要以ColⅠ为主,ColⅢ含量会减少,与人肝组织纤维化进程相似。肝组织内ColⅠ和ColⅢ检测显示(见图5c,d),整个试验阶段,Ⅰ型胶原和Ⅲ型胶原含量均高于正常小鼠,与血清检测结果一致。
转化生长因子β(TGF-β)是肝组织纤维化的关键调节因子,TGF-β家族包括TGF-β1,TGF-β2和TGF-β3,其中TGF-β1在促进肝星状细胞向成纤维细胞转化中起着重要的作用,肝组织纤维化过程中,TGF-β1在肝星状细胞和内皮细胞内会显著增加,TGF-β1的增加,同时又会伴随金属蛋白酶组织抑制因子(TIMP-1)的增加,因此TGF-β1和TIMP-1常被用作检测肝组织纤维化的指标。我们也测定了AAV8-1.2HBV注射小鼠的TGF-β1和TIMP-1的变化。
ELISA方法检测AAV8-1.2HBV注射小鼠血清中TGF-β1和TIMP-1变化,结果显示(见图5),建模2周模型小鼠血清中即可观察到TGF-β1(图5e)和TIMP-1(图5f)的增加,1个月血清中TGF-β1和TIMP-1浓度达到峰值,1个月后血清中TGF-β1和TIMP-1浓度稳定分别维持在126ng/mL和19ng/mL,与对照小鼠相比,TGF-β1和TIMP-1浓度分别增加了40%和38.46%,差异显著(P<0.05)。
同时我们利用RT-qPCR的方法测定了AAV8-1.2HBV注射小鼠肝组织内TGF-β1和TIMP-1mRNA的相对表达量,结果如图5所示,与对照小鼠相比,肝组织中TGF-β1(图5g)在建模1个月就有9倍的增加,6个月时TGF-β1mRNA相对表达量是对照的27倍。1个月的时候TIMP-1mRNA未见明显增加,但是6个月TIMP-1mRNA能检测到2倍增加(图5h)。结果与血清中检测一致,肝组织纤维化间接指标均显示AAV8-1.2HBV小鼠能诱导肝组织纤维化的发生。
Claims (9)
1.一种建立HBV持续性感染和纤维化小鼠模型的方法,其特征在于,利用重组病毒AAV2/8‐HBV1.2建立的,包括如下步骤:使用重组病毒AAV2/8‐HBV1.2作为载体,其中,AAV2的末端145bp的重复序列,含有病毒复制和包装信号,包装衣壳蛋白是AAV8的衣壳蛋白,被包装的序列是HBV 1.2倍的基因组;上述重组病毒通过三质粒共转染法在293T细胞中包装后,通过氯化铯密度梯度离心纯化后,然后通过尾静脉注射到小鼠体内,构建成HBV持续性感染和纤维化小鼠模型,其中,用三质粒共转染法制备AAV8-1.2HBV:转染试剂为PEI,所用质粒为pSSV9-1.2HBV,pXR8,pXX680,使用氯化铯密度梯度离心纯化,使用Q-PCR法检测制备病毒的滴度,Q-PCR的引物如下:上游引物q-1.2HBV-f,GCGGGCGTTTTATCATCTTCCTC;下游引物q-1.2HBV-r,GAAAGCCCTACGAACCACTGAAC。

6.根据权利要求1的小鼠模型的建立方法,其特征在于,其中小鼠为C57BL/6小鼠。
7.根据权利要求1的小鼠模型的建立方法,其特征在于,其中,重组病毒AAV2/8‐HBV1.2的制备方法如下:将1.2倍基因组的HBV片段克隆到去除Rep和Cap基因的含有腺相关病毒2型TR结构的pSSV9骨架质粒中,获得质粒载体pAAV-HBV1.2。
8.根据权利要求1的小鼠模型的建立方法,其特征在于,其中,尾静脉注射到小鼠体内的重组病毒的量为2.0x1011vg。
9.根据权利要求1的小鼠模型的建立方法,其特征在于,注射后两周血清中可以检测到病毒血症的发生,也可以检测到HbsAg和HbeAg,肝脏组织可以检测到HBV的复制、转录和表达,模型小鼠不产生急性炎症反应,但存在肝脏组织的慢性病理变化,包括少量淋巴细胞浸润、脂肪变性、胶原蛋白的累积以及肝脏显微结构的改变,纤维化分子标示显著升高,该模型小鼠没有形成HBV抗原的血清学转化。
