背景技术
免疫系统通过识别和杀灭病原体(例如细菌、病毒和寄生虫)以及癌细胞来保护有机体抵御疾病。为了识别和中和病原体,进化出多种机制。典型的脊椎动物免疫系统包括数条提高特异性的防御途径。最简单地,物理屏障防止病原体进入有机体。如果病原体破坏这些屏障,则先天免疫系统提供直接、但非特异性的应答。如果病原体成功逃避先天免疫应答,脊椎动物拥有第三层保护——通过先天免疫应答活化的获得性免疫系统。这里免疫系统在侵染过程中调整其应答,以提高其病原体识别。然后,该提高的应答在已清除病原体后以免疫学记忆的形式得以保留,并使获得性免疫系统在每次遇到该病原体时作出更快且更强的反应。所述免疫学记忆也是免疫背后的原则。在本发明的上下文中,尤其关注先天免疫系统。
先天免疫系统起到以非特异性方式保护宿主免于入侵微生物的作用。通过活化,例如通过巨噬细胞、单核细胞、嗜中性粒细胞和树突细胞对细菌、病毒或原生动物的吞噬作用,先天免疫系统引发一系列宿主防御应答(Akira等人,2006.Cell124:783-801)。这些应答的核心机制为产生和/或释放促炎细胞因子、抗微生物防御分子、水解酶、活性氧类别(ROS)或活性氮类别(RNS)。不同的微生物病原体组分,称为病原体相关分子模式(PAMP),被先天免疫系统的模式识别受体(PRR)所识别。这些受体中有Toll样受体(TLR)、NOD样受体(NLR)、RIG样受体(RLR)和肽聚糖样识别蛋白(PGRP),均为针对病原体的早期宿主防御作出贡献(Creagh和O'Neill.2006.Trends Immunol.27:352-357;Dziarski和Gupta.2006.Cell Microbiol.8:1059-1069;Ishii et al.,2008.Cell Host.Microbe3:352-363)。例如,细菌PAMP鞭毛蛋白——鞭毛的单体亚单位——结合于TLR5并活化转录因子NF-κB(Hayashi等人,2001.Nature410:1099-1103),最终导致促炎细胞因子以及包括牛皮癣素(S100A7c)和人β-防卫素2(hBD-2)的抗微生物肽在人原代角质形成细胞(KC)中的表达(Abtin等人,2008.FASEB J.7:2168-2176;Akira和Takeda.2004.Nat.Rev.Immunol.4:499-511;Miller等人,2005.J.Immunol.174:6137-6143)。

因此,在一个方面,本发明涉及菌蜕(BG)提高先天免疫应答的用途。
根据本发明,出人意料地发现菌蜕(BG)的包被结构被KC识别并提高先天免疫应答。特别地,已经发现具有天然或近乎天然包被结构的菌蜕可以提高先天免疫应答。
BG为细菌、特别是革兰氏阴性菌的非活细胞包被制备物,去除了胞质成份。可以通过例如细菌中质粒编码的裂解基因的受控表达、例如通过革兰氏阴性菌中噬菌体φX174的质粒编码的裂解基因E的表达来产生BG。基因E编码膜蛋白,其能够融合内膜和外膜,从而形成E-特异性裂解通道,所有胞质成份均通过该通道排出(Witte等人,1990.Biochimie72:191-200;Witte et al.,1990.J.Bacteriol.172:4109-4114)。作为非活细菌包被的BG保持了天然细菌的全细胞形态学。在BG中保留了所有细胞表面结构,包括外膜蛋白、粘附素、脂多糖(LPS)和肽聚糖(Mayr等人,2005.Adv.Drug Deliv.Rev.57:1381-1391)。
先前已表明,BG能够刺激获得性免疫系统。特别地,BG可以用于针对其自身包被结构的免疫,或者作为外来靶抗原的抗原递送系统使用(Jalava等人,2003.Expert.Rev.Vaccines.2:45-51;Mayr et al.,2005.Adv.Drug Deliv.Rev.57:1381-1391)。在兔动物模型中使用霍乱弧菌(Vibrio cholerae)菌蜕进行免疫,防御以全毒性霍乱弧菌攻击后的腹泻和死亡(Eko等人,2003.Vaccine21:3663-3674),或者在大肠杆菌BG表面上整合乙型肝炎病毒的核心抗原导致小鼠中针对该核心抗原的显著免疫应答(Jechlinger等人,2005.Vaccine23:3609-3617)。BG也可以用作活性物质例如阿霉素的递送载体(Paukner等人,2006.Expert.Opin.Drug Deliv.3:11-22)或者作为DNA(Kudela等人,2007.Cancer Lett.1:54-63)和酶(Huter等人,1999.Journal of ControlledRelease61:51-63.)的载体。
在本发明中,发现菌蜕也增强先天免疫防御系统、特别是皮肤的先天免疫防御系统。因此,菌蜕(BG)用于提高先天免疫应答。
在本发明的优选实施方案中,菌蜕表面不包含鞭毛蛋白单体。
在另一优选实施方案中,菌蜕表面不包含鞭毛蛋白。
鞭毛蛋白为约30,000至60,000道尔顿的蛋白质,其聚合形成细菌鞭毛的丝。蛋白质鞭毛蛋白为细菌鞭毛的主要成分,大量存在于几乎所有鞭毛细菌中。在哺乳动物中,经常对鞭毛抗原、从而对鞭毛细菌激起获得性免疫应答(即T细胞和抗体应答)。这可能是因为鞭毛蛋白为鞭毛细菌中极丰富的蛋白质并且进一步存在识别鞭毛蛋白TLR5的特异性先天免疫受体这一事实。
在表面上不包含鞭毛蛋白的菌蜕可以来自不能够表达鞭毛蛋白的菌株,例如鞭毛蛋白缺失突变体。或者,所述菌蜕可以来自不能够将鞭毛蛋白运送到细胞表面的菌株。为了获得在表面上不包含鞭毛蛋白的菌蜕,可以使用任何合适的本领域已知的基因工程方法。例如,缺失编码鞭毛蛋白蛋白质的基因产生鞭毛蛋白缺失突变体。或者,可以通过任何本领域合适的方法,例如使用RNAi,抑制鞭毛蛋白的表达。此外,可以将突变——例如预成熟终止密码子——引入编码鞭毛蛋白的基因中,导致表达截短的蛋白质。缺失和/或突变鞭毛蛋白基因的表达调控序列也可以导致所述基因功能的丧失,因此可以产生不能够表达鞭毛蛋白的菌株。还可以通过例如缺失和/或突变鞭毛蛋白基因的各序列也可以获得不能够将鞭毛蛋白输送到细胞表面的突变体菌株。所述技术对于本领域技术人员而言是熟知的。
知晓鞭毛蛋白为对于刺激先天免疫应答以及获得性免疫应答重要的细菌组分,更加出人意料的是去除鞭毛蛋白的BG也对于提高先天免疫应答有效。
优选通过诱导至少一种先天免疫调节子的表达和/或释放提高先天免疫应答。
免疫调节子在例如免疫强化、免疫抑制或诱导免疫耐受中参与将免疫应答调节到所需的水平。免疫强化,即增强免疫应答,根据本发明是需要的。可以通过提高免疫应答发展的速度和程度以及延长免疫应答持续时间来增强免疫应答。
先天免疫调节子是免疫调节子,如前所定义,其参与先天免疫应答。先天免疫调节子的实例包括促炎细胞因子、活性氮类别(RNS)和活性氧类别(ROS)。
根据本发明,优选选自抗微生物防御分子、促炎细胞因子、活性氮类别(RNS)和活性氧类别(ROS)的至少一种先天免疫调节子的表达和/或释放被诱导。
抗微生物防御分子,也已知作为抗微生物肽或宿主防御肽,是一类强广谱抗生素。已表明所述分子杀死革兰氏阴性菌和革兰氏阳性菌、分枝杆菌、包膜病毒、真菌、甚至转化的或癌性细胞。不同于大部分常规抗生素,抗微生物肽也可以通过作为免疫调节子起作用而具有增强免疫性的能力。抗微生物防御分子的实例包括牛皮癣素和人β防卫素-2(hBD-2)。
根据本发明,增强了选自牛皮癣素和人β防卫素-2(hBD2-)的至少一种抗微生物防御分子的表达。
促炎细胞因子为提高系统炎症的细胞因子。促炎细胞因子的实例包括TNF-α、白介素-1(IL-1)、白介素-6(IL-6)和白介素-8(IL-8)。
根据本发明,增强选自白介素-6(IL-6)和白介素-8(IL-8)的至少一种促炎细胞因子的表达和/或释放。
活性氮类别(RNS)是衍生自分别通过诱导型一氧化氮合酶(iNOS)和NADPH氧化酶的酶活性产生的一氧化氮(NO)和超氧化物(O2-)的抗微生物分子家族。在经细胞因子和微生物产物、特别是干扰素-γ(IFN-γ)和脂多糖(LPS)诱导后,iNOS主要在巨噬细胞中表达。然而,一氧化氮(NO)的产生不仅仅是吞噬细胞的特征。此外,其它参与免疫反应的细胞,例如上皮细胞或角质形成细胞,也能够产生这些自由基(Bogdan,C.,2001.Nat Immunol2(10):p.907-16;Bogdan,C.,2000ImmunolRev.173:p.17-26;Bogdan et al.,2000Curr Opin Immunol.12(1):p.64-76)。

活性氮类别与活性氧类别(ROS)一起发挥作用而损伤细胞,引起亚硝化应激。因此,这两个类别经常统称为ROS/RNS。

白藜芦醇为多酚化合物,以顺式或反式异构体形式天然存在于很多植物物种,尤其存在于葡萄、花生和浆果的皮中,表现出广谱免疫调节活性。很多研究指出其抗癌、抗氧化和心脏保护性质以及延长多种有机体的寿命(Aggarwal等人,2004.Anticancer Res.24(5A):p.2783-840;Gao等人,2003.Biochem Pharmacol.66(12):p.2427-35;Falchetti等人,2001.Life Sci.70(1):p.81-96;Saiko等人,2008.MutatRes.658(1-2):p.68-94)。此外,白藜芦醇具有治疗动物和人类的多种传染性疾病的巨大治疗潜能。已表明其抗皮肤真菌(Chan,2002.Biochem Pharmacol.63(2):p.99-104)和病毒如单纯疱疹病毒(Herpessimplex)(Faith等人,2006.Antiviral Res.72(3):p.242-51)的活性。已针对广泛范围的细菌物种——包括造成急性呼吸道感染的胞内肺炎衣原体(Chlamydia pneumoniae)——证实了白藜芦醇的生长抑制活性(Chan,2002.Biochem Pharmacol.63(2):p.99-104;Schriever等人,2003Atherosclerosis.171(2):p.379-80)。此外,实现了引起鱼疾病的原生动物物种降低的感染性(Leiro等人,2004Dis Aquat Organ.59(2):p.171-4;Leiro等人,2004Antimicrob Agents Chemother.48(7):p.2497-501),并且更近期,表明白藜芦醇清除造成皮肤利士曼病的胞内皮肤病原体——硕大利什曼原虫(L.major)(Kedzierski等人,2007.Parasitol Res.102(1):p.91-7;Kedzierski等人,2009.Curr Med Chem.16(5):p.599-614)。
根据本发明所用的菌蜕可以来自多种细菌。优选地,菌蜕来自非病原性革兰氏阴性菌,更优选大肠杆菌菌株。可以使用任何大肠杆菌菌株。适宜的大肠杆菌菌株的例子为大肠杆菌Nissle1917。
如上所概述,表面屏障,例如皮肤,是作为第一道感染防御的机械屏障的实例。此外,皮肤和呼吸道分泌抗微生物肽,例如β-防御素,因此起到抵御感染的化学屏障的作用。因此,优选在皮肤和/或粘膜(mucosa,其也命名为黏膜)中提高先天免疫应答。粘膜的实例包括口粘膜、颊粘膜、食管粘膜、胃粘膜、肠粘膜、呼吸道粘膜、支气管粘膜、鼻粘膜、嗅粘膜、子宫粘膜、阴茎粘膜和子宫内膜。
皮肤以及粘膜由上皮细胞组成。上皮细胞形成上皮,其代表沿着遍布身体的结构的腔和表面的细胞组成的组织。
角质形成细胞是表皮即皮肤外层的主要成分。角质形成细胞是至关重要的免疫调节子,通过在休眠时分泌细胞因子如IL-4和TGFβ维持免疫应答的完整性,但当引发时,角质形成细胞将通过TNFα和IL-1β分泌而刺激皮肤炎症和朗氏细胞活化。
根据本发明,优选在上皮细胞中诱导至少一种先天免疫调节子的表达和/或释放。优选地,上皮细胞是角质形成细胞。
认为刺激先天免疫应答以非特异性方式增强宿主对其它有机体感染的防御。因此,根据本发明,优选地,在哺乳动物、优选人、更优选易感染个体或者具有受损免疫系统的个体中提高先天免疫应答。
在本发明进一步的方面,菌蜕施用于哺乳动物、优选人。优选菌蜕施用于皮肤和/或粘膜。
施用菌蜕的合适模式包括经口、局部、粘膜、鼻和/或肺施用。
合适的药物剂型为,例如,胶、吸入剂、鼻腔喷雾、皮肤贴剂、膏剂、洗剂、气雾剂、栓剂、灌洗剂、凝胶液、悬液、牙膏的形式。
根据本发明,可以将菌蜕施用于任何个体。优选地,菌蜕施用于婴儿、幼儿、老人、慢性病个体、具有受损免疫系统的个体例如AIDS患者、移植接受者。
在另一方面,本发明涉及提高先天免疫应答的方法,其中将菌蜕(BG)施用于患者。菌蜕的适宜施用模式和适宜药物形式/剂型/配方如上所述。

实施例
实施例1:菌蜕提高人角质形成细胞中先天免疫应答
关于BG诱导上皮细胞、特别是人KC的先天免疫调节子表达的能力知之甚少。在本研究中,研究了大肠杆菌BG诱导人原代KC中抗微生物肽和促炎细胞因子表达的能力。获得的结果表明:BG表面上鞭毛蛋白的存在增强了人类KC中抗微生物牛皮癣素和hBD-2的表达以及白介素(IL)-6和IL-8的释放。
材料&方法
细胞培养
从新生儿包皮制备的人原代KC自Clonetics(San Diego,CA,USA)获得,并如前所述(Rendl等人,2002.J.Invest Dermatol.119:1150-1155),培养于无血清角质形成细胞生长培养基(KGM,Clonetics)。为了刺激,在12孔组织培养板(Corning Incorporated,Corning,NY,USA)中培养第三代KC,并于60-70%的汇合时使用。在角质形成细胞基础培养基(KBM,Clonetics)中进行刺激。
RNA分离和qRT-PCR
刺激后,细胞用磷酸盐缓冲盐水(PBS)进行洗涤,并根据制造商说明使用Reagent(Invitrogen,Carlsbad,CA,USA)分离RNA。对于cDNA合成,使用基因Amp RNA PCR试剂盒(Applied Biosystems,Foster City,CA,USA)和寡dT引物(Roche Diagnostics,Basel,Switzerland),以MuLV-反转录酶对RNA进行反转录。研究的所述基因的cDNA序列获得自GenBank。使用来自Whitehead Institute forBiomedical Research(Cambridge,MA,USA)的PRIMER3软件设计引物。使用以下正向(F)和反向(R)内含子-跨越引物:

使用用于扩增和检测的Fast Start SYBR Green I试剂盒(RocheDiagnostics)通过LightCycler技术进行qRT-PCR。在所有测定中,使用标准程序(10min变性步骤和55个循环的95℃5sec;65℃15sec,和72℃15sec;采用0.1℃步进的熔点分析;最后的冷却步骤)扩增cDNA。各LightCycler毛细管加载1.5μl DNA Master混合物(MasterMix);1.8μl MgCl2(25mM);10.2μl H2O;和0.5μl各引物(10μM)。使用Pfaffl的数学模型(Pfaffl.2001.Nucleic Acids Res.29:e45)确定靶基因表达的相对量化和扩增效率。将靶基因的表达标准化为看家基因β-2-微球蛋白的表达。所有实时PCR一式三份进行。通过PCR产物测序证实PCR反应的特异性。
BG产生和KC刺激
大肠杆菌NK9373(wt)和大肠杆菌NK9375(ΔfliC)(具有fliC基因内的框缺失的鞭毛蛋白-缺陷型菌株(Bates et al.,2005.Mol.Microbiol.57:380-391))由David Bates博士(Baylor College of Medicine,Houston,Texas)友情提供。于35℃在含有庆大霉素(gentamycin)(20μg/ml)的非动物源Lennox肉汤(LBv;10g/l sojapeptone(豆胨),5g/l酵母提取物,5g/l NaCl)中培养具有裂解质粒pGLysivb(未发表)的大肠杆菌菌株。将两升培养基用4ml甘油(glycerine)储液接种,由一个单一转化子集落传下并用作过夜温育后发酵的预培养物。使用Techfors S发酵器(Infors Ag,Bottmingen,Switzerland)在20l培养基中进行发酵。存档以下参数:温度、流动、搅拌子、pH、pO2、发泡以及用于酸、碱的抗泡的泵。培养和裂解细菌,接着进行:测量光密度(OD600)、使用螺旋接种仪(WASP system,Don Whitley Scientific Limited,WestYorkshire,UK)确定集落形成单位和周期取出样品的显微术。在通气和搅拌下(通过程序化顺序控制流动和搅拌)在设置为pH7.2的LBv培养基中培养细菌至中对数生长期。通过温度上移至42℃诱导裂解蛋白E的表达。在完全裂解过程(达到pO2水平的平台)后,通过加入β-丙内酯(propiolacton)(BPL)杀死剩余的完整细菌。时间间隔30min以两份相等剂量加入总计0.075%BPL。对于与BPL温育,搅拌速率设置为600rpm。对于通过分离机(CTC1,GEA Westfalia Separator GmbH,Oelde,Germany)进行收获,温度设置为16℃,并使用约200ml/min的流速。在重悬浮BG团块之前,以5l无菌蒸馏水漂洗所述系统,随后使用Hermle ZK401离心机(Hermle Labortechnik GmbH,Wehingen,Germany)于4℃以8,000min-1、15min,以总体积为7.5l的蒸馏水通过5重悬浮/离心循环洗涤BG。将最终的团块重悬浮于200ml蒸馏水,等分至冻干瓶中,并存储于-80℃。使用Lyolab B(LSLSecfroid,Aclens,Switzerland)冻干器将样品冻干约60h。
在施用于KC前,将来自大肠杆菌NM522、大肠杆菌NK9373(wt)和大肠杆菌NK9375(ΔfliC)的冻干BG重悬浮于KBM培养基中。对于体外测定,使用重组IL-1α(R&D Systems,Minneapolis,MN,USA)和来自鼠伤寒沙门氏菌(S.typhimurium)的纯化鞭毛蛋白(Invitrogen)。如Abtin等人(FASTEB J22(2008),2168-2176)中所述制备wt和ΔFliC大肠杆菌的培养物上清液。在刺激KC之前,将培养物上清液以1:100稀释于KBM。
细胞因子测量
从刺激的KC的培养物上清液通过离心除去分离的细胞或细胞片段,并贮存于-20℃直至分析。根据制造商说明书,通过酶联免疫吸附测定(ELISA;R&D Systems)测定IL-6和IL-8的浓度。
免疫测定分析
为了分析蛋白质表达,将KC裂解于SDS-PAGE上样缓冲液(50mM Tris,pH7.4,2%SDS)。经超声处理后,通过离心除去不溶细胞残余物,并通过BCA(二辛可酸)方法(Pierce,Rockford,IL,USA)方法测量蛋白质浓度。如前所述(Mildner等人,2006.Biochem.Biophys.Res.Commun.348:76-82)进行Western印迹分析。通过膜的丽春红S(Ponceau S)染色证实蛋白质裂解物的等量加样。使用以下第一步骤抗体:小鼠单克隆IgG1抗-牛皮癣素克隆47C1068(稀释1:500;Abcam,Cambridge,UK)。根据制造商说明书,使用Chemiglow试剂(Alpha Innotech,San Leandro,CA,USA)将膜显色。
应用基于ImageJ软件的分析来定量通过免疫印迹分析获得的条带的密度(Rasband,W.S.,ImageJ,U.S.NIH,Bethesda,Maryland,USA;http://rsb.info.nih.gov/ij/,1997-2009)。将以多种颗粒浓度的wt和ΔFliC E.coli BG刺激KC后获得的牛皮癣素表达的值标准化为以重组IL-1α刺激KC后获得的牛皮癣素表达的值。
异硫氰酸荧光素(FITC)-标记的BG摄取
如前所述(Kudela等人,2007.Cancer Lett.1:54-63;Kudela et al.,2005.J.Immunother.28:136-143)测量人原代KC的胞吞活性的效率。简言之,将24孔板(2x105细胞/孔)中培养的原代KC于+37℃与FITC-BG(1000/细胞)温育2小时。温育后,以PBS洗涤温育细胞三次,以除去过量的BG。最后,将细胞使用TrypLETM Express(Invitrogen)分离,以PBS洗涤两次,并在PBS中的冷1.5%多聚甲醛(Sigma-Aldrich,St.Louis,MO)进行固定,在BD FACSCantoTM流式细胞仪(BD Biosciences,Pharmingen,San Jose,USA)上分析。
统计学分析
通过GraphPad Prism第5版(GraphPad Software,La Jolla,CA)分析获得的结果。通过斯氏t检验评价两组之间差异的统计学显著性,并通过一维ANOVA分析多于两组之间差异的统计学显著性。p<0.05认为差异是显著的。
结果
通过BG在人原代KC中表达抗微生物肽
已报道大肠杆菌培养物上清液诱导上皮KC中抗微生物肽牛皮癣素和hBD-2的表达(Abtin等人,2008.FASEB J.7:2168-2176;Glaser等人,2005.Nat.Immunol.6:57-64)。为了研究BG引发上皮细胞先天免疫应答的能力,以不同颗粒浓度的获自wt大肠杆菌的BG刺激人原代KC24小时。通过鞭毛蛋白(10ng/ml)和IL-1α(10ng/ml)刺激KC作为所分析的抗微生物肽上调的阳性对照,而将未处理细胞作为阴性对照。通过定量实时PCR(qRT-PCR)测定抗微生物牛皮癣素的相对mRNA上调。如图1所描绘,与BG温育后,KC中牛皮癣素的mRNA表达上调。刺激的牛皮癣素mRNA产生依赖于BG颗粒,并且于2x108BG颗粒/ml观察到最强上调,低于2x108颗粒/ml的BG浓度没有作用(图1)。
我们先前报道了KC中牛皮癣素的诱导依赖于大肠杆菌的鞭毛蛋白表达(Abtin等人,2008.FASEB J.7:2168-2176)。为了研究BG的鞭毛蛋白是否具有相似效果,我们生成了来自野生型(wt)NK9373和同基因鞭毛蛋白-缺陷型(ΔfliC)NK9375大肠杆菌菌株的BG。作为阳性对照,以鞭毛蛋白、IL-1α和来自wt大肠杆菌的上清液刺激KC。将在非条件化细菌培养基存在下温育的KC、在来自ΔFliC E.coli的培养物上清液存在下温育的KC和在无刺激下温育的KC作为阴性对照。刺激KC48小时,其后通过免疫印迹分析牛皮癣素蛋白质生成。不同于mRNA数据,在蛋白质水平于2x107BG颗粒/ml检测到牛皮癣素的模糊条带(图2A)。于2x108颗粒/ml观察到牛皮癣素的强诱导,其甚至比于2x109颗粒/ml通过wt(NK9373)大肠杆菌BG观察到的诱导更强。对于同基因ΔfliC(NK9375)菌株,从2x108BG颗粒/ml开始检测到牛皮癣素的模糊条带,于2x109BG颗粒/ml表达水平突出(图2A,B)。
以BG处理后在人原代KC中细胞因子的表达
通过ELISA研究与BG温育48小时后促炎细胞因子IL-6和IL-8通过KC的分泌。与wt(NK9373)和ΔfliC(NK9375)大肠杆菌BG温育后,KC释放IL-6和IL-8依赖于BG来源和所用的颗粒数(图3A,B)。所得的结果表明wt(NK9373)大肠杆菌BG对IL-6和IL-8释放的作用从浓度2x107颗粒/ml开始可检测到。尽管低于2x107颗粒/ml的浓度几乎是无效的,2x109颗粒/ml分别从2pg/ml(未处理)至600pg/ml强烈增强IL-6分泌和从60pg/ml(未处理)至1350pg/ml强烈增强IL-8分泌(图3A,B)。然而,当使用2x109颗粒/ml时,ΔfliC(NK9375)大肠杆菌BG仅分别将两种细胞因子的分泌提高至134pg/ml和750pg/ml的IL-6和IL-8,而低于2x109颗粒/ml的BG浓度则对两种细胞因子的释放无显著作用(图3A,B)。此外,将KC与来自wt大肠杆菌和ΔFliC大肠杆菌的培养物上清液温育后检测到与鞭毛蛋白存在相关的IL-6和IL-8分泌的显著差异,此时与来自wt大肠杆菌的培养物上清液相比,在ΔFliC大肠杆菌存在下温育KC后细胞因子产生低近一个数量级(图3A,B)。这些结果一起强调了BG的完整包被表面结构的重要性以及鞭毛蛋白在刺激先天免疫系统中的作用。
人原代KC对wt(NK9373)和ΔfliC(NK9375)大肠杆菌BG的胞吞作用
如上所提及,KC对抗微生物牛皮癣素的上调和促炎细胞因子的增强释放依赖于BG表面上鞭毛蛋白的存在。为了进一步研究鞭毛蛋白在KC对BG的摄取中的作用,比较并分析了wt(NK9373)和ΔfliC(NK9375)大肠杆菌BG的胞吞作用。在BG表面上缺失鞭毛蛋白引起KC结合并胞吞BG的能力显著减低(低于~6-倍)。FAC分析清楚地表明对于FITC-标记的wt大肠杆菌BG(图4A)和对于ΔFliC大肠杆菌BG(图4B)内化之间的差异。如图4C所描绘,由于完整鞭毛蛋白的存在,观察到KC结合并内化wt大肠杆菌BG的能力与ΔFliC大肠杆菌BG相比提高约6倍。
讨论
BG为来自革兰氏阴性菌的非活细胞包被制备物,去除胞质成分,而其细胞形态学和天然表面抗原结构得以保留[40]。在本研究中,使用人原代KC测定了BG对先天免疫调节子的调节的效应。将产生自非病原性大肠杆菌的BG与人KC温育上调了抗微生物牛皮癣素的表达。这与先前报道一致,在先前报道中大肠杆菌菌株的培养物上清液或破坏的细胞增强了抗微生物肽的表达[2]。因此,本研究证实了包含鞭毛蛋白的BG包被结构是完全有功能的和完整的,并被KC识别,与先前研究中所用的细菌化合物类似提高了先天免疫应答[2]。
如前所报道,KC对大肠杆菌的应答性通过TLR5及其配体鞭毛蛋白介导[2]。wt大肠杆菌表现出以BG浓度-依赖性诱导牛皮癣素产生(图1);而ΔFliC大肠杆菌在比wt菌株高一个数量级的BG浓度处才显示牛皮癣素表达(图2)。因此,从促炎细胞因子IL-6和IL-8释放获得的数据表明BG制备物中与ΔFliC突变体菌株相比时鞭毛蛋白存在的重要性(图3)。从先前以大肠杆菌上清液作为shad鞭毛蛋白和/或纯化鞭毛蛋白来源的研究,明显的是鞭毛蛋白是人KC中牛皮癣素的主要诱导子[2]。然而,由于ΔFliC BG能够诱导牛皮癣素,似乎存在另外的独立于FliC的途径。
此外,来自wt大肠杆菌和ΔFliC大肠杆菌的KC对BG摄取的研究表明来自具有鞭毛的菌株的BG被摄取比ΔFliC BG好大概一个数量级(图4)。该观察提示BG表面上鞭毛蛋白的存在有助于KC对BG的结合与内化。此外,在与ΔFliC大肠杆菌BG温育或与ΔFliC大肠杆菌培养物上清液温育后在制备这些蛋白质中KC应答的降低反映了鞭毛蛋白在牛皮癣素产生以及IL-6和IL-8分泌中的关键作用。问题依旧是ΔFliC大肠杆菌BG表面上的其它成分例如LPS是否具有对于抗微生物肽和/或促炎细胞因子产生的信号转导活性。然而,KC释放IL-6和IL-8对鞭毛蛋白存在更加敏感,因为不能检测到与BG颗粒数的直接关联(图3)。在与ΔFliC BG温育后与IL-6相比KC对IL-8的较高分泌(图3)以及来自ΔFliC大肠杆菌的BG的部分内化(图4)可能与BG壳上存在LPS相关。来自培养物上清液中存在的破坏细菌细胞的LPS的存在可以解释在与来自ΔFliC大肠杆菌的培养物上清液温育后检测到KC对IL-8的低分泌(图3B)。先前表明LPS活化皮肤黑色素瘤细胞引起增强的IL-8产生和细胞粘附(Malteni等人,CancerLett.235(2006),75-83)。尽管已报道TLR4不参与内皮细胞的细胞LPS摄取,但LPS活化TLR4-介导的表皮KC与其吞噬活性之间的联系尚需确认(Dunzendorfer等人,J.Immunol.173(2004),1166-1170)。尽管一些研究者观察到TLR4在KC表面上的功能表达而在其它研究中却未观察到的事实[2],BG包被上的完整LPS仍可能有助于包括KC的人皮肤细胞识别细菌成分的复杂过程。因此,仍需确定在通过LPS信号转导活化表皮KC中涉及的确切作用和机制仍需确定。
也可能的是,wt大肠杆菌BG中存在的鞭毛在KC的溶酶体区室中的酸性降解导致单体鞭毛蛋白,其能够结合TLR5并刺激细胞因子IL-6和IL-8表达以及牛皮癣素产生。由于结合于TLR5的鞭毛蛋白[2]不能与ΔFliC BG发生并且因此通过该结合诱导的信号转导不能发生,从而导致牛皮癣素的NF-κB诱导的表达的内在信号转导的其它途径必须通过KC中的BG诱导。无论鞭毛蛋白表达可能感知BG或BG组分存在的可能的胞内受体为NLR,例如NOD1或NOD2。近来的研究已经报道了KC中肽聚糖片段对NOD1[22]和NOD2[2]的功能性表达。具体而言,NOD1介导含有氨基酸间-二氨基庚二酸的肽聚糖片段的感知,而NOD2介导胞壁酰二肽的感知[2],这些片段均为仍完整的大肠杆菌BG肽聚糖的降解产物[60]。已报道了胞壁酰二肽(MDP)活化NOD2与原代KC中提高的抗微生物肽产生之间的联系[2],在我们的研究中已表明了BG诱导牛皮癣素的用途。因此,获得的结果表明非存活且安全的BG在治疗方法中增强皮肤的先天免疫防御系统的理想用途。可以通过将药物或其它生物活性物质包装到也可以胞内递送的BG中来合并BG的潜在治疗效果,以刺激另外的有益健康效果[2]。
实施例2:加载白藜芦醇的菌蜕在RAW264.7中NO产生的调节作用
如先前研究所表明,BG对如细胞因子分泌和抗微生物肽表达的先天免疫系统参数有很大影响。考虑到iNOS诱导先天免疫性中主要参与分子NO的产生由LPS刺激,我们检查了BG刺激的自由基释放的程度。此外,针对调节化学物质引起的抗炎(减低NO产生)活性以及抗增殖活性,研究了负载RV的菌蜕的作用。
材料&方法
化学物质
若不另外陈述,所有化学物质获得自Sigma Aldrich。
细胞培养物
在添加2mM L-谷氨酰胺(Lonza Bio Whittaker)和10%热灭活的胎牛血清(Gibco;Invitrogen)的含有4,5g/L葡萄糖的Dulbecco改性必需培养基(DMEM;Lonza BioWhittaker;W/O Glut P-red)中培养从美国典型培养物保藏中心(ATCC,USA)获得的RAW264.7。此外,将100U Pen-Strep(Lonza)加入培养基。对于评估存活性,使用了另一种培养基。其中,细胞培养于添加10%热灭活的胎牛血清(FCS)、青霉素(104μg/ml)、链霉素(100μg/ml)、200mM L-谷氨酰胺、HEPES缓冲液(10mM)、10%非必需氨基酸(NEAA)和原生质素(5μg/ml,Lonza)的Roswell Park Memorial Institute Medium(RPMI)1640。
培养细胞45-48h(37℃;5%CO2),直至其达到其汇合状态。用于分析各种实验的培养基仅含抗生素。
菌蜕制备
通过噬菌体衍生的裂解蛋白质E的受控表达,制备来自大肠杆菌NM522(pGLysivb;240106-5/6)的BG,如其它地方所述(Witte等人,1992.Arch Microbiol.157(4):p.381-8;Mayr等人,2005.InfectImmun73(8):p.4810-7)。通过加入抗生素灭活未裂解的细菌。将冻干的BG存贮于室温(1mg冻干重量含有1.27*1010颗粒)。在处理实验前将BG重悬浮于测试培养基。
用白藜芦醇负载BG
将12-24mg量的冻干BG悬浮于不同浓度的白藜芦醇(1-35mgRV/ml甲醇)并伴随剧烈震荡(800rpm)于28℃温育30min。通过13000rpm离心15min收集负载的BG,并用水洗涤团块三次。1mg BG等分试样贮存于-20℃,直至使用。
亚硝酸盐测定
将RAW264.7细胞接种于96孔板,并培养两天。然后,以200μl定义浓度的空BG(阳性对照)或以加载白藜芦醇的菌蜕(46μgRV/1x1010BG)、或以空菌蜕加外部给予的定义浓度的白藜芦醇刺激约3*105巨噬细胞/孔20min。其后,通过以PBS洗涤两次除去菌蜕悬液,并于37℃5%CO2黑暗中温育细胞另外20小时。为了研究纯LPS(Fluka;大肠杆菌血清型055:B5)的刺激作用,采用多种LPS浓度进行长期温育实验20小时。
作为NO产生的一个指示,通过使用Griess反应(Green等人,1982Anal Biochem.126(1):p.131-8)在巨噬细胞上清液中测量亚硝酸盐浓度。简言之,将100μl各上清液与90μl5%H3PO4中的1%氨苯磺胺(Fluka)和90μl水中的N-(1-萘基)乙二胺二盐酸盐混合。使用ELISA读数器(Tecan Sunrise)确定550nm处的吸光度。
摄取研究
向RAW264.7应用并检测FITC-标记的BG
作为胞吞白藜芦醇递送的指示,在被分析的细胞系中测定FITC-标记的BG的摄取。为了该目的,将冻干的菌蜕5-15mg重悬浮于1.5ml0,1M Na2CO3;pH~9.0。其后,向BG悬液加入25μl FITC储液(1ml DMSO中2mg FITC)并于16℃在黑暗中震荡2h。用PBS的五个洗涤步骤(5min;14000rpm)并检查阳性标记后,在碳酸钠缓冲液中重悬浮菌蜕团块,并贮存于-20℃直至使用。
RAW264.7的培养和处理与对于亚硫酸盐测定中的培养和处理相同,但有微小调整。与定义量的空BG温育20min后,以PBS洗涤巨噬细胞两次,在空的临近孔中收集洗涤溶液。然后于激发和发射波长485/535(增益40)处使用Tecan Geniospro荧光计记录荧光。取总荧光值为施用的菌蜕量的100%,由所得的荧光/巨噬细胞计算摄取。
细胞毒性测定
中性红测定用于检测空BG或RV负载的BG以及化学物质本身对RAW264.7细胞存活性的影响。中性红(3-氨基-m-二甲氨基-2甲基-吩嗪盐酸盐)在活细胞溶酶体中选择性积累(Repetto等人2008.NatProtoc,2008.3(7):p.1125-31),并因此提供存活性的定量评估。
将96孔板的每孔接种1,25x105细胞,并使其粘附过夜。用含有不同BG与细胞比例(10,100,1000)的加载RV的BG和未加载的BG(均重悬浮于sf-培养基中)的200μl培养基处理或者单独用RV处理巨噬细胞20min,然后进行20小时的回收期或24小时。以两种RV浓度(15μM和30μM)进行相同的条件。Triton X-100(0.01%)用作阳性对照,而以测试培养基处理的细胞代表阴性对照。处理后,以PBS洗涤细胞两次,并以培养基温育20小时,或直接以100μl NR(80μg/ml终浓度)温育另外两小时(37℃;5%CO2)。其后,弃去燃料,并以PBS洗涤孔两次。通过加入100μl酸性脱色液(1ml乙酸、73ml96%乙醇和26ml去离子水)完成染料提取。将平板震荡10min,通过平板读数器(Dynex OpsysMR)于570nm(参照波长690nm)测量显色的颜色。
统计学
使用GraphPad Prism(第5版,GraphPad Software,Inc;SanDiego;CA,USA)分析所有结果。数据表示为平均值+SD。使用斯氏t检验进行统计学分析。P-值<0.05认为统计学上显著。
结果
将BG加载RV
通过将BG简单重悬浮于所需的RV-溶液中,将冻干的大肠杆菌NM522BG加载RV。为了测定BG中RV的量,进行乙醇提取并通过HPLC分析。结果描绘于图5A。可以观察到RV的加载浓度与回收的RV之间清楚的相关性(相关系数r2=0.9980)。将冻干的BG悬浮于35mg/ml RV溶液后获得最高加载效率。提取1x1010BG后,平均可检测46μg RV。当考虑开始时冻干的菌蜕的量时,回收约4%的储液。
鼠巨噬细胞粘附并摄取FITC-标记的BG
进行荧光测定定量实验以研究BG的量,所述BG在短时间的共温育后或者是细胞结合的、或者由鼠巨噬细胞摄取。图5B描绘了在20min共温育和两个洗涤步骤后通过RAW264.7和大肠杆菌NM522BG的荧光测量获得的结果。可以看到应用的和细胞结合的菌蜕清楚的剂量相关性,导致近乎完美的线性回归(相关系数r2=0.998)。此外,在96孔板或24孔板进行实验时未观察到差异(数据未显示)。在短暂的温育时间后,平均25%的应用菌蜕与巨噬细胞结合。
为了澄清在该期间BG是否也已经被鼠巨噬细胞摄取,进行共聚焦激光扫描显微术(CLSM)研究,其中可见地检查内化。为了该目的,在将FITC-标记的大肠杆菌NM522菌蜕与RAW264.7细胞温育20分钟和40分钟后,进行z-堆叠(z-stack)。巨噬细胞以选择性结合细胞的F-肌动蛋白骨架的Texas-Red鬼笔环肽进行染色。在图6A和图6B中描绘了显示多个光学切面的代表性单z-堆叠的图像。可见已经被染为红色的细胞内化的FITC-标记的BG显示黄色。着绿色的BG代表没有被吞入并在外部粘附至细胞的那些BG。此外,在BG-细胞比例为1000时,我们在每个巨噬细胞中观察到摄入的BG以及内化的BG簇。
诱导NO生成
因为iNOS由LPS诱导,在用不同浓度的LPS刺激RAW264.7细胞20小时后,研究NO生成。如图7A所描绘,以1-1000ng/ml LPS处理导致具有始于1ng/ml的显著敏感性的NO生成。
由于BG完全保持其结合于包被内的LPS分子的事实,进行实验以测定BG能够刺激NO产生至何种程度。结果是菌蜕与细胞比为10已经足够显著诱导NO生成(P=0.0005),水平类似于1ng/ml游离LPS。
当细胞用MOI为100-1000的空BG处理时,达到清楚的颗粒依赖性刺激NO生成。然而,更高浓度的BG(2000颗粒/细胞和5000颗粒/细胞)未显示任何进一步的效果。
通过BG自身和加载RV的BG与外部给予的RV对比刺激NO的调节能力
如图7B所示,当将BG以BG与细胞比为100-1000施用于巨噬细胞时,BG能够以剂量依赖方式刺激NO生成。
因为许多研究表明RV能够降低LPS-诱导的NO-生成(Saiko等人,2008.Mutat Res.658(1-2):p.68-94;Tsai等人,1999Br J Pharmacol.126(3):p.673-80),进行实验来研究RV调节BG诱导的NO生成的潜能。为了该目的,将大肠杆菌NM522BG以500的比例施用于细胞。同时,将三种不同浓度的RV加入细胞培养物中。20min共温育和两个洗涤步骤后,在20小时后测量NO生成。结果描绘于图8A。可见用空大肠杆菌NM522菌蜕与0.3μM和0.3μM RV的同时处理对于BG诱导的NO生成没有影响。在用30μM RV处理后可观察到统计学显著差异(P=0,0078)。
在20min处理后,基于BG中RV的捕获量(图5A)以及与巨噬细胞结合(粘附于细胞和被细胞摄取)的BG的数目(图5B)可以测定BG递送的白藜芦醇颗粒。由于以MOI100和1000使用具有最高RV-产量(46μg RV/1x1010BG)的BG进行刺激RAW264.7细胞中NO-诱导,由BG计算的RV递送值将对应于0.48+0.25μM和8.86+2.21μM白藜芦醇(5.8x1013和1.07x1015RV-颗粒/200μl)。
为了研究BG中结合的RV是否具有对NO生成的调节模式,分析相同量的空菌蜕相比加载RV的菌蜕的影响。此外,评估了由于同时应用空BG和3μM RV(3.61x1014RV颗粒/200μl)的自由基释放。
可见,外部应用的RV与空大肠杆菌NM522BG一起没有与负载RV的菌蜕一样有效地降低自由基释放。我们的发现表明,以BG与细胞比为100使用加载RV的菌蜕发现硝酸盐显著降低(P=0.0048),而3μM RV没有效果(图8B)。另一方面,在以MOI1000处理巨噬细胞后获得的结果对于采用两种变体均导致降低。然而,与用单独的BG处理相比,当RV-菌蜕被应用于RAW264.7细胞时以及采用3μM RV同时施用空BG所产生的NO的量显著较低(P=0.0002vs.P=0.0158)(图8C)。
确定细胞存活性
为了研究空BG和负载RV的BG(46μg RV/1x1010BG)对RAW264.7巨噬细胞存活性的作用,使用三种不同的BG与细胞比(10,100,1000)检查两种不同的处理条件。以对于NO-测量相同的方式进行第一测试,即20min处理接着20小时测量。第二分析涉及长期温育24小时的效果。如图9A所绘,大肠杆菌NM522菌蜕本身在该期间没有细胞毒性作用。然而,以MOI10的BG温育巨噬细胞24小时导致显著的生长刺激作用(P=0.0016),而较高的BG-浓度没有不同于无血清培养基处理的对照。尽管在以加载RV的菌蜕或化学物质本身短时间共温育后未见到效果(数据未显示),在24小时后以MO10和100的RV-BG处理后发现增强的代谢活性(P=0.0008和0.0111)(图9B)。
因为我们没有测定24小时温育后与细胞结合的BG的量,不能对BG递送的RV浓度作出充分的陈述。
理论上,如果所有应用的MOI1000的BG均被巨噬细胞摄取,可以达到30μM的水平。然而,我们由显微镜研究知道这是不正确的。假定一半量的BG将在该期间被吞入,获得15μM白藜芦醇的计算浓度。因此,进行了比较实验,其中研究了15μM和30μM RV对细胞存活性的作用,相比以BG中的结合RV(46μg RV/1x1010BG),其以菌蜕与细胞比为1000应用。然而,最低化学浓度根本没有影响,在用30μM RV处理后细胞存活性快速下降(81.23+10.6%)。以RV-BG也观察到细胞存活性的显著降低(P=0.0462)。平均而言,10%的细胞由于递送的化学物质而死亡(图9B)。
讨论
在本研究中,我们已经证明BG诱导巨噬细胞的NO-形成,并且该活性可以由负载RV的BG调节。
已经在数个研究中证明了依赖于iNOS活性和NO-释放来治愈病原体-引起的疾病,例如利士曼病或结核(Bogdan,C.,2000ImmunolRev.173:p.17-26;Sharma等人,2007.Inflammopharmacology15(6):p.252-9;Chakravortty和Hensel,2003.Microbes Infect.5(7):p.621-7)。
一般而言,知晓病原体被先天免疫系统的细胞所吞噬。来自其它研究的结果表明BG优选地被那些细胞吞入,并且已经报道了多种巨噬细胞细胞系和树状细胞的摄取(Haslberger等人,2000.J Biotechnol.83(1-2):p.57-66;Kudela等人,2005.J Immunother.28(2):p.136-43;Paukner等人,2003J Drug Target11(3):p.151-61)。根据先前发现,在BG短期共温育后,可以证实在RAW264.7巨噬细胞细胞系中的摄取。有趣的是,使用CLSM进行显微镜观察显示巨噬细胞能够吞入单一菌蜕以及菌蜕的簇。
因为BG如其天然对应物一样具有所有其病原体相关分子模式(PAMP),它们被其相应的宿主先天免疫受体例如Toll样受体(TLR)所识别。因此,细菌LPS与TLR4以及鞭毛蛋白与TLR5相互作用(Ishii等人,2008Cell Host Microbe.3(6):p.352-63)。在鼠RAW264.7巨噬细胞细胞系中BG的摄取主要与TLR4相关,因为他们几乎不表达TLR5(Applequist等人,2003.Int Immunol.14(9):p.1065-74)。另一方面,在实施例1中,表明大肠杆菌BG通过依赖鞭毛蛋白的TLR5途径以及其它摄取机制进入原代角质形成细胞。
使用人巨噬细胞的来自Panaro等人的研究表明LPS诱导的NO生成之间的直接相关性。当L–NMMA——i-NOS的竞争性抑制剂存在时,功效在两个参数方面均显著降低(Panaro等人,1999.Int J ClinLab Res.29(3):p.122-7)。
因为LPS以通过与TLR-4相互作用诱导iNOS而闻名(Bogdan,C.,2001.Nat Immunol2(10):p.907-16),我们在RAW264.7细胞中进行了以游离LPS长期温育(20小时)和以各种量的BG短期温育(20min,然后在20小时后测定NO释放)之间的比较实验。结果是以MOI10的大肠杆菌菌蜕处理巨噬细胞表现出与100ng/ml LPS相同的效果,Panaro等人发现其是寄生虫去除的原因(Panaro等人,1999.Int J ClinLab Res.29(3):p.122-7)。此外,我们分析了观察到的颗粒依赖性BG-细胞结合与NO释放程度之间的关系,并发现显著的正相关(Spearmanr=1;P值=0.0167)。
除了NO-生成,如细胞因子的其它因素引发针对胞内病原体的先天免疫应答。很多细胞产生I型干扰素如IFNγ,并表现出强抗微生物活性。在早期研究中,表明BG具有刺激细胞因子产生的特征。例如,Ebensen等人表明应用溶血性曼氏杆菌(M.haemolytica)BG在被免疫的小鼠中引起脾IFN-γ-产生细胞的量增加(Ebensen等人,2004.JImmunol.172(11):p.6858-65)。在另一动物研究中,在用大肠杆菌0157:H7BG免疫后,脾IFNγ水平显著提高(Mayr等人,2005.InfectImmun73(8):p.4810-7)。因为已知IL-12提高IFNγ产生并在Th1细胞的发展中起重要作用,Haslberger等人进行了使用BG的研究并报道了在抗原呈递细胞中有效刺激IL-12产生(Haslberger等人,2000.JBiotechnol.83(1-2):p.57-66)。
回到NO-生成,来自使用白藜芦醇的研究的结果表明其对iNOS诱导的抑制特征(Saiko等人,2008.Mutat Res.658(1-2):p.68-94;Tsai等人,1999Br J Pharmacol.126(3):p.673-80)。因此,在本研究中,测定了白藜芦醇(或者与空BG共温育,或者胞内结合于BG)对BG诱导RAW264.7巨噬细胞产生NO的作用。与其空白版本相比,施加结合RV的BG获得了高度显著降低的自由基释放。而且,浓度水平为3μM的外部添加的纯化合物——比由MOI100的结合细胞的菌蜕递送的浓度高约6倍——对NO生成没有作用,而加载BG的应用则降低自由基释放约15%。
据我们所知,白藜芦醇的细胞摄取机制尚不清楚。至今,还没有发现药物的特定受体。由于其结构,白藜芦醇能够与细胞表面受体如雌激素受体或整合素相互作用(等人,2008.Mutat Res.658(1-2):p.68-94;Lin等人,2006Faseb J.20(10):p.1742-4)。然而,由于我们的发现以及在巨噬细胞摄取后加载RV的BG胞内递送白藜芦醇的事实,存在内部白藜芦醇受体是很有可能的。我们建议为这样的假定受体命名为BGRV。
已知细菌LPS负责对广泛种类的哺乳动物细胞的多种病理生理学作用。在最坏情况下,内毒素性休克和多种器官衰竭继之以死亡可能是后果。该内毒素性通过活化宿主免疫和炎性细胞、特别是单核巨噬细胞而介导,单核巨噬细胞产生大量生活活性调节子,包括肿瘤坏死因子α(TNF-α)、白介素1、IL-6和一氧化氮(Hirohashi和Morrison,1996.Infect Immun.64(3):p.1011-5)。
因为革兰氏阴性菌内毒素内容物也存在于BG中,在过去的十年用BG进行了安全和毒性研究。进行实验,其中通过使用标准鲎变性细胞溶解物(Limulus amoebocyte lysate,LAL)测定和2-酮-3-脱氧辛酸盐(KDO)测定研究来自病原性大肠杆菌O26:B6和鼠伤寒沙门氏菌C5的菌蜕制备物(Haslberger等人,2000.J Biotechnol.83(1-2):p.57-66;Mader等人,1997.Vaccine.15(2):p.195-202)。平均而言,与马流产沙门氏菌(S.abortus equi)的游离LPS相比,两种菌蜕种均表现出仅2-5%的内毒素活性。使用RAW264.7细胞的细胞培养物实验表明:与游离LPS相比,对于分泌肿瘤坏死因子α(TNFα)和前列腺素E2(PGE2)合成需要多100倍的BG。作者还强调当以<250ng kg-1的剂量(该剂量足够在大鼠中诱导可测量的免疫应答)静脉内施用BG时,在兔中没有记录到显著更低的免疫应答(Mader等人,1997.Vaccine.15(2):p.195-202.)。
在本研究中也进行了细胞毒性研究。即使在进行高达24小时的温育时,也没有观察到大肠杆菌BG本身的杀伤作用。因此,清楚地表明:对于iNOS表达和产生高量的NO的BG相关的连续刺激没有在体外引起细胞死亡。
然而,当细胞与低BG MOI10温育时,观察到生长刺激作用。在相同的处理条件下,当将RV负载于颗粒内部时,该阳性作用甚至更加被增强。然而,在1000BG/细胞施用24小时后,可以进一步表明BG递送的RV引起的细胞毒性作用。产生约10%的细胞存活性降低,它表明BG有效递送RV的另一证据。
除了抗炎作用即iNOS抑制,还表明RV引起体外抗微生物活性(Chan,2002.Biochem Pharmacol.63(2):p.99-104;Faith等人,2006.Antiviral Res.72(3):p.242-51;Schriever等人,2003Atherosclerosis.171(2):p.379-80;Kedzierski等人,2007.Parasitol Res.102(1):p.91-7),并且已经在多种研究中发现其癌化疗能力(综述请参见Saiko等人,2008.Mutat Res.658(1-2):p.68-94)。
