Abstract
本发明禽霍乱蜂胶菌苗及制备方法是一种畜禽疫病制剂及其制备方法,它由A型多杀性巴氏杆菌与蜂胶制成,其含菌量为100亿/ml,含蜂胶干物质10-15ml/ml,它主要通过下列步骤生产,一.制备菌液,1.选苗种;2.育种,3.接种,4.保存,5.灭活,6.保存;二.制备蜂胶液,1.选取蜂胶,2.溶胶,3.配液,4.保存;三.配制菌苗,上述两液按比例混合,由于菌苗佐剂与浓度的改变故使菌苗的保护率提高到80-100%,免疫期延长到6个月,产生作用时间缩短到5天,仅需一次注射即可达到安全有效。

本发明禽霍蜂胶菌苗及制备方法属禽疫病制剂及其制备方法。
目前家禽霍乱主要用药禽霍乱菌苗存在的问题是免疫期短(一般为3个月左右)使用次数多(因介质为AL(OH),浓度为20~40亿/mL),产生作用的时间长(14天),保护率低(50~75%)等问题,因此,给家禽饲养业带来的危害深重,且造成经济损失。
本发明的目的是改变菌苗的介质,提供一种安全性好,无副作用,保护率高,免疫期长,产生作用时间短,使用方便的禽霍乱菌苗及制备方法。
一种禽霍乱蜂胶菌苗及制备方法,其特征在于工艺步骤为,一.制备菌液,1.选取菌种,2.育种,3.接种,4.配液,5.灭活,6.保存;二.制蜂胶液,1.选取蜂胶,2.溶胶,3.配液,4.保存;三.配制菌苗,取菌液保存液与蜂胶保存液混和,使含菌量为100亿/mL,含蜂胶干物质为10~15mg/mL的黄色混悬液,无菌分装后,置4~8℃冰箱内保存备用。
本发明可按下述方法实施:取菌种为由中国兽医药品监察所鉴定,保管和供应的A型多杀性巴氏杆菌,育种在血液马丁琼脂平板上划线培养(37℃,20~22小时)后,取4~6个标准菌落接种于血液马丁琼脂培养管斜面上培养(37℃18~24小时),于培养管中加入无菌生理盐水2ml,洗出菌苔到含血红素马丁琼脂的大扁瓶内培养(37℃,18~25小时),另用经12~18小时培养的马丁汤培养物或生理盐水洗下扁瓶内,培养好的菌苔,调整到菌液浓度为160~200亿活菌/ml,加入福尔马林使最终浓度为0.1~0.3%,再在37~39℃中灭活24小时,制成灭活菌液。
蜂胶佐剂的配制是如下进行的,取已知纯度的蜂胶磨碎,过筛后溶于95%的乙醇溶液中,再根据要求配比成适当浓度的溶液于4℃下保存。
再把上述方法配成的菌液与蜂液佐剂混合配成含菌量为100亿/ml,蜂胶干物量为10~15mg/ml的混悬液,无菌分装于100或500ml的菌苗瓶内,密封保存,检验合格后于4~8℃下保存备用。
本发明的优点在于,佐剂与浓度改变后使菌苗的保护率提高至80~100%,免疫期可延长到6个月,产生作用的时间短到5天,仅需注射一次即可达到安全有效。
Claims (2)
1、一种禽霍乱蜂胶菌的制备方法,其特征在于主剂为A型多杀性巴杆菌、其含菌量为100亿/mL,佐剂为蜂胶,蜂胶干物质含量为10-15mg/mL。
2、根据权利要求1所述的禽霍乱蜂胶菌苗的制备方法,其特征在于菌液制备由下述步骤完成:选取经鉴定所保管的A型多杀性巴氏杆菌育种在血液马丁琼脂平板上划线培养37℃,20-22小时,再取出4-6个标准菌落接种于血液马丁琼脂培养管斜面上培养(37℃20-22小时),用2mL无菌生理盐水把培养管中的菌苔洗出,接种含血红素马丁琼脂的大扁瓶内培养(37℃,18-24小时),再用经12-18小时培养的马丁汤培养物或生理盐水洗出扁瓶内培养好的菌苔,调整到菌液浓度为160-200亿菌/mL,再加入福尔马林使最终浓度为0.1-0.3%,放在37-39℃中灭活24小时制成灭活菌液备用。
