在皮肤病学中使用的皮肤或透皮配制剂开发过程中,体外模型具有巨大的重要性,因为活性成分的吸收和命运仅仅在皮肤中测定,具有很大的难度。治疗效果的定量表征同样是复杂的任务。与对动物的节制和保护有关的法律和伦理规范的一般性认可导致明显减少使用动物模型。但是,在那些旨在每日在皮肤释放恒定量的配制剂的情况下,对活性成分释放的检测具有巨大的重要性。
与活性成分释放有关的检测在下述方面具有显著的重要性:

在软膏剂或其它皮肤药物的治疗应用过程中,三个过程先后或同时出现:(1)活性成分从媒介物中的释放;(2)活性成分渗透进入皮肤的外层;(3)随后渗透进入活皮肤(living skin),在该过程之后有时候活性成分吸收进入循环。当配制剂旨在表面或皮肤使用时,最后的过程通常并非所需,但是在配制剂被设计用于全身效应时,该过程可以起到非常重要的作用。

活性成分的溶解主要取决于温度、活性成分的浓度及其分散度(粒径、形状、粒径分布)。
分布过程主要受到活性成分的溶解度的控制。像溶解性质、分布介质、溶解-改进助剂、温度和溶解方法这类因素可以在所述过程中起到重要作用。
扩散过程主要受到介质的粘度、凝胶结构和物理-化学结构和活性成分的性质及其制备方法的控制。
根据现有技术的膜装置,使用适用于提供平衡扩散的池。这些随后被适合于模拟体内条件的流动池所替代。

根据本发明的装置适用于预测透皮药物配制剂的体内性质。它良好地适用于近似人类皮肤的生理学条件和透皮药物组合物可以在与那些实时应用的那些类似的环境条件下检测。使用本发明的装置,有可能在透皮制剂应用过程中研究表面作用。根据本发明的方法,检测和体外模拟任意透皮制剂,包括但不限于具有高浓度挥发性组分的组合物,例如凝胶、乳剂、外部应用的溶液、悬浮液、粉末、气溶胶等。
根据本发明的装置的系统和它们的联系描述于图4。膜渗透系统由具有开放样品隔室的膜渗透池(C1)、用于控制环境因素的系统、用于受体相流出(stream)的递送系统(1)、取样和分析系统(3)组成。装置受到模块(block)类型或微处理器类型控制器(4)控制。
根据本发明的进一步方面,提供了具有用于检测配制剂的开放样品隔室的膜渗透池。考虑到受体相和受试制剂的性质,根据本发明的池可以由现有技术已知的任意适合的惰性材料制成,例如钢。膜位于池体的上部。膜适用于固定(fastening)的设备,例如螺纹(thread)固定环或弹性固定环原位保持(held)。提供具有受体相入口和出口的池体(其中定位受体相的室),和任选提供夹套适用于附着用于温度调整和控制的液体恒温器(图2,图3)。任选地,在池体外侧提供绝热物(thermal insulation)。通过测量和/或校准精确测定膜侧面的池体横切面面积。池的一个可能的实施方案显示于图2。同样装载乳膏剂样品的池显示于图3。
在描述于图4的流程图中的根据本发明装置的一个实施方案中,具有开放样品隔室的膜渗透池位于在每侧配有玻璃门的柜中。池的一部分是池体(C1),其具有与之附着的膜和弹性固定环和适合作为受体相的入口和出口的流体连接器(connections)。池体由供有用于传感器的镗孔(borings)的AISI 36级别不锈钢模块制成。池体可以从装置移除用于清洁和消毒。池的重量约500g,它显著重于现有技术中已知的常规池的重量。由于池相对大的重量、热容量和紧密设计,不需要对受体相单独温度控制,其原因在于流体流出已经在入口恢复池的温度。精确测定池的横切面面积和体积(分别10.00cm2和3,00cm3),由此有助于计算结果。
池的温度受到由镀镍的黄铜制成的对流散热器(convectiveradiator)(C3)控制,所述对流散热器通过液体恒温器(C4)递送的传热介质加热/冷却。传热介质防冻剂。恒温器是电加热的和如果需要,通过外部冷却单元提供传热介质的冷却。温度可以以0.1℃的精密度(precision)设定和控制。温度传感器(C2)位于池内部。递送与池温度成正比的信号以控制回路(图4,C)。系统的控制速率非常快(时间常量小于30秒)。加热受到可变的宽度平方(width square)波长信号控制。加热元件经固态继电器转换。当需要冷却时,尽可能按需要加热预冷却的传热介质以维持预设温度。
在装置中,使用具有2升甘油-蒸馏水(50∶50,v/v)传热介质的LABOR-MIM ultratermostat(C4,Labor MIM,Esztergom,Hungary)。温度传感器(C2)是NIVELCO CTFP-121-1(Nivelco,Budapest,Hungary)Pt/100/B铂热传感器,通过模块类型OMRON E5CSV-R1T-500(日本)温度控制器提供温度控制(图4,C)。
适用于检测目的的任何类型的膜可以在根据本发明的装置中使用。可能使用天然的膜(例如人类来源或获自哺乳动物的膜,例如猪、大鼠、兔子皮肤或其层,借助于细胞繁殖等获得的膜)或人工膜(生物学或合成获得的材料,例如聚硅氧烷(Silastic)、纤维素或其衍生物、醋酸乙烯酯或丙烯酸酯聚合物制成的膜)。根据本发明的装置适用于使用皮肤膜或包含皮肤层(例如表皮、上真皮、角质层等)的膜,所述膜通过皮肤雾化(dermatomization)、热处理、化学或酶处理等制备。适用于吸收-渗透研究的膜的制备通过现有技术已知的方法进行。
受试配制剂的样品位于用于模拟皮肤或其它生物学屏障的膜得上表面上。样品的量为约0.001-0.1g/cm2计算的膜面积。
根据本发明样品可暴露于具有可控制的流速,温度和如果需要,控制的湿度的空气流。根据本发明的装置适合在预定的温度和湿度通过下述系统提供具有控制的流速的气流:空气递送系统(压缩空气筒或中央压缩器、通气机(ventillator)、HVAC设备等)、任选空气清洁设备,温度控制系统和湿度控制系统。还可能根据现有技术已知的方法测量和记录离开装置的空气的焓和湿度。
根据本发明的一个实施方案,通过电压控制、可变的rpm、低电压通风机(A1)提供空气流。气流的温度借助于电加热单元、空气温度传感器(A2)和控制系统控制(图4,A)。通过用预冷却的气流向通气机进料来冷却,和通过上述方法对其加热。通过变化可能快速控制速率。

通过通气机的电压控制使空气流速控制在0.5-2.5m/s。空气流作为膜表面上的控制电压的函数来校正。
加热元件(A3)的温度受到温度传感器(A2,NIVELCO CTFP-121-1Pt/100/B铂加热传感器)的信号的控制。控制器(C)是OMRONE5CSV-R1 T-500单元。空气流通过受LM7815电压控制器控制的INCA199411()通气机来提供。通气机的电压可以持续在3-15伏特调整。在一个实施方案中,进入装置的空气相对湿度与环境空气相同。但是,空气湿度可以使用现有技术已知的任意方法控制。
应用至膜表面的受试样品可以暴露于光。这同样有助观察在样品中出现的物理-化学变化。根据一个方法,通过与日光发射光谱类似的光发射二极管(L1)(YOLDAL YZ-WS5S20,Sunny-White,最大光强度11.000mcd)提供光照。光暴露通过传感器(L2)测量和L1的发射因此是受控制的。光强度可以使用内部建造的控制器通过L1的激发电压的频率控制来调整。在这种方式中,LED的发射光谱不取决于光强度。
在那些当受试制剂经历具有与正常日光不同的强度或发射光谱的光线(例如当受试制剂在高于海平面的高海拔(其中紫外线辐射的强度显著高于海平面)应用时)的情况下,可能用具有与实时使用环境下所预期相似的发射光谱和强度的不同光源代替所述光源。
如果需要,使用随后同时地或替代地目测评价的成像方法,可以记录来自暴露于环境因素的受试制剂的改变。
渗透的测量如下进行:提供具有恒定流速、组成和温度的受体相的流出,导致所述流出的受体相流过渗透池和测定离开具有开放样品隔室的膜渗透池(1)的排出物流出中的活性物或受试制剂的特征性成分的浓度,所述浓度作为受体相体积或时间的函数(图4)。
活性成分的浓度或受试制剂的特征性组分的浓度可以如下在线进行:测定分析物浓度,其中使用在线分析系统(3),例如分光光度法,使用配有流通比色皿的装置。可供选择地,使用任意分析方法,所述分析物的测定可以通过在预定时间段或排出物体积对排出物受体相流出取样和对分析物浓度测定来进行(图4)。优选地,使用液相测定。还优选使用测定方法,提供选择测定分析物。可以优选使用色谱方法。
在出于在线测定的目的的取样过程中,重要地是保持池出口与膜的相同高度以防止压力在膜的积累。
受体相的递送优选通过泵出所述液体进行。出于此目的,可以使用适用于以需要的流速递送具有所需精密度的受体相的任何泵。受体相在渗透检测实验过程中的流速降至体液,例如血流量的流速的量度(magnitude)内。但是,受体相的流速可受到检测过程中使用的膜的压阻力的影响。在实验过程中,受体相的流速通常为0.5-3ml/min,优选0.1-0.5ml/min。流速的波动相对于实际流速必需不超过1%。优选地,可以使用体积置换泵。池的入口和出口孔可以用合适的螺纹来提供以允许使用低死体积的配管和连接器,形成高效液相色谱系统。
例如,可以使用液相色谱系统的部件用于递送受体相。可以使用洗脱槽作为受体相的贮器(E1)。优选地,可以使用耐热和化学玻璃瓶,例如TORAX玻璃瓶。通过液相色谱系统的高压泵将受体相递送至渗透池。在高效液相色谱系统中使用的泵(E2)可以有利地使用,由于它们的高精密度和低流动波动(图4)。
特别重要地是,防止任意压力在池中积累,其原因在于压力差干扰膜扩散过程。在这种顺序中,保持入口配管的横切面面积小于出口配管的横切面面积,优选入口管的横切面面积是出口管横切面面积的四分之一。
受体相通常是适用于模拟生理学液体的溶液。适合的受体相是纯净水、盐水、磷酸盐缓冲盐水或林格溶液。
根据现有技术已知的方法之一,受体相在使用前脱气,即声处理或气体吹扫。优选地,使用氩吹扫。如果可用,可以使用高效液相色谱设备的在线脱气模块。
本申请发明人使用的作为洗脱递送模块的Shimadzu LC-6A液相色谱或Agilent(Hewlett Packard)HP G 1311A高压泵,其附属于ERMA ERC-3312或Agilent(Hewlett Packard)G 1322A脱气单元适用于受体相递送。
具有出于液相色谱的目的建造的流动池的检测器(S1)还可以用作在线测定分析物的设备(图4)。出于此目的,本申请的发明人使用Agilent(Hewlett Packard)G1315A光电二极管阵列检测器或Shimadzu UV-分光光度检测器以及已经成功使用Beckman荧光检测器。可以根据分析物的性质选择检测参数,诸如检测波长或激发/发射波长。在这情况下,不存对分析物的特殊检测的设备,可以使用质量-敏感的检测器(例如曲光指数或蒸发激光散射检测器)。使用计算机化的数据采集系统获得和评价检测器信号。
装置的控制任选地可以如下进行:通过包括临界参数的测量和控制、数据采集和分析的计算机化的过程控制系统进行,而不使用单个电子或电子机械控制系统。
还可能将多个池和取样/分析系统建造进入一个装置,因此有助于同时检测。
根据本发明的进一步的方面,提供了体外模拟活性成分从药物或化妆品受试制剂释放,特别是模拟吸收-渗透过程的方法。
方法如下进行。受体相过滤和脱气,池脱气和将膜与之附着。在流速、温度、空气流和光强度(通常1-24小时)稳定后,在1-3秒将受试制剂均匀地转移至膜。受试配制剂的量优选是在实时条件下应用的量,或实际上在检测下的制剂的一个剂量单位。
此后,受体相的流动重新开始和在在线分析的情况下,记录制剂的活性成分的浓度或制剂的特征性组分。作为受体相的体积或消逝时间的函数的实际浓度的计算可以通过外部校正来进行。从这些数据,测定溶解速率。可供选择地,在离线分析的情况下,收集级分和在每个级分中的分析物通过现有技术已知的适合的分析方法来测定。从这些数据,测定在相应的级分中存在的分析物的量和计算溶解速率。
实施例1
曲克芦丁5%凝胶的膜渗透检测
受试制剂含有5%曲克芦丁作为活性成分。在检测中,0.9重量%氯化钠溶液用作受体相。受体相的流速为1ml/min。在检测中使用的膜塞璐玢,10cm宽,10cm长。池的温度为34℃。池暴露于天然的白天光照。在第一个(I)检测中,没有应用人工的空气流。在第二个检测组(II)中,线形空气流速为2m/sec。活性成分的浓度通过在349nm的波长经紫外分光法在线测定。
膜连同装置允许稳定60分钟。此后,在2-3秒内,将通过分析精密度测量的约300mg的量的受试制剂均匀地转移至膜上。此后受体相的流动重新开始和记录在受体相中溶解的活性成分的浓度。
在检测组(I)和(II)中溶解的活性成分的量显示在表1(相对于应用至膜的活性成分的量)。

实施例2
使用塞璐玢和人类皮肤膜的包含吡罗昔康的乳膏剂的比较性膜渗透检测
受试制剂含有1%吡罗昔康作为活性成分。在检测中,受体相包含0.9重量%氯化钠溶液。受体相的流速为0.3ml/min。在第一个检测系列(I)过程中使用塞璐玢膜,在第二个检测系列(II)使用人类皮肤膜。池的温度保持在34℃。在两个检测系列过程中,池暴露于天然的日光。没有应用空气流。
膜允许稳定60分钟。
在稳定时间段后,在2-3秒内,将通过分析精密度测量的300-400mg的受试制剂均匀地应用至膜。受体相的流动重新开始和收集每个30分钟时间段的排出物级分。通过高效液相色谱测定每个级分的吡罗昔康和尼泊金甲酯浓度。使用每个分析物的体积和浓度,计算每个级分的吡罗昔康和尼泊金甲酯的量。
使用外标法在反相系统中进行高效液相色谱分析。使用NucleosilC18(250mmx4,6mm i.d.)柱和乙腈-甲醇缓冲液(1000∶660∶340,v/v/v)流动相进行分离。分离在40℃温度进行。流动相的流速为1.0ml/min。检测通过紫外线光电二极管阵列检测器在波长250-450nm进行。在检测系列(I)和(II)中释放的吡罗昔康和尼泊金甲酯(甲基-4-羟基苯甲酸酯)与应用至膜上的量的相对量显示在表2。

Claims (10)
1.用于检测旨在透皮使用的化妆品或药物配制剂的装置,包括受体相调节和递送系统、具有渗透池、优选开放样品隔室膜渗透池的膜渗透系统、取样和任选分析系统和控制系统。
2.膜渗透系统,包括具有开放样品隔室的渗透池、温度控制系统和适用于在膜渗透池的开放样品隔室的表面调整和控制一种或多种环境因素的设备。
3.根据权利要求2的膜渗透系统,其特征在于在膜渗透池的开放样品隔室的可调整的和可控制的环境变量选自光强度、光波长、空气流速、空气温度和空气湿度。
4.具有开放样品隔室的渗透池,包括进料和排出物受体相的入口和出口、池体、膜、用于使膜固定于池体的设备和任选可控制的温度夹套和绝热物。
5.根据权利要求4的池,其特征在于在膜的侧面其体积和横切面面积经过高精密度的测定。
6.用于检测旨在透皮使用的化妆品或治疗药物制剂的方法,包括将样品转移至附着于权利要求3的渗透池表面的膜,使所述样品暴露于预选择的和控制的环境因素,同时将控制流动的恒温受体相递送通过池体和测定排出物中的药物活性成分或受试样品的特征性组分的浓度。
7.根据权利要求6的方法,其特征在于选自温度、光强度、光波长、空气流速、空气温度、空气湿度的一种或多种环境因素在膜渗透池的开放表面和任选膜渗透池的附近受到控制。
8.根据权利要求6或7的方法,其中作为受体相,使用水、盐水、林格溶液、磷酸盐缓冲盐水、表面活性剂的水溶液或蛋白质、优选人血清白蛋白的水溶液。
9.根据权利要求6-8中任一项的方法,其中检测在恒温下进行。
10.根据权利要求1或2的装置,其中使用开放样品隔室膜渗透池。
