据估计约25%的世界作物生产由于微生物腐败而损失,其中真菌导致的腐败是迄今为止最为重要的原因。从经济学角度以及人道主义角度来看,预防食物制品腐败都非常重要。毕竟世界上许多地方的人们都在遭受饥饿。
为了预防农产品例如植物、作物和水果的真菌损伤,目前例如通过喷雾施加杀真菌剂。目前应用的杀真菌剂伴随着若干缺点。
首先,由于所述杀真菌剂的频繁使用,若干真菌已经发展出对某些杀真菌剂的部分或甚至完全的抗性。因此,尽管用目前使用的杀真菌剂进行广泛的处理,但是腐败问题仍然可能发生。

纳他霉素是多烯大环内酯抗霉菌剂,其已作为防腐剂使用超过40年,用于预防由与某些食物(例如乳酪、香肠和饮料如果汁和葡萄酒)相关的酵母或霉菌生长导致的腐败。通过使用Streptomyces natalensis发酵生产的该天然防腐剂在全世界被广泛用作食物防腐剂,并且在食品工业中具有较长的安全使用历史。其非常有效地抵抗所有已知的食物腐败霉菌。尽管纳他霉素在例如乳酪工业中被使用多年,但是迄今为止从未观察到抗性霉菌物种的出现。
纳他霉素在水中具有低溶解度(30-50mg/l),并且只有溶解的级分具有抗真菌活性。因为纳他霉素对大部分真菌而言具有低于10mg/l的MIC(最小抑制浓度),所以溶解的浓度在大部分情况下足以预防真菌的发展。例如在正常条件下对乳酪进行表面处理的情况下,溶解的纳他霉素的变性通过未溶解的纳他霉素的溶解和例如通过喷雾或乳酪涂层供应的纳他霉素在表面上的扩散来补偿。
已有报道称,纳他霉素可以被单独或组合地用于控制农产品中的真菌生长。WO 2005/074687公开了纳他霉素适用于保护香蕉植物抵抗真菌疾病。WO 97/47202中公开了与真菌细胞壁降解酶组合的纳他霉素适用于农产品的抗真菌处理,而WO 2008/009657中表明,与亚磷酸盐/酯组合的纳他霉素能保护农产品抵抗真菌疾病。
在养殖业(farming industry)或农业中使用时,纳他霉素主要作为溶液通过喷雾应用。然而,也可以通过浸渍(dipping)、浸没(immersion)或涂抹(painting)来应用。存在与(例如通过喷雾)在农产品上应用纳他霉素相关的若干缺点。首先,由于纳他霉素溶液的低剪切粘度,纳他霉素可能变得不均匀地分布在农产品表面上。因此,产品的一些部分的表面上可能不具有足够的纳他霉素,而在其他部分中所应用的纳他霉素可形成池。纳他霉素可能甚至流出农产品的表面。其次,纳他霉素可能由于下雨或灌溉而从农产品例如植物上被冲掉。第三,存在随风漂流的风险,即被喷雾在农产品上而不分散的能力较差,导致达到农产品的纳他霉素量减少。所有这些缺点导致纳他霉素效力的降低,因而导致缺乏农产品抵抗真菌腐败的保护。
可以通过对纳他霉素溶液添加增稠剂来消除一些上述缺点。增稠剂例如黄原胶的使用,能提高纳他霉素溶液的粘度,并可导致纳他霉素在产品上更好和更均匀的分布(见EP 0 867 124和US 5,552,151)。
尽管本领域中提出了一些解决方案,但是仍然需要下述纳他霉素组合物,所述纳他霉素组合物在粘附至产品例如农产品表面的方面具有经改进的特性,并因此导致这些产品的微生物腐败的减少。
发明内容
本发明通过提供新的抗微生物剂(例如抗真菌剂)组合物,解决了上述问题,所述组合物包含多烯抗真菌剂和增稠剂。在一个实施方案中,多烯抗真菌剂选自由纳他霉素、制霉菌素(nystatin)、两性霉素B、三烯菌素(trienin)、意北霉素(etruscomycin)、菲律宾菌素(filipin)、钦氏菌素(chainin)、制皮菌素(dermostatin)、lymphosarcin、杀念珠菌素(candicidin)、金色制霉素(aureofungin)A、金色制霉素B、哈霉素(hamycin)A、哈霉素B或鲁斯霉素(lucensomycin)构成的组。在一个优选的实施方案中,多烯抗真菌剂是纳他霉素。在一个实施方案中,组合物也可含有两种或更多不同的多烯杀真菌剂。应当理解,多烯杀真菌剂的衍生物(包括但不限于多烯杀真菌剂的盐或溶剂合物)或经修饰形式的多烯杀真菌剂也可以在本发明的组合物中应用。含有纳他霉素的商业产品的一个例子是商品名为Delvocid的产品。Delvocid由DSM FoodSpecialties(荷兰)生产,其含有50%(w/w)纳他霉素。所述商业产品可以被掺入本发明的组合物中。
本发明的抗微生物组合物通常包含从约0.005g/l到约100g/l的多烯杀真菌剂,优选地,从约0.01g/l到约50g/l。优选地,所述量为0.01g/l到3g/l。在一个优选的实施方案中,多烯杀真菌剂是纳他霉素。
在一个实施方案中,本发明的组合物包含甲基羟基乙基纤维素(MHEC)作为增稠剂。根据本发明的组合物中MHEC的量以组合物中干物质的总量为基础,优选地为按重量计0.05%到4%,尤其是按重量计0.2%到2%。可以使用具有高(例如3000mPas)、中(例如1500mPas)或低(例如400mPas)平均粘度的MHEC。具有高、中或低平均粘度的MHEC类型是本领域技术人员公知的(例如TylopurMHB3000P或WalocelMW400)。通常在20℃下,在2%(w/w)水性溶液中用Hoeppler滚球式粘度计测定平均粘度值。本发明的组合物也可包含两种或更多不同的增稠剂,只要所述增稠剂之一是MHEC即可。在一个优选的实施方案中,组合物仅包含MHEC作为增稠剂。
根据本发明的抗微生物组合物可还包含至少一种UV吸光剂(UV吸收剂)。本发明的组合物可包含从约0.01g/l到约100g/l、优选地从约0.02g/l到约25g/l、尤其是从约0.03g/l到约10g/l的UV吸收剂。在一个实施方案中,上述量存在于即用型溶液中。
本发明的组合物可具有从3.5到8、优选地从5到7的pH。它们可以是固体例如粉末组合物,或者可以是液体。本发明的组合物可以是水性或非水性的即用型组合物,但是也可以是水性或非水性的浓缩组合物/悬浮液,或在使用前必须用合适的稀释剂例如水或缓冲体系稀释的储存组合物、悬浮液和/或溶液。或者,本发明的组合物也可以被用于制备涂层乳液。本发明的组合物也可以具有浓缩干产品(例如粉末、颗粒和片剂)的形式。它们可以被用于制备用于对产品进行浸没或喷雾的组合物,所述产品例如是农产品,包括植物、作物、蔬菜和/或水果。

增稠剂的例子包括但不限于,基于胶乳的产品如Prolong(HollandFyto B.V.,荷兰)和Bond(Loveland Industries Ltd),基于pilonene/萜的产品如Nu-film(Hygrotech Saad)和Spray-Fast(Mandops)和长链多糖。或者,增稠剂可以是聚合物或来自一种聚合物的共聚物,例如聚丙烯酸酯和聚乙烯,例如Neocryl(DSM,荷兰)。本发明的组合物也可包含两种或更多不同的增稠剂。
表面活性剂、去垢剂、乳化剂、铺展剂和湿润剂的例子包括但不限于,阴离子表面活性剂,例如月桂基硫酸钠,或聚乙烯烷基醚或聚氧乙基醚,例如Tween60、61或65。有用的表面活性剂的其他例子是有机硅酮、磺基丁二酸盐、醇乙氧基化物、脂肪酸乙氧基化物、脂肪酸丙氧基化物和商业产品Zipper(Asepta BV,荷兰)。本发明的组合物也可包含两种或更多不同的上述试剂。
合适的防腐剂和抗微生物剂的例子包括但不限于,弱酸防腐剂例如山梨酸、乳酸、苯甲酸、丙酸、柠檬酸、乙酸或它们的碱金属盐或碱土金属盐;无机酸例如盐酸;咪唑;硼砂;亚硫酸氢钙;EDTA钙二钠;脱氢乙酸;异噻唑(例如cit/mit);亚硫酸盐(sulphitites)或硫二噁英(sulphurdioxine);盐;二甲基碳酸氢盐、硝酸盐/酯或亚硝酸盐/酯;和本领域已知的经修饰的空气包装组合物。

合适的分散剂的例子包括但不限于,Morwet D-425、Pluronic P和Silwet L-77。本发明的组合物也可以包含两种或更多不同的分散剂。
合适的流动添加剂的例子包括但不限于,高岭土、滑石、Wacker-BelsilPMS MK、Wacker-BelsilTMS 803、三硅酸镁、铝硅酸钠(sodiumaluminosilicate)、膨润土和聚二甲基硅氧烷。本发明的组合物也可包含两种或更多不同的流动添加剂。
在一个实施方案中,本发明的组合物包含纳他霉素、含亚磷酸根的化合物、MHEC、磺基丁二酸盐和任选地包含防腐剂。
经有效量的根据本发明的抗微生物组合物处理的产品是本发明的另一方面。组合物可存在于产品上和/或产品中。优选地,产品通常对真菌腐败易感,例如是食物产品、饲料产品;农产品;织物;皮革;纸;纤维;涂料;和涂层。

本发明提供了用本发明的组合物处理的产品。经处理的产婆可含有包含本发明组合物的涂层。在一个实施方案中,经处理的产品在其表面上包含从0.000001到200mg/dm2,优选地从0.00001到100mg/dm2,更优选地从0.00005到10mg/dm2的多烯杀真菌剂,例如纳他霉素。在又一个实施方案中,它们在其表面上包含0.05到4mg/dm2,优选地从0.2到2mg/dm2的MHEC(在将10μm的潮湿涂层应用在其表面上的情况下)。还在又一个实施方案中,它们在其表面上包含0.5到40mg/dm2,优选地从2到20mg/dm2的MHEC(在100μm的潮湿涂层应用在其表面上的情况下)。

实施例
实施例1
在橙上,增稠剂对纳他霉素的抗真菌效力的影响
在实验中,检查了包含纳他霉素和不同增稠剂的抗微生物组合物的抗真菌活性。被检查的增稠剂是黄原胶、MHEC(甲基羟乙基纤维素,TylopurMHB3000P)、HPMC(羟丙基甲基纤维素,Methocel)和CMC(羧甲基纤维素,Blanose)。
如下制备含有增稠剂和抗真菌剂纳他霉素的组合物。通过搅拌过夜,将增稠剂溶于水中。之后向含有增稠剂的溶液中添加含有纳他霉素的悬浮液,获得包含最终浓度0.4-0.6%(w/v)的增稠剂和最终浓度1000μg/ml的纳他霉素的组合物。
人工污染新鲜的未经处理的橙。在每个橙上破坏果皮表面区域上的两个点,并用霉菌物种Penicillium roqueforti Thom(CBS 479.84,IBT 21543)的孢子悬浮液污染。使用公知的方法制备霉菌孢子。针对每种抗微生物组合物测试六个橙。污染后将橙在新鲜制备的抗微生物组合物中浸渍,滴干(dripped off)并晾干。将橙于室温下储存在封闭盒中暗处,用透明层板覆盖以便于连续观察。使用公知方法,通过添加水将每个盒顶部空间中的相对湿度提高至95-100%。
结果清楚地证实,与包含纳他霉素和HPMC、CMC或黄原胶的抗微生物组合物相比,含有纳他霉素和增稠剂MHEC的抗微生物组合物更好地保护橙抵抗霉菌生长(见表1)。
在又一个实验中,在橙上检查包含1000μg/ml纳他霉素和0.2%(w/v)上述增稠剂的抗微生物组合物的抗真菌活性。橙得自荷兰的本地零售商。用霉菌Penicillium italicum(ATCC 36041)的孢子处理橙。使用公知的方法获得霉菌孢子。根据Lapere de Bellaire and Dubois(见Plant disease81:1378-1383,(1987))所述方法损伤橙。使用木塞打孔器将橙损伤三次,之后每个创伤用含有106个孢子/ml的5μl P.italicum孢子悬浮液污染每个创伤。在室温下孵育1小时后,将橙在新鲜制备的抗微生物组合物中浸渍1分钟,滴干并晾干。实验一式九份地进行。在如上文所述的条件下将橙储存23天,并视觉判断霉菌发展。
表2中的结果清楚地表明,与包含纳他霉素和增稠剂黄原胶、HPMC或CMC的抗微生物组合物相比,包含纳他霉素和增稠剂MHEC的抗微生物组合物更好地保护橙抵抗霉菌生长。使用包含0.4%(w/v)增稠剂的抗微生物组合物观察到类似的结果(数据未显示)。

实施例2
在橙上,增稠剂对包含纳他霉素和亚磷酸盐的组合物的抗真菌效力的影响
在该实验中,如实施例1中所述在橙上检查包含纳他霉素、亚磷酸盐和上述增稠剂的抗微生物组合物的抗真菌活性,其中橙被储存了13天。
组合物包含处于35mM K2CO3的缓冲液中的最终浓度0.4%(w/v)的增稠剂、1000μg/ml的纳他霉素和240mM亚磷酸钾。

实施例3
草莓上,增稠剂对纳他霉素抗真菌效力的影响
在该实验中,在草莓上检查包含纳他霉素和上述增稠剂的抗微生物组合物的抗真菌活性。
如实施例1中所述制备含有增稠剂和抗真菌剂纳他霉素的组合物。组合物包含终浓度0.6%(w/v)的增稠剂和1000μg/ml的纳他霉素。
用5μl新鲜制备的霉菌物种Botrytis cinerea(ATCC 12481)孢子(106个孢子/ml)的浓缩悬浮液人工污染来自荷兰本地零售商的新鲜的、未经处理的草莓。使用公知方法制备霉菌孢子。污染后一个小时,将草莓在新鲜制备的抗微生物组合物中浸渍1分钟,滴干并晾干。实验一式九份进行。在实施例1所述条件下将草莓储存6天,并视觉判断霉菌发展。
表4中的结果清楚地证实,与包含纳他霉素和黄原胶、HPMC或CMC的抗微生物组合物相比,含有纳他霉素和增稠剂MHEC的抗微生物组合物更好地保护草莓抵抗霉菌生长。

新鲜咖啡豆上,增稠剂对纳他霉素抗真菌效力的影响

如实施例1中所述制备含有增稠剂和抗真菌剂纳他霉素的组合物。组合物包含终浓度0.6%(w/v)的增稠剂和1000μg/ml的纳他霉素。

表5中的结果清楚地证实,与包含纳他霉素和黄原胶、HPMC或CMC的抗微生物组合物相比,含有纳他霉素和增稠剂MHEC的抗微生物组合物更好地保护咖啡豆抵抗霉菌生长。

实施例5
香蕉上,增稠剂对包含纳他霉素和亚磷酸盐的组合物抗真菌效力的影响
在该实验中,在香蕉上检查包含纳他霉素、亚磷酸盐和增稠剂MHEC、HPMC和CMC的抗微生物组合物的抗真菌活性。
如实施例2中所述制备含有增稠剂、抗真菌剂纳他霉素和亚磷酸钾的组合物。组合物在35mM K2CO3的缓冲液中包含最终浓度0.6%(w/v)的增稠剂、1000μg/ml的纳他霉素和240mM亚磷酸钾。
在该实验中使用来自荷兰本地零售商的有机香蕉。用致植物病的霉菌Colletotrichum musae(CBS 19231)的孢子处理香蕉。使用公知方法获得霉菌孢子。根据Lapere de Bellaire and Dubois(see Plant disease 81:1378-1383,(1987))所述的方法损伤香蕉。使用木塞打孔器将香蕉损伤三次,之后每个创伤用含有106个孢子/ml的5μl C.musae孢子悬浮液污染每个创伤。在室温下孵育1小时后,将香蕉在新鲜制备的抗微生物组合物中浸渍1分钟,滴干并晾干。实验一式九份地进行。在如实施例1所述的条件下将橙储存13天,并视觉判断霉菌发展。
表6中的结果清楚地证明,与包含纳他霉素和亚磷酸盐和增稠剂HPMC或CMC的抗微生物组合物相比,包含纳他霉素和亚磷酸盐和增稠剂MHEC的抗微生物组合物更好地保护香蕉抵抗霉菌生长。

另外,还基于炭疽病(antracnose)视觉判断香蕉,所述疾病可以最好地被描述为受影响的创伤周围棕色侵蚀的蔓延(腐败)。结果展示于表7中并且与增稠剂MHEC的组合给出了最好的结果。

Claims (14)
1.抗微生物组合物,其包含纳他霉素和甲基羟乙基纤维素。
2.根据权利要求1所述的抗微生物组合物,其特征在于,其包含从约0.005g/l到约100g/l的纳他霉素。
3.根据权利要求1或2所述的抗微生物组合物,其特征在于,其包含从约0.5g/l到约40g/l的甲基羟乙基纤维素。
4.根据权利要求1到3中任一项所述的抗微生物组合物,其特征在于,所述组合物还包含至少一种选自下组的另外的化合物,所述组由UV-吸收剂、粘着剂、表面活性剂、乳化剂、去垢剂、防腐剂、稳定剂、铺展剂、抗氧化剂、抗成泡剂、湿润剂、其他抗微生物剂、填充剂、喷雾油、分散剂和流动添加剂组成。
5.根据权利要求1到4中任一项所述的抗微生物组合物,其特征在于,是所述组合物还包含亚磷酸盐/酯。
6.根据权利要求1到5中任一项所述的抗微生物组合物,其特征在于,所述组合物是固体或液体。
7.经有效量的、根据权利要求1到6中任一项所述的抗微生物组合物处理的产品。
8.根据权利要求7的产品,其为农产品。
9.根据权利要求7或8所述的产品,其通常对真菌腐败易感。
10.用于处理产品的方法,所述方法包括下述步骤:向所述产品中掺入和/或在所述产品上应用根据权利要求1到6中任一项所述的抗微生物组合物。
11.根据权利要求10所述的方法,其中所述产品中和/或所述产品上的真菌生长被预防。
12.根据权利要求10或11所述的方法,其中所述产品的储存期限被提高。
13.根据权利要求1到6中任一项的组合物用于预防产品中和/或产品上真菌生长的用途。
14.根据权利要求10到12中任一项所述的方法或根据权利要求13所述的用途,其特征是所述产品是农产品。
