Abstract

发明背景
本发明涉及含金属的、治疗的、抗微生物的和抗真菌的化合物。更具体而言,本发明涉及N-杂环卡宾的金属配合物及其作为抗微生物剂、抗真菌剂和放射性药物组合物的用途。
长久以来,银由于其抗微生物性能而被使用。这种使用早于对其机制的科学或医学了解。例如,古希腊和罗马人使用银币保持水的纯度。现今,银仍然被美国国家航天航空局(NASA)在其航天飞机上用于同样的目的。在1800年前,就开始使用硝酸银治疗很多医学状况。现今,1%硝酸银溶液仍被广泛使用,在婴儿出生后预防淋菌性眼炎(gonorrheal opthalmia)。自从至少在19世纪的后期,银以很多不同的形式被施用,以治疗和预防很多类型与细菌有关的病痛。
其它治疗,例如将银箔施用于术后伤口以预防感染,在欧洲作为医疗实践保留到20世纪80年代,而且硝酸银依然被用作局部抗微生物剂。在20世纪60年代,开发了非常成功的烧伤治疗银配合物,磺胺二嗪银(silver sulfadiazine),示于在下面式1中。商业已知为Cream 1%,该配合物一直是用于预防二度和三度烧伤感染的最有效治疗之一。已经显示磺胺二嗪银对很多革兰氏阳性和革兰氏阴性细菌具有优良的抗微生物性能。据认为,银在表皮创伤区域的缓慢释放负责治愈的过程。对外科受伤鼠的研究表明了硝酸银和磺胺二嗪银对帮助治愈过程的有效性。尽管还未了解这些现象的完整机制,然而通过使用这些普通的银抗微生物剂,减少了伤口的炎症和肉芽形成。1磺胺二嗪银
最近开发的银涂布技术已导致被称作Acticoat的烧伤敷料的产生。该敷料的目的是在提供抗感染屏障的同时避免粘着到伤口。一些临床试验也已经证明,对比用硝酸银处理的传统的伤口敷料,该敷料可轻松除去。Acticoat已经显示在抗微生物功能上的提高,超过硝酸银和磺胺二嗪银。Acticoat由纳米晶体银颗粒组成。抗生素抗性菌株已发展到抗硝酸银和磺胺二嗪银,但还未发展到抗纳米结晶银。纳米结晶银更广泛的活性范围明显是由于银阳离子和不带电银种类的释放。由于传染原(infectious agent)的抗生素抗性菌株不断出现,对新型抗生素存在需求。
在其它治疗应用中,金属化合物也起着重要的作用。在放射性药物领域可以看到金属的有用性的例子。使用放射疗法破坏肿瘤细胞是众所周知的,但在治疗后肿瘤可以再现。肿瘤内的缺氧细胞(hypoxiccell)对X射线辐射的抵抗性是其它肿瘤细胞的2.5至3倍。因此,这些细胞更有可能在放射疗法或化学疗法后存活,并导致肿瘤再次出现。将放射性核素靶向缺氧细胞将作为使它们可见的方法。
γ射线辐射体例如99Tc的配合物作为显象剂特别有用,而且治疗性放射性药物如89Sr、153Sm、186Re和166Ho在骨肿瘤的治疗中是重要的。Rh-105发射319keV(19%)的γ射线,该射线允许在活体内跟踪并允许剂量测定法计算。通过使用整个周期表,可以利用很多更具放射性的核,以构建诊断或治疗剂。
放射性金属的配合物的有效性高度取决于螯合配体的本质。成功的金属药物必须既靶向特定组织或器官又可从其它组织中快速清除。另外,对显象和肿瘤治疗而言,靶器官或组织必须对放射性药物具有最佳的暴露。因此,对设计用于结合放射性金属的新型配体系统存在需求。
发明概述
虽然以前已知一些N-杂环卡宾金属配合物(metal complex ofN-heterocyclic carbenes),然而,N-杂环卡宾银配合物用作抗微生物剂还没有被认识到。同样,N-杂环卡宾和放射性金属的配合物可以被用作放射性药物也没有被认识到。强螯合配体,例如在此所述的螯状N-杂环卡宾(pincer N-heterocyclic carbenes),可以提供用于产生放射性药物配合物的可替代的更具优势的路线。
因此,提供抑制微生物生长的方法是本发明的一个方面。通过使微生物暴露于N-杂环卡宾的银配合物(silver complex ofN-heterocyclic carbene)而抑制微生物生长。
提供治疗癌细胞的方法也是本发明的一个方面。通过使癌细胞暴露于N-杂环卡宾和放射性金属的配合物而治疗癌细胞。因此,提供新型N-杂环卡宾也是本发明的一个方面,当其与银络合时用作抗微生物剂是有用的,而且当其与放射性金属络合时,作为放射性药物是有用的。
提供合成放射性药物的方法是本发明进一步的方面。提供合成抗微生物化合物的方法也是本发明的一个方面。
上述方面的至少一种或多种,及其超越与治疗感染有关的已知技术的优势通过这里所描述的和要求保护的本发明得以实现,这些优势从随后的说明书可见是明显的。
一般地,本发明提供用于抑制微生物生长或真菌生长的方法,包括施用有效量的N-杂环卡宾的银配合物这一步骤。

本发明也提供用于合成抗生素化合物的方法,包括:使咪唑鎓盐与过渡金属或碱反应而生成N-杂环卡宾;和使N-杂环卡宾与银化合物反应而生成具有N-杂环卡宾的银配合物。
本发明也提供用于治疗癌细胞的方法,包括施用有效量的N-杂环卡宾和放射性金属之配合物的步骤。

在本说明书和所附权利要求中,单数形式“一个(a)”,“一个(an)”和“该(the)”包括复数指代,除非上下文另外清楚指示。除非另外被定义,在此所使用的所有技术和科学术语具有与本发明所属领域中的普通技术人员所一般理解的相同的含义。

在一个实施例中,N-杂环卡宾为由式4或5表示的双齿卡宾,其中R1为一个C1-C6烷基或C1-C6羟烷基,R2为氢原子。在一个具体实施例中,N-杂环卡宾由式4或5表示,其中R1为一个C2-C3羟烷基,R2为氢原子。在另一个实施例中,N-杂环卡宾由式4表示,每个相邻的R1和R2一起形成一个取代烷基。

一般地,咪唑鎓盐为N-杂环卡宾的直接前体(immediateprecusor)。可以使用几个步骤,以将咪唑鎓盐转化为相应的N-杂环卡宾。通过在溶剂例如THF和液态氨中与碱例如KOtBu、KH和NaH的脱质子化作用,可以从咪唑鎓盐产生N-杂环卡宾。可分离的N-杂环卡宾可以代替很多过渡金属配合物上的二电子给体(例如四氢呋喃,一氧化碳,腈,膦和吡啶),生成N-杂环卡宾过渡金属配合物。然而,分离卡宾不总是可行的。
在合适的过渡金属配合物存在下,由相应的咪唑鎓盐的脱质子化作用,原位生成N-杂环卡宾,也可以获得N-杂环卡宾配合物。在金属配合物上的碱性配体,例如氢化物,醇盐或乙酸盐,可以使咪唑鎓盐脱质子而形成容易结合金属上的空配位点的N-杂环卡宾。例如,已经显示Pd(OAc)2与很多咪唑鎓盐(imidazolium salt)反应,形成钯-卡宾配合物。
也可以用无机或有机碱处理咪唑鎓盐而生成卡宾。已经显示,咪唑鎓盐与含碱性取代基的金属的反应对卡宾的过渡金属配合物的合成非常有帮助。碱性氧化物Ag2O与咪唑鎓盐的结合可以被用于生成银-卡宾配合物。使用银-卡宾配合物作为卡宾转移剂(carbene transferreagent)已被用于提供金(I)和钯(II)的卡宾配合物。已经以这种方式使用银-卡宾配合物以提供具有Pd-卡宾和Cu-卡宾键的配合物。使用卡宾转移剂,形成过渡金属-卡宾键在很多情况中是有利的,原因在于这些反应是在温和条件下,而且不使用强碱的情况下进行的。
例如,2当量n-丁基咪唑或甲基咪唑和1当量二碘甲烷在回流THF中的缩合以高收率提供了如式8a或8b所示的咪唑鎓盐。如式8a或8b所示的物质在水中与Ag2O结合,分别形成了水溶性银二聚体9a和9b。9a和9b的阳离子部分的热椭圆体图(thermal ellipsoid plot)被表示如下。
通过N-丁基咪唑与2,6-双(卤甲基)吡啶分别以2∶1的摩尔比反应可容易地获得螯状配体(pincer ligand)二卤化2,6-双-(n-丁基咪唑鎓甲基)吡啶(2,6-bis-(n-butylimidazoliummethyl)pyridine dihalide)(化合物16a和16b)。在CH2Cl2中配体16a与Ag2O容易反应,产生银卡宾配合物17。配合物17在空气和光中是稳定的。17的TEP
螯状N-杂环卡宾与作为桥接单元的吡啶的合成概况如下表述。二当量咪唑钾与2,6-双(溴甲基)吡啶反应,以70%的收率产生化合物19。由式18表示的化合物与2-溴乙醇或3-溴丙醇结合,分别产生了19a和19b。19a或19b的Br盐与等摩尔量Ag2O结合,分别产生银双卡宾聚合物20a和20b。化合物20a已经被晶体学表征。由20a和20b表示的溴化盐易溶,而且在水中缓慢分解,在含有该任一化合物的烧瓶壁上生成银镜。20a及其丙醇类似物20b是有效的杀菌剂。这些配合物的衍生物可以使用组氨酸作示例性前体进行合成,如下所述,以改进它们的抗微生物性能(antimicrobial property)。20a的TEP
参照硝酸银,使用LB肉汤稀释技术在酵母和真菌(白色念珠菌(Candida albicans),黑曲霉(Aspergillus niger),毛霉菌属(Mucor),酿酒酵母(Saccharomyce cerevisiae))上研究了水溶性银(I)N-杂环卡宾20a的抗微生物活性,并在具有临床重要性的细菌(大肠杆菌(E.coli),金黄色葡萄球菌(S.aureus),绿脓杆菌(P.aeruginosa))上做了同样的研究。采用Kirby-Bauer琼脂扩散(滤纸片)过程的银化合物的敏感性试验显示银(I)N-杂环卡宾在所有细菌上表现出和硝酸银一样有效的抗微生物活性,该试验是通过测量生长抑制区而进行的,使用银化合物溶液浸渍的滤纸片,滤纸片被放置在琼脂板上的生物体菌苔上。考察了包含多种银化合物浓度和细菌或真菌的过夜培养物的生长。对于每种生物体而言,含每种银化合物的最小抑制浓度(MIC)的试管被用于接种琼脂板,以证实在该培养物中没有能活下去的生物体。与硝酸银相比,在7天的试验期间,化合物20a在更低的浓度下对细菌和真菌是有效的,而且具有更长的银活性期。对大鼠的毒性试验显示,配体19a、20a的前体和20a降解时形成的物质是低毒的,而且在两天之内通过肾清除,这是通过尿的质谱分析而测定的。

通过咪唑鎓盐前体44与氧化银的相互作用,可以合成单齿N-杂环卡宾银配合物,例如那些由式48表示的化合物。如上所述,可以选择侧链R,以便修改配合物的水溶性、亲油性和其它性能。例如,R可以为氢或选自烷基、环烷基、链烯基、环烯基、炔基、芳基、芳基烷基、烷基芳基、杂环和烷氧基及其取代衍生物的C1-C12有机基团。例如那些由式46和47表示、分别由组胺和组氨酸合成的银配合物可以被合成并被用作抗微生物化合物。因为组胺和组氨酸存在于体内,因此当其衍生物被用作局部抗微生物剂时,被期望可引起最少的皮肤刺激,并且被期望作为具有优异毒物学性能的内部抗微生物剂而产生极有限的问题。
下面提供了具有桥接两个N-杂环卡宾(参见式3)的亚甲基(methene)或亚甲基(methylene)并具有附加取代基的螯状N-杂环卡宾的合成。可以选择取代基,目的是为总配合物提供足够的溶解性、亲油性或其它性能。在下面所讨论的步骤中,吡啶环和咪唑担当基本的结构单元(building block)。基于上述化合物8a和8b的合成,两当量化合物58将与二碘甲烷结合,形成化合物59。化合物59与HCl反应,开环生成化合物60。因为伯胺比咪唑氮更活泼,所以一当量烷基卤将容易地加成到化合物60的伯胺上,形成化合物61。第二个烷基卤将加成到化合物61的第二咪唑氮上,形成双咪唑鎓盐阳离子,如化合物62所示。双咪唑鎓盐阳离子62可以与Ag2O结合,形成如式63所示的银配合物,类似于上述化合物9a和9b。
可以用HCl处理化合物27,生成化合物64,然后使化合物64与含增溶取代基的衍生烷基卤接触,生成化合物65。用羧酸和二环己基碳二亚胺(DCC)也可以使化合物64衍生而形成酰胺键。化合物65在较高温度下与类似地含有增溶取代基的衍生烷基卤结合,将生成如式66所示的咪唑鎓盐双阳离子,其可以进一步与金属例如铑络合。
银-卡宾配合物也可以被用作卡宾转移剂(carbene transferreagent),以产生其它卡宾配合物。使用卡宾转移剂,在很多情况下对过渡金属-卡宾键的形成是有利的,因为反应是在温和的条件下而且不需要使用强碱而进行的。例如,8b与Pd(OAc)2在DMF中结合,然后在乙腈中用NaI处理,导致式8c所示化合物的形成。该化合物的热椭圆体图如下所示。类似地,8b与PtCl2和乙酸钠在DMSO中结合,以50%的收率生成由式8d表示的化合物。化合物8d的X射线热椭圆体图被显示如下。8c的TEP8d的TEP
由式8a所示的咪唑鎓盐与[(1,5-环辛二烯)RhCl]2在回流MeCN中在NaOAc和KI存在下结合,以80%收率生成铑卡宾8e。已通过1H和13C NMR以及X射线晶体学表征了该化合物。该铑配合物在延长的时间期间内是水稳定的。相关的螯合双-卡宾铑配合物已经被合成,而且已显示足够稳定,可用在催化过程中。8e的TEP

对于吡咯桥接的螯状N-杂环卡宾的合成,在甲基上具有离去基团的2,5-双二甲基吡咯在本发明的合成方法中是特别有用的。氯化二甲铵在甲醛水溶液中与吡咯进行曼尼希反应,生成2,5-双二甲基氨甲基吡咯,由式70表示。二碘甲烷加入到在TNF中的吡咯70中,生成二碘化2,5-双(三甲基氨甲基)吡咯(2,5-bis(trimethylaminomethyl)pyrrolediiodide)(式71)。
含有2-硝基咪唑基的分子被认为是被靶向于缺氧细胞。这些化合物在硝基咪唑基上被还原,并被捕获到具有低氧环境的细胞内。2-硝基咪唑基与螯状N-杂环卡宾结合而形成由式73表示的化合物,可以如下进行。期望由式72表示的化合物与双咪唑以2∶1的比例缩合而生成由式73表示的化合物。类似地,可以合成具有多种连接物片段的2-硝基咪唑的其它衍生物。连接物基团的种类,包括聚环氧乙烷(PEO),将考虑到相对于靶向基团定位螯合剂时的柔性,以及考虑到化合物的辛醇/水的分配系数的变化,这与通过肾的清除相关。也可想象类似73的铑配合物的形成。可以使用类似的步骤合成含有硝基咪唑和增溶取代基的75和76的衍生物。
可以使用在此所示的金属的同位素作为N-杂环卡宾配合物的组分,以形成放射性药物。例如,可以使用105Rh代替Rh。105Rh具有1.5天的合宜的半衰期,而且也放射相对低水平的γ辐射。铑的此种同位素通过β发射分解为105Pd,一种钯的稳定的天然存在的同位素。其它可利用的同位素可以选自过渡金属、来自镧系的元素和来自锕系的元素。优选的同位素为Ag、Rh、Ga和Tc。
如上所述,本发明包括金属N-杂环卡宾配合物,其可以从几种N-杂环卡宾前体,咪唑鎓盐制备。从生物类似物例如嘌呤碱得到的咪唑鎓盐可以容易地与氧化银(I)在合适的溶剂中反应,获得银-N-杂环卡宾配合物,嘌呤碱包括黄嘌呤、次黄嘌呤、腺嘌呤、鸟嘌呤及其衍生物。可以容易地将咪唑鎓盐阳离子(imidazolium cation)分类为单-咪唑鎓盐阳离子,例如那些由式77-81表示的阳离子,双-咪唑鎓盐阳离子,例如那些所表示的阳离子。
优选的单-咪唑鎓盐阳离子包括那些由式48-52表示的阳离子:其可以被用于形成优选的单齿N-杂环卡宾银配合物,例如那些分别具有式53-57的配合物。通过咪唑鎓盐前体(imidazolium precursor)48-52分别与氧化银的相互作用,可以合成在式53-57中所示的卡宾银配合物。
桥接的双-咪唑鎓盐阳离子可以由Z表示。其中Z可以为亚甲基、杂环基(heterocyclic group)、二甲基杂环基、二甲基环烷基、二甲基取代杂环基、芳基、二甲基取代芳基。双-咪唑鎓盐阳离子可以通过Z1和Z2的桥接而形成环(环芳),其中Z1和Z2每个可以独立或结合起来,并选自杂环、C1-C12取代杂环、芳基、C1-C12取代芳基、C3-C12取代酮和C1-C12链烯基。每个R基团;R1、R2、R3和R4官能度和咪唑鎓盐的抗衡阴离子(counter anion)X可以被修饰,以改善化合物的亲油性。X抗衡阴离子可以来自卤化物、碳酸盐、乙酸盐、磷酸盐、六氟磷酸盐(hexafluorophosphate)、四氟硼酸盐、硝酸盐、硫酸二甲酯、氢氧化物和硫酸盐。每个R基团(R1,R2,R3和R4)可以独立地或结合起来选自氢、C1-C12烷基、C1-C12取代烷基、C1-C12烷氧基、C1-C12环烷基、C1-C12取代的C1-C12环烷基、C1-C12链烯基、C1-C12环烯基、C1-C12取代环烯基、C1-C12炔基、C1-C12芳基、C1-C12取代芳基、C1-C12芳基烷基、C1-C12烷基胺、C1-C12取代烷基胺、C1-C12烷基戊糖磷酸盐(alkylpentose phosphate)、C1-C12酚和C1-C12酯。在一些它的药物学应用中,对R1、R2、R3和R4官能度的选择是期望的。
作为携带银的卡宾前体,也对嘌呤进行了研究。特别感兴趣的是鸟嘌呤,其为DNA中的核碱基(nucleobases)之一。鸟嘌呤91具有类似于咖啡碱95的环状系统。因为鸟嘌呤无毒,因此7,9-二甲基鸟嘌呤具有低毒性似乎是合理的。这使得二甲基鸟嘌呤配体对于囊性纤维化(cystic fibrosis)研究非常有吸引力,因为我们正在寻找可用作银阳离子载体的无毒而且小的配体。
鸟嘌呤91与硫酸二甲酯二甲基化作用,然后用氢氧化铵处理,生成水不溶性的7,9-二甲基鸟嘌呤两性离子92。HBr加入到两性离子92中生成溴化盐93。该溴化盐溶于水中,而且使用THF可被沉淀出来。通过使溴化盐悬浮在DMSO中,将Ag2O加入到该溶液中并在60-80℃加热大约6小时而形成银配合物。
黄嘌呤已经用作支气管扩张药(bronchodilator)很多年,用于治疗囊性纤维化(cystic fibrosis)患者身上的导气管梗阻(airwayobstruction)。因为黄嘌呤含有咪唑环,我们假定使它们烷基化而形成咪唑鎓盐及最终形成银卡宾配合物应当是可能的。因为它们用作支气管扩张药,我们也假定它们的甲基化衍生物会是相对无毒的。可能最熟知的黄嘌呤为咖啡碱95。我们已经考察了形成甲基化咖啡碱的咖啡碱的烷基化和用咖啡碱作为卡宾前体的银卡宾配合物的形成。甲基化咖啡碱已经证明比咖啡碱毒性更小。
通过咖啡碱95与硫酸二甲酯在硝基苯中反应,生成甲基化咖啡碱的硫酸甲酯,1,3,7,9-四甲基黄嘌呤盐(1,3,7,9-tetramethylxanthanium),96a。在水中使用NH4PF6的阴离子交换产生96b。
配体96a是水溶的,而且在水中与Ag2O反应生成配合物97a。97a在水中稳定存在达5天。C-107Ag和C-109Ag连接物(couplings)的缺乏在13C NMR时标上显出立体易变行为(fluxional behavior),正如对很多银(I)配合物的观察。同样,96b与Ag2O在DMSO中反应形成97b,已经通过X射线晶体学对97b进行结构表征。96b和97b的阳离子部分的热椭圆体图(TEP)被显示如下。96b的TEP97b的TEP
咖啡碱1,3,7-三甲基黄嘌呤为一种黄嘌呤衍生物,黄嘌呤衍生物一般在医药中被用作利尿药、中枢神经系统兴奋剂和环腺苷一磷酸(c-AMP)磷酸二酯酶的抑制剂。使用修改的文献步骤合成碘化1,3,7,9-四甲基黄嘌呤鎓盐(1,3,7,9-tetramethylxanthinium iodide)(甲基化咖啡碱),其为咪唑鎓盐,并通过1H,13C NMR,质谱法和X射线晶体学进行表征。98的TEP
二当量碘化1,3,7,9-四甲基黄嘌呤鎓盐与三当量氧化银(I)在甲醇中室温下反应,生成化合物99。100的TEP
99在甲醇和乙酸乙酯的混合物中结晶,生成化合物100,其为无色晶体,溶于水而且是空气稳定的。通过1H,13C NMR和质谱法对化合物99和100进行表征。X射线晶体学被用于确定100的分子结构,其热椭圆体图如上所示。使用滤片试验(filter disk test)和标准MIC技术评价100的抗微生物性能。已发现化合物100对金黄色葡萄球菌(S.aureus),绿脓杆菌(P.aeruginosa)和大肠杆菌(E.coli)具有有效的抗微生物活性。评价对化合物98的剂量反应效应(dose-responseeffect),以测定该化合物对鼠的毒性。毒性研究为用于测定杀死半数动物(鼠)所需的致死量(LD 50)的标准方案(standard protocol)。对化合物98的LD 50评估值为每Kg鼠2.37g。用在本研究中的方案被Institutional Animal Care and Use Committee(IACUC),University ofAkron批准。
施用有效量的N-杂环卡宾的过渡金属配合物用于活体外(in-vitro)和活体内(in-vivo)医学应用的给药方法由气溶胶、生物可降解聚合物、聚合胶束(polymeric micelles)、水凝胶型材料(hydrogeltypes materials)、树枝状聚合物(dendrimer)和改性C-60富勒烯(C-60fullerenes)组成。
为了展示本发明的实践,合成了两个N-杂环卡宾101和102,并如下所述对其抗微生物性能进行试验。这些化合物可以参照式4所示。其中R1为羟乙基或羟丙基,R2为氢原子。通过使2,6-双-(咪唑甲基)吡啶与2-碘乙醇或3-溴丙醇反应而合成这些卡宾101和102,以提供式101和102的化合物。
这些化合物的IR光谱在3325cm-1处显示O-H伸缩谱带振动。从在硝基苄基基体(nitrobenzyl matrice)中的这些化合物所得到的FAB-MS光谱在m/z 456处显示[51][I]+(C17H23N5O2I),在m/z 436处显示[52][I]+(C19H27N5O2Br)。这些化合物可容易地与Ag2O反应,以高收率形成银-双(卡宾)钳状配合物(silver-bis(carbene)pincer complex)103和104。
在这些化合物的1H NMR光谱中9.13ppm、9.36ppm处咪唑鎓盐质子(imidazolium proton)的损失,以及在这些化合物的13C NMR中181ppm处共振的出现,证实了化合物103和104的形成。关于化合物103形成及结构的进一步证据由X射线晶体学提供。
通过缓慢蒸发化合物103的甲醇溶液,获得化合物103的无色晶体。有趣的是,化合物103在甲醇水溶液中经历了完全的阴离子交换,用氢氧离子代替碘离子。在固态状态,化合物103以一维线性聚合物存在,如在图1中所示。图1为化合物103的热椭圆体图,其热椭圆体在30%的概率水平(probability level)下被绘制。为清晰起见,氢原子从图1中被省略。
在银原子处的几何形状接近线性,其中C5-Ag1-C15的键角为174.7(4)°,Ag1-C5和Ag1-C15的键长分别为和质谱法表明,在溶液和气相中,化合物103以单体存在,而X射线晶体学显示,化合物103在晶体中是聚合的。
化合物103与六氟磷酸铵水溶液的阴离子交换反应导致化合物105形成。在固体状态,化合物105以二聚体存在,如在图2中所示。图2为化合物105的热椭圆体图,其热椭圆体在30%概率水平下被绘制。为清晰起见,氢原子从图2中被省略。银原子的几何形状接近线性,其中键角:C32-Ag1-C5(175.7(4)°),C22-Ag2-C17(174.6(3)°),键长: 阴离子的性质对化合物103相对化合物105的结构变化是重要的,而且阴离子的选择对这些化合物的溶解性具有显著的影响。例如,化合物103在水介质中是可溶的,而化合物105则不是。表1给出了这两种化合物的晶体数据总结。表1
评价了化合物103和105作为抗微生物剂的有效性。使用标准琼脂板覆盖(agar plates overlay)方法,获得如在表2中所示的敏感性数据。在该试验中,用20μl已知浓度的银化合物浸透直径为6mm的滤纸盘,并将其放置在琼脂板上的微生物的菌苔上。过夜培养后,测量微生物的生长被试验溶液所抑制的区域的直径,作为对银化合物的相对抗微生物活性的测量。试验微生物为大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphylococcus aureus)和绿脓杆菌(Pseudomonasaeruginosa)。硝酸银为所用参考标准,而化合物101和102作负对照。表2.银化合物的抗微生物活性
数据证实,化合物103和105具有抗微生物性能,其水平比得上硝酸银,如在表2中所示。螯状配体(pincer ligand)化合物101和102被发现没有抗微生物活性。
根据最小抑制浓度测定方法(MIC)也对银化合物进行了试验。MIC是用于评价抗微生物剂的抑菌活性的标准微生物技术。在该情况下,MIC基于可得到的银的总量,而不是基于银离子的浓度。试验了每种银化合物103和105的0.5%(w/v)溶液。在将银配合物溶解在培养基(LB肉汤)中时,在所有样品中都观察到AgCl沉淀。评价银配合物溶液的上清部分的稀释系列的活性,其中每天加入恒定体积的新生长的微生物(20μl)。再次使用大肠杆菌(Escherichia coli),金黄色葡萄球菌(Staphylococcus aureus)和绿脓杆菌(Pseudomonasaeruginosa)作为试验微生物。通过目测检查培养物的生长(+)或无生长(-),获得MIC,如在表3中所汇报。在表3中,DF为稀释因子。从结果可以推断,与硝酸银相比,化合物103和105与氯离子(chlorideion)不易结合,原因在于Ag-C给体配位键的稳定性。因此,化合物103和105显示出比硝酸银更好的抗微生物活性。当考虑用于活体内应用的银化合物时,这是化合物103和105的期望性能。可注意到,尽管使用等重的银化合物,然而,由化合物103和105所释放的银离子的量比由硝酸银所释放的银离子的量低大约2.7倍。表3银化合物的上清液的MIC结果(较少氯化银)
尽管不希望在任何具体理论上以可获得专利为条件,但据信,化合物103和105的活性和稳定性以及其在水中的溶解性可以归功于Ag-C给体配位键随时间分解为银金属和银离子的相对缓慢。

测定最小致死浓度,以评价由式103和105表示的化合物的杀菌性能。使用具有最低Ag化合物浓度的培养基的透明(无生长)部分,通过使用灭菌环在琼脂板上划线0.01ml的溶液,随后在37℃培养24-48小时。目测计数菌落,其中最小杀菌浓度(MBC)的终点被看作在琼脂板上无生长。长达培养和MBC试验的第七天,与硝酸银比较,试验化合物显示出改进的杀菌效应,对于银化合物而言,在培养和试验的第十天之后观察到无生长。尽管事实是在整个培养期内,新生长的微生物每天都被加入到含银化合物的培养基中。103和105的杀菌和抑菌性能被认为是由于Ag-C给体(卡宾)配位键随时间缓慢分解为银金属、银离子、AgCl,以及它们在水中的溶解性。
烷醇N-官能化银卡宾配合物103和105在水介质中是可溶的。另外,它们已经证明是有用的抗微生物剂,而且它们在水中的可溶性使它们成为可以用于活体内应用的极佳的银化合物。银配合物在氯化物溶液中的溶解性和稳定性已经成为限制银配合物用于活体内应用的关键因素。
根据本发明的另一个方面,合成了银(I)咪唑环芳偕二醇配合物(silver(I)imidazole cyclophane gem diol complex)106[Ag2C36N10O4]2+2(x)-,其中,x=OH-,CO3 2-。MIC试验显示,在具有大约等量银情况下,水溶液形式的106的抗微生物活性比0.5%AgNO3的效果小2倍(fold)。当通过静电纺丝(electrospinning)(技术)将106包封到中以获得由纳米纤维制作的层(mat)时,106的抗微生物活性得以增强。纤维层(fiber mat)释放银纳米粒子的聚集体,并在很长一段时间内维持纤维层的抗微生物活性。通过包囊,106的杀菌活性速率被极大地改善,而且所用银量被减少很多。具有75%(106/tecophilic)的106的纤维层含有2mg银,其比16mg(0.5%)的AgNO3低8倍,比1%磺胺二嗪银膏(10mg)低5倍。已发现纤维层以与0.5%AgNO3相同的速率杀死金黄色葡萄球菌(S.aureus),其中在琼脂板上为零菌落,而且比磺胺二嗪银膏快大约6小时。在上面进行试验并被发现有效的接种物为大肠杆菌(E.coli)、绿脓杆菌(P.aeruginosa)、金黄色葡萄球菌(S.aureus)、白色念珠菌(C.albicans)、黑曲霉(A.niger)和酿酒酵母(S.cerevisiae)。使用透射电子显微镜和扫描电子显微镜检术表征纤维层。通过对鼠静脉内给药,评价配体(二氯化咪唑鎓盐环芳偕二醇(imidazolium cyclophane gem dioldichloride))的急性毒性,其LD 50为100mg/Kg鼠。
本发明的静电纺丝纤维(electrospun fiber)可以密封银(I)N-杂环卡宾配合物。水溶银(I)卡宾配合物107,氢氧化银(I)-N-螯合2,6-双(羟乙基咪唑甲基)吡啶(silver(I)-N-pincer2,6-bis(hydroxylethylimidazolemethyl)pyridine hydroxide)在一些临床上重要的细菌上的抗菌活性被描述如上。化合物107为少量溶于纯乙醇中但完全溶于甲醇中的化合物的实例。通过改变与双(咪唑甲基)吡啶化合物的亲核端基耦合的官能化基团,1型银(I)卡宾配合物在乙醇中的溶解性得到改善。尽管在方程1中显示的是其中m=2和m=3的实施方案,但m可以是至少为1的任何正整数,优选地,m为大约1至大约4范围内的值。此外,在不背离本发明的范围的情况下,可以使用上述替代的原材料或前体,生成期望的银(I)卡宾配合物。下面所说明和描述的具体实施方案在描述本发明中被用于阐明性目的。方程1
静电纺丝(Electrospinning)是一种通用的方法,其用于通过产生聚合物溶液或聚合物熔体的静电荷喷雾,拉长并凝固,生成具有从几纳米至大于微米的直径的纤维。所形成的纤维可以被用于过滤材料、涂布模板(coating template)、防护衣、生物医药应用、创伤敷料、药物传输、太阳能帆船(solar sail)、太阳能电池、催化剂载体及复合材料的增强剂。
通过使2,6-双(咪唑甲基)吡啶与1,3-二氯丙酮反应,可以制备咪唑鎓盐(NHC)环芳偕二醇盐(imidazolium cyclophane gem-diol salt)108。如下面的方程2所示。不期望具有吸电子基团存在的盐108作为偕二醇(gem-diol)优先于碳酰形式形成。并非束缚于理论,据认为盐108作为偕二醇的形成是通过用被观测到具有5-6的pH范围的微酸溶液的酸催化过程而进行的。方程2
1H NMR光谱显示了偕O-H作为宽峰出现在7.65ppm处,在13CNMR和红外光谱中都观察到在盐108中没有C=O。在3387cm-1处,观察到O-H伸缩振动,而在1171cm-1处观察到C-O伸缩振动,在91ppm处观察到13C NMR化学位移。X射线晶体学进一步提供了证据,108的结构如下图所示:盐108的TEP,其热椭圆体图在50%概率水平下被绘制。为清晰起见,省略了抗衡阴离子。
依照在方程3中所阐明的反应方案,氧化银(I)与盐108在甲醇中结合,以高收率生成配合物106,其为空气及光稳定的黄色固体,通过在1H NMR光谱的9.35ppm处咪唑鎓盐质子的损失而得以证实。配合物106的质子NMR显示具有复杂峰的宽信号,该峰不容易被确定。同样,并非束缚于理论,这可以归因于该化合物在NMR时标上的立体易变行为(fluxional behavior)。方程3
咪唑碳(NCN)的共振信号从138ppm向184和186ppm处的13C NMR谱低磁场(downfield)的移动表明了Ag-C键的稀少耦合。所观测到的大Ag-C耦合常数值(JAgC=211Hz)与所报道的109Ag核耦合的204Hz-220Hz范围一致。一般观察到109Ag耦合,原因在于与107Ag相比,其具有更高的灵敏度。X射线晶体学证实了配合物106的结构,其被显示在式II中,其中键长: 和观察到弱的Ag1...Ag2相互作用,其键长为比通常报告的范围的Ag...Ag键长,但比的Ag...Ag范德华半径短。在银金属中,已知Ag...Ag键长为C-Ag-C的键角几乎是线性的,其中C15-Agl-C31键角为175.20(18)°,C21-Ag2-C5键角为170.56(18)°。式II配合物106的热椭圆体图,其热椭圆体图在50%概率水平下被绘制。为清晰起见,省略了抗衡阴离子。
通过透射电子显微镜(TEM)和扫描电子显微镜检术(SEM)表征源自和银配合物的静电纺丝纤维。在初纺纤维(as-spunfiber)中,没有观察到明显的相分离,如图3中所示,这表示和银配合物总体上均匀混合。通过扫描电子显微镜检术(SEM),分别测量具有纯(100微米),25∶75银配合物106/(30微米)和75∶25配合物106/(60微米)的纤维层的厚度。配合物106被聚合物包封阻碍了银配合物快速分解为水介质中的银离子或银颗粒。当静电纺丝纤维被暴露于水时,在聚合物基体中已观察到纳米级银颗粒的形成。透射电子显微镜研究显示,纳米-银颗粒在纤维中的活化是在一段时间内逐渐发生的过程。通过使初纺纤维暴露于水中,配合物106分解并释放银离子,银离子集聚成为纳米级大小的银颗粒。在暴露于水蒸气30分钟之内已经观察到银颗粒聚集体的形成(如图4中所示)。在水存在下,银离子与吸附在纤维表面上的聚集体的聚集被认为是简化的机制,通过该机制,纤维层释放了银的活性形式,以实现其抗微生物活性。配合物106的纤维在光和空气中稳定数月,但其对高湿度环境敏感。图3由106和以25比75的重量比制备的静电纺丝纤维。(a)初纺纤维(b)通过将初纺纤维暴露于水而形成的银颗粒。图4TEM图:显示了通过将配合物106和(重量比50∶50)的纤维暴露于水蒸气环境的银颗粒的释放;(a)初纺纤维,(b)在水蒸气环境中65小时的纤维。杀菌效果
使用修改的Kirby Bauer技术,将包封配合物106的静电纺丝纤维层和作为对照的纯静电纺丝纤维放置在琼脂板中的微生物的菌苔上,并在35℃下过夜培养。使用的接种物为临床感兴趣的革兰式阳性和革兰氏阴性原核生物(大肠杆菌(Escherichiacoli)、绿脓杆菌(Pseudomonas aeruginosa)和金黄色葡萄球菌(Staphylococcus aureus))。使用的真菌为白色念珠菌(Candidaalbicans)、黑曲霉(Aspergillus niger)、酿酒酵母(Saccharamycescerevisiae)。琼脂板在35℃过夜培养后,杀菌活性显示了在纤维层周围和纤维层内的透明抑制区(clear zone of inhibition)。在25℃培养48小时后,观察到杀霉菌活性(fungicidal activity)。作为对照的纯纤维层显示无生长抑制(参见图5)。当纤维层的组分从75%的配合物106和25%的变为25%的配合物106和75%的时,观察到在纤维层周围的透明抑制区的直径上没有明显差别。75(配合物106/)纤维层的抑制区的直径为4.00mm,而25%(配合物106/)纤维层的抑制区的直径为2.00mm。这两种类型的纤维层之间的抑制区直径的差别与该两种纤维层中存在的银(3∶1比例)的量没有线性关系。这些结果进一步显示,Kirby Bauer技术作为测定药物抗微生物活性的定量工具具有局限性。银离子的扩散能力可能已经被二级银化合物的形成所限制。已知离子化银与生物配体例如核酸,蛋白质和细胞膜进行配体交换反应。图5包封配合物106的纤维层的敏感性试验,与纯纤维层的杀菌活性比较。(a)配合物106/(25∶75)(b)纯(c)配合物106/(75∶25)
当将一片纤维层放在5ml蒸馏水中并曝光4天后,在试管的底部观察到一些银粒子的沉淀。银粒子从纤维层表面沥滤到溶液中是随时间逐渐发生的。通过将配合物106的初纺纤维层与硝酸银和磺胺二嗪银1%膏或磺胺嘧啶银(silvadene)(SSD)(其为广泛使用的临床药物)进行比较,纳米银粒子从配合物106的初纺层中释放到水介质中导致对配合物106的初纺纤维层关于时间的杀伤(杀菌活性)动力学的考察。用在本研究中的两种类型的纤维层组合物75∶25(Ag含量=424μg/mL)和25∶75(Ag含量=140μg/mL)都显示比SSD(Ag含量=3020μg/mL)更快的杀伤率。含3176μg/mLAg的硝酸银(0.5%)与浓度比其低8倍的配合物106/tecophilic 75∶25(Ag=424μg/mL)显示出大约相同的杀伤率(参见图6)。银化合物对绿脓杆菌(P.aeruginosa)的杀菌活性比金黄色葡萄球菌(S.aureus)快。纤维层杀死细菌比磺胺嘧啶银快。图6银化合物在金黄色葡萄球菌上的CFU(菌落形成单位)对时间(小时)图,表达了每种被测银化合物的杀菌活性动力学。
研究了配合物106的初纺纤维层的抑菌活性和杀菌活性作为接种生物体体积的函数的时间依赖性。在一周多的时间内,配合物106的纤维层对每天用新生长生物体接种(25μL)的绿脓杆菌(P.aeruginosa)、大肠杆菌(E.coli)和金黄色葡萄球菌(S.aureus)显示出有效的杀菌活性。这表明,初纺纤维层在长的时间期间内保持不断地释放活性银种类。用作对照的纯层在接种24小时之内显示无抗微生物活性。在用200μL(2×107)多的新生长生物体接种后,在2周多的时间内,具有75%配合物106/tecophilic组合物的配合物106的初纺纤维层比25%配合物106/tecophilic对绿脓杆菌(P.aeruginosa)显示出更好的杀菌效果。在琼脂板上每天用LB肉汤溶液划线10天后,观察对金黄色葡萄球菌(S.aureus)和大肠杆菌(E.coli)的抑菌活性。目测检查被培养溶液,显示无生物体生长。
使用最小抑制浓度(MIC)试验,研究了108、配合物106和AgNO3在含水LB肉汤中的杀菌活性。培养24小时后,在配合物106和AgNO3的杀菌活性和MIC上一般没有差别,如在表5中所示。然而,在培养48小时之后,硝酸银在比配合物106低2倍的浓度(838μg/mL)下显示更好的抗微生物活性。比较AgNO3和106的活性的MIC结果,此两种化合物具有大约相同的银含量。表5.比较具有大约相同银含量的AgNO3和配合物106的活性的MIC结果。DF为稀释因子(1mL)。+=生长,-=无生长。每种化合物的每mL银含量(μg)被计算为(Ag的分子量/化合物的分子量)×wt%。
未测定磺胺二嗪银的MIC值,因为其溶液混浊,所用的108的浓度显示无抗微生物活性。在MIC试验中,观察到具有最小浓度的配合物106(209μg/mL)和AgNO3(216μg/mL)的稀释液在24小时培养之后,在琼脂板上显示相同数量的金黄色葡萄球菌(S.aureus)菌落生长。25%配合物106/tecophilic纤维层具有最小浓度的银,为140μg/mL(参见表6),而且持续释放生物可利用的银种类数天。观察到在体积每天增加的接种物上无生物体生长。

因此,通过包封在合适的聚合物纤维中,配合物106的抗微生物活性在非常低的Ag粒子浓度下在更长的期间被增强。含424μg/mL银的纤维层75%(配合物106/tecophilic)的杀菌活性在银浓度上比AgNO3(3176μg/mL)低8倍,其不仅显示了与硝酸银同样快的杀伤率,而且保持了LB肉汤的原始颜色,透明黄色溶液,不像硝酸银,其污染了LB肉汤颜色,并使其变为黑褐色。磺胺二嗪银膏不易溶于含水LB肉汤中,因此影响了其杀菌活性的速率。
包封配合物106的纤维层的抗微生物活性可以被认为是活性银种类的组合,其可以包括AgCl2 -离子、Ag+离子簇、AgCl和自由Ag+离子。理论上,活性银粒子在溶液中的缓慢释放导致氯化银的快速生成。更多的氯化物离子作为主要的抗衡离子存在将进一步导致负电荷[AgyClx]n-离子种类的形成(其中y=1,2,3...等;x=2,3...(y+1);n=x-1)。已经形成[AgI3]2-、[Ag2I4]2-、[Ag4I8]4-和[Ag4I6]2-型的阴离子银配合物。阴离子氯化银种类的形成可以不限于在溶液中的银颗粒的沥滤聚集体(leached aggregate),也可以被发现存在于纤维层的表面上,如在图8的SEM图中所示。已知阴离子二氯化银(silver dichloride)溶于水介质中,因此是生物可利用的。据报告,阴离子卤化银对敏感和抗性菌株都是有毒的。在层中的纤维网络上所吸附的活性银种类是有利的,因为纤维层较水合银离子的常规使用不得不增加了活性银种类的表面积。该机制可以说明纤维层在水介质中具有有效杀菌活性,甚至在与配合物106的未包封形式相比非常低的银浓度下。尽管配合物106在水中不易溶,然而已经观察到其在水介质中发生快速分解。因此,配合物106的杀菌活性被减小,原因在于LB肉汤培养基中活性银种类的差的可利用性,这可能是由于形成了次级银化合物(secondary silvercompound),特别是AgCl的缘故。图8.在LB肉汤培养基中,两周的抗微生物活性之后,配合物106和(75∶25)的静电纺丝纤维。(a)纤维片段的立体图;(b)封装在Tecophilic中的银粒子的大聚集体(400nm);(c)在基体中的银聚集体(直径为200nm至300nm)和银颗粒(直径为10nm至20nm);(d)具有银颗粒聚集体的纤维层的俯视图。急性毒性评价
通过静脉内给药108而进行LD 50评价,将108溶于缓冲盐溶液中,经鼠的尾部施用。使用平均重量为500g的成年鼠。每周逐步施用0.3ml剂量(5mg,50mg)。认真检查鼠的剂量反应效应(doseresponse effect)。当50%的鼠在死亡之前显示有力的抽搐时,施用50mg108后10分钟出现死亡。尸体解剖报告显示有肺出血,并在死亡鼠的脑部发现出血,诊断为中风(stoke)。观察到存活的鼠体重损失,在饮食上有显著减少,以及低尿排出量。LD 50评价值被发现为100mg/Kg鼠。
具有官能化基团的108的合成有助于使银(I)咪唑环芳偕二醇适合包裹进纳米纤维。已经显示纤维层在接种物上具有改善的银(I)-n-杂环卡宾配合物抗微生物活性,其在LB肉汤培养基中在比磺胺嘧啶银低8倍的浓度下具有比磺胺嘧啶银更快的杀伤率。银(I)-n-杂环卡宾配合物的胶囊化增加了活性银种类的生物利用率,并减少了所用银的量。已经证明,与以水合形式提供银比较,纳米纤维中的胶囊化银(I)卡宾配合物是有前景的物质,适合于在较长时间期间内银离子的持续和有效的传输,其具有最大的杀菌活性。未封装形式经常涉及0.5%硝酸银中银的量,与其相比,使用该胶囊化技术,抗微生物活性所需的银的量被减少。此外,纤维层可保持LB肉汤原始颜色的能力是主要的整形有利因素。对配体在鼠上的急性毒性的评价显示LD 50为100mg/Kg鼠,该值被认为是中等毒性的。
除了有用的抗微生物或抗菌性能,据认为本发明可以抑制真菌生长,也可以抑制病毒生长。本发明的物质组合物和方法也考虑了银经由任何已知载体传输到位置,载体包括但不限于通过肺的吸入,液体直接施用于眼或任何其它类型的局部应用。概括性试验
氧化银(I)、磺胺二嗪银和1,3-二氯丙酮购自Aldrich。丙酮、乙腈、甲醇、乙醇、六氟磷酸铵和生物体;酿酒酵母(S.cerevisiae)(ATCC2601)、白色念珠菌(C.albicans)(ATCC 10231)、黑曲霉(A.niger)(ATCC 16404)、大肠杆菌(E.coli)(ATCC 8739)、绿脓杆菌(P.aeruginosa)(ATCC 9027)、金黄色葡萄球菌(S.aureus)(ATCC6538)购自Fisher。未经纯化而使用所有的试剂。红外光谱图记录在Nicolet Nexus 870 FT-IR分光计上。1H和13C NMR数据被记录在VarianGemini 300MHz仪器上,所获得的光谱以氘代溶剂为参考。质谱分析数据被记录在带有阳离子极性的ESI-QIT Esquire-LC上。TEM图被记录在120KV的FEI TE CNAI-12透射电子显微镜(TEM)上。二氯化咪唑鎓盐环芳偕二醇(imidazolium cyclophane gem-dioldichloride)的合成
将在60ml乙腈中的含0.24g(1.0毫摩尔)2,6-双(咪唑甲基)吡啶和0.254g(2.0毫摩尔)1,3-二氯丙酮的溶液在75℃搅拌8小时,过滤后得到为褐色固体的108。收率:0.9毫摩尔,89.6%。通过从乙腈/水中缓慢蒸发获得108的PF6盐的无色晶体。Mp:175-178℃。1HNMR(300MHz,DMSO-d6):δ4.68(s,4H,CH 2 C(OH)2CH 2 ),5.67(s,4H,CH2),7.40(s,2H,NC(H)CH),7.47(d,2H,J=7.8Hz,m-pyr),7.65(s,2H,C(OH)2),7.89(s,2H,NCHC(H)),7.94(t,1H,J=7.8Hz,p-pyr),9.34(s,2H,NC(H)N)。13C NMR(75MHz,DMSO-d6):δ51.8,55.2,91.1,120.5,122.0,123.9,138.0,138.8,152.6。ESI-MS m/z:384[M2+2Cl-],348[M2+Cl-]。FT IR(Nujol,cm-1):3387,3105,1597,1564,1439,1346,1171,1085,996,755。分析计算:C,48.54;H,4.41;N,16.94;Cl,17.13。观测:C,48.33;H,4.32;N,16.7;Cl,16.76。氢氧化双核银卡宾环芳偕二醇(dinuclear silver carbene cyclophanegem-diol hydroxide)的合成
在室温下,将0.232g(1.0毫摩尔)氧化银(I)与0.366g(0.9毫摩尔)108在70ml甲醇中的组合物搅拌50分钟。浓缩滤液,获得为黄色固体的配合物106。通过缓慢扩散,从乙醇中得到配合物106的单晶,包含针状碳酸盐(a spike ofcarbonate)。收率:0.618g,0.738毫摩尔,82%。Mp:202-204℃。ESI-MS m/z:400[0.5M2+],801[2M+],837[2M+2OH-]。FTIR(Nujol,cm-1):3415,3105,1596,1564,1439,1344,1169,1084,1028,996,758。13C NMR(75MHz,DMSO-d6):δ48.6,51.1,53.8,92.1,119.9(J=1.4Hz),121.6,128.6,137.8(J=2.4Hz),154.2,184.9(J卡宾-Ag=211Hz)。分析计算:Ag,24.54;C,43.79;H,4.20;N,15.24。观测:C,43.15;H,4.22;N,14.89。静电纺丝纤维(electrospun fiber)
将溶解在比例为9比1的乙醇和四氢呋喃的混合物中。使预制的溶液与配合物106的乙醇溶液混合。制备具有配合物106与之间的不同重量比的溶液。比例为0/100,25/75和75/25。将配合物106和的溶液置于吸移管中。在溶液滴的表面和接地收集器之间施加15KV的电位差,距离为大约20cm。使用透射电子显微镜(TEM)和扫描电子显微镜检术(SEM)表征初纺纤维(as-spun fiber)和暴露于水的纤维。抗微生物试验
在无菌试管中测量灭菌LB肉汤(5mL)。将菌环量稳定期培养微生物(大肠杆菌、绿脓杆菌、金黄色葡萄球菌)引入含LB肉汤溶液的试管中。在35℃,在振荡培养箱中过夜培养该混合物。对稳定期培养的真菌(白色念珠菌、酿酒酵母、黑曲霉)进行同样的步骤,并在室温下无振荡培养72小时。纤维层试验
将恒定体积(25μl)的新生长的生物体放在LB琼脂板上,使其生长,以获得生物体的菌苔。将配合物106和纯tecophilic的纤维层(2.0cm×2.0cm)放在LB琼脂板的细菌(大肠杆菌、绿脓杆菌、金黄色葡萄球菌)菌苔上,并在35℃过夜培养。通过目测检查在纤维层区域中及其周围的生长和无生长情况而观察杀菌活性。将大约相同尺寸的纤维层放置在真菌(白色念珠菌、酿酒酵母、黑曲霉)的菌苔上,并在室温下培养48小时。测量亮区(clear zone)的直径。最小抑制浓度(MIC)试验
通过将1mL银化合物的新制备储备溶液(具有相同量银颗粒)转移到含有2mL LB肉汤的无菌培养管中,标记为A,进行系列稀释,以获得一系列浓度。将1mL混合好的A的溶液转移到含LB肉汤的培养管B中。重复相同步骤,得到试管C、D和E的稀释溶液。通过目测检查用25μl生物体接种的标记为A-E的上述浓度银化合物的生长/无生长情况,测定MIC。在35℃过夜培养、生物体无生长之后,第二天,将另外80μl新生长生物体加入每个培养物中,并在相同温度下培养。杀菌活性的动力学试验
测量相同体积(5mL)的LB肉汤,放入无菌培养管中,并用100μl金黄色葡萄球菌接种到含有硝酸银(12.8mg,25mg),磺胺二嗪银(20mg),11.3mg配合物106/tecophilic(25∶75)和11.4mg配合物106/tecophilic(75∶25)纤维层的每个试管中。在35℃培养该混合物,通过在琼脂板上划线菌环量的每种混合物而检查一段时间内的杀菌活性。然后在37℃将琼脂板过夜培养,并计数所形成的生物体的菌落数量。使用100μl绿脓杆菌重复同样的步骤。动物研究
雄性Sprague Dawley(Harlen Sprague Dawley,Indianapolis,IN)成年鼠(体重400-500g)被放在University ofAkron的动物设备(animalfacility)中。温度和湿度保持恒定,光/暗循环为6.00am-6.00pm:光,6.00pm-6.00am:暗。随意提供食物(Lab diet 5P00,Prolab,PMI nutrition,Intl.,Bretwood,Mo.)和水。用乙醚麻醉动物,目的是将化合物注射到尾静脉中,使用27规格的注射器针,在0.3ml体积的灭菌盐水中。配体的剂量为5mg和50mg。在试验的最后剂量,杀死动物,将肝,肺,肾和心脏组织取出并在-70℃冷冻。每天收集尿样,用于较后检查化合物分布。这些研究经过University ofAkron Institutional Animal Care andUse Committee(IACUC)批准。X射线晶体结构测定
晶体数据和结构精修参数(structure refinement parameter)包含在支持性信息中。108和配合物106的晶体都被涂布石蜡油,安装在kyro环上,并将其在氮气流下放在测角计上。使用Mo Kα放射 在BruckerApex CCD衍射计上,在100K温度下,收集X射线数据。使用SAINT软件积分强度数据,使用SADABS进行经验吸收纠正。通过直接的方法解析108和配合物106的结构,并使用全矩阵最小二乘法(full-matrix least square procedure)使108和配合物106的结构完善。用各向异性位移法(anisotropic displacement)使所有非氢原子精修。
很显然,在通过施用N-官能化银卡宾配合物而提供抑制微生物生长的方法上,本发明非常有效。因此,应当理解,在不背离在此所公开和描述的本发明精神的情况下,可以确定明显落在所要求保护发明的范围之内的任何变动,以及由此对具体组分元素的选择。
Claims (2)
1.由下式任一个表示的N-杂环卡宾的银配合物:
其中R独立选自氢、C1至C12烷基、C1至C12取代烷基、C1至C12烷氧基、C1至C12环烷基、C1至C12取代环烷基、C1至C12链烯基、C1至C12环烯基、C1至C12取代环烯基、C1至C12炔基、C1至C12芳基、C1至C12取代芳基、C1至C12芳基烷基、C1-C12烷基胺、C1-C12取代烷基胺、C1-C12烷基戊糖磷酸盐、C1-C12酚和C1-C12酯、C1至C12烷基芳基、C1-C12吡咯、C1-C12吡啶或C1-C12噻吩,并且其中X选自卤化物、碳酸盐、乙酸盐、磷酸盐、六氟磷酸盐、四氟硼酸盐、硝酸盐、硫酸二甲酯、氢氧化物或硫酸盐。
