在使用较短PEG链的实施方案中,所得CS纳米颗粒具有更小的直径。在本文所述的PEG包覆的CS纳米颗粒的使用方法的一个实施方案中,相对于较大直径的CS纳米颗粒,相对较小的直径允许用于肾排泄或改善肾排泄。例如,在另外的分离步骤后,获得较短PEG包覆的CS纳米颗粒,其流体动力学半径为4nm,并且通过荧光相关光谱法测定窄的粒径分布。
参照图1和U.S.专利公开No.2004/0101822A1中所述,包含荧光染料的非PEG包覆的CS纳米颗粒的每染料亮度(per dye brightness)在游离染料在水溶液中的过程中是改善的。本文所述的PEG纳米颗粒包覆的另一个优点是相对于未包覆的CS纳米颗粒,其观察到每染料荧光亮度进一步改善。在使用纳米颗粒的许多体外和体内的方法中,甚至相对于未包覆的CS纳米颗粒的信号噪音比改善是有利的。
除了改善的信号,PEG包覆的CS纳米颗粒显著降低了试验测试受试者中的死亡率。例如,静脉内注射未包覆的10nm以下的二氧化硅纳米颗粒可导致试验动物死亡。例如,在一个试验中,当注射200μl剂量的2.7mg/ml的未包覆的点溶液(dot solution)时,组中的5只小鼠死亡。相反,注射类似剂量的[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷包覆的CS纳米颗粒的5只小鼠的死亡率为零。
2.制备包覆的CS纳米颗粒的方法
本文所述的制备包覆的CS纳米颗粒的方法可通过制备[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷包覆的CS纳米颗粒的下列示例性方法来理解。
用于所述应用的CS纳米颗粒通过Wiesner和Ow在US专利公开No.2004/0101822A1中所述的方法来合成,使得根据动态光散射法来测定它们的直径低于10nm。在一个实施方案中,制得的包覆的CS纳米颗粒保持总直径低于10nm。所得CS纳米颗粒针对甲醇透析。在这些步骤后它们浓缩为约10mg/ml。
随后CS纳米颗粒包覆[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷。必须量通过下列方式来计算:如Tripp和Hair、Tripp,C.P.和M.L.Hair(1995).Reaction of Methylsilanolss with Hydrated Silica Surfaces:TheHydrolysis of Trichloro-,Dichloro-,and Monochloromethylsilanes and theEffects of Curing.LANGMUIR 11(1):149-155中所述(其通过引用方式并入本文),首先估计在给定体积的纳米颗粒溶液中的表面硅醇的总量。知道表面硅醇的量和因此单层覆盖表面所需要的硅烷化合物(包覆材料)的量,然后将过量10倍的包覆化合物[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷稀释到甲醇体积中,所述甲醇体积是待包覆的纳米颗粒溶液的体积的两倍。向该溶液中加入氨,以获得最终浓度为0.2摩尔。在恒定搅拌下将纳米颗粒滴定到甲醇/氨/[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷混合物中,并在室温下继续搅拌12小时。最终,在超纯水中透析纳米颗粒。
在一个实施方案中,PEG包覆化合物以杂-双官能PEG化合物的形式提供。所述官能图可以为(但不限于)马来酰亚胺官能团、酯官能团、和羟基官能团。杂-双官能PEG化合物的一种官能团可以反应以形成用于与CS纳米颗粒的二氧化硅的壳偶联的硅烷。第二官能图可以反应以连接配体。所述配体可以是U.S.专利公开No.2004/0101822A1中所述的任意配体。在一个实施方案中,配体包括能够识别靶分子或物质的靶向部分。
在按照下列方式进行包覆过程的实施方案中:使用非常短的亲水性化合物(如硅烷-PEG,例如至多10个单体单元),并使用乙酸盐缓冲盐作为催化剂和仅使用水作为溶剂,这导致即使在加入纳米颗粒前短PEG-硅烷也快速絮凝。这使得包覆过程无效。较小催化剂氨的使用和几乎不含水,或者在水和醇的混合物中,这样的反应条件解决了该问题。
包覆的(和未包覆的)CS纳米颗粒可包括太大而无法通过肾脏的颗粒或聚集体。纳米颗粒尺寸分布可通过使用市售过滤旋转柱(例如PallCorporation(10KD或30KD尺寸的Jumbo-、Macro-、Micro-和Nanosep柱)中的那些或其他供货商(如Millipore)的产品)经过过滤来变窄。滤出物可进一步通过类似但具有更小的孔径的产品(例如1KD或3KD尺寸的Jumbo-、Macro-、Micro-和Nanosep柱)在体外浓缩。CS纳米颗粒还可以使用由Simpore开发的超薄膜(其可能用于更大的流量和更低的孔的损失(由于它们稀疏的横截面))来过滤。图2示出CS纳米颗粒包覆和使用两种过滤器过程的过滤的示例性方法。
我们通过荧光相关光谱法(FCS)分析使用[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷包覆而制备的典型纳米颗粒的两种尺寸部分(3KD滞留物和30KD滞留物)。我们发现对于较小的3KD滞留物而言,铃动力直径为4nm并且尺寸分布非常窄,对于较大的30KD滞留物,直径为16nm。
在另外的实施方案中,CS颗粒的核通过Wiesner和Ow在US专利公开No.2004/0101822A1(其通过引用的方式并入本文)中所述的方法来合成,使得通过荧光相关光谱法测定的核的直径小于5nm。随后使所得核包覆PEG包覆化合物(例如[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷)或杂-双官能PEG化合物的壳,使得制得的CS颗粒的直径小于7nm。通过该方法合成的CS纳米颗粒具有非常窄的尺寸分布。无需另外的合成后过滤过程来使尺寸分布变窄。图3示出三个不同批次中的CS颗粒的荧光相关光谱法尺寸表征。CS颗粒尺寸分布分别以6nm、4nm和3nm为中心。图4示出所得CS颗粒在各种缓冲盐溶液中14天后的稳定性。
用于包封Cy5.5染料的6nm、4nm和3nm CS颗粒的染料前体的制备
在氮气惰性化的手套箱中将1mg的Cy5.5马来酰亚胺染料溶解在1mL的二甲基亚砜(DMSO)中。在Cy5.5马来酰亚胺染料完全溶解在DMSO中后,将3-巯基丙基三甲氧基硅烷(MPTMS)以50∶1MPTMS∶Cy5.5马来酰亚胺的摩尔比加入所述溶液中。在磁力搅拌板上在室温下在黑暗中搅拌反应至少12小时。
包封Cy5.5染料的基于二氧化硅的富集染料的6nm、4nm和3nmCS颗粒的制备:
在干净的圆底玻璃烧瓶中加入合适量的乙醇或甲醇溶剂。试剂浓度如下表所示。以下列顺序加入试剂:水、染料前体、原硅酸四乙酯(TEOS)、2.0M在乙醇中的氨。在磁力搅拌板上在室温下搅拌反应至少12小时。
使用PEG包覆化合物包覆二氧化硅富集染料的核来制备6nm、4nm和3nm的CS颗粒:
向如上所述合成的含有基于二氧化硅的富集染料的核的混合物中加入硅烷化PEG化合物(例如[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷),以产生包围核的壳。加入的PEG化合物的量如下表所示。为了制备具有窄的尺寸分布的颗粒,使用剂量容积式流量吸液管以较少的等分试样间歇(例如每10至15分钟小于5mM)来加入PEG化合物,并且连续搅拌。
在加入所有PEG化合物后,将反应混合物在黑暗中搅拌12小时。收集所得CS颗粒,并针对溶剂甲醇或乙醇通过透析方法来纯化,从而除去未反应的加入物。另外,针对去离子水来透析CS颗粒,从而交换溶剂。然后水中的CS颗粒可在不同缓冲液溶液中再生以用于成像应用。

3.使用方法
在腹部手术中输尿管的意外损坏是并发症和医疗差错的主要原因。二氧化硅纳米颗粒允许外科医生使用特定装备的腹腔镜使输尿管和膀胱通过组织而可视化。可视化帮助外科医生在血管、泌尿管、神经和腹部处理中避免意外损坏这些结构。尽管可将支架插入输尿管中以在手术过程中显示这些结构,但是这种方法本身就可能损坏复杂的结构。另外,包括泌尿科医生进行该方法的成本大大降低了其经济上的适用性。
使用荧光染料来使输尿管可视化的概念由Udshamadshuridze提出,N.S.Udshamadshuridze,Intraoperative Visualization of the Ureterswith Fluorescein Sodium Z.Urol.Nephr.,Vol.81;pp.635-639(其通过引用方式并入本文)。该研究揭示出使用荧光素,批准用于临床的非毒性染料。然而,荧光素发射出波长无法明显透过(脂肪)组织的光。因此,尽管其出版于1988年,但是未见临床采用该研究。
在用于使受试者的输尿管可视化时,CS纳米颗粒、特别是具有近红外荧光化合物的PEG包覆的CS纳米颗粒可提供许多优点。例如,通过包封近红外荧光染料而获得的亮度增强使得它们优于相同浓度的游离染料。组织的吸收系数被认为在近红外光区(650nm-900nm)更小,使得光可更深地透过数厘米厚的组织。另外,染料与CS纳米颗粒的二氧化硅网络的共价键合会避免染料泄露到周围组织并且在其他器官或组织中积累。这种泄露将降低目标器官和周围组织的对比度。在注射到体内后保持强度的荧光染料有利于其作为成像助剂在临床中的应用。
因此,CS纳米颗粒、特别是PEG包覆的CS纳米颗粒可静脉内注射到人或动物中(用于人时,将使用FDA批准、按照GMP生产并因此具有相应的孔径的过滤器)。CS纳米颗粒在通过肾脏后不会降低荧光并浓缩在尿液中。这使正在进行腹部手术的外科医生以尿液从肾脏流向膀胱的方式来观察输尿管。使用特定装备的腹腔镜这些结构(输尿管和膀胱)是通过脂肪组织可视化的,因此避免对于这些结构的意外损坏,如图5所示。
除了使输尿管成像,二氧化硅纳米颗粒可加入传感器系统中以赋予观察者时间和空间信息。例如,主要由Wiesner et al所述的pH传感器证据基于二氧化硅纳米颗粒,该二氧化硅纳米颗粒加入环境敏感性染料和用于公制传感的参照染料(“纳米颗粒传感器”)。该主要pH传感器的证据已经证实可扩展测定其他生理参数,例如金属状态、氧的状态、氧化还原状态等,这些生理参数可涉及染料发射情况的变化。通过将纳米颗粒传感器或其他基于纳米颗粒的传感装置注射到机体内,研究者和临床医生可使机体成像并获得其他重要的生理数据。
加入体内的纳米颗粒的分布是影响它们体内应用可能性的关键问题。期望有快速试验,其中在目标位置观察(使用可透过组织的NIR成像系统)注射的点,并且在提供测量或其他功能后快速清除。一个接受FDA批准用于注射诊断性纳米颗粒的关键问题是它们从机体清除。通过确保快速的肾脏清除、低的残留物质量和物质在体内的强度,通过使用本文所述的包覆的CS纳米颗粒可开发出更安全更精确的试验。
包覆的CS纳米颗粒的其他优点和特征从下列与未包覆的CS纳米颗粒的比较中将是显而易见的。
参照图6,可见未包覆的CS纳米颗粒和[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷包覆的CS纳米颗粒的体内分布在注射到小鼠中2小时后是不同的。可见在2小时后在脾脏和肝脏中积累未包覆的CS纳米颗粒。尿液浓度表现出与端点分析(在注射CS纳米颗粒2小时后)中的相同。然而,PEG包覆的CS纳米颗粒即使在2小时后也仍然保留在血流中,因此仍可通过肾脏分泌。
参照图7,未包覆(A)和[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷包覆C的点(B)在两个独立试验中静脉内注射到麻醉的猪中。在两个试验中,随时间对血液和尿液进行取样,并分析CS纳米颗粒含量。明显地,包覆的C的点(B)停留在血流中而不是像未包覆的点(A)一样从其中耗尽。还可以注射比未包覆的CS纳米颗粒更高剂量的PEG包覆的CS纳米颗粒,而不会存在CS纳米颗粒聚集的风险,这可在使用[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷包覆的CS纳米颗粒(B)获得的更高尿液浓度中看到。在血液中注射更高剂量的CS纳米颗粒直接转变为更高的尿液浓度。期望在尿液中获得更高浓度的CS纳米颗粒,因为作为使输尿管可视化的基础的检测的荧光信号将会更强。
参照图8,[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷包覆的纳米颗粒的尺寸分布已经表明影响肾脏排泄。包覆的纳米颗粒通过30K的柱过滤。滤出物在3K的柱上重新浓缩。两部分(滞留物和重新浓缩的滤出物)匹配相同的荧光并且单独注射(5只小鼠/部分)。在2小时后,取出尿液和血液并且分析荧光(RFU=相对荧光单位)。较之滞留物部分(较大的纳米颗粒),30K滤出物部分(较小的纳米颗粒)在尿液中在更高程度上清晰。另外,在注射较小的纳米颗粒部分的小鼠中,血液中检测的荧光明显更低(p<0.05),原因在于荧光纳米颗粒的排泄。对照组示出未注射纳米颗粒的小鼠的背景信号。
Claims (11)
1.一种荧光纳米颗粒,包含:
基于二氧化硅的核,包含:
含有巯基取代基的有机官能团;和
有机荧光化合物;
二氧化硅的壳;以及
硅烷-PEG化合物;
其中所述二氧化硅的壳包封所述基于二氧化硅的核;并且所述硅烷-PEG化合物与所述二氧化硅的壳偶联。
2.权利要求1所述的纳米颗粒,其中所述荧光纳米颗粒的直径为10nm或更小。
3.权利要求1所述的纳米颗粒,其中所述硅烷-PEG化合物包含25或更少的重复的PEG单元。
4.权利要求3所述的纳米颗粒,其中所述硅烷-PEG化合物选自:[甲氧基(聚氧乙烯)丙基]-三甲氧基硅烷、[甲氧基(聚氧乙烯)丙基]-二甲氧基硅烷和[甲氧基(聚氧乙烯)丙基]-单甲氧基硅烷。
5.权利要求1所述的纳米颗粒,其中所述纳米颗粒在激发时能够发射波长大于650nm的荧光。
6.权利要求1所述的纳米颗粒,其中所述二氧化硅的壳包含硅醇;并且其中所述硅烷-PEG化合物与所述硅醇偶联。
7.权利要求1所述的荧光纳米颗粒,还包含适于与靶分子或底物关联的配体。
8.一种组合物,包含:
多个权利要求1所述的纳米颗粒;
其中所述多个纳米颗粒中少于10%的纳米颗粒具有直径大于10nm的二氧化硅的壳。
9.一种荧光纳米颗粒,包含:
基于二氧化硅的核,包含有机荧光化合物;
二氧化硅的壳;
适于与靶分子或底物关联的配体;以及
硅烷-PEG化合物;
其中所述二氧化硅的壳包封所述基于二氧化硅的核,并且所述荧光纳米颗粒的直径在1nm和100nm之间。
10.权利要求9所述的荧光纳米颗粒,其中所述硅烷-PEG化合物与所述二氧化硅的核以及适于与靶分子或底物关联的配体偶联。
11.一种荧光纳米颗粒,包含:
基于二氧化硅的网络,包含有机荧光材料;
聚合物配体,其通过包含有机官能团的连接剂与所述基于二氧化硅的网络的外表面偶联;
适于与靶分子或底物关联的配体;
包含有机官能团的连接剂;以及
硅烷-PEG化合物,其与所述基于二氧化硅的网络的外表面偶联。
