胰酪胨肉毒梭菌增菌肉汤
技术领域  本发明涉及一种胰酪胨肉毒梭菌增菌肉汤,其主要成分是胰酪蛋白胨、蛋白胨、酵母浸粉、葡萄糖、硫乙醇酸钠、蒸馏水、1.5%胰蛋白酶(1∶250)溶液按比例混合经加温、溶解、混合、冷却、分装、高温灭菌等工艺而成。主要用于肉毒梭菌的增菌培养。常与疱肉培养基同时使用。已有培养基剂品种很多,不具有切实可靠的方法;对培养成分有破坏作用,亦影响其物理性状。随着消毒研究的进展,国内外对培养基的研究也在不断的发展中。生物实验工作者一直希望能有一种能克服上述培养基不足的胰酪胨肉毒梭菌增菌肉汤。
背景技术  本发明的目的在于提供一种能克服上述培养基不足的新型产品,用于肉毒梭菌的增菌培养。常与疱肉培养基同时使用,本发明的胰酪胨肉毒梭菌增菌肉汤克服了上述的不足,快速检测肉毒梭菌的毒素,经酶激化后显示毒力,便于进行测毒动物试验,可弥补疱肉培养基的不足。方法简单,使用方便,结果可靠。
发明内容  本发明的要点在于选择合适的组分,并进行合理混配,经加温、溶解、混合、冷却、高温灭菌、分装、等工艺而成一种胰酪胨肉毒梭菌增菌肉汤。
本发明选择的配方如下(重量百分比%):胰酪蛋白胨50-60g;蛋白胨0.5-1.0g;酵母浸粉20-30g;葡萄糖4.0-5.0g;硫乙醇酸钠1.0-1.5g;蒸馏水1000mL;1.5%胰蛋白酶(1∶250)溶液67mL。
酿成的培养基呈黄色透明液体。将肉毒梭菌48h培养物,稀释成1∶1000菌悬液,接种0.01mL,应能生长。肉毒梭菌的毒素,经酶激化后显示毒力,便于进行测毒动物试验,可弥补疱肉培养基的不足。
本发明产品的制备方法是:
(1).将胰酶消化液加温到80℃,用200g/L氢氧化钠溶液调至pH7.0-7.2。加入内浸液、磷酸氢二钠、磷酸二氢钾,煮沸10min后,再调pH,韦氏梭菌pH为8.2-8.4,脓毒梭菌pH为8.0-8.2,煮沸20min,过滤透明,分装于无菌瓶内;培养韦氏梭菌瓶中按12%加新鲜洗净牛肉渣,培养脓毒梭菌瓶中按0.1%加活性炭,113℃30min高压灭菌备用;
(2).配制250g/L葡萄糖,113℃30min高压灭菌,种菌前按1%(V/V)无菌操作加入。
本发明具有以下优点:胰酪胨肉毒梭菌增菌肉汤克服了以往培养基的不足,能快速准确检测肉毒梭菌,常与疱肉培养基同时使用。肉毒梭菌的毒素,经酶激化后显示毒力,便于进行测毒动物试验,可弥补疱肉培养基的不足。培养基清澈,方法简单,使用方便,结果可靠。

1.一种胰酪胨肉毒梭菌增菌肉汤,其主要成分是胰酪蛋白胨、蛋白胨、酵母浸粉、葡萄糖;硫乙醇酸钠、蒸馏水、1.5%胰蛋白酶(1∶250)溶液按比例混合,经加温、溶解、混合、冷却、分装、高温灭菌等工艺而成;其特征在于具有如下配方:胰酪蛋白胨50-60g;蛋白胨0.51.0g;酵母浸粉20-30g;葡萄糖4.0-5.0g;硫乙醇酸钠1.0-1.5g;蒸馏水1000mL;1.5%胰蛋白酶(1∶250)溶液67mL。
2.一种权利要求1所述胰酪胨肉毒梭菌增菌肉汤的制备方法,其特征在于具有如下的步骤:
