Abstract

Description
本发明涉及测定有机阳离子转运蛋白(OCT)活性的方法,涉及在无细胞电生理传感器芯片的帮助下测定调节OCT活性的化合物的活性或鉴定所述化合物的方法,所述无细胞电生理传感器芯片含有固体支持传感器电极和脂质层,该脂质层含有定位于传感器电极最接近空间附近的OCT,然而传感器电极相对于所用的溶液和脂质层是电绝缘的,还涉及传感器芯片本身和含有该传感器芯片的试剂盒。
人有机阳离子转运是有机阳离子跨细胞转运的重要机制。因此,有机阳离子转运蛋白(OCT)不仅是直接影响疾病相关异常的潜在药物靶标,而且是允许潜在药物生物利用率参数变化的潜在ADMET(吸附、分配、新陈代谢、排泄和毒性)靶标。
OCT属于包括单向转运蛋白、同向转运蛋白和反向转运蛋白的超家族,如广谱抗药蛋白、易化扩散系统和质子反向转运蛋白。它们不依赖钠和质子梯度来介导具有不同分子结构的小阳离子的转运。通过人OCT(hOCT)的底物特异的、不依赖钠的转运机制已经在肝、肾、小肠和神经系统中有所描述(Pritchard JB&Miller DS(1993),Physiol.Rev.73(4)765-796)。在1997年已克隆到了人有机阳离子转运蛋白hOCT1(Zhang,L.等人(1997)Mol.Pharmacology 51(6),913-921)。
当经过它的转运循环时,OCT变换电荷。该变换可能起源于带电底物的运动或携带(部分)电荷蛋白质部分的运动。可以通过radiofluxes和标准的双电极电压钳电生理学来监测OCT的活性,任一方法的常见缺点如时间分辨率差、灵敏度低、阻断剂和竞争底物间难分辨、假阳性和假阴性等等(Arndt等人(2001)Am J Physiol Renal Physiol,281,F454-F468)。
在某些其它情况下,转运蛋白相关的电流可以在生理环境中通过膜片钳实验或在人工“黑脂质膜”中直接监测。在后一种情况下,脂双层在两个缓冲储存器间的小孔中形成,每个缓冲储存器包含Ag/AgCl电极。蛋白质掺合进双分子层后,可以例如通过ATP衍生物的光激活来触发生物活性(例如酶活性)。然而,由于它缺乏稳定性,利用该体系不能进行快速的缓冲液交换实验,而将所述体系限制在光活化底物上。可以通过将含蛋白质的颗粒固定在传感器表面或传感器芯片上来克服稳定性的缺乏。
无细胞电生理传感器芯片通常基于含转运蛋白的膜碎片或小泡产生,该膜碎片或小泡通常电偶联到镀金生物芯片上。膜碎片通常吸附到传感器芯片表面,所述表面在薄金膜上优选具有修饰的脂质层。膜碎片通常形成能够维持跨膜离子梯度的腔。用合适的底物激活后,离子或带电底物被跨膜转运。因为吸附的膜碎片和覆盖的电极表面行为像电容器,如果将参比电极置于周围溶液中,那么运动中的离子代表可检测的变化电流。
本发明的问题涉及虽然hOCT1的膜片钳实验失败了,但是用此类传感器芯片能否特异地并灵敏地检测到OCT活性的问题。
令人惊奇的是,已发现可以建立无细胞测定,其显示出需要的灵敏度以便OCT激活时检测特异信号。尤其令人惊奇的是因为OCT在没有细胞背景,即无细胞内物质、细胞骨架等的情况下可以在本发明无细胞测定中起作用。尤其是,本发明的测定法可以在具有独特优势的宽pH和/或高离子浓度范围内进行。
因此,本发明的第一个实施方案涉及测定OCT活性的方法,包括以下连续步骤:
(a)提供含有固体支持传感器电极和脂质层的无细胞电生理传感器芯片,其中所述脂质层含有位于传感器电极最接近空间附近的OCT,而所述传感器电极相对于所用的溶液和脂质层是电绝缘的,
(b)用含离子的非活化溶液处理传感器芯片,
(c)用含离子和底物的活化溶液处理传感器芯片,并
(d)测定电信号。
OCT例如选自SLC22A1(OCT1)、SLC22A2(OCT2)、SLC22A3(OCT3)、SLC22A4(OCTN1)和SLC22A5(OCTN2)。通常它是哺乳动物来源,尤其是来自大鼠、小鼠、兔、猪、豚鼠、黑腹果蝇、秀丽隐杆线虫或人。优选地它是人OCT1。
电极通常包含金属材料或导电金属氧化物,尤其是金、铂、银或铟锡氧化物。
固体支持的传感器电极通常是玻璃或聚合物支持的传感器电极,尤其是浮法硼硅玻璃(borofloat)支持的传感器电极,尤其是浮法硼硅玻璃支持的金电极。在优选的实施方案中,脂质层通过化学键,尤其通过组氨酸标签偶联或链霉抗生物素蛋白-生物素偶联,或通过疏水、亲水或离子力附着到电极上。
电极还是电绝缘的,例如,通过一个或更多绝缘单层,尤其通过一个或更多绝缘的两亲性有机化合物,更尤其是通过一个或更多绝缘的膜单层,最尤其是通过硫醇层,特别是十八烷基硫醇(octadecyl thiol)作为面向电极的下层,并且膜单层作为远离电极的上层进行电绝缘。
传感器芯片尤其包含固相支持体,该固相支持体携带传感器电极和带孔的盖板,形成与滴定平板的那些孔相似的孔。玻璃或聚合物平板用作合适的支持体。在玻璃支持体,例如,玻璃平板的情况下,电极优选包含薄的平版印刷结构的金膜,在其表面已经例如通过硫醇化学修饰过,然而对于聚合物支持体,也可以使用被修饰的厚膜金电极。由于合适底物的范围,可以制造单传感器芯片及传感器带或甚至具有96或384个传感器的传感器阵列板。特别是基于聚合物的传感器具有低成本大量生产的潜力。
通常对于所有的传感器类型,在已进行表面修饰并用水溶液将孔填满后,金表面变成电容器。可以通过带电的参比电极如Pt/Pt或Ag/AgCl、氧化铟锡或其它与溶液接触的电极来测定该电容器的性质。此外,传感器表面优选为非常亲水的,即对膜碎片和小泡是粘性的。因此,保持在它天然或类似天然的环境中,例如在生物膜片、小泡或蛋白脂质体中的OCT容易吸附到亲水传感器表面上,形成区室,该区室的内部空间和它的溶液与金表面及孔内周围溶液电绝缘。如果插入到小杯中,芯片的孔确定流动室的内水体积,使得能够在传感器表面上进行快速的溶液交换。
用于本发明的无细胞电生理传感器芯片例如在WO02/074983中有所描述,尤其是在权利要求和/或图1和/或2(包括所述PCT申请的附图描述)中,将其引入本文作为参考,除非在本发明中另作说明。也可从IonGateBiosciences GmbH,Frankfurt/Main,德国获得(以SURFE2R生物传感器系统名称出售)。
如果将不含底物或OCT活化剂的溶液变换成含底物或OCT活化剂的溶液,可诱导电极的可测量瞬时充电电流,其范围通常在100pA到4nA间。因此,用活化溶液即含底物的溶液替换非活化溶液将触发OCT的活性。随后反向替换溶液可将传感器芯片恢复到它的初始状态。根据本发明,含离子溶液的独特优势在于将假象减到最小,导致特异且灵敏的信号。
可以在PC或另外在受控工作站中调整进行溶液交换实验所必需的所有组分。在常规体系中,非活化(即不含底物)溶液和活化溶液通常保存在玻璃瓶中。通常应用于瓶子的气压推动溶液通过电机操作的阀系统并通过流动室。或者,可以使用自动进样器以自动模式处理几种溶液。
在使用传感器芯片前,优选用含离子的洗涤溶液洗涤电极。
在任何情况下,本发明的含离子溶液优选包含选自Na+、K+、Mg2+和/或Ca2+的单价和二价离子。
含离子溶液中离子的总浓度优选为从约100mM到约1000mM,尤其是从约200mM到约500mM,更尤其是从约300mM到约500mM,最尤其是约435mM。含离子溶液中单价离子的浓度优选为从约300mM到约400mM,并且含离子溶液中二价离子的浓度优选为从约2mM到约10mM,尤其是从约5mM到约8mM,更尤其是约5mM。
在另一优选的实施方案中,含离子溶液还包含缓冲液,尤其是HEPES/NMG,30±10mM,pH 7.0±1.0缓冲液。
含离子溶液的实例为:
(a)洗涤溶液:
30±10mM的缓冲液,例如HEPES/NMG,pH 7.0±1.0,
300±100mM的单价离子,例如NaCl,
4±2mM的二价离子,例如MgCl2。
(b)非活化溶液:
30±10mM的缓冲液,例如HEPES/NMG,pH 7.0±1.0,
300±100mM的单价离子,例如NaCl,
4±2mM的二价离子,例如MgCl2,和
0.5-100mM的单价离子,例如NaCl,其应该与活化溶液中底物的浓度等摩尔。
(c)活化溶液:
30±10mM的缓冲液,例如HEPES/NMG,pH 7.0±1.0,
300±100mM的单价离子,例如NaCl,
4±2mM的二价离子,例如MgCl2,和
0.5-100mM的底物,例如氯化胆碱。
活化溶液的底物通常是有机阳离子,尤其是阳离子药物、阳离子异生素和/或阳离子维生素,更尤其是伯胺、仲胺、叔胺或季胺,最尤其是胆碱、乙酰胆碱、烟碱、N1-甲基烟酰胺、吗啡、1-甲基-4-苯基吡啶鎓、普鲁卡因酰胺、四乙铵、三丁基甲基铵、异喹胍或生物胺如肾上腺素、去甲肾上腺素(norpeinephrine),或肉碱,或亲脂化合物如奎宁、奎尼丁,或类固醇如皮质酮,或有机阴离子如对氨基马尿酸、丙磺舒。
通常,使用电流和/或电势测定法测定电信号,并且将步骤(b)到(d)进行至少2次,尤其进行2到4次。
该发明的上下文中,术语“电信号”或“电流”将指响应活化溶液替换非活化溶液的峰电流,包括但不局限于最大峰电流。电流振幅通常在10到100ms内上升,接着在约2秒内较慢的衰减。电流的极性可以是正的或负的,取决于运输的离子的极性和/或蛋白质转移部分的极性及它们跨区室膜或在区室膜内的转运或转移的矢量方向。对于OCT活性的测定,通常不考虑由非活化溶液替换活化溶液引起的电流或由洗涤溶液替换非活化溶液引起的电流。优选选择流速和间隔,使得响应活化溶液替换非活化溶液的电流不受其它替换步骤引起的电流响应的影响而保持不偏倚。
本发明的方法也可以在化合物,尤其是OCT的刺激剂(活化剂)或抑制剂的存在下进行。
因此,本发明也涉及鉴定调节OCT活性的化合物的方法,包括以下连续步骤:
(a)进行本发明的方法,并
(b)鉴定所述化合物。
所述化合物通常为有机阳离子,尤其是阳离子药物、阳离子异生素和/或阳离子维生素和/或生物胺,更尤其是伯胺、仲胺、叔胺或季胺,其中化合物通常是OCT的刺激剂或抑制剂。化合物例如可存在于化合物文库中。
本发明的另一主题是含有OCT的无细胞电生理传感器芯片本身,如上详细描述。根据本领域技术人员通常所知的方法和/或如实施例中特别描述的方法将OCT结合到传感器芯片上。
传感器芯片还包含用于从电极获取测量数据的数据获取装置,和任选地使交换和/或混合含离子溶液可行的交换和/或混合手段。传感器芯片也可以是微量培养板或微量滴定板的形式。
本发明的另一主题是装置,其包含本发明传感器芯片、参比电极、用于从电极获取测量数据的数据获取装置、用于使交换和/或混合含离子溶液可行的交换和/或混合手段、流动分析装置、电源、计算机和自动进样器。参比电极优选为Pt/Pt、Ag/AgCl或氧化铟锡电极。
本发明的另一主题是试剂盒,该试剂盒含有
(a)本发明无细胞电生理传感器芯片或本发明装置,
(b)至少一种如上定义的含离子溶液,和任选地
(c)如上定义的底物。

材料

测定方法
(a)膜的制备
通过离心从病毒转染的Sf9或HighFive悬浮细胞系或稳定转染的贴壁CHO细胞系中收集细胞后,将约2g湿重细胞的等分试样在液氮中快速冷冻并储存于-80℃用于进一步的制备。
将细胞沉淀在冰上融化并将其转移到冰预冷的缓冲液中(0.25M蔗糖,5mM Tris pH 7.5,2mM DTT,每50ml含一种完整的蛋白酶抑制剂混合物片剂(Roche Diagnostics GmbH,Mannheim,德国))。
通过细胞破裂制备膜碎片。利用Parr Cell Disruption Bomb(ParrInstrument Company,Illinois,美国)通过氮细胞破碎方法或利用DounceHomogenisator(7ml,来自Novodirect GmbH,Kehl/Rhein,德国)通过Dounce匀浆方法对细胞进行匀浆,并在4℃和680g下将悬浮液离心10分钟,在4℃和6100g下将悬浮液离心10分钟。收集上清液并在SW41甩平式转头中于4℃和100,000g下再次离心1小时。
在约2ml的5mM Tris pH 7.5中将沉淀悬浮。用87%的蔗糖(在5mMTris中)将悬浮液调整到56%。然后在底部以2ml的56%级分,接着是3ml 45%蔗糖、3ml 35%和2ml 9%蔗糖开始建立蔗糖梯度。
在4℃和100000g下再次离心2.5小时(或甚至更多),用巴氏吸管小心吸取梯度带并将其收集在含5ml 300mM NaCl、5mM MgCl2、30mMHepes pH 7.5或10mM Tris/HCl pH7.5的新管中。
接着是另一离心步骤:在150000g,4℃下1小时。
将得到的沉淀在300mM NaCl、5mM MgCl2、2mM DTT、30mMHepes pH 7.5、10%甘油中重悬浮。
(b)生物传感器的制备
根据以下方案制备生物传感器。
1.向生物传感器加入30μl硫醇溶液(异丙醇中含2%的硫醇)
2.温育时间:15分钟
3.用3×70μl异丙醇冲洗
4.真空干燥生物传感器
5.干燥时间:30分钟

用“活化”缓冲液和“非活化”缓冲液填满生物传感器体系的缓冲液容器A、B和C后,将模型(dummy)装到传感器固定器上并用所有的缓冲液冲洗体系以从整个流体系统中去除气泡。然后用标准的基于玻璃的传感器替换空传感器或盲(blind)传感器,其中用含有hOCT2的CHO膜碎片预加载所述基于玻璃的传感器(3mm直径的化学修饰的金表面,IonGateBiosciences GmbH,Frankfurt/M.,德国)。通过将气压应用到缓冲液容器中来完成经过流体系统(包括传感器流动室)的液体转运。
测定通常在250毫巴超压下进行,该压力导致约300μls-1的流速。对于它的活性的测定,通过“非活化”和“活化”溶液连续处理包含OCT蛋白质的膜。随后以相反顺序替换溶液将传感器芯片恢复到它的初始状态。通过控制软件定义顺序(见图1),其中“非活化”缓冲液流经传感器表面,接着是“活化”缓冲液和“非活化”缓冲液。在整个顺序中,将电流响应数字化(2000样品s-1)并保存为数据文件。对于剂量反应实验,分别将抑制剂溶解在“非活化”和“活化”缓冲液中。所有的化学试剂为分析级或更高级别。
数据分析
高对照:抑制前用100mM氯化胆碱激活后的谷值电流;
低对照:抑制后用0mM氯化胆碱激活后的谷值电流;
结果由校正的原始数据计算而来。
结果
1.图1A和1B显示的是抑制前(黑色标记线)和后(灰色标记线),向传感器(分别具有包含rOCT2和hOCT2的固定膜)加入含有胆碱的活化溶液后的电响应。峰振幅等同于转运蛋白的初始活性;衰减归因于生物传感器夹层结构的电容的充电。
2.图2A和2B显示的是分别在包含rOCT2和hOCT2的膜上胆碱浓度对电响应值(高对照)的影响。
根据胆碱浓度滴定法的结果,在以下试验中使用100mM的胆碱浓度,因为这允许测定具有高振幅的信号。
3.测定的pH依赖性表明在pH 7.4时具有最高的蛋白质活性,其因此被用于后续试验中(图3)。对于抑制实验,将胆碱浓度降低至10mM(在检测竞争性抑制剂效应的KM值范围内)。测定OCT的标准抑制剂(TBA)的IC50分别为3.5μM(对于rOCT2)(图4A)和2.9μM(对于hOCT2)(图4B)。
4.通过使用上面定义的参数比较来自重组细胞系的不同膜制剂。用CHO细胞系获得最佳结果。昆虫细胞制剂产生高品质的信号,然而具有减弱趋势,不适合于IC50的测定。
5.通过人工膜片钳电生理学进一步监测CHO细胞系,所述人工膜片钳电生理学被认作离子转运蛋白研究的最佳标准。对于rOCT2,hOCT2的电流几乎检测不到,并且不能测定IC50值(图5A和5B)。
6.对于信号灵敏度的进一步评估,检测了其他底物和抑制剂。图6A和6B显示了这些实例。
连同此处报道的对于OCT2其他家族成员,例如hOCT1或hOCT3的测定一起,克隆和产生了构建体。利用Invitrogen’s Flpln-和T-REXSystem(目录号R758-07)产生了细胞系。
Claims (53)
1.无细胞电生理传感器芯片在制备用于通过用含离子的非活化溶液处理所述传感器芯片、用含离子和底物的活化溶液处理所述传感器芯片、然后测定电信号而测定有机阳离子转运蛋白(OCT)活性的试剂盒中的用途,其中
所述无细胞电生理传感器芯片含有固体支持的传感器电极和脂质层,所述脂质层含有位于传感器电极最接近空间附近的所述OCT,而所述传感器电极相对于所用的溶液和所述脂质层是电绝缘的。
2.权利要求1的用途,其中所述OCT选自OCT1(SLC22A1)、OCT2(SLC22A2)、OCT3(SLC22A3)、OCTN1(SLC22A4)、OCTN2(SLC22A5)。
3.权利要求1或2的用途,其中所述OCT为哺乳动物来源或者来自黑腹果蝇或秀丽隐杆线虫。
4.权利要求3的用途,其中所述哺乳动物为大鼠、小鼠、兔、猪、豚鼠或人。
5.权利要求3的用途,其中OCT为人OCT1(SLC22A1)。
6.根据权利要求1或2的用途,其中所述电极包含金属材料或导电金属氧化物。
7.根据权利要求6的用途,其中所述电极包含金、铂、银或铟锡氧化物。
8.根据权利要求1或2的用途,其中所述固体支持的传感器电极是玻璃或聚合物支持的传感器电极。
9.根据权利要求8的用途,其中所述固体支持的传感器电极是浮法硼硅玻璃支持的传感器电极。
10.根据权利要求8的用途,其中所述固体支持的传感器电极是浮法硼硅玻璃支持的金电极。
11.根据权利要求1或2的用途,其中所述脂质层通过化学键,或通过疏水、亲水或离子力附着在电极上。
12.根据权利要求11的用途,其中所述脂质层通过组氨酸标签偶联或链霉抗生物素蛋白-生物素偶联附着在电极上。
13.根据权利要求1或2的用途,其中所述电极被一种或更多绝缘单层电绝缘。
14.根据权利要求13的用途,其中所述电极被被一种或更多绝缘的两亲性有机化合物电绝缘。
15.根据权利要求13的用途,其中所述电极被一种或更多绝缘膜单层电绝缘。
16.根据权利要求13的用途,其中所述电极被硫醇层作为面向电极的下层并且膜单层作为远离电极的上层进行电绝缘。
17.根据权利要求16的用途,其中所述硫醇是十八烷基硫醇。
18.根据权利要求1或2的用途,其中测定所述活性包括首先用含离子的洗涤溶液洗涤所述电极的步骤。

31.根据权利要求30的用途,其中所述有机阳离子是阳离子药物、阳离子异生素和/或阳离子维生素,所述亲脂化合物是奎宁、奎尼丁,或类固醇,所述有机阴离子是对氨基马尿酸、丙磺舒。
32.根据权利要求31的用途,其中所述有机阳离子是胆碱、乙酰胆碱、烟碱、N1-甲基烟酰胺、吗啡、1-甲基-4-苯基吡啶输、普鲁卡因酰胺、四乙铵、三丁基甲基铵、异喹胍,所述类固醇是皮质酮。
33.根据权利要求30的用途,其中所述生物胺是肾上腺素、去甲肾上腺素或肉碱。
34.根据权利要求1或2的用途,其中使用电流和/或电势测定手段测定所述电信号。
35.根据权利要求1或2的用途,其中用含离子的非活化溶液处理所述传感器芯片、用含离子和底物的活化溶液处理所述传感器芯片、然后测定电信号进行至少2次。
36.根据权利要求35的用途,其中用含离子的非活化溶液处理所述传感器芯片、用含离子和底物的活化溶液处理所述传感器芯片、然后测定电信号进行2到4次。
37.根据权利要求1或2的用途,其中用含离子的非活化溶液处理所述传感器芯片、用含离子和底物的活化溶液处理所述传感器芯片、然后测定电信号在化合物的存在下进行。
38.根据权利要求37的用途,其中所述化合物为OCT抑制剂。
39.无细胞电生理传感器芯片在制备用于通过用含离子的非活化溶液处理所述传感器芯片、用含离子和底物的活化溶液处理所述传感器芯片、然后测定电信号而测定化合物调节OCT活性之活性的试剂盒中的用途,其中:
所述无细胞电生理传感器芯片含有固体支持的传感器电极和脂质层,所述脂质层含有位于传感器电极最接近空间附近的所述OCT,而所述传感器电极相对于所用的溶液和所述脂质层是电绝缘的。
40.权利要求39的用途,其中测定所述活性在活化溶液的底物存在和/或不存在下进行。
41.无细胞电生理传感器芯片在制备用于通过用含离子的非活化溶液处理所述传感器芯片、用含离子和底物的活化溶液处理所述传感器芯片、然后测定电信号而鉴定调节OCT活性的化合物的试剂盒中的用途,其中:
所述无细胞电生理传感器芯片含有固体支持的传感器电极和脂质层,所述脂质层合有位于传感器电极最接近空间附近的所述OCT,而所述传感器电极相对于所用的溶液和所述脂质层是电绝缘的。
42.权利要求41的用途,其中所述化合物为有机阳离子、生物胺、亲脂化合物、有机阴离子。
43.权利要求42的用途,其中所述有机阳离子是阳离子药物、阳离子异生素和/或阳离子维生素。
44.权利要求42的用途,其中所述化合物为伯胺、仲胺、叔胺或季胺。
45.权利要求41的用途,其中所述化合物为OCT的抑制剂。
