Abstract

按照一个安全范围达到106细菌的有效的灭菌方法对一种源于动物或人类的生物材料灭菌,所述材料被用做植入物或者移植物,通过该方法的4个连续步骤,有害微生物被物理地分离和化学地杀菌,所述步骤开始是在抗生素,然后是在脱氧胆酸,再然后是在表面活性物质并且最终是在初级醇中进行杀菌,分别伴随着在步骤2-4之后用水溶液漂洗的步骤。安全范围大于106细菌的灭菌生物心脏瓣可以不经任何进一步处理植入到体内,没有钙化倾向,并确保长的服务年限。

Description

生产可植入或可移植生物材料的灭菌方法
技术领域
本发明涉及一种生产源于动物或人类的可植入或可移植生物材料的灭菌方法。
背景技术
长期以来,已有多种对异种或外源物质进行灭菌并将其作为植入物或移植物的成果。例如,已知从猪,牛,或者马获得的外源材料制造生物的心脏瓣,使用戊二醛以封闭表面并且也起到杀菌消毒的作用。以这种方法生产的生物心脏瓣满足强制的杀菌要求但是却包含阻碍,即依赖于患者的年龄,可能发生逐渐的钙化作用,这可能造成生物心脏瓣失效。
技术内容
本发明的目的是说明一种对源于动物或人类的生物材料进行灭菌的方法,以提供经国际标准验证,不需费力,并在有害微生物或随后的钙化作用方面没有风险的生物植入物或移植物。
这一目的是根据包含本发明权利要求1所描述的特征的方法而实现的。从属权利要求公开了本发明进一步的特征和有益的改善。
基本的发明思想是用4个连续的过程步骤结合处理动物源的材料,每个步骤以特定的时间间隔实施,首先置于抗生素溶液中,然后放到脱氧胆酸溶液,然后放入表面活性物质中,和最后放入初级醇中。每步所使用的灭菌物质对各自的微生物同时具有物理的灭菌效果,例如,分离和移除,以及化学灭菌效果。第二到第四步的处理步骤每一步都跟随着一个使用水溶液漂洗的步骤,优选使用氯化钠溶液,以除去灭菌剂和分离出的微生物。以特定的顺序和时间间隔进行灭菌和漂洗步骤是很重要的。
所述的灭菌方法保证对材料源的灭菌满足国际标准并达到大于106的病菌的安全辐度(margin of safety)。以这种方法消毒的生物物质可以植入(或移植到)人体而没有任何安全风险。因为根据本发明的杀菌方法也从材料中将导致钙化的磷脂分离出来,使得钙化的风险明显降低。植入物或移植物的年限增加。
具体实施方式
通过从猪源材料生产心脏瓣植入物这一实例将更详细的解释根据本发明的一个具体实施方式的方法。
在第一步中,取猪主动脉瓣,剪切并除去脂肪,在抑制细菌和真菌生长的抗生素溶液中保存特定的时间。同时,完成了物理分离作用以减少微生物的数量。抗生素溶液可以含有,例如,青霉素,链霉素和两性霉素。
在随后的第二步中,将之前用抗生素处理的主动脉瓣放入含脱氧胆酸的生理盐水溶液。Sodium deoxylate具有去细胞化的作用,即杀死和分离存在于组织中的细胞的作用,以及进一步阻止微生物生长的抗生素作用。
在上述两步骤之后,用水溶液多次冲洗主动脉瓣以洗掉脱氧胆酸和表面的细菌。
在随后的步骤(第三步)中,将主动脉瓣放入表面活性物质中,即具有溶血,抗病毒和抗菌特性,和-除了其化学作用-含有可影响微生物结合和分离的物理作用的脂蛋白。
脂蛋白处理后,也用水溶液进行多次冲洗的步骤以洗去前述步骤中任何残留的洗涤剂并同时减少细菌的数量。
接下来的步骤-第四步-包括用例如70%乙醇的初级醇处理主动脉瓣,其作用于细菌和真菌的营养细胞以及含有包膜的病毒。另外,可能仍旧存在的并且主要导致生物植入物钙化的磷脂被从细胞膜上除去。当猪心脏瓣放入用于处理的化学品中时其化学作用再一次伴随着物理分离过程。
第四步后,再一次进行水溶液多次冲洗步骤以平衡由乙醇脱去的组织中的水并利用稀释作用进一步减少细菌和真菌部分。
作为上述猪主动脉瓣杀菌处理步骤的结果,得到了按照欧洲标准消毒的,并且可以在不进行任何额外处理的情况下被植入的生物心脏瓣。
通过授权的检定中心对所述过程的杀菌效果以及遵照基于欧洲标准ISO14160的分别的无菌限度进行了确认。这一灭菌方法的一个测试发现由于在每步处理中使用的化学杀菌剂的联合作用-化学(杀死)和物理(分离和移除)-使得获得了按照欧洲标准大于106病菌的安全辐度(margin of safety)。

Claims (5)

1.一种生产源于动物或人类的可移植或可植入生物材料的灭菌方法,其特征在于所述方法包括4个灭菌步骤,其按照下述顺序一个接一个的进行并具有一个联合的化学和物理作用:
1)在抗生素溶液中处理原材料以阻止微生物的生长并分离它们;
2)进一步在脱氧胆酸中处理上述材料以阻止微生物的生长并分离它们,并结合接着的第一次在水溶液中漂洗的过程,以除去脱氧胆酸并从表面洗去被分离的微生物;
3)进一步用表面活性脂蛋白溶液处理材料,所述脂蛋白对于微生物具有溶血,抗细菌,抗病毒和分离的作用,并为了进一步除去脱氧胆酸,结合随后的第二次在水溶液中漂洗的过程;和4)用具有杀真菌的,杀细菌的和抗病毒作用的初级醇进一步处理材料以进一步结合被分离并彻底破坏的微生物,结合随后的第三次在水溶液中漂洗的过程。
2.根据权利要求1的方法,其中初级醇是70%的乙醇。
3.根据权利要求1的方法,其中一个漂洗过程包括多次连续的漂洗步骤。
4.根据权利要求1的方法,其中在灭菌步骤1-4中所用的具有化学和物理作用的灭菌剂溶于钠盐溶液。
5.根据权利要求1的方法,其中将含水钠盐溶液用作洗涤剂。
