Abstract

本发明涉及一种显微持卵管。该显微持卵管的吸持端为斜面开口,即持卵管的吸持端开口的端面与吸持端纵轴的夹角的锐角小于90°,且最好在25°-85°之间。本发明有利于从卵子或胚胎的侧面而非正面用显微注射针或辅助孵化针顺利刺破透明带而不造成卵子或胚胎受压变形,对多种显微操作有利。这一新持卵管及其相应的操作方式具有减少胚胎受损,提高胚胎健康发育能力的意义。

Description

吸持开口斜面型显微持卵管
技术领域
本发明涉及一种显微持卵管。
背景技术
显微持卵管(micro-holding pipette)是一种用于显微操作的负压吸持固定器,可用于显微镜下固定人或动物的卵、胚胎等。到目前为止,显微持卵管的开口端端面均为垂直端,即开口端面与开口端纵轴夹角为90°,(参见图1、图2)这种设计已广泛用于卵子的胞浆内单精子显微受精和胚胎的辅助孵化、胚胎移植前遗传诊断等的显微操作。但是这种设计使吸持端吸持卵子或胚胎仅限于9:00位置,这就限定了对卵子或胚胎的操作模式,即吸持端对侧操作针(如显微注射针,辅助孵化针或取单卵裂球针)只有在3:00位置进针穿入透明带内最容易,而在其它位置如5:00时则由于吸持端和对侧操作针端作用力不同轴而使操作困难,造成操作时间长和操作时卵或胚胎受压变形,这些都不利于胚胎健康发育。
然而,胚胎机械法辅助孵化等一些显微操作需要对4:00-6:00位置进行操作,而且发明者在实践中还发现,当使用传统显微持卵管时,对侧操作针即使在3:00位置进针穿入透明带内操作上最容易,但仍然因为对侧操作针直接对卵或胚胎细胞用力而在操作时使卵或胚胎受压变形。若在4:00-6:00位置先穿入透明带再行其它操作,则可避免对侧操作针直接对卵或胚胎细胞用力,而只对其侧面的透明带用力,这样对卵子或胚胎机械挤压少,但这一操作若用常规显微持卵管吸持,则会有前述的由于双侧操作微管作用力不同轴而使操作困难。
发明内容
本发明需要解决的技术问题是:显微持卵管能方便地吸持卵子或胚胎的8:45-7:00位置,使对侧操作针能方便地在4:00-5:30位置进行操作。
本发明所采用的技术方案是:该显微持卵管的吸持端的开口为斜面开口,即持卵管的吸持端开口的端面与吸持端纵轴的夹角的锐角小于90°,且最好在25°-85°之间。
本发明的有益效果:发明人已实验证明该型持卵管有利于从卵子或胚胎的侧面而非正面用显微注射针或辅助孵化针顺利刺破透明带而不造成卵子或胚胎明显受压变形,对多种显微操作有利。这一新持卵管及其相应的操作方式具有减少胚胎受损,提高胚胎健康发育能力的意义。

具体实施方式
现以胚胎机械法透明带打孔中的穿刺透明带的步聚为实施例说明如下:
参见图3-图4,本发明吸持开口斜面型显微持卵管1的吸持端的开口2为斜面开口,即持卵管的吸持端的开口端面5与吸持端纵轴6的夹角的锐角小于90°,最好在25°-85°之间。
参见图5,当显微持卵管1为斜面开口时,卵子3被吸持于约8:00的位置,此时对侧操作针4可以方便地从约5:00的位置穿入卵子3外围的透明带,而得到如图6所示结果。此过程中吸持端与对侧操作针端的作用力基本上同轴使得穿刺顺利,不造成卵子或胚胎受压变形。而常规显微持卵管为垂直型开口,胚胎或卵总被吸持于9:00的位置,则上述同样操作会由于吸持端与对侧操作针端的作用力不同轴而使操作困难,造成操作时间长和操作时卵或胚胎受压变形。
使用本发明吸持开口斜面型显微持卵管,使机械法透明带打孔得到了如下进步:1、操作便捷,速度提高,减少了胚胎体外操作时间从而避免体外长时间操作带来的多种影响;2、减少了常规法对胚胎造成的挤压而有利于避免胚胎受损。另外,本发明吸持开口斜面型显微持卵管的出现,将打破传统的多种胚胎显微操作模式,带来许多新的技术创新,发明者除将该吸持开口斜面型显微持卵管用于胚胎机械法透明带打孔(包括辅助孵化和胚胎移植前遗传诊断)外,还正在研究将其用于显微授精等新方法。

Claims (2)

1.一种吸持开口斜面型显微持卵管,其特征在于持卵管的吸持端的开口为斜面开口,即持卵管的吸持端开口的端面与吸持端纵轴的夹角的锐角小于90°。
2.根据权利要求1所述的一种吸持开口斜面型显微持卵管,其特征在于持卵管的吸持端开口的端面与吸持端纵轴的夹角的锐角的范围在25°-85°之间。
