Description
Verfahren zur Darstellung von Tropinon monocarbonsäureäthylester. Es ist bekannt (vergl. Journal of the Che- mical Society of London, Bd. 111112 (1917), S. 762 ff.), dass durch Behandlung des Kon densationsproduktes von Succindialdehyd, Acetondicarbonsäurediäthylester und Metbyl- amin mit Säuren Tropinon erhalten werden kann.
Es wurde nun gefunden, dass xnan durch Verseifung des Tropinondicarbonsäurediäthyl- esters unter milden Bedingungen zu dem Tropinonmonocarbonsäureäthylester gelangen kann, welcher als Ausgangsstoff für Cocain- synthesen technische Bedeutung besitzt. Es war nicht vorauszusehen, dass einerseits die Verseifung nur der einen Estergruppe glatt gelingen würde, und-'dass anderseits die frei gewordene Carboxylgruppe sich ohne Schädi gung des Restmoleküls abspalten lassen würde.
Denn die durch halbseitige Verseifung des Diesters zunächst entstehende Verbindung, welche eine stark basische Gruppe und eine freie Carboxylgruppe enthielt, hätte infolge innerer Salzbildung der Abspaltung von Kohlen- säure einen ähnlichen Widerstand entgegen setzen können, wie z. B. Clykokoll öder Ecgonin.
<I>Beispiel 1:</I> 28 Teile Tropinondicarbonsäui@ediäthyl- ester, welches ein dickes, schwach basisches Öl ist, welches durch Kondensation von Aoe- tondicarbonsäureester mit Succindialdehyd und lylethylamin in alkalischer Lösung gewonnen werden kann, werden in 50 Teilen Alkohol gelöst. Dazu fügt man 22 Teile Kalilauge <B>(l.</B> l) und erwärmt kurze Zeit bis zum Sieden.
Nach dem Erkalten fügt man etwas Eis hin zu, säuert mit Schwefelsäure an; übersättigt mit Ammoniak und extrahiert völlig mit Äther oder gechlorten Kohlenwasserstoffen. <I>Beispiel 2</I> 5 Teile Tropinondicarbonsäurediäthylester werden mit 30 Teilen Salzsäure (25 >ige) 20 Minuten lang am Rückflusskühler gekocht, wobei Kohlendioxyd entweicht, Nach d.em Abkühlen wird die Lösung mit etwas Tier kohle gerührt und filtriert. Man versetzt das Filtrat unter Kühlung mit konzentriertem Ammoniak im Überschuss und rührt schnell hintereinander mehrmals in einer Trommel mit Äther aus.
Bei den letzten Auszügen wird zwecks Aussalzung etwas Chlorammo nium oder Kaliumcarbonat zugefügt.
Der Äther wird mit wenig Soda getrocknet und abdestilliert. Der Rückstand ist ein dickes Öl, welches beim Verrühren mit wenig Wasser das Hydrat des Tropinonmonocarbonsäure- äthylesters liefert.
Der wasserfreie Ester bildet ein Öl, wel ches in kleinen Mengen unzersetzt destillier bar ist. Beim Verreiben mit Wasser nimmt es 2 Mol. H.a0 auf und geht in ein festes Hydrat über, welches bei 63 schmilzt. Dieses ist mit Wasser ziemlich leicht, in Alkohol in beliebiger Menge löslich. Das Pikrat ist in Benzol leicht löslich und schmilzt bei 135 bis<B>136'.</B> In wässeriger Kalilauge löst sich der Ester leicht und wird so fest gebunden, dass er durch Ausschüttelung, mittelst Äther aus der alkalischen Lösung nicht gewonnen werden kann.
Process for the preparation of tropinone monocarboxylic acid ethyl ester. It is known (cf. Journal of the Chemical Society of London, Vol. 111112 (1917), p. 762 ff.) That tropinone can be obtained by treating the condensation product of succindialdehyde, diethyl acetone dicarboxylate and methylamine with acids .
It has now been found that by saponification of the tropinone dicarboxylic acid diethyl ester under mild conditions, the tropinone monocarboxylic acid ethyl ester can be obtained, which is of industrial importance as a starting material for cocaine syntheses. It was not foreseeable that, on the one hand, the saponification of only one ester group would succeed smoothly and, on the other hand, the carboxyl group that had become free could be split off without damaging the remaining molecule.
Because the compound initially formed by one-sided saponification of the diester, which contained a strongly basic group and a free carboxyl group, could have offered a similar resistance to the splitting off of carbonic acid as a result of internal salt formation. B. Clycol or Ecgonine.
<I> Example 1: </I> 28 parts of tropinondicarboxylic acid diethyl ester, which is a thick, weakly basic oil, which can be obtained by condensation of aoetondicarboxylic acid ester with succindialdehyde and ethylamine in alkaline solution, are dissolved in 50 parts of alcohol solved. Add 22 parts of potassium hydroxide solution (left) and heat it to boiling for a short time.
After cooling, add a little ice, acidify with sulfuric acid; oversaturated with ammonia and fully extracted with ether or chlorinated hydrocarbons. <I> Example 2 </I> 5 parts of tropinondicarboxylic acid diethyl ester are boiled with 30 parts of hydrochloric acid (25> ige) for 20 minutes on the reflux condenser, during which carbon dioxide escapes. After cooling, the solution is stirred with a little animal charcoal and filtered. Concentrated ammonia in excess is added to the filtrate, while cooling, and it is stirred several times in a drum with ether in rapid succession.
In the last extracts, a little chlorine ammonium or potassium carbonate is added for the purpose of salting out.
The ether is dried with a little soda and distilled off. The residue is a thick oil which, when stirred with a little water, yields the hydrate of the ethyl tropinone monocarboxylate.
The anhydrous ester forms an oil which can be distilled in small amounts without decomposition. When rubbed with water, it absorbs 2 moles of H.a0 and turns into a solid hydrate, which melts at 63. This is fairly easy with water, and can be dissolved in any quantity in alcohol. The picrate is easily soluble in benzene and melts at 135 to <B> 136 '. </B> The ester dissolves easily in aqueous potassium hydroxide solution and is so firmly bound that it cannot be extracted from the alkaline solution by shaking it out using ether can.
