Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Darstellung einer kol loidal löslichen Metallpyrophosphatkasein-Ver- bindung, welches darauf beruht, dass man das Kasein in Alkalipyrophosphat löst und zu dieser Lösung ein Calciumsalz zugibt.
Metallpyrophosphatkasein - Verbindungen sind bereits in der Therapie zur Anwendung gekommen. Von diesen Präparaten wird mit Rücksicht auf den besonderen Verwendungs zweck gefordert, dass sie in Alkali gut lös lich sind; denn nur solche Verbindungen werden vermittelst des alkalischen Darm saftes vollständig und ohne Schwierigkeiten resorbiert. Die bekannten Metallpyrophosphat- kasein-Verbindungen entsprechen diesen Be dingungen nicht oder nur teilweise.
Nach den bekannten Verfahren zur Her stellung von Metallpyrophosphatkasein-Ver- bindungen wird zum Beispiel Kasein in Al kali gelöst und die alkalische Lösung mit Natriumphosphat, sowie Metallsalzlösungen behandelt. Beispielsweise wird nach einem Verfahren empfohlen, die sauren Valenzen des Kaseins durch Aufnehmen in einem ÜTber- schusse von Alkali oder Erdalkali vollständig abzusättigen und den 11letaphosphorsäurerest durch Zufügen von Metaphosphorsäure zur alkalischen Lösung dem Eiweissmolekül an zulagern.
In gleicher Weise wird nach einem weiteren Verfahren zur Herstellung von Tri- calciumphosphateiweiss-Verbindungen vorge gangen. Beide Verfahren haben den Nach teil, dass die darnach hergestellten Verbin dungen nicht vollständig alkalilöslieh sind. Um die Alkalilöslichkeit zu erhöhen, wird ernpfohleri, in grosser Verdünnung zu arbeiten, was in der Technik wenig vorteilhaft ist. Ferner ist auch schon der Vorschlag gemacht worden, Pyrophosphorsäure und ihre Salze für die Herstellung alkalilöslicher Calcium- und Eisen phosphörsäureeiweiss-Verbindungen zu verwen den.
Es wird aber auch bei diesen Verfahren zur Erlangung der gewünschen Produkte auf den Zusatz von freiem Alkali besonderer Wert gelegt. Neben der unvollkommenen Alkali löslichkeit ist an den bisher bekanntgewor denen Verbindungen noch auszusetzen, dass sie -- wohl infolge des Zusatzes von freiem Alkali bei der Einführung des Phosphorsäure- re,tes in das Eiweissmolekül - nur einen geringen Phosphorsäuregehalt aufweisen.
Nach dem neuen Verfahren wird das Ka sein, ohne es vorher in Alkali zu lösen, in Alkalipyrophosphat aufgenommen und die entstandene Lösung mit einem Calciumsalz umesetzt. Durch diese einfache Massnahme des, Lösens in Alkalipyrophosphat wird gleich zeitig der Eintritt des Phosphorsäurerestes in das Eiweissmolekül bewirkt. Das Reaktions- gemisch zeigt nach dem Zusatze des Metall salzes eine saure Reaktion,
wodurch sich die Unisetzung von den bisher bekannten Ver fahren, bei welchen in einer alkalischen Lö sung gearbeitet wird, wesentlich unterschei det. Das Verfahren hat vor den bekannten den Vorteil, dass die Reaktion in grösserer Konzentration vorgenommen werden kann. Die kolloidal lösliche Metallpyrophosphat- kasein-Verbindung besitzt einen hohen Gehalt an Metall und Phosphorsäure, sowie vollstän dige Alkalilöslichkeit. Die verdünnten alkali schen Lösungen sind beständig.
Die neue Verbindung soll in der Therapie Verwendung finden. Beispiel.: 66 Teile Kasein werden in einer Lösung von 112 Teilen kristallisiertem Natriumpyro- phosphat und 800 Teilen Wasser gelöst und unter Rühren mit 156 Teilen einer vierfach normalen Calciumchloridlösung versetzt. Durch Zugabe von etwas Alkohol wird die neue Verbindung gefällt.
Nach dem Waschen wird sie bei<B>500</B> getrocknet. Das Präparat gibt in verdünntem Natriunibikarbonat oder in ver- dünnter Sodalösung eine beständige Lösung.

According to the known method for producing metal pyrophosphate casein compounds, for example, casein is dissolved in alkali and the alkaline solution is treated with sodium phosphate and metal salt solutions. For example, one method recommends completely saturating the acidic valences of casein by absorbing an excess of alkali or alkaline earth and adding metaphosphoric acid to the protein molecule by adding metaphosphoric acid to the alkaline solution.
In the same way, a further process for the production of calcium phosphate white compounds is proceeded. Both processes have the disadvantage that the compounds produced in accordance with them are not completely soluble in alkali. In order to increase the alkali solubility, it is recommended to work in great dilution, which is not very advantageous in technology. Furthermore, the proposal has already been made to use pyrophosphoric acid and its salts for the preparation of alkali-soluble calcium and iron phosphoric acid protein compounds.

This simple measure of dissolving in alkali pyrophosphate causes the phosphoric acid residue to enter the protein molecule at the same time. The reaction mixture shows an acidic reaction after the addition of the metal salt,
whereby the unisposition of the previously known Ver drive, in which work is carried out in an alkaline solution, significantly different. The method has the advantage over the known ones that the reaction can be carried out in greater concentration. The colloidally soluble metal pyrophosphate casein compound has a high content of metal and phosphoric acid, as well as complete solubility in alkali. The diluted alkaline solutions are stable.
The new compound should be used in therapy. Example: 66 parts of casein are dissolved in a solution of 112 parts of crystallized sodium pyrophosphate and 800 parts of water, and 156 parts of a four-fold normal calcium chloride solution are added while stirring. The new compound is precipitated by adding a little alcohol.
After washing, it is dried at <B> 500 </B>. The preparation gives a permanent solution in diluted sodium bicarbonate or in dilute soda solution.
