BESCHREIBUNG
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Aminofluoranverbindungen, welche mindestens eine aralkylierte Aminogruppe aufweisen.
Die Aralkylgruppe wird in der Regel durch Umsetzen der Aminoverbindung mit einem Aralkylhalogenid, wie z. B. Benzylchlorid oder Benzylbromid eingeführt, wobei die Umsetzung sowohl in polaren Medien als auch in nicht-polaren Lösungsmitteln in Gegenwart von Alkaliverbindungen durchgeführt wird.
Diese Methoden haben jedoch den Nachteil, dass bei polycyclischen Aminoverbindungen das Endprodukt in manchen Fällen in ungenügender Ausbeute erhalten wird.
Der vorliegenden Erfindung liegt die Aufgabe zugrunde, ein verbessertes Verfahren zu entwickeln, welches eine Steigerung der Ausbeute verspricht und gleichzeitig zur praktischen Verwendung geeignete Produkte mit hoher Reinheit ohne komplizierte Reinigungsstufen ergibt.
Gegenstand der vorliegenden Erfindung ist daher ein Verfahren zur Herstellung von N-aralkylierten Aminofluoranverbindungen, welches dadurch gekennzeichnet ist, dass man eine Fluoranverbindung der Formel

in der der Ring A unsubstituiert oder durch Halogen, Nitro, Amino, Mononiederalkylamino oder Diniederalkylamino substituiert ist, Rl, R2 und R3, unabhängig voneinander, je Wasserstoff, Halogen, Niederalkyl oder Niederalkoxy,
X1, X2, Y1 und2, unabhängig voneinander, je Wasserstoff, unsubstituiertes oder durch Halogen, Hydroxy, Cyano oder Niederalkoxy substituiertes Alkyl mit höchstens 12 Kohlenstoffatomen, Cycloalkyl, Tetrahydrofurfuryl, Aryl oder Acyl oder die Substituentenpaare (Xl und X2) und (Y1 und Y2), unabhängig voneinander,
je zusammen mit dem gemeinsamen Stickstoffatom einen fünf- oder sechsgliedrigen, vorzugsweise gesättigten, heterocyclischen Rest bedeuten, wobei mindestens in -NX,X2 oder -NYlY2 oder im Ring A eine primäre oder sekundäre Aminogruppe vorhanden ist, mit einem Arylsulfonsäure-Aralkylester umsetzt.
Halogen bedeutet beispielsweise Fluor, Brom, Jod oder vorzugsweise Chlor.
Niederalkyl und Niederalkoxy stellen bei der Definition der Reste der Fluorane in der Regel solche Gruppen oder Gruppenbestandteile dar, die 1 bis 5, insbesondere 1 bis 3 Kohlenstoffatome aufweisen, wiez. B. Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, sek.Butyl, tert.Butyl oder Amyl, bzw. Methoxy, Ethoxy, Isopropoxy, tert.Butoxy oder tert.Amyloxy.
Acyl ist besonders Formyl, Niederalkylcarbonyl, wie z. B.
Acetyl oder Propionyl oder Benzoyl. Weitere Acylreste können Niederalkylsulfonyl, wie z. B. Methylsulfonyl oder Ethylsulfonyl sowie Phenylsulfonyl sein.
Cycloalkyl bedeutet z. B. Cyclopentyl und vor allem Cyclohexyl.
Wenn die Substituentenpaare (X1 und X2) und (Yl und Y2) zusammen mit dem gemeinsamen Stickstoffatom einen heterocyclischen Rest darstellen, so ist dieser beispielsweise Pyrrolidino, Piperidino, Pipecolino, Morpholino, Thiomorpholino oder Piperazino, wie z. B. Methylpiperazino. Bevorzugte gesättigte hete- rocyclische Reste für -NXlX2 und -NY1Y2 sind Pyrrolidino, Piperidino oder Morpholino.
Der Ring A ist vorzugsweise nicht weiter substituiert. Falls er Substituenten aufweist, ist er in erster Linie durch Halogen oder Diniederalkylamino substituiert.
Besonders geeignet unter den Ausgangsstoffen sind die Fluoranverbindungen der angegebenen Formel, in der entweder die Gruppe -NXlX2 oder die Gruppe -NY1Y2 eine primäre Aminogruppe (-NH2) darstellt.
Besonders gute Ergebnisse im erfindungsgemässen Verfahren werden erhalten, wenn man eine Fluoranverbindung der angegebenen Formel verwendet, in der -NYlY2 -NH2 und R2 Wasserstoff oder vor allem Niederalkyl oder insbesondere Methyl bedeuten.
Als Arylsulfonsäure-Aralkylester kommen solche in Frage, bei denen sowohl der Arylrest als auch der Aralkylrest unsubstituiert oder ringsubstituiert sind. Bevorzugte Substituenten sind hierzu Halogene, Nitro, Trifluormethyl, Niederalkyl, besonders Methyl oder Niederalkoxy wie Methoxy.
Besonders geeignet unter diesen Sulfonsäureestern sind Benzolsulfonate, p-Toluolsulfonate, p-Brombenzolsulfonate oder p Nitrobenzolsulfonate.
Als Beispiele für zur Einführung der Aralkylgruppe geeignete Sulfonsäureester seien genannt: p-Toluolsulfonsäurebenzylester,
Benzolsulfonsäurephenethylester, p-Toluolsulfonsäurephenethylester, p-Toluolsulfonsäurephenpropylester, p-Toluolsulfonsäure-a-methylbenzylester,
Benzolsulfonsäurephenpropylester, p-Toluolsulfonsäurephenisopropylester, p-Toluolsulfonsäure-4-chlorbenzylester, p-Toluolsulfonsäure-4-methylbenzylester, p-Toluolsulfonsäure-2,5-dimethyl-benzylester, p-Toluolsulfonsäure-2 ,5-dichlorbenzylester, p-Nitrobenzolsulfonsäurebenzylester, p-Brombenzolsulfonsäurebenzylester, p-Toluolsulfonsäure-2 ,4-dimethyl-benzylester, p-Toluolsulfonsäure-4-trifiuormethylbenzylester, p-Toluolsulfonsäure-4-methoxybenzylester.
Bevorzugter Sulfonsäurearalkylester ist p-Toluolsulfonsäurebenzylester.
Die Aminofluoranverbindung und der Sulfonsäureester werden im allgemeinen in etwa stöchiometrischen Mengen eingesetzt, wobei pro Aminogruppe eine oder zwei Aralkylgruppen eingeführt werden . Ein Überschuss von z. B. 10-15 % an einer der Komponenten ist jedoch ebenfalls möglich.
Die Umsetzung erfolgt zweckmässigerweise bei erhöhter Temperatur, vorzugsweise bei 80 bis 120 "C und in Anwesenheit eines säurebindenden Mittels.
Geeignete säurebindende Mittel sind z. B. Alkalimetallhy- droxide, Erdalkalimetallhydroxide, Alkalimetallhydrogencarbonate, Alkalimetallcarbonate oder tertiäre Stickstoffbasen, wie z. B. Pyridin, N-Methylpiperidin oder Trialkylamine oder auch Mischungen dieser Verbindungen. Bevorzugt ist Natriumcarbonat.
Die Reaktion findet vorzugsweise in einem nicht polaren, vor allem aromatischen Lösungsmittel, wies. B. Benzol, Toluol, Xylol, Chlorbenzole wie Dichlorbenzol oder Trichlorbenzol, oder Nitrobenzol statt. Bevorzugtes Lösungsmittel ist Toluol.
Nach der Kondensationsreaktion wird die N-aralkylierte Aminofluoranverbindung auf übliche Weise isoliert, beispielsweise indem man das Lösungsmittel durch Wasserdampfdestillation entfernt, danach die N-aralkylierte Aminofluoranverbin- dung abfiltriert und trocknet. Nötigenfalls kann die aralkylierte Aminofluoranverbindung durch Umkristallisieren z. B. in Isopropanol gereinigt werden.
In den nachfolgenden Beispielen bedeuten Prozente Gewichtsprozente.
Beispiel 1
12 g 2-Amino-3-methyl-6-diethylaminofluoran (gemäss DE AS-2 001864, Beispiel 6) werden in 28 g Toluol mit 8,7 g Natriumcarbonat verrührt und auf 100 "C erwärmt Hierauf lässt man eine Lösung von 23,6 g p-Toluolsulfonsäurebenzylester (hergestellt gemäss Monatshefte-Chemie 82, S. 452-459, 1951) in Toluol bei 10105 "C im Verlaufe von 2 h zutropfen. Man rührt noch8 h bei 100 nach, trägt auf 100 gWasseraus, worauf die Toluolphase abgetrennt und 3 mal mit 50 g Wasser gewaschen wird. Nach Trocknung über Natriumsulfat und Filtrierung wird das Toluol unter reduziertem Druck abdestilliert.
Das Rohproduktwird in wenig Isopropylalkohol aufgenommen und als kristallines Produkt bei 20 "C abfiltriert. Man erhält nach Trocknung 12 g einer Fluoranverbindung der Formel

Auf säuremodifiziertem Kieselgel ergibt diese Verbindung sofort eine rote Farbe.
Beispiel 2
12 g 2-Amino-3-methyl-6-diethylaminofluoran werden in 60 mlToluol mit 12,4 g Kaliumcarbonat auf 100 "C erwärmt.
Hierauf werden 39,3 g p-Toluolsulfonsäurebenzylester, gelöst ion 51 gToluol, innerhalb von 1 h bei 100-105 "Czudosiert. Eine dem Reaktionsgemisch entnommene Analysenprobe zeigt im Liquid Chromatogramm (LC) bereits 68,8% Kondensationsprodukt der Formel (11). Die Reaktion wird während 1 h bei Rückfluss gehalten. Die jetzt gezogene Analysenprobe ergibt 99,2% Reaktionsprodukt.
Das Reaktionsproduktwird vom anorganischen Salz klarfiltriert, worauf das Filtrat eingeengt und mit 75 ml Isopropanol verrührt wird. Das auskristallisierte Kondensationsprodukt wird abfiltriert und wiegt nach Trocknung 10,5 g. Im Filtrat sind gemäss LC-Analyse weitere 2,1 g der Dibenzylaminofluoranverbindung gemäss der Formel (11), jedoch keine entsprechende Monobenzylaminofluoranverbindung.
Vergleichsbeispiel 12 g 2-Amino-3-methyl-6-dimethylaminofluoran werden in 60 ml Toluol mit 12,4 g Kaliumcarbonat auf 100 "C erwärmt.
Hierauf wird eine Lösung von 19 g Benzylchlorid in 51 g Toluol innerhalb von einer Stunde bei 100-105 OC zudosiert. Eine dem Reaktionsgemisch entnommene Analysenprobe zeigt im LC noch kein Kondensationsprodukt der Formel (11). Die Reaktion wird während 1 hbei Rückiluss gehalten. Eine erneut gezogene Analysenprobe enthält immer noch keine Dibenzylaminofluoranverbindung der Formel (11), sondern lediglich 3,2% der entsprechenden N-monobenzylierten Aminofluoranverbindung.
Das Reaktionsgemisch wird vom anorganischen Salz klarfiltriert, worauf das Filtrat eingeengt und mit 75 ml Isopropanol verrührt wird. Auf diese Weise werden durch Abfiltrierung des auskristallisierten Produktes 8,3 g des als Ausgangsstoff eingesetzten 2-Amino-3-methyl-6-diethylaminofluoran zurückgewonnen.
Durch Aufkonzentrieren des Filtrates schreitet die Kondensation leicht voran. Die LC-Analyse ergibt 1,7 g Monobenzylamino- und 1,1 g Dibenzylaminofluoranverbindung.
DESCRIPTION
The present invention relates to a process for the preparation of aminofluorane compounds which have at least one aralkylated amino group.
The aralkyl group is typically obtained by reacting the amino compound with an aralkyl halide, such as. B. benzyl chloride or benzyl bromide, the reaction being carried out both in polar media and in non-polar solvents in the presence of alkali compounds.
However, these methods have the disadvantage that, in the case of polycyclic amino compounds, the end product is in some cases obtained in insufficient yield.
The object of the present invention is to develop an improved process which promises an increase in yield and at the same time results in products of high purity suitable for practical use without complicated cleaning steps.
The present invention therefore relates to a process for the preparation of N-aralkylated aminofluorane compounds, which is characterized in that a fluoran compound of the formula

in which ring A is unsubstituted or substituted by halogen, nitro, amino, mono-lower alkylamino or di-lower alkylamino, R 1, R 2 and R 3, independently of one another, are each hydrogen, halogen, lower alkyl or lower alkoxy,
X1, X2, Y1 and2, independently of one another, are each hydrogen, unsubstituted or substituted by halogen, hydroxy, cyano or lower alkoxy alkyl having a maximum of 12 carbon atoms, cycloalkyl, tetrahydrofurfuryl, aryl or acyl or the substituent pairs (Xl and X2) and (Y1 and Y2 ), independently of each other,
each together with the common nitrogen atom represent a five- or six-membered, preferably saturated, heterocyclic radical, with at least one of —NX, X2 or —NYlY2 or in ring A having a primary or secondary amino group, reacted with an arylsulfonic acid aralkyl ester.
Halogen means for example fluorine, bromine, iodine or preferably chlorine.
Lower alkyl and lower alkoxy in the definition of the radicals of the fluoranes are generally groups or group components which have 1 to 5, in particular 1 to 3, carbon atoms, such as. B. methyl, ethyl, n-propyl, isopropyl, n-butyl, sec-butyl, tert-butyl or amyl, or methoxy, ethoxy, isopropoxy, tert-butoxy or tert-amyloxy.
Acyl is especially formyl, lower alkylcarbonyl, such as. B.
Acetyl or propionyl or benzoyl. Other acyl radicals can be lower alkylsulfonyl, such as. B. methylsulfonyl or ethylsulfonyl and phenylsulfonyl.
Cycloalkyl means e.g. B. cyclopentyl and especially cyclohexyl.
If the substituent pairs (X1 and X2) and (Yl and Y2) together with the common nitrogen atom represent a heterocyclic radical, this is, for example, pyrrolidino, piperidino, pipecolino, morpholino, thiomorpholino or piperazino, such as, for. B. Methylpiperazino. Preferred saturated heterocyclic radicals for -NXlX2 and -NY1Y2 are pyrrolidino, piperidino or morpholino.
Ring A is preferably not further substituted. If it has substituents, it is primarily substituted by halogen or di-lower alkylamino.
Particularly suitable among the starting materials are the fluoran compounds of the formula given, in which either the group -NXlX2 or the group -NY1Y2 represents a primary amino group (-NH2).
Particularly good results in the process according to the invention are obtained if a fluoran compound of the formula given is used, in which -NYlY2 -NH2 and R2 signify hydrogen or especially lower alkyl or especially methyl.
Arylsulfonic acid aralkyl esters are those in which both the aryl radical and the aralkyl radical are unsubstituted or ring-substituted. Preferred substituents for this are halogens, nitro, trifluoromethyl, lower alkyl, especially methyl or lower alkoxy such as methoxy.
Particularly suitable among these sulfonic acid esters are benzenesulfonates, p-toluenesulfonates, p-bromobenzenesulfonates or p nitrobenzenesulfonates.
Examples of suitable sulfonic acid esters for introducing the aralkyl group are: p-toluenesulfonic acid benzyl ester,
Phenyl benzenesulfonic acid, phenyl p-toluenesulfonate, phenyl p-toluenesulfonate, a-methylbenzyl p-toluenesulfonate,
Phenolsopropyl benzenesulfonic acid, phenopropyl p-toluenesulfonate, 4-chlorobenzyl p-toluenesulfonate, 4-methylbenzyl p-toluenesulfonate, 2,5-dimethyl-p-toluenesulfonate, p-toluenesulfon-2, 5-dichlorobenzyl ester, p-nitrobenzene, p-nitrobenzene Benzyl bromobenzenesulfonic acid, 2-p-toluenesulfonic acid, 4-dimethyl-benzyl ester, 4-trifluoromethylbenzyl p-toluenesulfonic acid, 4-methoxybenzyl p-toluenesulfonic acid.
The preferred aryl sulfonic acid is benzyl p-toluenesulfonate.
The aminofluorane compound and the sulfonic acid ester are generally used in approximately stoichiometric amounts, one or two aralkyl groups being introduced per amino group. An excess of e.g. B. 10-15% of one of the components is also possible.
The reaction is expediently carried out at elevated temperature, preferably at 80 to 120 ° C. and in the presence of an acid-binding agent.
Suitable acid-binding agents are e.g. B. alkali metal hydroxides, alkaline earth metal hydroxides, alkali metal bicarbonates, alkali metal carbonates or tertiary nitrogen bases, such as. B. pyridine, N-methylpiperidine or trialkylamines or mixtures of these compounds. Sodium carbonate is preferred.
The reaction preferably takes place in a non-polar, especially aromatic, solvent. As benzene, toluene, xylene, chlorobenzenes such as dichlorobenzene or trichlorobenzene, or nitrobenzene instead. The preferred solvent is toluene.
After the condensation reaction, the N-aralkylated aminofluorane compound is isolated in a conventional manner, for example by removing the solvent by steam distillation, then filtering off the N-aralkylated aminofluorane compound and drying it. If necessary, the aralkylated aminofluorane compound can be recrystallized, e.g. B. be cleaned in isopropanol.
In the following examples, percentages mean percentages by weight.
example 1
12 g of 2-amino-3-methyl-6-diethylaminofluoran (according to DE AS-2 001864, Example 6) are stirred in 28 g of toluene with 8.7 g of sodium carbonate and heated to 100 ° C. Then a solution of 23. 6 g of benzyl p-toluenesulfonate (prepared according to the monthly magazine Chemie 82, pp. 452-459, 1951) in toluene at 10105 ° C. in the course of 2 h. The mixture is stirred for a further 8 hours at 100, poured onto 100 g of water, whereupon the toluene phase is separated off and washed 3 times with 50 g of water. After drying over sodium sulfate and filtration, the toluene is distilled off under reduced pressure.
The crude product is taken up in a little isopropyl alcohol and filtered off as a crystalline product at 20 ° C. After drying, 12 g of a fluoran compound of the formula are obtained

This compound immediately gives a red color on acid-modified silica gel.
Example 2
12 g of 2-amino-3-methyl-6-diethylaminofluoran are heated to 100 ° C. in 60 ml of toluene with 12.4 g of potassium carbonate.
39.3 g of benzyl p-toluenesulfonate, dissolved in 51 g of toluene, are then metered in at 100-105 "C. within 1 h. An analysis sample taken from the reaction mixture already shows 68.8% condensation product of the formula (11) in the liquid chromatogram (LC). The reaction is held at reflux for 1 h and the analytical sample now taken gives 99.2% of the reaction product.
The reaction product is filtered off from the inorganic salt, whereupon the filtrate is concentrated and stirred with 75 ml of isopropanol. The condensation product which has crystallized out is filtered off and weighs 10.5 g after drying. According to LC analysis, the filtrate contains a further 2.1 g of the dibenzylaminofluoran compound according to formula (11), but no corresponding monobenzylaminofluoran compound.
Comparative Example 12 g of 2-amino-3-methyl-6-dimethylaminofluoran are heated to 100 ° C. in 60 ml of toluene with 12.4 g of potassium carbonate.
A solution of 19 g of benzyl chloride in 51 g of toluene is then metered in at 100-105 ° C. within one hour. An analysis sample taken from the reaction mixture does not yet show any condensation product of the formula (11) in the LC. The reaction is held at reflux for 1 h. A freshly drawn analytical sample still contains no dibenzylaminofluoran compound of the formula (11), but only 3.2% of the corresponding N-monobenzylated aminofluoran compound.
The reaction mixture is filtered clear of the inorganic salt, whereupon the filtrate is concentrated and stirred with 75 ml of isopropanol. In this way, 8.3 g of the 2-amino-3-methyl-6-diethylaminofluorane used as the starting material are recovered by filtering off the crystallized product.
By concentrating the filtrate, the condensation progresses slightly. LC analysis shows 1.7 g of monobenzylamino and 1.1 g of dibenzylaminofluoran compound.
