Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von N-aryloxyalkylierten Aminofluoranverbindungen, welches dadurch gekennzeichnet ist, dass man eine 2-Aminofluoranverbindung der Formel

X1 und X2 unabhängig voneinander, je Wasserstoff, unsubstituiertes oder durch Halogen, Hydroxy, Hyano oder Niederalkoxy substituiertes Alkyl mit höchstens 12 Kohlenstoffatomen, Cycloalkyl, Tetrahydrofurfuryl, Aryl oder Acyl oder (X1 und X2) zusammen mit dem gemeinsamen Stickstoffatom einen fünf- oder sechsgliedrigen, vorzugsweise gesättigten, heterocyclischen Rest bedeuten, mit einem Arylsulfonsäureester der Formel

worin Z einen Arylrest und
Y einen Aryloxyalkylrest bedeuten, umsetzt.
Halogen bedeutet beispielsweise Fluor, Brom, Jod oder vorzugsweise Chlor.
Niederalkyl und Niederalkoxy stellen bei der Definition der Reste der Fluorane in der Regel solche Gruppen oder Gruppenbestandteile dar, die 1 bis 5, insbesondere 1 bis 3 Kohlenstoffatome aufweisen, wie z.B. Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, sek.Butyl, tert.Butyl oder Amyl, bzw. Methoxy, Ethoxy, Isopropoxy, tert.Butoxy oder tert. Amyloxy.
"Alcyl" ist besonders Formyl, Niederalkylcarbonyl, wie z.B. Acetyl oder Propionyl oder Benzoyl. Weitere Acylreste können Niederalkylsulfonyl, wie z.B. Methylsulfonyl oder Ethylsulfonyl sowie Phenylsulfonyl sein. Cycloalkyl bedeutet z.B. Cyclopentyl und vor allem Cyclohexyl. Aryl ist besonders Phenyl oder Tolyl.
Wenn (X1 und X2) zusammen mit dem gemeinsamen Stickstoffatom einen heterocyclischen Rest darstellen, so ist dieser beispielsweise Pyrrolidino, Piperidino, Pipecolino, Morpholino, Thiomorpholino oder Piperazino, wie z.B. Methylpiperazino. Bevorzugte gesättigte heterocyclische Reste für -NX1X2 sind Pyrrolidino, Piperidino oder Morpholino.
Der Ring A ist vorzugsweise nicht weiter substituiert. Falls er Substituenten aufweist, ist er in erster Linie durch Halogen oder Diniederalkylamino substituiert.
Besonders gute Ergebnisse im erfindungsgemässen Verfahren werden erhalten, wenn man eine Fluoranverbindung der angegebenen Formel verwendet, in der R2 Wasserstoff oder vor allem Niederalkyl oder insbesondere Methyl bedeutet.
Als Arylsulfonsäureester der Formel (2) kommen solche in Frage, bei denen sowohl der Arylrest Z als auch der Aryloxyalkylrest Y unsubstituiert oder ringsubstituiert sind. Bevorzugte Substituenten sind hierzu Halogene, Nitro, Trifluormethyl, Niederalkyl, besonders Methyl oder Niederalkoxy wie Methoxy. Besonders geeignet sind p-Toluolsulfonsäure-2-phenoxyethylester
und Benzolsulfonsäure-2-phenoxyethylester.
p-Toluolsulfonsäure-2-phenoxyethylester ist bevorzugt.
Die Aminofluoranverbindung und der Sulfonsäureester werden im allgemeinen in etwa stöchiometrischen Mengen eingesetzt, wobei pro Aminogruppe eine oder zwei Aryloxyalkylgruppen eingeführt werden. Ein Überschuss von z.B. 10-15 % an einer der Komponenten ist jedoch ebenfalls möglich.
Die Umsetzung erfolgt zweckmässigerweise bei erhöhter Temperatur, vorzugsweise bei 80 bis 120 DEG C und in Anwesenheit eines säurebindenden Mittels.
Geeignete säurebindende Mittel sind z.B. Alkalimetallhydroxide, Erdalkalimetallhydroxide, Alkalimetallhydrogencarbonate, Alkalimetallcarbonate oder tertiäre Stickstoffbasen, wie z.B. Pyridin, N-Methylpiperidin oder Trialkylamine oder auch Mischungen dieser Verbindungen. Bevorzugt ist Natriumcarbonat.
Die Reaktion findet vorzugsweise in einem nicht polaren, vor allem aromatischen Lösungsmittel, wie z.B. Benzol, Toluol, Xylol, Chlorbenzole wie Dichlorbenzol oder Trichlorbenzol, oder Nitrobenzol statt. Bevorzugtes Lösungsmittel ist Toluol oder Xylol.
Nach der Kondensationsreaktion wird die N-aryloxyalkylierte Aminofluoranverbindung auf übliche Weise isoliert, beispielsweise indem man das Lösungsmittel durch Wasserdampfdestillation entfernt, danach die N-aryloxyalkylierte Aminofluoranverbindung abfiltriert und trocknet. Nötigenfalls kann die aryloxyalkylierte Aminofluoranverbindung durch Umkristallisieren z.B. in Isopropanol gereinigt werden.
In den nachfolgenden Beispielen bedeuten Prozente Gewichtsprozente.
Beispiel 1:
10 g 2-Amino-3-methyl-6-diethylaminofluoran (gemäss DE-AS 2 001 864, Beispiel 6) werden in 40 ml Xylol (Isomerengemisch) mit 10,4 g Kaliumcarbonat und 21,9 g 2-Phenoxyethyl-p-toluolsulfonat (hergestellt gemäss J. Org. Chem. 9 (1944), 235, 238-240) während 11 Stunden bei 137-140 DEG C unter Rückfluss gerührt. Dann versetzt man mit weiteren 50 ml Xylol und extrahiert mehrmals mit je 100 ml Wasser bei 50 DEG C. Alsdann wird die Xylolphase aufkonzentriert bis das Kondensationsprodukt auskristallisiert. Das Produkt wird abfiltriert, mit Isopropanol gewaschen und getrocknet. Man erhält 3,1 g der Verbindung der Formel

in Form von farblosen Kristallen. Schmelzpunkt 189-191 DEG C. Auf säuremodifiziertem Kieselgel erzeugt eine Toluollösung dieses Produktes eine instante schwarze Farbe.

Das gemäss Beispiel 1 erhaltene Filtrat ergibt nach Säulenchromatographie (Kieselgel, Eluiermittel: Toluol/Essigsäureethylester (20:1)) 2,8 g der Fluoranverbindung der Formel

welche, aus Isopropanol/Toluol (10:1) umkristallisiert, einen Schmelzpunkt von 117-119 DEG C aufweist.
Auf säuremodifiziertem Kieselgel erzeugt diese Verbindung sofort eine rote Farbe.
Beispiel 3:
9,6 g 2-Amino-6-diethylaminofluoran werden in 50 ml Xylol mit 6,9 g Kaliumcarbonat und 14,6 g 2-Phenoxyethyl-p-toluolsulfonat unter gutem Rühren erwärmt und während 22 Stunden bei 133-137 DEG C unter Rückfluss gehalten. Das Reaktionsgemisch wird mit 50 ml Xylol und bei 80 DEG C mit 100 ml Wasser versetzt. Hierauf wird die Xylolphase abgetrennt, mehrmals mit Wasser gewaschen und eingeengt. Man erhält 20 g des rohen Reaktionsgemisches. Mittels Säulenchromatographie (Kieselgel, Eluiermittel: Toluol/Essigsäureethylester (10:1)) werden die Fluoranverbindungen der Formeln (5) und (6) aufgetrennt und jeweils aus Isopropanol umkristallisiert.
