PATENTANSPRÜCH E 1. Verfahren zur Herstellung eines ss-Ketols der Formel (I):

gekennzeichnet durch die Umsetzung von 6-Methoxy-2-naphthaldehyd mit Aceton in Gegenwart einer Base bei herabgesetzten Temperaturen.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die Umsetzung bei Temperaturen unter 5 C ausgeführt wird.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass die Umsetzung in wässerigem Aceton ausgeführt wird.
4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass die Umsetzung in Gegenwart von Natriumhydroxid ausgeführt wird.
5. Verwendung des nach dem Verfahren gemäss Anspruch 1 erhaltenen ss-Ketols der Formel (I) zur Herstellung von 4-(6-Methoxy2-naphthyl)-butan-2-on, dadurch gekennzeichnet, dass besagtes p- Ketol durch katalytische Hydrierung reduziert wird.
6. Verwendung nach Anspruch 5, dadurch gekennzeichnet, dass der Hydrierungskatalysator Palladium aufweist.
7. Verwendung nach Anspruch 5 oder 6, dadurch gekennzeichnet, dass die Hydrierung unter Anwendung eines Wasserstoffdruckes im Bereich von 0,5 bis 4 bar ausgeführt wird.
8. Verwendung nach einem der Ansprüche 5 bis 7, dadurch gekennzeichnet, dass die Hydrierung bei einer Temperatur von 20 bis 700 C ausgeführt wird.
9. Verwendung nach einem der Ansprüche 5 bis 8, dadurch gekennzeichnet, dass die Hydrierung in einem inerten organischen Lösungsmittel, inerten organischen Lösungsmittelgemisch oder wässerigen Medium ausgeführt wird.
10. Verwendung nach einem der Ansprüche 5 bis 9, dadurch gekennzeichnet, dass die katalytische Hydrierung des ss-Ketols der Formel (I) in situ, ohne Isolierung erfolgt.
BESCHREIBUNG
Diese Erfindung bezieht sich auf ein neues Verfahren zur Herstellung einer Zwischenproduktverbindung, die für die Herstellung von 4-(6-Methoxy-2-naphthyl)-butan-2-on brauchbar ist.
Die US-PS 4061 779 beschreibt 4-(6-Methoxy-2-naphthyl)-butan-2-on (Nabumetone) und seine Verwendung bei der Behandlung von rheumatischen und arthritischen Affektionen. Eine Anzahl von Verfahren zur Herstellung der Verbindung werden ebenfalls beschrieben. Die US-PS 4 741 und 4247709 beschreiben weitere Verfahren zur Herstellung von Nabumetone.
Ein Verfahren zur Herstellung eines ss-Ketol-Zwischenproduktes, das zur Herstellung von Nabumetone brauchbar ist, wurde nun gefunden; dieses Verfahren ist durch Kondensation von 6-Methoxy-2naphthaldehyd mit Aceton gekennzeichnet.
Demgemäss bezieht sich die vorliegende Erfindung auf ein Verfahren zur Herstellung eines ss-Ketols der Formel (I):

das durch die Umsetzung von 6-Methoxy-2-naphthaldehyd mit Aceton in Gegenwart einer Base bei herabgesetzten Temperaturen gekennzeichnet ist.
Das ss-Ketol der Formel (I) kann dann durch katalytische Hydrierung zu Nabumetone reduziert werden.
Vorzugsweise wird die Kondensation von 6-Methoxy-2-naphthaldehyd mit Aceton bei einer Temperatur unter 5 C ausgeführt.
Bei höheren Temperaturen, z. B. bei Umgebungstemperatur, besteht die Neigung, dass Wasserabspaltung eintritt, wodurch das entsprechende a,ss-ungesättigte Keton erzeugt wird, wie in Beispiel 4 der GB-PS Nr. 1 474377 beschrieben.
Die Umsetzung wird zweckmässig in wässerigem Aceton unter Verwendung von Natriumhydroxid als Base ausgeführt und das Produkt mit Ether extrahiert.
In einer bevorzugten Ausführungsform können die Kondensation von 6-Methoxy-2-naphthaldehyd mit Aceton und die anschliessende Reduktion in einer in situ-Herstellung kombiniert werden. Die basische Lösung der Verbindung der Formel (I) wird mit Säure neutralisiert und die anschliessende Hydrierung und die Isolierung des p-Ketols der Formel (I) ausgeführt.
Die Hydrierung der Verbindung der Formel (I) kann unter Anwendung eines niedrigen, mittleren oder hohen Wasserstoffdrucks, z. B. von ca. 0,5 bis 4 bar Wasserstoff, ausgeführt werden, aber im allgemeinen wird es bevorzugt, einen Druck von ca. 0,9 bis 1,5 bar Wasserstoff, z. B. einen Druck von 1 bar, anzuwenden.
Die Hydrierung wird in Gegenwart eines Katalysators, z. B.
eines Edelmetallkatalysators, wie Palladium, das bevorzugt wird, ausgeführt. Geeignete Formen von Pailadiumkatalysatoren umfassen Palladium auf Kohle, Palladium auf Bariumsulfat, Palladium auf Calciumcarbonat. Palladium auf Kohle wird bevorzugt
Geeignete inerte Lösungsmittel für die Umsetzung umfassen niedere Alkohole, schwache Säuren (wie Essigsäure), Ester (wie Ethylacetat), Halogenkohlenwasserstoffe und ketonische Lösungsmittel, wie Methylisobutylketon oder Aceton, oder ein geeignetes Gemisch von zwei oder mehreren dieser Lösungsmittel. Ein Gemisch von Ethanol/Ethylacetat oder Essigsäure/Aceton ist besonders geeignet. Die Umsetzung kann auch in einem wässerigen Medium stattfinden.
Die Umsetzung kann bei Umgebungstemperatur oder einer erhöhten Temperatur, z. B. 20 bis 700 C und zweckmässiger 40 bis 60 C, vorzugsweise ca. 55" C, ausgeführt werden.
Nachdem die Reaktion beendet ist (z. B. gemäss Beurteilung durch HPLC, TLC oder Aufhören der Wasserstoffaufnahme), kann die gewünschte Verbindung durch Abfiltrieren des Katalysators und Entfernung des Lösungsmittels, z. B. durch Eindampfen unter vermindertem Druck, erhalten werden.
Gewünschtenfalls kann die so erhaltene Verbindung durch Umkristallisation, z. B. aus wässerigem Ethanol, weiter gereinigt werden.
Die folgenden Beispiele erläutern die Erfindung.
Beispiel 1 4-Hydroxy-4- (6-methoxy-2-naphtltyl)-butan-2-on
6-Methoxy-2-naphthaldehyd (15,0 g) in Aceton (150 ml) wurde in einem Eis/Wasser-Bad gekühlt, und 2-normales Natriumhydroxid in Wasser (6,4 ml) wurde zugesetzt. Nach 2,5stündigem Rühren in der Kälte wurde der pH-Wert mit verdünnter Säure auf 8 herabgesetzt. Die Lösung wurde mit Wasser (250 ml) und Ether (500 ml) geschüttelt. Die etherische Schicht wurde mit Wasser gewaschen, getrocknet und unter vermindertem Druck destilliert, bis sich ein kristalliner Niederschlag von 4-Hydroxy-4-(6-methoxy-2-naphthyl)butan-2-on (16,1 g, Schmelzpunkt 106 bis 107 C) bildete. Die Umkristallisation aus Ethylacetat ergab farblose nadelförmige Kristalle (Schmelzpunkt 107,5 bis 108 C).
IR-Spektrum (Suspension): 1605, 1795 und 3450 cm-'.
90-MHz-NMR (CDCl3): 2,15 (s, 3H), 2,9 (m, 2H), 3,4 (d, H), 3,85 (s, 3H), 5,25 (m, H), 7,05-7,75 (m, 6H) ppm.

Erfordert: C 73,8 H 6,6% Beispiel 2 4- (6-Metlzaxy-2-naphthyl) -butan-2-on
4-Hydroxy-4-(6-methoxy-2-naphthyl)-butan-2-on (3,66 g) in Ethanol (60 ml) und Ethylacetat (40 ml) mit einem Katalysator von lO%igem Palladium auf Kohle (0,5 g) wurden in einer Wasserstoffatmosphäre eine Stunde lang kräftig gerührt, während man gerade bis zur Rückflusstemperatur erhitzte. Der Katalysator wurde entfernt und die Lösung bei vermindertem Druck eingedampft, wobei eine Reihe von Ausbeuten eines kristallinen Produktes vom Gesamtgewicht 2,80 g erhalten wurde. Durch IR- und NMR-Spektroskopie wurde gezeigt, dass dieses Produkt 4-(6-Methoxy-2-naphthyl)-butan-2-on war, das etwas unveränderte Hydroxyverbindung enthielt.
Beispiel 3 4Hydroxy-4-(6-methoxy-2-naphthyl)-butan-2-on (156 mg) in Aceton (2,0 ml), Wasser (0,05 ml) und Essigsäure (1,0 ml) mit einem Katalysator von l0%igem Palladium auf Kohle (20 mg) wurden in einer Wasserstoffatmosphäre unter Rühren auf 55" C erhitzt. In Abständen wurden Proben entnommen und durch HPLC gegen ein Standardgemisch von Ausgangsmaterial (A), 2-Hydroxy-4-(6methoxy-2-naphthyl)-butan (B) und 4-(6-Methoxy-2-naphthyl)butan-2-on (C) untersucht. Die Menge an (C) in der Reaktion stieg nach 6stündiger Hydrierung bei 55 C auf ein Maximum (94%). In diesem Zeitpunkt betrugen die Mengen an (A) und (B) 3% bzw. 4%.
Beispiel 4 4(6-Methoxy-2-napAthyl) -butan-2-on
6-Methoxy-2-naphthaldehyd (6,0 g) in Aceton (100 ml) wurde in einem Eis/Wasser-Bad gekühlt, und eine 2-normale Lösung von Natriumhydroxid in Wasser (2,5 ml) wurde zugesetzt. Nach 2,5stündigem Rühren in der Kälte wurden Essigsäure (50 ml) und ein 10%der Palladium-auf-Kohle-Katalysator (1,0 g) zugesetzt. Die Suspension wurde in einer Wasserstoffatmosphäre ca. 6 Stunden lang kräftig bei 55" C gerührt.
Als die Reduktion beendet war, wurde der Katalysator entfernt und die Lösung mit Wasser (200 ml) und Ether (150 ml) geschüttelt.
Die wässerige Schicht wurde abgetrennt und erneut extrahiert (50 ml Ether). Die etherischen Extrakte wurden vereinigt, mit einem Überschuss an Natriumbicarbonatlösung in Wasser gewaschen und dann über Natriumsulfat getrocknet. Das Lösungsmittel wurde entfernt, wobei das rohe, im Titel genannte Produkt in einer Reihe von Ausbeuten (Gesamtgewicht 6,2 g) erhalten wurde.
Nach Umkristallisation aus heissem Ethanol wurde 4-(6-Methoxy-2-naphthyl)-butan-2-on (5,0 g) vom Schmelzpunkt 81 bis 83,5 C erhalten.

DESCRIPTION
This invention relates to a novel process for the preparation of an intermediate compound useful for the preparation of 4- (6-methoxy-2-naphthyl) butan-2-one.
US-PS 4061 779 describes 4- (6-methoxy-2-naphthyl) -butan-2-one (nabumetone) and its use in the treatment of rheumatic and arthritic affections. A number of methods of making the compound are also described. U.S. Patent Nos. 4,741 and 4,247,709 describe other processes for making nabumetones.
A process for the manufacture of an ss-ketol intermediate useful for the manufacture of nabumetone has now been found; this process is characterized by the condensation of 6-methoxy-2naphthaldehyde with acetone.
Accordingly, the present invention relates to a process for the preparation of an ss-ketol of the formula (I):

which is characterized by the reaction of 6-methoxy-2-naphthaldehyde with acetone in the presence of a base at reduced temperatures.
The ss-ketol of formula (I) can then be reduced to nabumetones by catalytic hydrogenation.
The condensation of 6-methoxy-2-naphthaldehyde with acetone is preferably carried out at a temperature below 5 ° C.
At higher temperatures, e.g. B. at ambient temperature, there is a tendency that dehydration occurs, whereby the corresponding a, ss-unsaturated ketone is produced, as described in Example 4 of GB-PS No. 1 474377.
The reaction is conveniently carried out in aqueous acetone using sodium hydroxide as the base and the product extracted with ether.
In a preferred embodiment, the condensation of 6-methoxy-2-naphthaldehyde with acetone and the subsequent reduction can be combined in an in situ preparation. The basic solution of the compound of formula (I) is neutralized with acid and the subsequent hydrogenation and isolation of the p-ketol of formula (I) is carried out.

6-Methoxy-2-naphthaldehyde (15.0 g) in acetone (150 ml) was cooled in an ice / water bath and 2 normal sodium hydroxide in water (6.4 ml) was added. 4-Hydroxy-4- (6-methoxy-2-naphthyl) butan-2-one (3.66 g) in ethanol (60 ml) and ethyl acetate (40 ml) with a catalyst of 10% palladium on carbon ( 0.5 g) were stirred vigorously in a hydrogen atmosphere for one hour while heating to the reflux temperature.
