Verfahren zur Herstellung von a, fl-ungesuttigten Aldehyden
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von aX ungesättigten Aldehyden, das dadurch gekennzeichnet ist, dass man einen Ester von 1, 1-disubstituiertem Propin- (2)-ol- (l) in Gegenwart eines eine Verbindung eines Metalles der Nebenreihe lb des periodischen Systems enthaltenden Katalysators mit einem sauren Mittel behandelt und das Reaktionsprodukt hydrolysiert.
Die Herstellung der Ausgangsstoffe kann in an sich bekannter Weise durch Kondensation von Acetylen mit einem Keton und Veresterung des gebildeten, tertiären Acetylencarbinols erfolgen. Aus Diketoverbindungen werden auf analoge Weise die entsprechenden zwei veresterte tertiäre Acetylencarbinol- gruppierungen aufweisenden Stoffe gebildet.
Für das erfindungsgemäl3e Verfahren besonders geeignete Ausgangsstoffe sind Verbindungen der Formel

worin R1 und R2 gesättigte oder ungesättigte aliphatische, alicyclische oder aromatische Reste oder zusammen eine Polymethylenbrücke darstellen (diese Reste können auch Sauerstoffunktionen, wie z. B.
Hydroxyl-, Ester-, Carboxyl-, Ather-, Carbonyloder Enolradikale tragen) und R3 eine veresterte Hydroxylgruppe bedeutet.

Zweckmässig werden z. B. die niederen Carbonsäureester der oben genannten Verbindungen als Ausgangsstoffe eingesetzt. Vorteilhafte Ester stellen z. B. die Benzoate, Formiate, Thioacetate sowie insbesondere die Acetate, dar. Wenn erwünscht, k¯nnen die Acetylencarbinole im gleichen Reaktions gefäss hergestellt werden, in welchem anschliessend die erfindungsgemässe Reaktion vorgenommen, wird.
In der ersten Stufe des erfindungsgemässen Verfahrens werden als Katalysatoren Kupfer-, Silberoder Goldverbindungen verwendet. Soll die Umsetzung unter Verwendung von Kupfersalz-Katalysatoren durchgeführt werden, kann dem sauren Medium auch das Metall als solches, z. B. in fein verteilter Form, gegebenenfalls auch auf einem Träger, zugesetzt werden, wobei die gewünschten Kupfersalze sich im Reaktionsmedium selbst bilden. Man kann als Katalysatoren Oxyde, wie z. B. Kupferoxyd, gegebenenfalls in Kombination mit einem andern Metall, z. B. Kupferchromoxyd, verwenden. Bevorzugt werden als Katalysatoren Salze eingesetzt, beispielsweise Kupfer-oder Silber-acetate,-carbonate, -oxalate,-nitrate oder-sulfate sowie Gemische dieser Salze.
Als saure Mittel können sowohl anorganische wie organische Säuren verwendet werden. Bevorzugt ist es allerdings, die Reaktion in Gegenwart einer niederen Carbonsäure, z. B. Propionsäure oder Buttersaure, insbesondere von Essigsäure, vorzunehmen, wobei das saure Mittel gleichzeitig als Lösungsmittel dienen kann. Wird die Umsetzung in Gegenwart von starken Säuren, z. B. Mineralsäuren, wie Schwefelsäure oder Phosphorsäure, vorgenommen, so werden diese Säuren vorteilhaft mit einem inerten Lösungsmittel verdünnt. Als Lösungsmittel können z. B.
Benzol, Toluol, Tetrachlorkohlenstoff, Essigester oder Aceton verwendet werden.
Die zur Reaktion gelangenden Propinylester kön- nen in gelöster Form, als Emulsion oder als Suspension, angewendet werden. Die Behandlung mit dem sauren Mittel kann in flüssiger Phase oder in der Dampfphase erfolgen.
Durch die erfindungsgemässe Behandlung unterliegen die Propinylester einer Umlagerung, welche, wie in einzelnen Fällen nachgewiesen werden konnte, anscheinend zunächst zur Bildung von Allenverbindungen der nachstehenden Formel führt :

Wird die erste Reaktionsstufe in Gegenwart einer Carbonsäure vorgenommen, kann daneben auch durch Anlagerung eines Mols dieser Säure eine a, fi- ungesättigte Diacylverbindung der Formel III entstehen :

Je nach den herrschenden Reaktionsbedingun- gen und des als Ausgangsmaterial verwendeten ter tiären Acetylencarbinolesters kann im erhaltenen Reaktionsgemisch das Mengenverhältnis der Reaktionsprodukte der Formeln II und III variieren.
Erfolgt die Behandlung unter besonders energi- schen Bedingungen, z. B. bei höheren Temperaturen oder während einer längeren Zeitdauer, kann unter Umlagerung der Doppelbindung als Nebenprodukt ein Enolester, z. B. der Formel IV, entstehen :

worin R1'den nach Abspaltung eines Wasserstoffatomes verbleibenden Rest R1 bedeutet.
Zweckmässig wird die Reaktion durch analytische Kontrolle so geleitet, dass vorwiegend ein Gemisch der beiden Komponenten II und III gebildet wird.
Beispielsweise kann durch Vornahme der Acetylenprobe mit Silbernitrat das Verschwinden des Aus gangsmaterials festgestellt werden. Der Umsetzungsgrad kann auch auf Grund der I. R.-Spektren verfolgt werden. Die Umsetzung wird mit Vorteil beendet, sobald praktisch kein Ausgangsmaterial mehr vorhanden ist.
In einer zweiten Reaktionsstufe wird das erhaltene Gemisch hydrolysiert. Die Hydrolyse kann sowohl alkalisch als auch sauer durchgeführt werden, wobei sich die bevorzugte Methode nach der Be ständigkeit des zu bildenden Aldehyds gegenüber dem die Hydrolyse bewirkenden Mittel richtet.
Zweckmässig wird unter milden Bedingungen hydrolysiert. Solche milde Hydrolysebedingungen stellen z. B. Erwärmen mit Soda in wässerigem Alkohol, z. B. Methanol, Behandlung mit alkoholischer Kalilauge bei tiefer Temperatur, Erwärmen mit verdünnter Essigsäure oder Behandlung mit verdünnter Schwefelsäure in Aceton bei Raumtemperatur dar.
Enthält das in der ersten Reaktionsstufe vorhandene saure Reaktionsmedium Wasser, so erfolgt unmittelbar nach der Bildung der Umlagerungs-bzw.
Säureanlagerungsprodukte deren mehr oder weniger vollständige Hydrolyse. Es gelingt, auf diese Weise die a,, -ungesättigten Aldehyde in einem Arbeitsgang herzustellen.
So kann man z. B. ein entsprechendes Propinylacetat in Gegenwart von Silberacetat in wässeriger Essigsäure erwärmen, wobei durch Umlagerung und Hydrolyse in einem Arbeitsgang die Bildung des Aldehyds erfolgt.
Man kann auch ein entsprechendes Propinylacetat in wasserfreier Essigsäure in Gegenwart von Silbercarbonat erwärmen und das gebildete Reaktionsprodukt nach Isolierung in einer zweiten Stufe durch Erwärmen mit Soda in wässerigem Methanol zum freien Aldehyd hydrolysieren.
Die Isolierung der gebildeten ungesättigten Al- dehyde kann nach an sich bekannten Methoden erfolgen. Beispielsweise können diese Verbindungen über die entsprechenden Bisulfitadditionsverbindun- gen isoliert werden.
Die nach dem erfindungsgemässen Verfahren er hältlichen Verbindungen stellen wertvolle zum Teil wohlbekannte Aldehyde von mannigfacher Bedeutung dar. Beispielsweise besitzen sie Bedeutung als Riechstoffe und/oder als Zwischenprodukte für die Herstellung von Riechstoffen, Carotinoidverbindun- gen, fettlöslichen Vitaminen oder Steroidhormonen.
Beispiel 1
25, 2 g 3-Methyl-3-acetoxy-butin- (1) [hergestellt aus Methylbutinol durch Umsetzung mit Essigsäure- anhydrid und Phosphorsäure ; Siedepunkt 78-79 ;
100 mm nid = 1, 4178] werden in 50 cm3 Eisessig gelöst und nach Zugabe von 0, 2 g Silbernitrat unter Stickstoff während 30 Minuten auf 100 erwärmt.
Man giesst das Reaktionsgemisch auf Eis, extrahiert mit Petroläther und wäscht 5mal mit Wasser. Nach dem Trocknen mit Natriumsulfat und Abdampfen des Lösungsmittels erhält man 25, 6 g eines Reaktionsproduktes mit folgenden Eigenschaften : n 22/D =
1, 4420 ; U. V.-Absorptionsmaximum bei 234 m, u, E = 170 (in Athanol). Das Produkt enthält nach dem I. R.-Spektrum kein Ausgangsmaterial mehr und besteht vorwiegend aus 3-Methyl-1, l-diacetoxy- buten-(2) nebst wenig 3-Methyl-buten- (2)-al- (l).
Durch Hochvakuumdestillation erhält man 17, 4 g reines Diacetat vom Siedepunkt 57¯/0, 03 mm ; n2D2= 1, 4360 ; Schmelzpunkt 19-200. Diese Sub- stanz besitzt kein U. V.-Absorptionsmaximum.
10 g dieses Diacetates werden in 100 cm3 Aceton gelöst, 10 g 3n Schwefelsäure zugesetzt und während d 2 Stunden bei 20 stehengelassen. Durch Extraktion mit Äther, Waschen mit gesättigter Natriumchloridl¯sung, Trocknen mit Natriumsulfat, Filtrieren und Eindampfen gewinnt man 4-5 g rohes 3-Methyl buten'- (2)-al- (l), das durch Destillation gereinigt wird. Siedepunkt 130131 1730 mm ; nD = 1, 4521 ; U. V.-Absorptionsmaximum bei 235 m, u. (in Athanol).
Das Semicarbazon dieses Aldehydes schmilzt bei 210 und besitzt ein U. V.-Absorptionsmaximum bei 271 mM (in Äthanol).
Beispiel 2
50 g 1-Athinyl-1-acetoxy-cyclohexan in 500 cm3 wässriger Essigsäure werden nach Zugabe von 0, 5 g Silbernitrat während 30 Minuten unter Stickstoff und am Rückfluss gekocht. Die Reaktionsmischung wird auf Eis gegossen, mit Petroläther (Siedeintervall 40 bis 60 ) extrahiert, wiederholt mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum eingedampft. Das erhaltene Reaktionsprodukt besitzt ein U. V.-Absorptionsmaximum bei 243 m, u, El = 1540 (in Athanol). Der daraus über die kristalline Bisulfitadditionsverbindung gewonnene Cyclohexylidenacetaldehyd hat folgende Eigenschaften : Siedepunkt 85 bis 90¯/12 mm ; nD = 1, 4835 ; U. V.-Absorptionsmaximum bei 240 mu (in Athanol).
Das Semi carbazon dieser Verbindung schmilzt bei 198-200 und hat ein U. V.-Absorptionsmaximum bei 273 m, u (in Athanol).
Durch fraktionierte Destillation des mit Bisulfit nicht in Reaktion getretenen Anteils erhält man reines 2- [Cyclohexen- (l)-yl]-vinyl- (l)-acetat ; Siede punkt 60-62010, 04 mm ; nD = 1, 5102 ; U. V.-Ab- sorptionsmaximum bei 242 mu (in Athanol).
In analoger Weise erhält man ausgehend von 1-Athinyl-1-acetoxy-cyclopentan (nD = 1, 4559) ein
Reaktionsprodukt, welches ein U. V.-Absorptions- maximum bei 243 m, u, E 1 = 1125 (in Athanol) auf weist ; n 22/D = 1, 5015. Der daraus über die kristalline
Bisulfitadditionsverbindung gewonnene Cyclopentyl- idenacetaldehyd wird in üblicher Weise in das Semi carbazon übergeführt, welches einen Schmelzpunkt von 199-200 und'ein U. V.-Absorptionsmaximum bei 272, 5 m, u, Ei = 1647 (in Äthanol) aufweist.
Beispiel 3
90 g 3, 7-Dimethyl-3-acetoxy-octen- (6)-in- (l) [hergestellt aus 3, 7-Dimethyl-octen- (6)-in- (l)-ol- (3) durch Behandlung mit Essigsäureanhydrid und
Phosphorsäure] werden zusammen mit 0, 9 g Silber carbonat und 300 cm3 Eisessig während einer Stunde bei 90 ger hrt. Hierauf giesst man das Reaktions gemisch in einen Scheidetrichter zu 900 cm3 Wasser und extrahiert das abgeschiedene 01 mit Petroläther.
Die Petrolätherlösung wird mit gesättigter Bicarbonat lösung gewaschen, mit Natriumsulfat getrocknet und bei 40 im Vakuum eingedampft. Man erhält 102 g eines gelben Öls, das im U. V.-Spektrum nicht absorbiert und gemäss dem I. R.-Spektrum aus etwa 60 to
3, 7-Dimethyl-1-acetoxy-octatrien-(1, 2, 6), 35"/o 3, 7 Dimethyl-1, 1-diacetoxy-octadien- (2, 6) und 50/o un verändertem Ausgangsmaterial besteht. Dieses Ge misch weist bei 20 eine Brechung von etwa 1, 469 und eine Dichte von etwa 0, 967 auf.
Dieses Gemisch wird in 500 cm3 Methanol auf genommen, mit 60 g Soda und 100 cm3 Wasser ver setzt und während 30 Minuten am Rückfluss unter
Rühren gekocht. Hierauf giesst man das Gemisch in einen Scheidetrichter zu 1500 cm3 Wasser und extrahiert mit Petroläther. Die getrocknete Petrol ätherlösung wird abgedampft, wobei rohes Citral als braungelbes Öl in einer Ausbeute von etwa 70 g zu rückbleibt. U. V.-Absorptionsmaximum bei 237 bis 238 m, a, El= 800 (in Athanol). Durch Reinigung über die Bisulfitadditionsverbindung lassen sich daraus 59 g reines Citral isolieren : Ausbeute etwa 84 /o.
Beispiel 4
330 g 37-Dimethyl-3-acetoxy-octen-(6)-in-(1) werden mit 900 cm3 Eisessig und 3 g Silbercarbonat während 2 Stunden bei 80 gerührt. Das Reaktions produkt wird mit 3000 cm3 Eiswasser vermischt und mit Petroläther (Siedeintervall 40-60 ) ausgeschüt telt. Die Petrolätherlösung wäscht man mit gesättigter
Natriumbicarbonatlösung und trocknet mit Natrium sulfat. Nun wird auf 1000 cm3 eingeengt, auf-30 bis-50 gekühlt, worauf nach einigem Stehen ein Produkt auskristallisiert, das kalt genutscht und mit kaltem Petroläther gewaschen wird.
Durch Destillation im Hochvakuum und erneutes Umkristallisieren erhält man reines 3, 7-Dimethyl-1, 1-diacetoxy- octadien- (2, 6), das bei 4 schmilzt ; Siedepunkt 87¯/0,05 mm; n 20/D = 1, 4588 ; Dichte 0, 999.
Die Behandlung dieses Produktes mit Soda wie in Beispiel 3 ergibt ein Citral, welches ein U. V.-Absorptionsmaximum bei 237 m, *"E I = 850 aufweist.
Aus der nach Abtrennung des Diacetates verbleibenden Mutterlauge erhält man nach dem Abdampfen des Petroläthers ein 01, das bei der Destillation eine Fraktion bei 58-620/0, 05 mm ergibt.
Das so gereinigte 01 enthält pro Molekül eine Acetylgruppe und stellt gemϯ I. R.-Spektrum 3, 7-Dimethyl- 1-acetoxy-octatrien- (1, 2, 6) dar ; nD = 1, 4810 ; Dichte 0, 927. Es zeigt keine Absorption im U. V.-Spektrum.
Wird das so erhaltene Produkt wie in Beispiel 3 mit Soda behandelt, erhält man ein Citral mit einem U. V.-Absorptionsmaximum bei 237 m, u, El= 780 (in Äthanol).
Beispiel S
20 g 3, 7-Dimethyl-3-acetoxy-octen- (6)-in- (1) werden mit 100 cm3 950/piger Essigsäure und 0, 2 g Kupferpulver während einer Stunde am Rückfluss gekocht. Aus dem Reaktionsprodukt lassen sich in üblicher Weise 16 g eines Ols gewinnen ; n 20/D = 1, 4932 ; U. V.-Absorptionsmaximum bei 237 mu.
Durch Reinigung über die kristalline Bisulfitaddi- tionsverbindung erhält man aus diesem 01 8 g reines Citral : Ausbeute etwa 50%.
Beispiel 6
20 g 3, 7-Dimethyl-3-acetoxy-octen-(6)-in-(1) werden in 200 cm3 Eisessig und 5 cm3 Wasser gelöst und nach Zugabe von 0, 4 g Goldchlorür während 2 Stunden unter Stickstoff am Rückfluss gekocht. Aus dem Reaktionsprodukt lassen sich in üblicher Weise 18, 4 g rohes Citral gewinnen ; nid = 1, 4782 ; U. V. Absorptionsmaximum bei 235 m, u, E i = 270. Durch Reinigung über die Bisulfitadditionsverbindung kann daraus reines Citral erhalten werden.
In analoger Weise erhält man bei Verwendung von 0, 1 g Kupferoxyd anstelle von Goldchlorür
19, 1 g rohes Citral ; n 20/D = 1, 4981 ; U. V.-Absorp- tionsmaximum bei 238 mu El= 538 (in Athanol).
Es kann über die Bisulfitadditionsverbindung gereinigt werden.
Beispiel 7
10 g 3, 7-Dimethyl-3-acetoxy-octen-(6)-in-(1) werden in 100 cm3 Propionsäure und 5 cm3 Wasser gelöst und nach Zugabe von 0, 5 g Kupferchromoxyd während 20 Minuten am Rückfluss gekocht. Aus dem Reaktionsprodukt lassen sich 9, 8 g rohes-Citral gewinnen ; n2D2= 1, 4881 ; U. V.-Absorptionsmaximum bei 234 mcc, El = 393 (in Feinspritlösung). Es kann nach der Bisulfitmethode gereinigt werden.
Beispiel 8
50 g 3, 7-Dimethyl-3-acetoxy-octen- (6)-in- (1) werden mit einem Gemisch von 0, 5 g Kupfersulfat, 10 cm3 konzentrierter Schwefelsäure und 500 cm3 Benzol während 15 Minuten ger hrt. Die Temperatur des Gemisches steigt ohne äussere Erwärmung auf 65 . Nach der üblichen Aufarbeitung wird das isolierte dunkle Reaktionsprodukt durch halbstün- diges Kochen mit Soda in verdünnter methylalkoho- lischer Lösung zu rohem Citral hydrolysiert ; U. V. Absorptionsmaximum bei 236 m//. Es wird über die Bisulfitadditionsverbindung gereinigt ; n20= 1, 4855 ; U. V.-Absorptonsmaximum bei 237-238 mol (in Athanol).
Beispiel 9
20 g 3, 7-Dimethyl-3-acetoxy-octen- (6)-in- (l) werden mit 80 cm3 Essigester unter Zusatz von 1 g Kupferacetat und 10 cm3 konzentrierter Schwefelsäure unter Rühren auf 703 erwärmt. Bei dieser Temperatur tritt plötzlich eine Reaktion ein, so dass das Reaktionsgemisch zum Aufsieden kommt. Man rührt noch 15 Minuten ohne weitere Erwärmung und erhält nach der üblichen Aufarbeitung und der Hydrolyse wie in Beispiel 8 rohes Citral als dunkles 01 ; nD = 1, 4884. Reines Citral kann daraus in bekannter Weise über die Bisulfitadditionsverbindung isoliert werden.
Beispiel 10
25 g 3, 7-Dimethyl-3-benzoxy-octen- (6)-in- (l) [welches aus 3, 7-Dimethyl-3-hydroxy-octen-(6)-in-(1) durch Behandlung mit Benzoylchlorid in Pyridin gewonnen wird ; nD = 1, 5147] werden in 250 cm3 Eisessig gelöst und nach Zugabe von 12, 5 cm3 Wasser und 0, 25 g Kupferpulver während 30 Minuten unter Stickstoff am Rückfluss gekocht. Man giesst das Reaktionsgemisch auf Eis, extrahiert mit Petroläther und wäscht den Extrakt mit Wasser, verdünnter Natronlauge und erneut mit Wasser. Nach dem Trocknen mit Natriumsulfat und Abdampfen des Lösungsmittels im Vakuum bei 405 erhält man 14, 1 g eines Rohproduktes ; nid = 1, 4970 ; U. V.-Absorptionsmaximum bei 236 mÁ, E 1/1 - 562 (in Athanol).
Über die Bisulfitadditionsverbindung kann daraus 8, 1 g reines Citral isoliert werden.
In analoger Weise erhält man aus 3, 7-Dimethyl- 3-formoxy-octen- (6)-in- (1) [welches aus 3, 7-Di methyl-3-hydroxy-octen- (6)-in- (1) durch Behandlung mit einem Gemisch von Ameisensäure und Essigsäureanhydrid erhalten wird ; n2l = 1, 4529] unter Verwendung von Kupferacetat als Katalysator ein Reaktionsprodukt, das im U. V.-Spektrum ein Absorptionsmaximum bei 235 mM(inÄthanol) besitzt und über die Bisulfitadditionsverbindung reines Citral liefert.
Beispiel 11 148 g 3, 6, 7-Trimethyl-octen- (6)-in- (l)-ol- (3) werden in einem 1-Literkolben mit einer Lösung von 1, 7 g Phosphorsäure in 114 g Essigsäureanhydrid während 14 Stunden bei 25-35 gerührt. Hierauf versetzt man das Gemisch mit 277 cm3 80''/piger EssigsÏure, 4, 45 g Soda und 1, 48 g Silbercarbonat und erwärmt unter Rühren während einer Stunde am Rückfluss.
Nun giesst man das Reaktionsgemisch auf Eis und extrahiert mit Petroläther. Die gewaschene und getrocknete Petrolätherlösung wird vom Lö sungsmittel befreit, wobei als Rückstand rohes ±-Methyl-citral in Form eines braunen Ols in einer Ausbeute von 156 g erhalten wird ; nid = 1, 4850 bis

In analoger Weise wird 3-Methyl-7-phenyl-3 hydroxy-hepten- (6)-in- (1) durch Behandlung mit Essigsäureanhydrid und Phosphorsäure in 3-Methyl 7-phenyl-3-acetoxy-hepten- (6)-in- (l) übergeführt (n24 = 1, 52. 93) und daraus 3-Methyl-7-phenyl heptadien- (2, 6)-al- (l) hergestellt ; U. V.-Absorptions- maximum bei 247 m, ce ; El = 698 (in Äthanol), nid = 1, 5599.
Beispiel 12
5 g 3, 7, 11-Trimethyl-3-acetoxy-dodecin-(1) [ge- wonnen aus 3, 7, 11-Trimethyl-3-hydroxy-dodecin-(1) durch Behandlung mit Essigsäureanhydrid und Phosphorsäure ; Siedepunkt 105¯/0, 05 mm ; nD = 1, 4430] werden in 25 cm3 Eisessig gelöst und nach Zugabe von 0, 1 g Silbernitrat während 30 Minuten unter Stickstoff auf 100 erwärmt. Die Aufarbeitung nach Beispiel 10 liefert 4, 9 g eines Rohproduktes ; n 21/D = 1,4599; U. V.-Absorptionsmaximum bei 238 mÁ; E 1/1 = 366 (in ¯thanol). Es wird in 50 cm3 Aceton und 5 cm3 3n Schwefelsäure während 20 Stunden bei 20 stehengelassen. Durch Extraktion mit PetrolÏther, Waschen mit Wasser und Entfernen des Lösungsmittels gewinnt man 4, 1 g rohes 3, 7, 11 Trimethyl-dodecen-(2)-al-(1); n 21/D = 1,4678; U.V.
Absorptionsmaximum bei 237 mÁ; E 1/1 = 553 (in Äthanol). Die in üblicher Weise gereinigte Substanz siedet bei 120 ss12 mm und liefert ein öliges Semicarbazon, welches ein U. V.-Absorptionsmaximum bei 272 tRa (in Athanol) besitzt. Der freie Aldehyd besitzt einen starken angenehmen Fettaldehydgeruch.
In analoger Weise wird 3, 7, 11-Trimethyl-3- acetoxy-dodecadien-(6, 10)-in-(1) (n24= 1, 4700) in 3, 7, 11 - Trimethyl - dodecatrien - (2, 6, 10)-al- (1) (Farnesal) übergeführt, U. V.-Absorptionsmaximum bei 237 mu ; 590 (in Äthanol) ; nD = 1, 4977.
Beispiel 13
40 g 3, 7, 11, 15-Tetramethyl-3-acetoxy-hexadecin- (1) [Dehydroisophytylacetat, hergestellt aus Dehydroisophytol mittels Essigsäureanhydrid und Phosphor- sÏure, n 23/D = 1,4493] werden in 100 cm3 Eisessig gel¯st und nach Zugabe von 0,4 g Silbercarbonat eine Stunde unter Stickstoff auf 90 erwärmt. Man giesst das Reaktionsgemisch auf Eis, extrahiert mit Petroläther und wäscht den Extrakt 5mal mit Wasser.
Nach dem Trocknen mit Natriumsulfat und Abdampfen des Lösungsmittels erhält man 38, 3 g eines Reaktionsproduktes, das im U. V.-Spektrum nur eine schwache Absorption bei 238 m, u besitzt ; Ei = 40 (in Athanol) ; nD = 1, 4589. 20 g des so erhaltenen Produktes werden in 100 cm3 Methanol gelöst und nach Zugabe von 10 g Soda in 50 cm3 Wasser eine Stunde unter Rückfluss in Stickstoffatmosphäre ge- kocht. Durch Extraktion mit PetrolÏther, Waschen mit Wasser, Entfernen des Lösungsmittels und Trocknen über Natriumsulfat erhält man 16, 1 g rohes 3, 7, 11, 15-Tetramethyl-hexadecen- (2)-al- (1) (Phytal) ; nD = 1, 4650 ; U. V.-Absorptionsmaximum bei 238 m, ; Et = 336 (in Athanol).
Durch Hochvakuumdestillation gewinnt man daraus reines Phytal als farbloses viskoses 01 vom Siedepunkt 15701 0, 03 mm.
Beispiel 14
10 g 3, 7-Dimethyl-3-acetoxy-8-methoxy-octen- (6)- in- (l) [gewonnen aus 3, 7-Dimethyl-3-hydroxy-8-meth- oxy-octen- (6)-in- (l) durch Behandlung mit Essigsäureanhydrid und Phosphorsäure ; n20= 1, 4632] werden in 25 cm3 Eisessig gelöst und nach Zugabe von 0, 1 g Silbercarbonat eine Stunde unter Stickstoff auf 90 erwärmt. Die Aufarbeitung nach Beispiel 13 liefert 9, 3 g eines Reaktionsproduktes, das im U. V. Spektrum eine schwache Absorption bei 240 m, besitzt ; Et = 50 ; nD = 1, 4737.
Das erhaltene Produkt wird in 50 cm3 Methanol gelöst und nach Zugabe von 5 g Soda in 25 cm3 Wasser während 30 Minuten unter Rückfluss in Stickstoffatmosphäre gekocht. Die Aufarbeitung nach Beispiel 13 ergibt 6, 6 g 3, 7-Dimethyl-8-methoxy-octadien-(2,6)-al-(1) ; nD = 1, 4865 ; U. V.-Absorptionsmaximum bei 237 mÁ; E 1/1 = 518 (in Athanol).
Beispiel 15
28 g Cyclohexylester der 4-Methyl-4-acetoxyhexin- (5)-sÏure [mittols Essigsäureanhydrid und Phosphorsäure aus Cyclohexylester von 4-Methyl-4- hydroxy-hexin- (5)-sÏure hergestellt] werden in 100 cm3 Eisessig mit 0, 3 g Silbercarbonat eine Stunde unter Stickstoff auf 90 erwärmt. Das nach der Aufarbeitung gemäss Beispiel 13 erhaltene Reaktionsprodukt wird während 3 Stunden mit 10 g Natriumbicarbonat, 200 cm3 Methanol und 20 cm3 Wasser erwärmt. Das Hydrolyseprodukt wird wie in Beispiel 13 aufgearbeitet und ergibt den Cyclohexyl- ester von 4-Methyl-hexen- (4)-al- (6)-säure als farblose ölige Flüssigkeit ; Siedepunkt 100-110¯/0,05 mm ; n2D2 = 1, 4710 ; U.
V.-Absorptionsmaximum bei 234 m, cc ; Ei = 380 (in Athanol).
Beispiel 16
10, 4 g 2, 6, 6-Trimethyl-1-äthinyl-1-acetoxy-cyclo- hexan [hergestellt nach J. Chem. Soc. (1952), 1102] werden in 25 cm3 Eisessig gelöst und nach Zugabe von 125 mg Silbercarbonat unter Stickstoff eine Stunde auf 90 erwärmt. Die Aufarbeitung nach Beispiel 13 ergibt 10, 1 g eines Reaktionsproduktes, das auf Grund des I. R.-Spektrums zur Hauptsache aus 1- [2, 6, 6-Trimethyl-cyclohexyliden]-vinylacetat- (2) besteht ; n23 = 1, 4927.
Nach 1/2stündigem Kochen in einer Lösung von 10 g Natriumbicarbonat in 100 cm3 Methanol erhält man nach üblicher Aufarbeitung 8, 2 g 2, 6, 6-Tri- methyl-cyclohexyliden-acetaldehyd ; n 25/D = 1, 4996 ; U. V.-Absorptionsmaximum in Petrolätherlösung bei 238 m, ; El = 780.
Das entsprechende Phenylsemicarbazon kristallisiert aus Athanol in derben Nadeln ; Schmelzpunkt 137-139 .
Beispiel 17
300 mg 17a-Sithinyl-34, 17}-diacetoxy-androsten- (5) [hergestellt nach Helv. 20 (1937), 1280] werden in 6 cm3 Eisessig gelöst und nach Zugabe von 15 mg Silbercarbonat unter Stickstoff während 20 Minuten auf etwa 95 erwärmt. Man extrahiert mit Ather, wäscht dreimal mit verdünnter Natronlauge, dreimal mit Wasser, trocknet über Natriumsulfat und d entfernt das Lösungsmittel am Vakuum bei 50 .
Der Rückstand (330 mg, amorph) wird in 5 cm3 Aceton gelöst und nach Zugabe von 1 cm3 3n Schwefelsäure 5 Stunden bei 27 stehengelassen. Die übliche Aufarbeitung liefert ein Rohprodukt, das aus Petroläther kristallisiert wird. 50 g of 1-ethynyl-1-acetoxy-cyclohexane in 500 cm3 of aqueous acetic acid, after adding 0.5 g of silver nitrate, are refluxed for 30 minutes under nitrogen. The reaction mixture is poured onto ice, extracted with petroleum ether (boiling interval 40 to 60), washed repeatedly with water, dried over sodium sulfate and evaporated in vacuo. The 01 purified in this way contains one acetyl group per molecule and, according to the I.R. spectrum, represents 3, 7-dimethyl-1-acetoxy-octatriene- (1, 2, 6); nD = 1.4810; Density 0.927. It shows no absorption in the U.V. spectrum.
If the product thus obtained is treated with soda as in Example 3, a citral is obtained with an U.V. absorption maximum at 237 m, u, E1 = 780 (in ethanol).

50 g of 3, 7-dimethyl-3-acetoxy-octene (6) -in- (1) are stirred for 15 minutes with a mixture of 0.5 g of copper sulfate, 10 cm3 of concentrated sulfuric acid and 500 cm3 of benzene. The temperature of the mixture rises to 65 without external heating. After the usual work-up, the isolated dark reaction product is hydrolyzed to crude citral by boiling for half an hour with soda in a dilute methyl alcohol solution; U.V. absorption maximum at 236 m //. It is purified via the bisulfite addition compound; n20 = 1.4855; U.V. absorption maximum at 237-238 mol (in ethanol).
Example 9
20 g of 3, 7-dimethyl-3-acetoxy-octene (6) -in- (l) are heated to 703 with 80 cm3 of ethyl acetate with the addition of 1 g of copper acetate and 10 cm3 of concentrated sulfuric acid while stirring. At this temperature a reaction occurs suddenly, so that the reaction mixture comes to the boil. The mixture is stirred for a further 15 minutes without further heating and, after the usual work-up and hydrolysis, as in Example 8, crude citral is obtained as dark oil; nD = 1.4884. Pure citral can be isolated therefrom in a known manner via the bisulfite addition compound.
Example 10
25 g of 3, 7-dimethyl-3-benzoxy-octen- (6) -in- (l) [which from 3, 7-dimethyl-3-hydroxy-octen- (6) -in- (1) by treatment with Benzoyl chloride is recovered in pyridine; nD = 1.5147] are dissolved in 250 cm3 of glacial acetic acid and, after addition of 12.5 cm3 of water and 0.25 g of copper powder, refluxed for 30 minutes under nitrogen. The reaction mixture is poured onto ice, extracted with petroleum ether and the extract is washed with water, dilute sodium hydroxide solution and again with water. After drying with sodium sulfate and evaporation of the solvent in vacuo at 405, 14.1 g of a crude product are obtained; nid = 1,4970; U.V. absorption maximum at 236 mA, E 1/1 - 562 (in ethanol).
8.1 g of pure citral can be isolated therefrom via the bisulfite addition compound.
In an analogous manner, from 3, 7-dimethyl-3-formoxy-octen- (6) -in- (1) [which from 3, 7-dimethyl-3-hydroxy-octen- (6) -in- ( 1) obtained by treatment with a mixture of formic acid and acetic anhydride; n2l = 1.4529] using copper acetate as a catalyst, a reaction product which in the U.V. spectrum has an absorption maximum at 235 mM (in ethanol) and delivers pure citral via the bisulfite addition compound.
EXAMPLE 11 148 g of 3, 6, 7-trimethyl-octen- (6) -in- (1) -ol- (3) are mixed with a solution of 1.7 g of phosphoric acid in 114 g of acetic anhydride in a 1 liter flask for 14 Stirred at 25-35 hours. The mixture is then treated with 277 cm3 of 80 ″ / piger acetic acid, 4.45 g of soda and 1.48 g of silver carbonate and heated to reflux with stirring for one hour.
The reaction mixture is then poured onto ice and extracted with petroleum ether. The washed and dried petroleum ether solution is freed from the solvent, crude ± -Methyl-citral being obtained as a residue in the form of a brown oil in a yield of 156 g; nid = 1.4850 to
1, 4890; U.V. absorption maximum at 235 m, u; Et = about 620 (in ethanol). By purification via the bisulfite addition compound, 97 g of pure E-methyl-citral can be obtained therefrom in a known manner; Yield 66 "/ e; U.V. absorption maximum at 236 m, C; EI = about 820 (in ethanol).
In an analogous manner, 3-methyl-7-phenyl-3 hydroxy-hepten- (6) -in- (1) by treatment with acetic anhydride and phosphoric acid in 3-methyl 7-phenyl-3-acetoxy-hepten- (6) - converted into (l) (n24 = 1, 52, 93) and from it 3-methyl-7-phenyl heptadiene- (2, 6) -al- (l) produced; U.V. absorption maximum at 247 m, ce; El = 698 (in ethanol), nid = 1.5599.
Example 12
5 g of 3, 7, 11-trimethyl-3-acetoxydodecin- (1) [obtained from 3, 7, 11-trimethyl-3-hydroxydodecin- (1) by treatment with acetic anhydride and phosphoric acid; Boiling point 105¯ / 0.05 mm; nD = 1.4430] are dissolved in 25 cm3 of glacial acetic acid and, after the addition of 0.1 g of silver nitrate, heated to 100 for 30 minutes under nitrogen. Working up according to Example 10 gives 4.9 g of a crude product; n 21 / D = 1.4599; U.V. absorption maximum at 238 mA; E 1/1 = 366 (in ethanol). It is left to stand in 50 cm3 of acetone and 5 cm3 of 3N sulfuric acid for 20 hours at 20. Extraction with petroleum ether, washing with water and removal of the solvent gives 4.1 g of crude 3, 7, 11 trimethyl-dodecen- (2) -al- (1); n 21 / D = 1.4678; U.V.
Absorption maximum at 237 mÁ; E 1/1 = 553 (in ethanol). The substance, purified in the usual way, boils at 120 ± 12 mm and yields an oily semicarbazone which has an U.V. absorption maximum at 272 tRa (in ethanol). The free aldehyde has a strong, pleasant odor of fatty aldehyde.
In an analogous manner, 3, 7, 11-trimethyl-3-acetoxy-dodecadiene- (6, 10) -in- (1) (n24 = 1, 4700) is converted into 3, 7, 11-trimethyl-dodecatriene - (2, 6, 10) -al- (1) (Farnesal) transferred, UV absorption maximum at 237 mu; 590 (in ethanol); nD = 1.4977.
Example 13
40 g of 3, 7, 11, 15-tetramethyl-3-acetoxy-hexadecyne- (1) [dehydroisophytylacetate, prepared from dehydroisophytol using acetic anhydride and phosphoric acid, n 23 / D = 1.4493] are gel¯ in 100 cm3 of glacial acetic acid st and after adding 0.4 g of silver carbonate, heated to 90 for one hour under nitrogen. The reaction mixture is poured onto ice, extracted with petroleum ether and the extract is washed 5 times with water.
After drying with sodium sulfate and evaporation of the solvent, 38.3 g of a reaction product are obtained which, in the U.V. spectrum, has only a weak absorption at 238 μm; Egg = 40 (in ethanol); nD = 1.4589. 20 g of the product obtained in this way are dissolved in 100 cm3 of methanol and, after the addition of 10 g of soda in 50 cm3 of water, boiled under reflux in a nitrogen atmosphere for one hour. Extraction with petroleum ether, washing with water, removing the solvent and drying over sodium sulfate gives 16.1 g of crude 3, 7, 11, 15-tetramethyl-hexadecen- (2) -al- (1) (phytal); nD = 1.4650; U.V. absorption maximum at 238 m; Et = 336 (in ethanol).
Pure phytal is obtained therefrom by high vacuum distillation as colorless, viscous oil with a boiling point of 15701 0.03 mm.
Example 14
10 g of 3, 7-dimethyl-3-acetoxy-8-methoxy-octene (6) - in (l) [obtained from 3, 7-dimethyl-3-hydroxy-8-methoxy-octene (6 ) -in- (l) by treatment with acetic anhydride and phosphoric acid; n20 = 1.4632] are dissolved in 25 cm3 of glacial acetic acid and, after the addition of 0.1 g of silver carbonate, heated to 90 for one hour under nitrogen. Working up according to Example 13 yields 9.3 g of a reaction product which, in the U.V. spectrum, has a weak absorption at 240 m; Et = 50; nD = 1,4737.
The product obtained is dissolved in 50 cm3 of methanol and, after the addition of 5 g of soda in 25 cm3 of water, boiled under reflux in a nitrogen atmosphere for 30 minutes. Working up according to Example 13 gives 6.6 g of 3,7-dimethyl-8-methoxy-octadiene- (2,6) -al- (1); nD = 1.4865; U.V. absorption maximum at 237 mA; E 1/1 = 518 (in ethanol).
Example 15
28 g of cyclohexyl ester of 4-methyl-4-acetoxyhexyne (5) acid [with acetic anhydride and phosphoric acid from cyclohexyl ester of 4-methyl-4-hydroxyhexyne (5) acid] are prepared in 100 cm3 of glacial acetic acid with 0, 3 g of silver carbonate heated to 90 for one hour under nitrogen. The reaction product obtained after working up according to Example 13 is heated for 3 hours with 10 g of sodium bicarbonate, 200 cm3 of methanol and 20 cm3 of water. The hydrolysis product is worked up as in Example 13 and gives the cyclohexyl ester of 4-methylhexen (4) al- (6) acid as a colorless oily liquid; Boiling point 100-110¯ / 0.05 mm; n2D2 = 1,4710; U.
V. absorption maximum at 234 m, cc; Ei = 380 (in ethanol).
Example 16
10.4 g 2, 6, 6-trimethyl-1-ethinyl-1-acetoxy-cyclohexane [prepared according to J. Chem. Soc. (1952), 1102] are dissolved in 25 cm3 of glacial acetic acid and, after addition of 125 mg of silver carbonate, heated to 90 for one hour under nitrogen. Working up according to Example 13 gives 10.1 g of a reaction product which, based on the I.R. spectrum, mainly consists of 1- [2,6,6-trimethyl-cyclohexylidene] -vinylacetate- (2); n23 = 1,4927.
After boiling for 1/2 hour in a solution of 10 g of sodium bicarbonate in 100 cm3 of methanol, after conventional work-up, 8.2 g of 2,6,6-trimethyl-cyclohexylidene-acetaldehyde are obtained; n 25 / D = 1.4996; U.V. absorption maximum in petroleum ether solution at 238 m,; El = 780.
The corresponding phenyl semicarbazone crystallizes from ethanol in coarse needles; Melting point 137-139.
Example 17
300 mg of 17a-sithinyl-34, 17} -diacetoxy-androsten- (5) [prepared according to Helv. 20 (1937), 1280] are dissolved in 6 cm3 of glacial acetic acid and, after addition of 15 mg of silver carbonate under nitrogen, to about 95 heated. It is extracted with ether, washed three times with dilute sodium hydroxide solution, three times with water, dried over sodium sulfate and the solvent is removed in vacuo at 50.
The residue (330 mg, amorphous) is dissolved in 5 cm3 of acetone and, after the addition of 1 cm3 of 3N sulfuric acid, left to stand at 27 for 5 hours. The usual work-up gives a crude product which is crystallized from petroleum ether.
