REVENDICATIONS
1. Adjuvant d'immunité non spécifique, caractérisé en ce qu'il contient au moins une fraction choisie parmi les ribosomes et les acides ribonucléiques (ARN) ribosomaux d'au moins une bactérie choisie parmi: Corynebacterium acnei, Corynebacterium parfum, Corynebacterium granulosum, Corynebacteriumflavidum, Mycobacte rium smegmatls. Mycobacterium chelonei, Nocardia asteroides, Nocardia brasiliensis, Nocardia rhodocrans, Nocardia opaca, Nocardia rubra.
2. Procédé de préparation de l'adjuvant selon la revendication 1, caractérisé en ce que, pour la préparation de ribosomes bactériens utiles, on centrifuge à 30 000 g en milieu aqueux un broyat de cellules bactériennes correspondant aux ribosomes à extraire et que l'on préléve la phase aqueuse surnageante, et que l'on précipite les ribosomes de cette phase aqueuse.
3. Procédé selon la revendication 2, caractérisé en ce que l'on précipite les ribosomes de la phase aqueuse par de l'alcool éthylique.
4. Procédé de préparation de l'adjuvant selon la revendication 1, caractérisé en ce que, pour la préparation d'ARN ribosomaux bactériens utiles, I'on traite une suspension aqueuse de ribosomes correspondant aux ARN à extraire, préparée par le procédé selon la revendication 2, par du phénol et que l'on récupère la phase aqueuse pour en précipiter les ARN.
5. Procédé selon la revendication 4, caractérisé en ce que l'on précipite les ARN de la phase aqueuse par de l'alcool éthylique.
6. Préparation immunothérapique contenant à titre de principe actif l'adjuvant selon la revendication 1 avec un support acceptable.
7. Préparation immunothérapique selon la revendication 6, ca ractérisée en ce qu'elle contient de 0,1 à 1% en poids d'adjuvant d'immunité non spécifique par rapport à la composition.
8. Préparation immunothérapique selon l'une des revendications 6 ou 7, caractérisée en ce qu'elle se présente sous forme de doses d'un poids compris entre 1 et 10 mg.

L'invention, réalisée au Centre d'immunologie et de biologie
Pierre-Fabre, concerne des adjuvants d'immunité non spécifique,
leur préparation et des préparations immunothérapiques comprenant à titre de principe actif au moins un adjuvant d'immunité non spécifique selon l'invention avec un support acceptable. Ces préparations immunothérapiques peuvent être utilisées par voie parentérale, orale, rectale ou locale.
La présente invention repose sur l'observation que les ribosomes
et l'acide ribonucléique (ARN) ribosomal de certaines bactéries pos
sèdent, en plus de leur pouvoir immunogénique spécifique, des propriétés immunostimulantes non spécifiques susceptibles de renforcer et de stimuler de façon générale les défenses immunitaires de l'organisme, par stimulation du système réticuloendothélial.
Le pouvoir immunogénique non spécifique peut être utilisé de
deux façons, soit pour renforcer les défenses générales de l'orga
nisme à l'égard des maladies infectieuses bactériennes, dans ce cas le
ou les adjuvants d'immunité sont utilisés à titre de principe actif principal dans la préparation pharmaceutique utilisée, soit pour potentialiser l'action d'un vaccin spécifique.
La présente invention concerne donc tout d'abord un adjuvant
d'immunité non spécifique, caractérisé en ce qu'il est constitué par
les ribosomes ou l'ARN ribosomal d'au moins une bactérie choisie patmi: Corynebacterium ainsi, Corynebacteriumparvum, Corynebac-
terium granulosum, Corynebacteriumflavidum, Mycobacterium smegmatis, Mycobacterium chelonei, Nocardia asteroides, Nocardia brasiliensis, Nocardia rhodocrans, Nocardia opaca, Nocardia rubra.
Ces adjuvants d'immunité non spécifique se présentent, de préférence, sous forme lyophilisée.
Dans le cas où les adjuvants d'immunité non spécifique constituent le principe actif unique des préparations immunothérapiques selon l'invention, ces compositions sont de préférence sous forme injectable et comprennent de préférence de 0,1 à 1% en poids d'adjuvants d'immunité non spécifique, chaque dose unitaire de composition pharmaceutique étant comprise de préférence entre 1 et 10 mg.
Le support thérapeutiquement acceptable de ces préparations peut être quelconque mais sera de préférence le mannitol, éventuellement dans un tampon, comme un tampon phosphate.
On donne ci-après des exemples de formulation de telles préparations.

. 5 mg Il est à remarquer que lorsque l'on utilise des ARN à titre d'ad- juvants d'immunité non spécifique, la quantité en poids par dose de la préparation peut être plus faible que lorsque l'on utilise la fraction ribosomale correspondante.
La formule type III désigne une préparation dans laquelle les ad
juvants d'immunité non spécifique selon la présente invention ont été associés à d'autres adjuvants d'immunité, en l'occurrence des glycopeptides membranaires. On peut également utiliser d'autres adjuvants d'immunité tels que des glycopeptides extraits de parois ou membranes bactériennes, des glycoprotéines extraites des membranes bactériennes ou des corps bactériens entiers, des polysaccharides de membranes externes et capsulaires de bactéries Gram négatif.
Dans le cas où les adjuvants d'immunité non spécifique sont utilisés pour potentialiser l'action de vaccins, la quantité en poids d'adjuvants d'immunité non spécifique sera comprise entre 10 et 100% du poids de la partie active du vaccin.

Ce vaccin peut être préparé notamment par le procédé décrit dans le brevet français No 73.43957.
Les préparations immunothérapiques selon la présente invention peuvent être utilisées aussi bien en médecine humaine qu'en médecine vétérinaire.
Les ribosomes et les ARN ribosomaux peuvent être préparés par des procédés connus, par exemple par les procédés décrits dans les brevets français Nos 73.43957 et 75.10252.
En particulier, on prépare les ribosomes bactériens utiles comme adjuvants d'immunité non spécifique selon la présente invention par centrifugation à 30 000 g d'un broyat de cellules bactériennes en solution aqueuse, prélèvement de la phase surnageante et précipitation des ribosomes contenus dans la phase surnageante prélevée; de préférence la précipitation est réalisée au moyen d'alcool éthylique.
Le précipité de ribosomes ainsi obtenu peut, si cela est nécessaire, être purifié par exemple par traitement au dodécylsulfate de sodium.
Le broyat peut être obtenu par n'importe quel procédé connu, par exemple par traitement d'une culture bactérienne par un homogénéiseur.
Les ARN ribosomaux sont préparés à partir des ribosomes en phase aqueuse par extraction au phénol, les ARN se trouvant en fin d'extraction dans la phase aqueuse. Ils peuvent être récupérés à partir de la phase aqueuse par précipitation à l'alcool éthylique.
Ils peuvent en outre être purifiés, si cela est nécessaire.
On donne ci-après, à titre d'exemple, un mode de préparation des ribosomes et des ARN qui ont été utilisés dans les formules précédentes.
La souche de Corynebacterium parvum est entretenue en gélose profonde viande-foie et repiquée tous les deux mois. Un tube de gélose est utilisé pour ensemencer le milieu de préculture. La préculture est réalisée dans 300 ml de milieu réducteur (thioglycolate/cystéine) en culture statique en étuve, à 37"C pendant 24 h.
La culture proprement dite est réalisée dans un fermenteur de 20 I contenant 15 I d'un milieu (extrait de viande, extrait de levure, tryptone, glucose, NaCI et thioglycolate de sodium). Le flacon de préculture est transvasé dans le fermenteur au moyen d'une surpression d'air stérile et la croissance est faite à 37 C sous agitation lente, avec une aération faible, le pH étant réglé à 7 par addition automatique de soude. Le temps de croissance est de 48 à 72 h et le contrôle est effectué par mesure, à intervalles réguliers, de la densité optique.
A la fin de la phase exponentielle de croissance, les cellules sont recueillies par centrifugation continue sur Sharples, le fermenteur étant raccordé stérilement au séparateur, et en surpression d'air stérile. Le milieu de culture est éliminé après décontamination et la biomasse lavée à +4"C, par centrifugation avec un tampon stérile (tris-HC1 0,01M, pH 7 contenant MgCI2 0,01M et NaCI 0,l5M). Le concentrat bactérien final est immédiatement congelé en attendant d'être utilisé. Un contrôle bactériologique à ce stade est réalisé pour identifier la souche et déterminer le nombre de germes revivifiables, ainsi que l'absence de contaminants.
Pour l'obtention de biomasses plus importantes, le fermenteur de 201 est utilisé comme préfermenteur pour un fermenteur de 600 1, la culture et la récolte des cellules étant conduites dans les mêmes conditions.
Préparation de la fraction ribosomale
La biomasse est mise en suspension dans le tampon tris-HCl, MgCl2, pH 7 pour avoir environ 1 g en cellules sèches dans 20 ml de suspension. Aprés une homogénéisation rapide à l'Ultra-turrax, la suspension est désintégrée par passage 3 fois de suite dans un homogénéiseur APV équipé de clapets de désintégration, à la pression maximale, soit 560 kg/cm2. La suspension est réfrigérée en continu par passage dans un échangeur à plaques.
Après broyage, la suspension est clarifiée par passage sur Shar ples à 10000 g pour éliminer les débris cellulaires.
Le surnageant est recueilli et soumis à une centrifugation de 45 min à 30 000 g et à 4 C afin d'éliminer les parois, membranes et autres fractions de haut poids moléculaire.
Le surnageant contenant les ribosomes est conservé et la fraction ribosomale brute est précipitée par addition de 0,8 volume d'alcool éthylique 96% préalablement refroidi à -20"C. On laisse au repos 30 min à 4"C et le précipité est recueilli sur Sharples; le surnageant est éliminé.
Le précipité de ribosomes bruts est remis en suspension dans 1 volume du tampon tris-HCI, MgCl2, NaCI, pH 7 à 15"C par agitation pendant 1 h et on ajoute alors du sodium dodécylsulfate (SDS) pour avoir une concentration finale de 0,5% (agitation vive pendant
I h à température ambiante).
Les ribosomes lavés sont à nouveau précipités par addition de 0,7 volume d'alcool éthylique à - 20 C. Repos pendant 30 min à température ambiante.
Le précipité de ribosomes lavés est recueilli sur Sharples à
10 000 g et le surnageant est éliminé.
Le précipité est repris dans le même tampon (0,4 volume du volume initial de suspension), par agitation douce, une nuit, à OC.
La reprise est clarifiée par une centrifugation de 60 min à 30 000 g et à OC, le surnageant contenant les ribosomes est conservé soit congelé soit lyophilisé après stérilisation sur membrane (porosité 0,22 Il).
Extraction de l 'ARN ribosomal
L'ARN est extrait de la préparation ribosomale précédente par le phénol, dans les conditions suivantes.
Une solution de phénol à 80% est préparée avec du tampon tris HCl 0,001 M, pH 7 contenant EDTA 0,001M et 0,5% de SDS. Cette solution est thermostatée à 65 C.
L'extraction est faite volume/volume pendant 10 min à 65"C sous agitation. Le mélange est ensuite refroidi rapidement à +4"C et la phase aqueuse est recueillie par passage sur Sharples équipé d'un bol séparateur. La phase phénol est écartée.
La phase aqueuse est réextraite deux fois dans les mêmes conditions par 1/2 volume de phénol. La phase aqueuse finale est recueillie et extraite par trois fois I volume d'éther pour éliminer le phénol résiduel. L'éther subsistant dans la phase aqueuse est chassé par barbotage d'azote.
On ajoute alors du chlorure de sodium à la phase aqueuse pour avoir une molarité de 0,1M. L'ARN est alors précipité par addition de 2 volumes d'alcool éthylique pendant 4 à -20"C.
Le précipité est recueilli par centrifugation à 10 000 g et à OOC sur Sharples et repris dans du tampon tris-HC1 0,025M, pH 8,1 contenant NaCI 0,025M à +2"C pour avoir environ 2 mg d'ARN par ml de solution.
Puriftcation de L'ART ribosomal
A ce stade, FARN ribosomal peut être encore contaminé par des traces de carbohydrates qui peuvent être éliminées de la façon suivante.
A la solution d'ARN précédente, on ajoute I volume d'une solution 2,5M de phosphate dipotassique et 1 volume de 2-méthoxyéthanol. Agitation énergique pendant 2 à 3 min puis centrifugation pendant 5 min à 10 000 g et à +4 C. Le surnageant clair est recueilli avec précaution et mélangé à 1 volume d'acétate de sodium 0,2M.
On précipite alors l'ARN par addition lente de 0,5 volume d'une solution aqueuse à 1% de bromure de cétyltriméthylammonium. On laisse reposer 5 min à 4"C puis le précipité est recueilli par centrifugation pendant 5 min à 40 C. Le précipité est lavé deux fois par un excès d'alcool à 70% contenant CH3COONa 0,1M et remis en solution dans du tampon tris-HC1 0,01M, pH 7 et dialysé une nuit à +20C contre 20 volumes de ce tampon.
Le dialysat est finalement lyophilisé.
Afin de mettre en évidence l'activité de l'adjuvant d'immunité non spécifique selon l'invention, on a étuidé l'activité immunologique des formules type V, VI, VII et du vaccin ORL seul. Chaque formule est étudiée sur 10 souris.
Chaque animal reçoit une demi-dose de préparation à chaque injection, par voie sous-cutanée, au rythme de 5 injections en 15 d.
Après 10 d de repos, l'animal est ponctionné par les sinus rétroorbitaires.
Les sérums sont étudiés en immunodiffusion (Ouchterlony) et en immunoélectrodiffusion (Laurell) contre les antigènes bactériens complets dont on a extrait les ribosomes.
Ici, le vaccin ORL contient des ribosomes de Klebsiella pneumoniae.
En immunoélectrodiffusion la hauteur des Rockets est proportionnelle à la concentration en anticorps spécifique de l'antigène (ici
Klebsiella pneumoniae 600 l).
On observe que les Rockets correspondant aux préparations immunothérapiques selon la présente invention, c'est-à-dire comportant des adjuvants d'immunité non spécifique, sont nettement plus importants que la Rocket correspondant au vaccin ORL seul et qu'en conséquence on observe une nette augmentation du pouvoir immunologique des vaccins selon l'invention par rapport au vaccin
ORL seul.
Les préparations immunothérapiques selon la présente invention peuvent être utilisées pour la prévention en général des maladies d'origine bactérienne, ou pour prévenir des maladies particulières dans le cas où elles contier;nent un vaccin.

It is also possible to use other immunity adjuvants such as glycopeptides extracted from bacterial walls or membranes, glycoproteins extracted from bacterial membranes or whole bacterial bodies, polysaccharides from external membranes and capsular of Gram negative bacteria.
In the case where non-specific immunity adjuvants are used to potentiate the action of vaccines, the amount by weight of non-specific immunity adjuvants will be between 10 and 100% of the weight of the active part of the vaccine.

This vaccine can be prepared in particular by the method described in French patent No 73.43957.
The immunotherapeutic preparations according to the present invention can be used both in human medicine and in veterinary medicine.
Ribosomes and ribosomal RNAs can be prepared by known methods, for example by the methods described in French patents Nos. 73.43957 and 75.10252.
In particular, the bacterial ribosomes useful as adjuvants of non-specific immunity according to the present invention are prepared by centrifugation at 30,000 g of a ground material of bacterial cells in aqueous solution, removal of the supernatant phase and precipitation of the ribosomes contained in the phase. supernatant removed; preferably the precipitation is carried out by means of ethyl alcohol.
The ribosome precipitate thus obtained can, if necessary, be purified for example by treatment with sodium dodecyl sulfate.
The ground material can be obtained by any known method, for example by treatment of a bacterial culture with a homogenizer.
Ribosomal RNAs are prepared from ribosomes in the aqueous phase by phenol extraction, the RNAs being found at the end of extraction in the aqueous phase. They can be recovered from the aqueous phase by precipitation with ethyl alcohol.
They can also be purified, if necessary.
An example is given below of a method of preparing the ribosomes and the RNAs which have been used in the preceding formulas.
The Corynebacterium parvum strain is maintained in deep meat-liver agar and transplanted every two months. An agar tube is used to inoculate the preculture medium. The preculture is carried out in 300 ml of reducing medium (thioglycolate / cysteine) in static culture in an oven, at 37 ° C. for 24 h.
The actual culture is carried out in a 20 I fermenter containing 15 I of a medium (meat extract, yeast extract, tryptone, glucose, NaCl and sodium thioglycolate). The preculture flask is transferred to the fermenter by means of an overpressure of sterile air and the growth is made at 37 ° C. with slow stirring, with low aeration, the pH being adjusted to 7 by automatic addition of sodium hydroxide. The growth time is 48 to 72 h and the control is carried out by measuring, at regular intervals, the optical density.
At the end of the exponential growth phase, the cells are collected by continuous centrifugation on Sharples, the fermenter being sterile connected to the separator, and under sterile air pressure. The culture medium is removed after decontamination and the biomass washed at +4 "C, by centrifugation with a sterile buffer (0.01M tris-HCl, pH 7 containing 0.01M MgCl2 and 0.15M NaCl). The final bacterial concentrate is immediately frozen while waiting to be used. A bacteriological check is carried out at this stage to identify the strain and determine the number of revivable germs, as well as the absence of contaminants.
To obtain larger biomass, the 201 fermenter is used as a prefermenter for a 600 1 fermenter, the culture and the harvesting of the cells being carried out under the same conditions.
Preparation of the ribosomal fraction
The biomass is suspended in the tris-HCl buffer, MgCl2, pH 7 to have approximately 1 g in dry cells in 20 ml of suspension. After rapid homogenization with Ultra-turrax, the suspension is disintegrated by passing 3 times in succession through an APV homogenizer equipped with disintegration valves, at maximum pressure, ie 560 kg / cm2. The suspension is continuously refrigerated by passage through a plate exchanger.
After grinding, the suspension is clarified by passing over Shar ples to 10,000 g to remove cellular debris.
The supernatant is collected and subjected to centrifugation for 45 min at 30,000 g and at 4 ° C. in order to remove the walls, membranes and other high molecular weight fractions.
The supernatant containing the ribosomes is preserved and the crude ribosomal fraction is precipitated by adding 0.8 volume of 96% ethyl alcohol previously cooled to -20 "C. It is left to stand for 30 min at 4" C and the precipitate is collected. on Sharples; the supernatant is removed.
The precipitate of crude ribosomes is resuspended in 1 volume of tris-HCl buffer, MgCl2, NaCl, pH 7 at 15 "C by stirring for 1 h and then sodium dodecylsulfate (SDS) is added to have a final concentration of 0 5.5% (vigorous shaking during
1 h at room temperature).
The washed ribosomes are again precipitated by adding 0.7 volume of ethyl alcohol at -20 C. Rest for 30 min at room temperature.
The precipitate of washed ribosomes is collected on Sharples at
10,000 g and the supernatant is removed.
The precipitate is taken up in the same buffer (0.4 volume of the initial volume of suspension), by gentle stirring, overnight, at OC.
The recovery is clarified by centrifugation for 60 min at 30,000 g and at OC, the supernatant containing the ribosomes is kept either frozen or lyophilized after sterilization on a membrane (porosity 0.22 l).
Ribosomal RNA extraction
The RNA is extracted from the preceding ribosomal preparation by phenol, under the following conditions.
An 80% phenol solution is prepared with 0.001 M HCl tris buffer, pH 7 containing 0.001 M EDTA and 0.5% SDS. This solution is thermostatically controlled at 65 C.
The extraction is carried out volume / volume for 10 min at 65 "C with stirring. The mixture is then rapidly cooled to +4" C and the aqueous phase is collected by passage over Sharples equipped with a separator bowl. The phenol phase is discarded.
The aqueous phase is reextracted twice under the same conditions with 1/2 volume of phenol. The final aqueous phase is collected and extracted with three times I volume of ether to remove the residual phenol. The ether remaining in the aqueous phase is removed by bubbling nitrogen through.
Sodium chloride is then added to the aqueous phase to have a molarity of 0.1M. The RNA is then precipitated by adding 2 volumes of ethyl alcohol for 4 at -20 ° C.
The precipitate is collected by centrifugation at 10,000 g and at OOC on Sharples and taken up in 0.025M tris-HCl buffer, pH 8.1 containing 0.025M NaCl at +2 "C to have approximately 2 mg of RNA per ml of solution.
Purification of ribosomal ART
At this stage, ribosomal FARN can still be contaminated with traces of carbohydrates which can be eliminated as follows.
To the preceding RNA solution is added I volume of a 2.5M solution of dipotassium phosphate and 1 volume of 2-methoxyethanol. Vigorous stirring for 2 to 3 min then centrifugation for 5 min at 10,000 g and at +4 C. The clear supernatant is collected carefully and mixed with 1 volume of 0.2M sodium acetate.
The RNA is then precipitated by the slow addition of 0.5 volume of a 1% aqueous solution of cetyltrimethylammonium bromide. The mixture is left to stand for 5 min at 4 "C. then the precipitate is collected by centrifugation for 5 min at 40 C. The precipitate is washed twice with an excess of 70% alcohol containing 0.1 M CH3COONa and redissolved in buffer 0.01M tris-HC1, pH 7 and dialyzed overnight at + 20C against 20 volumes of this buffer.
The dialysate is finally lyophilized.
In order to demonstrate the activity of the non-specific immunity adjuvant according to the invention, the immunological activity of the formulas V, VI, VII and the ENT vaccine alone was studied. Each formula is studied on 10 mice.
Each animal receives a half-dose of preparation for each injection, subcutaneously, at the rate of 5 injections in 15 d.
After 10 d of rest, the animal is punctured by the retroorbital sinuses.
The sera are studied in immunodiffusion (Ouchterlony) and in immunoelectrodiffusion (Laurell) against the complete bacterial antigens from which the ribosomes have been extracted.
Here, the ENT vaccine contains ribosomes of Klebsiella pneumoniae.
In immunoelectro diffusion the height of the Rockets is proportional to the concentration of antibody specific to the antigen (here
Klebsiella pneumoniae 600 l).
It is observed that the Rockets corresponding to the immunotherapeutic preparations according to the present invention, that is to say comprising adjuvants of non-specific immunity, are clearly greater than the Rocket corresponding to the ENT vaccine alone and that consequently a marked increase in the immunological power of the vaccines according to the invention compared with the vaccine
ENT alone.
The immunotherapeutic preparations according to the present invention can be used for the general prevention of diseases of bacterial origin, or to prevent particular diseases in the case where they contain a vaccine.
