2. Verfahren nach Anspruch 1 zur Herstellung von 8'-(3 tert.-Butylamino-2-hydroxypwpoxy)-spiro[cydopentan 1,2'(1'H)-naphthalin]-4'(3'H)-on, dadurch gekennzeichnet, dass man eine Verbindung der Formel II, in der R2 und R3 Tetramethylen und n die Zahl 1 bedeuten und Rx die in Anspruch 1 angegebene Bedeutung besitzt, mit tert.-Butylamin umsetzt.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die in Anspruch 1 definierten Verbindungen der Formel I, in denen R2 und R3 identisch sind und die (S) Konfiguration am asymmetrisch substituierten Kohlenstoffatom der 3-Amino-2-hydroxypropoxy-Seitenkette besteht, herstellt.
Die Erfindung betrifft den in den Ansprüchen definierten Gegenstand.
Das anhand des Disclaimers in Anspruch 1 nicht Beanspruchte ist bereits bekannt.
R1 bedeutet vorzugsweise Alkyl. R2 und R3 stehen vorzugsweise für Alkyl, insbesondere identische Alkylgruppen oder bedeuten zusammen vorzugsweise Alkylen mit 4 oder 5, insbesondere 4 Kohlenstoffatomen.
Steht R1 für Alkyl, so ist dieser Rest vorzugsweise verzweigt, insbesondere in a-Stellung zum Stickstoffatom, an das er gebunden ist. Besonders bevorzugte Alkylreste sind Isopropyl, tert.-Butyl, 3-Pentyl und tert.-Pentyl, insbesondere tert.-Butyl.
Steht R1 für gegebenenfalls substituiertes Phenylalkyl, Phenoxyalkyl oder Phenylthioalkyl mit mehr als 2 Kohlenstoffatomen im Alkylteil, so ist dieser Alkylteil ebenfalls vorzugsweise verzweigt, insbesondere in a-Stellung zum Stickstoffatom, an das er gebunden ist. Beispiele solcher Reste sind a,a-Dimethylphenäthyl, a,a-Dimethylphenoxyäthyl und a,a-Dimethylphenylthioäthyl.
Alkyl (ausser wie oben für Rl erläutert) und Alkoxy enthalten vorzugsweise 1 oder 2, insbesondere 1 Kohlenstoffatom. Halogen steht vorzugsweise für Chlor oder Brom, insbesondere für Chlor.
n steht vorzugsweise für die ganze Zahl 1.
Die Verbindungen der Formel I können in freier Form oder in Säureadditionssalzen vorliegen. Aus den Verbindungen der Formel I in freier Form lassen sich in bekannter Weise Säureadditionssalze gewinnen und umgekehrt. So bilden die erfindungsgemässen Verbindungen der Formel I in freier Form Säureadditionssalze mit Säuren wie z.B. Maleinsäure.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I, indem man eine entsprechende Verbindung der Formel II, worin
R2, R3 und n obige Bedeutung besitzen und
Rx für einen Rest steht, der bei der Umsetzung mit einem substituierten Amin eine N-substituierte 2-Amino-l- hydroxyäthylgruppe ergibt, mit einer Verbindung der Formel III, worin R1 obige Bedeutung besitzt, umsetzt.
Die erfindungsgemässe Umsetzung ist eine Aminierung durch ein primäres Amin. Sie kann unter Verwendung von für die Herstellung bekannter 3-Amino-2-hydroxypropoxyaryl-Verbindungen bekannten Bedingungen erfolgen. Als Gruppe Rx verwendet man beispielsweise die Gruppe

worin Y Halogen, vorzugsweise Chlor oder Brom, oder eine Gruppe Ry-SO2-O-, worin Ry Phenyl, Tolyl oder niederes Alkyl bedeutet. Y steht insbesondere für Chlor. Die Umsetzung wird vorzugsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z. B. in einem Ather wie Dioxan durchgeführt. Gegebenenfalls verwendet man als Lösungsmittel die Verbindung der Formel III im Überschuss. Zweckmässig wird die Umsetzung auch in der Schmelze durchgeführt. Die Reaktionstemperatur liegt zwischen etwa 20 und etwa 200 "C, vorzugsweise bei Siedetemperatur des Reaktionsgemisches.
In den Verbindungen der Formel list das Kohlenstoffatom der Seitenkette, das die Hydroxygruppe trägt, asymmetrisch substituiert; sie können daher in Form von Racematen oder Enantiomeren auftreten. Falls R2 und R3 nicht identisch sind, treten sie als Diastereoisomerengemisch auf.
Bevorzugt sind die Verbindungen der Formel I, in denen R2 und R3 identisch sind, und die (S)-Konfiguration am asymmetrisch substituierten Kohlenstoffatom der 3-Amino-2hydroxypropoxy-Seitenkette besteht.
Die Enantiomeren bzw. Diastereoisomeren der Verbindungen der Formel I können auf an sich bekannte Weise erhalten werden, z. B. durch Durchführung des erfindungsgemässen Verfahrens unter Verwendung der entsprechenden Enantiomeren bzw. Diastereoisomeren der Verbindungen der Formel II, oder durch Aufspaltung mit optisch aktiven Säuren.
Die Ausgangsprodukte können analog zu bekannten Methoden erhalten werden:
Die Verbindungen der Formel II erhält man z. B. durch Ätherspaltung der entsprechenden Verbindungen der Formel IV, worin n, R2 und R3 obige Bedeutung besitzen und R4 Methyl oder Benzyl bedeutet, und anschliessende O-AI- kylierung der OH-Gruppe der so erhaltenen Phenole zu einer Gruppe-cH2-Rx-
Die Verbindungen der Formel lava, worin R4 obige Bedeutung besitzt und n1, R21 und R31 die oben für n, R2 und R3 angegebene Bedeutung besitzen mit der Einschränkung, dass R21 und R31 nicht gleichzeitig Wasserstoff bedeuten, erhält man z. B. durch Cyclisierung der Verbindungen der Formel V, worin R21, R31, R4 und nl obige Bedeutung besitzen, mit z.B.
Polyphosphorsäure.
Die Verbindungen der Formel Verhält man z.B. durch Hydrolyse der Verbindungen der Formel VI, worin R21, R31, R4 und n1 obige Bedeutung besitzen und Rs Alkyl mit 1 bis 4 Kohlenstoffatomen bedeutet, und anschliessende CO2-Abspaltung aus dem Reaktionsprodukt.
Die Verbindungen der Formel VI erhält man z. B. durch Grignard-Umsetzung der Verbindungen der Formel VII, worin R4 und nl obige Bedeutung besitzen und Z Chlor oder Brom bedeutet, mit den Verbindungen der Formel VIII, worin R21, R31 und R5 obige Bedeutung besitzen.
Die Verbindungen der Formel IVb, worin R4 obige Be deutung besitzt, erhält man z. B. durch Oxidation der ent sprechenden Benzosuber-5-ole mit z. B. Chromsäure oder Mangandioxid.
Soweit die Herstellung der benötigten Ausgangsmateri alien nicht beschrieben wird, sind diese bekannt oder können nach bekannten Verfahren bzw. analog zu den hier bzw. in den Beispielen beschriebenen oder analog zu an sich bekann ten Verfahren hergestellt und gereinigt werden.
In den nachfolgenden Beispielen, welche die Erfindung näher erläutern, erfolgen alle Temperaturangaben in Celsiusgraden, ohne Korrekturen.
Beispiel 1 8'-(3-tert.-Butylamino-2-hydroxypropoxy)-spiro[cyclö- pentan-1,2'(1 'H)-naphthalin]-4'(3'H)-on
7,5 g rohes 8'-(3-Chlor-2-hydroxypropoxy)-spiro[cyclo ?entan-1,2'(1'H)-naphthalin]-4'(3'E{)-on werden in 50 ml Dioxan und 30 ml tert.-Butylamin im Autoklaven während t0 Stunden bei 1300 gekocht, eingeengt und zwischen 10%iger wässriger Weinsäurelösung und Äther verteilt. Der vässrige Teil wird alkalisch gestellt, mit Äther extrahiert, die trganischen Phasen eingeengt und die verbleibende Titelverbindung in ihr Hydrogenmaleinat überführt (Smp. des Hydrogenmaleinats der Titelverbindung: 188-190 - aus Äthanol).
Das Ausgangsmaterial kann wie folgt erhalten werden: a) Zu einer Lösung von 2-Methoxybenzylmagnesiumchlorid (hergestellt aus 5,3 g Magnesium und 31,2 g 2-Meth oxybenzylchlorid in 400 ml Äther) wird eine Lösung von 39,9 g Cyclopentylidencyanessigsäureäthylester in 350 ml Tetrahydrofuran zugetropft und während 2 Stunden bei Raumtemperature nachgerührt. Die Mischung wird dann mit 300 ml l5%iger Ammoniumchlorid-Lösung versetzt, das Produkt mit Äther extrahiert und die Ätherlösung eingeengt.
Man erhält das rohe a-Cyano-(l -(2-Methoxybenzyl)cyclo- pentan}-essigsäure-äthylester, das bei 140-180 /0,002 mm destilliert wird (roh weiterverarbeitet).
b) Zu einer Lösung von 32,4 g a-Cyano-(l -(2-methoxy- benzyl)cyclopentan)essigsäure-äthylester in 300 ml Äthylenglykol wird 33,6 g Kaliumhydroxid zugegeben und die Lösung 40 Stunden bei 1800 gerührt. Die Lösung wird gekühlt, auf Eis-Wasser gegossen und die neutralen Nebenprodukte mit Äther wegextrahiert. Der wässrige Teil wird mit Salz Säure angesäuert und mit Äther extrahiert. Nach Einengen des Lösungsmittels bleibt die [1 -(2-Methoxybenzyl)cyclo- pentan]essigsäure (Smp. 85-87 - aus Hexan) zurück.
c) 5,7 g [1 -(2-Methoxybenzyl)cyclopentan]essigsäure wird unter Rühren bei 110 auf 30 g Polyphosphorsäure gegeben und 10 Minuten nachgerührt. Die Mischung wird gekühlt und 250 ml Wasser zugesetzt. Durch Extraktion mit Äther und Einengen des Äther-Extraktes erhält man das 8' Methoxy-spiro[cyclopentan- 1,2'(1 'H)-naphthalin]-4'(3'H)on (Smp. 62-66 - direkt weiterverarbeitet).
d) 5,0 g 8'-Methoxy-spiro[cyclopentan-1,2'(I'H)-naph- thalin]-4'(3'H)-on wird mit 50 ml Essigsäure und 10 ml 48%iger Bromwasserstofflösung 20 Stunden am Rückfluss gekocht, dann die Lösung eingeengt, mit Wasser verdünnt und mit Äther extrahiert. Nach Entfernen des Lösungsmittels bleibt das 8'-Hydroxy-spiro[cyclopentan- 1,2'(1'H)-naph- thalin]-4'(3'H)-on (Smp. 163-165 - aus Toluol) zurück.
e) Zu einer Lösung von 4,6 g 8'-Hydroxy-spiro[cyclo pentan-l ,2'(1'H)-naphthalin]-4'(3'H)-on in 30 ml Epichlorhydrin werden 3 Tropfen Piperidin zugegeben und die Mischung 4 Stunden bei 100" gerührt. Die Lösung wird eingedampft, in Äther aufgenommen, filtriert und eingeengt, wobei das rohe 8'-(3-Chlor-2-hydroxypropoxy)-spiro[cyclopentan-1,2'(1'H)-naphthalin]-4'(3'H)-on zurückbleibt.
Analog Beispiel 1 erhält man die folgenden Verbindun gen der Formel I durch Umsetzung entsprechender Verbin dungen der Formel II, worin Rx für

Die Verbindungen der Formel I, d. h. die Verbindungen der Formel I in freier Form oder in Form ihrer physiologisch verträglichen Säureadditionssalze, zeichnen sich durch interessante pharmakodynamische Eigenschaften aus. Sie können daher als Heilmittel verwendet werden.
Sie zeigen am spontanschlagenden, isolierten Meerschweinchenvorhof eine Hemmung der positiv-inotropen Adrenalinwirkung, wobei diese antagonistische Wirkung bei Badkonzentrationen von ca. 0,005 bis ca. 2,5 ml/l auftritt.
Am narkotisierten Ganztier (anästhesierten Hund) führen sie mit einer wirksamen kumulativen Dosis von ca. 0,02 bis ca. 0,6 mg/kg i.v. zu einer starken Hemmung der durch Iso proterenol [1 -(3,4-Dihydroxyphenyl)-2-2-isopropylamino- äthanol] bedingten Tachycardie und Blutdrucksenkung.
Sie besitzen demnach eine Blockwirkung auf die adrenergischen ss-Rezeptoren und können daher u. a. zur Prophylaxe und Therapie von Koronarerkrankungen, insbesondere zur Behandlung von Angina pectoris, Verwendung finden.
Aufgrund ihrer antiarrhythmischen Wirkung sind sie ausserdem zur Behandlung von Herzrhythmusstörungen geeignet.
Sie zeigen ausserdem interessante Stoffwechselwirkungen, indem sie die durch psychischen Stress induzierte Mobilisation von Glucose und Fettsäure im Blut hemmen. So bewirken sie an isolierten Fettzellen des epididymalen Fettgewebes von Ratten [in vitro Methode nach M. Rodbell, J.
Biol. Chem. 239 (1964) 375-380] bei einer Konzentration von ca. 0,1 bis ca. 10 mg/l eine bedeutende Hemmung der durch Isoproterenol stimulierbaren Glycerinfreisetzung und bei der Ratte hemmen sie die durch Isoproterenol stimulierte Lipolyse (Glycerinausschüttung) und Glycogenolyse (Plasmaglucoseanstieg) mit Dosen von ca. 0,01 bis ca. 5 mg/kg s.c.
Aufgrund ihrer metabolischen Wirkung können sie bei Zuständen, die zu einer durch psychischen Stress verursachten, unerwünschten Mobilisation von Fettsäuren und Glucose führen, eingesetzt werden.
Für die obige Anwendungen variieren die zu verwendenden Dosen naturgemäss je nach Art der verwendeten Substanz, der Verabreichung und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate bei Testtieren mit einer täglichen Dosis von ca. 0,01 mg/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden.
Die Tagesdosis liegt bei etwa 1 bis 200 mg. Für orale Applikationen enthalten die Teildosen etwa 0,25 bis etwa
100 mg der neuen Verbindungen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Ausserdem besitzen sie salidiuretische Wirkungen, wie sich in Tierversuchen, beispielsweise an der Ratte [Methode von E. Flückiger, Schweiz. med. Wochenschrift 93, (1963) 1232] mit Dosen von ca. 0,1 bis ca. 10 mg/kg p.o. zeigt.
Aufgrund dieser Wirkung können sie als Salidiuretika beispielsweise zur Behandlung von Oedema und Hypertonie verwendet werden.
Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Verabreichung und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ungefähr 0,1 bis ungefähr 10 ml/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 4 Anteilen verabreicht werden. Die Tagesdosis liegt bei 1 bis 50 mg. Für orale Applikationen enthalten die Teildosen etwa 0,25 bis etwa 20 mg der neuen Substanzen neben festen oder flüssigen Trägersubstanzen.
Bevorzugt sind das 8'-(3-tert.-Butylamino-2-hydroxy- propoxy)-spirotcyclopentan- 1,2'(1 H)-naphthalinj-4'(3'H)- on und seine physiologisch verträglichen Säureadditionssalze.
Die Verbindungen der Formel 1 bzw. ihre physiologisch verträglichen Säureadditionssalze können allein oder in geeigneter Dosierungsform verabreicht werden. Die Arzneiformen, z. B. eine Tablette, können analog zu bekannten Methoden hergestellt werden.

Insofar as the preparation of the required starting materials is not described, these are known or can be prepared and purified by known methods or analogously to the methods described here or in the examples or analogously to methods known per se.
In the following examples, which explain the invention in more detail, all temperatures are given in degrees Celsius without corrections.
Example 1 8 '- (3-tert-Butylamino-2-hydroxypropoxy) -spiro [cyclopentane-1,2' (1 'H) -naphthalene] -4' (3'H) -one
7.5 g of crude 8 '- (3-chloro-2-hydroxypropoxy) -spiro [cyclo? Entan-1,2' (1'H) -naphthalene] -4 '(3'E {) - one are in 50 ml of dioxane and 30 ml of tert-butylamine in an autoclave for 30 hours at 1300, boiled, concentrated and partitioned between 10% aqueous tartaric acid solution and ether. The aqueous part is made alkaline, extracted with ether, the trganic phases are concentrated and the remaining title compound is converted into its hydrogen maleate (mp. Of the hydrogen maleate of the title compound: 188-190 - from ethanol).
The starting material can be obtained as follows: a) To a solution of 2-methoxybenzylmagnesium chloride (prepared from 5.3 g of magnesium and 31.2 g of 2-methoxybenzyl chloride in 400 ml of ether) is a solution of 39.9 g of ethyl cyclopentylideasanate in 350 ml of tetrahydrofuran was added dropwise and the mixture was stirred at room temperature for 2 hours. The mixture is then mixed with 300 ml of 15% ammonium chloride solution, the product is extracted with ether and the ether solution is concentrated.
The crude ethyl a-cyano- (1 - (2-methoxybenzyl) cyclopentane} acetic acid is obtained, which is distilled at 140-180 / 0.002 mm (further processed raw).
b) 33.6 g of potassium hydroxide are added to a solution of 32.4 g of ethyl a-cyano- (l - (2-methoxy-benzyl) cyclopentane) in 300 ml of ethylene glycol and the solution is stirred at 1800 for 40 hours. The solution is cooled, poured onto ice-water and the neutral by-products are extracted with ether. The aqueous part is acidified with salt acid and extracted with ether. After concentration of the solvent, the [1 - (2-methoxybenzyl) cyclopentane] acetic acid (mp. 85-87 - from hexane) remains.
c) 5.7 g of [1 - (2-methoxybenzyl) cyclopentane] acetic acid are added to 30 g of polyphosphoric acid with stirring at 110 and the mixture is subsequently stirred for 10 minutes. The mixture is cooled and 250 ml of water are added. Extraction with ether and concentration of the ether extract gives the 8 'methoxy-spiro [cyclopentane-1,2' (1 'H) -naphthalene] -4' (3'H) one (mp 62-66) directly processed).
d) 5.0 g of 8'-methoxy-spiro [cyclopentane-1,2 '(I'H) -naphthaline] -4' (3'H) -one is mixed with 50 ml of acetic acid and 10 ml of 48% The hydrogen bromide solution was refluxed for 20 hours, then the solution was concentrated, diluted with water and extracted with ether. After removal of the solvent, the 8'-hydroxy-spiro [cyclopentane-1,2 '(1'H) -naphthalene] -4' (3'H) -one (mp. 163-165 - from toluene) remains .
e) 3 drops of piperidine are added to a solution of 4.6 g of 8'-hydroxy-spiro [cyclopentane-l, 2 '(1'H) -naphthalene] -4' (3'H) -one in 30 ml of epichlorohydrin added and the mixture stirred for 4 hours at 100 ". The solution is evaporated, taken up in ether, filtered and concentrated, the crude 8 '- (3-chloro-2-hydroxypropoxy) -spiro [cyclopentane-1,2' (1 'H) -naphthalene] -4' (3'H) -one remains.

The compounds of formula I, i.e. H. the compounds of formula I in free form or in the form of their physiologically tolerable acid addition salts are distinguished by interesting pharmacodynamic properties. They can therefore be used as a remedy.
They show an inhibition of the positive inotropic adrenaline effect on the spontaneously striking, isolated guinea pig atrium, this antagonistic effect occurring at bath concentrations of approx. 0.005 to approx. 2.5 ml / l.
On the anesthetized whole animal (anesthetized dog) they lead with an effective cumulative dose of approx. 0.02 to approx. 0.6 mg / kg IV. to strong inhibition of tachycardia caused by iso proterenol [1 - (3,4-dihydroxyphenyl) -2-2-isopropylaminoethanol] and lowering of blood pressure.
They therefore have a blocking effect on the adrenergic ss receptors and can therefore u. a. for the prophylaxis and therapy of coronary diseases, in particular for the treatment of angina pectoris.
Due to their antiarrhythmic effect, they are also suitable for the treatment of cardiac arrhythmias.
They also show interesting metabolic interactions by inhibiting the mobilization of glucose and fatty acid in the blood induced by psychological stress. They act on isolated fat cells of the epididymal adipose tissue of rats [in vitro method according to M. Rodbell, J.
Biol. Because of their metabolic effects, they can be used in conditions that lead to undesired mobilization of fatty acids and glucose caused by psychological stress.
For the above applications, the doses to be used naturally vary depending on the type of substance used, the administration and the condition to be treated. In general, however, satisfactory results are obtained in test animals with a daily dose of approximately 0.01 mg / kg body weight; if necessary, this dose can be administered in 2 to 4 portions or as a slow-release form.
The daily dose is around 1 to 200 mg. For oral applications, the partial doses contain approximately 0.25 to approximately
100 mg of the new compounds in addition to solid or liquid carriers or diluents.

These are preferably 8 '- (3-tert-butylamino-2-hydroxy-propoxy) -spirotcyclopentan- 1,2' (1 H) -naphthalinj-4 '(3'H) - one and its physiologically tolerated acid addition salts.
The compounds of formula 1 or their physiologically compatible acid addition salts can be administered alone or in a suitable dosage form. The dosage forms, e.g. B. a tablet can be prepared analogously to known methods.
