Als Reste R2 sind insbesondere solche bevorzugt, die sich von Alkanoylsäuren mit 1-18 Kohlenstoffatomen, vorzugsweise 2-8Kohlenstoffatomen ableiten, wies. B. monobasischeAlka- noylsäuren, wie die Ameisen-,Essig-, Propion-, Butter-, Isobutter-, a-Äthylbutter-, Pivalin-, Valerian-, Isovalerian-, a-Äthylvalerian-, Trimethylessig-, 2-Methylbutter-, 3-Äthylbutter-, Capron-, Triäthylessig-, Önanth- oder Caprylsäure, oder cyclische Säuren, vorzugsweise cycloaliphatische Säuren wie die Cyclopropylidenessig-, Cyclobutylcarbon-, Cyclopentylcarbon-, Cyclopentylessig-, ss-Cyclopentylpropion-, Cyclohexylcarbonoder Cyclohexylessigsäure oder auch carbocyclische Aryl- oder Aralkylsäuren wie die Benzoe, 2-,

Da der chemische Charakter der Acylgruppe für die Eigenschaften der erfindungsgemäss herstellbaren Verbindungen nicht kritisch ist, solange die Acylgruppe nicht toxisch wirkt oder die entsprechende Säure mit der primären Hydroxygruppe einen Ester bildet, sind auch andere aliphatische oder aromatische unsubstituierte und substituierte, mono-, di- und polybasische Carbonsäuren, gesättigte und ungesättigte aliphatische, araliphatische und aromatische Carbonsäuren mit bis zu 18 Kohlenstoffatomen, vorzugsweise bis zu 8 Kohlenstoffatomen, geeignet.
Genannt seien beispielsweise die Undecyl-, Dodecan-, Tetradecan-, Hexadecan-, Octadecan-, Palmitin-, Stearin- und (3- Cyclohexylpropionsäure, 2,3-. 2,4-, 2,6-, 3,4-und3,5-Dimethyl- benzoe-, Athylbenzoe-, Naphthoe-, 3-Methyl-a-naphthoe-, (3- Phenylpropion-, Diphenylessig- und & aphthylessigsäure oder zweibasische Alkanoylsäuren,wie z. B.
Oxal-, Malein-, Fumar-, Berastein-, Malon-, Glutar-, a-Methylglutar-, (3-Methylglutar-, ss,3- Dimethylglutar-, Adipin-, Pimelin- und Sebacinsäure, zweibasische aromatische Säuren, wie solche, die innere Anhydride bilden können, wie die Phthalsäure, Carbaminsäurenwie die Carbamin-, Phenylcarbamin-, n-Butylcarbamin-, Dimethylcarbamin-, Diäthylcarbamin- und Allophansäure oder heterocyclische Säuren, wie die (3-Furylcarbon-, Pyrrolcarbon-, ss-Pyrrolidi- nopropion-, N-Methylpyrrolidino-2-carbon-, 6-Hydroxy-indo- lyl-3-essig-, N-Methylmorpholino-2-carbon- und Pyrrol-2-carbonsäure oderSulfonsäuren mit 1-18 Kohlenstoffatomen,
vorzugsweise 1-12 Kohlenstoffatomen, wie Alkansalfonsäuren, beispielsweise Methan- und Äthansulfonsäure, und Arylsulfonsäuren, beispielsweise Benzol- und p-Toluolsulfonsäure.
Die in den erfindungsgemäss herstellbaren Verbindungen enthaltenen Acylreste können auch mit einem oder mehreren Substituenten substituiert sein.
Genannt seien als Substituenten beispielsweise folgende Reste: Hydroxy, Halo, Allroxy, Acyloxy, Sulfonyloxy, Amido, Sulfato, Nitro, Mercapto und Cyano, wie z. B. Reste der Glykol-, Milch-, Zitronen-, Wein-, Malein-, Glycerin-, Mannon-, Glucon- und Salicylsäure oder Reste von Aminosäuren wie Glycin, Aminopropion-, Diglykolamino- und Triglykolaminosäure, Methylglycin, Dimethylglycin, Diäthylglycin, p-Aminosalicyl-, p-Aminobenzoe-, Äthylmercaptoessig-, Benzylmercaptoessig-, Chloressig-, Fluoressig-, Trichloroessig-, Trifluoroessig-, Thioglycol-, m-Nitrobenzoe-, 2,3 ,4-Trimethoxybenzoe-, Phenoxyessig- und a-Naphthyloxyessigsäure.
Insbesondere geeignet sind vor allem die zweibasischen gesättigten und ungesättigten Carbonsäuren.
Die Salze leiten sich gewöhnlich von den entsprechenden Hemiacylaten dieserzweibasischen Säuren ab. Als Kation kommen insbesondere die Alkalimetalle Natrium und Kalium sowie Ammonium in Frage. Geeignet sind aber auch die zweiwertigen Erdalkalimetalle wie Calcium, wobei in der Regel auf ein Mol äquivalent Calcium dann 2 Moläquivalente Hemiacylat kommen.
Die erfindungsgemäss herstellbaren Verbindungen besitzen entweder selbst pharmakologisch wertvolle Eigenschaften oder
sie sind Zwischenprodukte zur Herstellung von anerkannt guten Arzneistoffen.
Die erfindungsgemäss erhältlichen Verbindungen sind u. a.
Diuretika vom Typ der Aldosteron-Antagonisten, d. h. sie keh- ren die Wirkung von Desoxycorticosteron auf die Natrium- und Kaliumausscheidung um.
Die Erfindung betrifft also ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I, das dadurch gekennzeichnet ist, dass man A4 6-3-Keto-steroide der allgemeinen Formel

mit Dimethylsufoxoniummethylid und Dimethylsulfoxid als Lösungsmittel methyleniert und erhaltene Verbindungen, in welchen R2 den Acylrest einer zweibasischen Säure bedeutet, gegebenenfalls in die entsprechenden Salze überführt.
Erhaltene Verbindungen, in welchen R2 ein Acylrest ist, können durch Verseifung in die entsprechenden Verbindungen überführt werden, worin R2 für Wasserstoff steht. Ist R2 Wasserstoff, so kann man die Ester in Gegenwart eines Veresterungskatalysators herstellen. Erhaltene Ester können umgeestert werden, indem man sie zuerst in entsprechende Hydroxylverbindungen überführt und dann mit der letztlich gewünschten Säure verestert.
Eine besondere Ausführungsform des erfindungsgemässen Verfahrens für den Fall, dass R2 den Rest einer zweibasischen Säure darstellt, besteht darin, dass das Hemiacylat in das Ammonium-, Alkali- oder Erdalkalimetallsalz überführt werden kann.
Bei Durchführung des crfindungsgemässen Verfahrens wird die Verbindung der Formel II mit Dimethylsulfoxoniummethylid methyleniert. Hierzu wird das A4 6-Steroid vorzugsweise unter Schutzgasatmosphäre, wie Stickstoff oder Argon, langsam entweder zu einer Suspension von Trimethylsulfoxoniumjodid mit Natriumhydrid in einem Mineralöl und Dimethylsulfoxid oder zu einer Lösung von Trimethylsulfoxoniumjodid und Natriumhydroxid in Dimethylsulfoxid zugegeben. Die Reaktion ist gewöhnlich bei 20-40"C nach 10-40 h beendet. Das Reaktionsprodukt kann extrahiert werden und durch Chromatographie kann die 6ss.6ss-Methylen- von der 6,7a-Methylenverbindung abgetrennt und gereinigt werden.
Zur Herstellung der neuen Verbindungen, in denen R2 einen Säurerest bedeutet, kann man entweder die bereits am B-Ring substituierten 17,-Ilydroxy-17a-(3-hydroxypropyl)-A4-3-keto- steroide oder die A4 6-3-Ketosteroide mit einem Alkansäureanhy drid umsetzen, wobei zuerst die primäre Hydroxygruppe verestert wird. Hierzu löst man zweckmässigerweise das Ausgangssteroid in einem basischen Lösungsmittel wie Pyridin, Piperidin.
Triäthylamin, Collidin oder Lutidin und gibt das entsprecllende Säureanhydrid hinzu. Günstig für die Veresterung ist aber auch der Zusatz eines Veresterungskatalysators wie Dimethylaminopyridin.
Die primäre Hydroxygruppe ist gewöhnlich bereits nach kurzer Zeit, d. h. nach 2-5 h, verestert. Die Reaktion kann natürlich durch Erwärmen, z. B. durch Erwärmen bis zur Siedehitze, beschleunigt werden.
Die Verseifung erfindungsgemäss erhaltener Verbindungen, in denen R2 Acyl ist, wird zweckmässigerweise unter milden Bedingungen, wie mit methanolischer Kalilauge in der Kälte, ausgeführt.
Falls z. Beine primäre 22-Hydroxygruppe mit einer zweibasischen Säure verestert wurde, kann das Hemiacylat in das gewünschte Alkalisalz durch Umsetzung beispielsweise mit einer methanolischen Kalium- oder Natriummethylat-Lösung erreicht werden. Zur Herstellung des Ammoniumsalzes verwendet man zweckmässigerweise eine Lösung von Ammoniak in Methanol.
Die erfindungsgemäss erhaltenen Reaktionsprodukte können nach an sich bekannten Methoden, wie Fällung, Filtration oder Extraktion, abgetrennt und beispielsweise durch Chromatographie und/oder Umkristallisation gereinigt werden.
Das als Ausgangsmaterial verwendete A4 6-ungesättigte 3- Keto-steroid kann beispielsweise so erhalten werden, dass man zuerst nach dem Verfahren derDT-OS 2 327 448 aus 3-Keto4- androsten-17-on das 17ss-Hydroxy-17 -(3-hydropropyl)-4-andro- sten herstellt und anschliessend die 86-Doppelbindungbeispiels- weise nach dem von Agnello beschriebenen Verfahren [Agnello et al., J. Amer. Soc. 82 (1960) 4293] einführt.
Ein bevorzugtes Ausgangsmaterial der Formel II kann wie folgt hergestellt werden:
Eine'Suspension von 6,85 g Kalium-tert.-butylat in 32 ml absolutem Tetrahydrofnran wird mit 50 g3-Methoxy-18-methyl- 1,3,5(10)-östratrien-17-on [C. Ruferu. a., Liebigs Ann. Chem.
752,1(1971)] versetzt und eine Lösung von 1,7 ml Propargylalkohol in 3,5 ml Tetrahydrofuran zugetropft, so dass die Innentem peratur 35 C nicht übersteigt. Man rührt 3 h bei 35" C nach und säuert dann das Reaktionsgemisch mit 13 ml 20%iger Scllwefel- säure an, bis pH - 3 erreicht ist und rührt 10 min unter Rückfluss nach. Das Reaktionsgemischwird dann in Eiswasser gegeben und der Niederschlag abfiltriert. Man nimmt ein Essigester auf, wäscht mit Wasser, trocknet und dampft ein. Man erhält 5,5 g 17ss- Hydroxy-17a-(3-hydroxypropinyl)-3-methoxy-18-methyl 1 ,3,5(10)-östratrien.
Man schüttelt eine Lösung von 5 g 1 7ss-Hydroxy-17a-(3-llydso- xypropinyl)-3-methoxy-18-methyl-l ,5( 10).östratrien in 50 ml Tetrahydrofuran mit 500 mg Pd/CaCo3 (5%mg) unter Wasserstoff bei Raumtemperatur und Normaldruck bis zur Beendigung der Wasserstoffaufnahme. Dann wird vom Katalysator abfiltriert und das Filtrat eingedampft. Man erhält 4,8 g 17ss-Hydroxy-17a- (3-hydroxypropyl)-3-methoxy-18-methyl-1 ,3 ,5(10)-östratrien.
Eine Lösung von 5 g 17ss-Hydroxy-17a-(3-hydroxypropyl)-3- methoxy-18-methyl-1,3,5(10)-östratrien in 200 ml absolutem Tetrahydrofuran mischl man mit 450 ml flüssigem Ammoniak bei -600C und fügt anschliessend 5 g Lithium in kleinen Stücken zu.
Die blaue Lösung wird 2,5 h bei -60 Cgerührtund dann durch Zutropfen von Äthanol bis zur Entfärbung zersetzt. Anschliessend lässt man das Ammoniak abdampfen, nimmt den Rückstand in Äther auf, wäscht mit einer Natriumchlorid-Lösung neutral, trocknet und dampft ein. Der Rückstand wird in 150 ml Methanol und 65 ml Methylenchlorid gelöst und mit 15 ml 3n Salzsäure 1 h zum Sieden erhitzt. Nach Spaltung des Enoläthers wird die Lösung eingeengt, der Rückstand in Methylenchlorid aufgenommen und die Lösung mit Natriumbicarbonatlösung neutral gewaschen, getrocknet und eingedampft. Man erhält 2 g 1 7(3-hydroxy-17 -(3-hydroxypropyl)-1 8-methyl-4-östren-3-on.
Eine Lösung von 2 g 17a-(3-hydroxypropyl)-18-methyl-17ss- hydroxy-4-östren-3-on 1,35 g Chloranil und 0,03 g p-Toluolsul- fonsäure in 200 ml Xylol wird 1 h zum Sieden erhitzt. Danach wird im Vakuum eingedampft und der Rückstand durch Gra dienten-Chrom atographie über SiO2 gereinigt. Man erhält 140 mg 17ss-Hydroxy-17a-(3-hydroxypropyl)-18-methyl4,6-östra- dien-3-on als amorphe Substanz. UV: e2ts5 = 22 000 (Methanol).
Die Ausgangsverbindung 17P-Hydroxy-l 7a-(3-hydroxypro- pyl)-4,6-östradien-3-on kann wie folgt hergestellt werden:
In900 ml flüssiges Ammoniak gibt man bei 600 C eine Lösung von 10 g 17a-(3-Hydroxypropyl)-3-methoxy-l ,3,5(10)- östratrien-1 7ss-ol [G. E. Arth u. a. ;J. Med. Chem. 6, 616 (1963)] in 400 ml absolutem Tetrahydrofuran. Anschliessend werden 10 g Lithium in kleinen Stücken zugegeben. Dann rührt man 2,5 h bei -60"C, worauf man durch langsame Zugabe von Äthanol die Lösung entfärbt und unter Rühren Ammoniak abdampfen lässt.
Den Rückstand nimmt man in Äther auf, wäscht mit einer Natriumchloridlösung neutral, trocknet und dampft ein. Der Rückstand wird in 300 ml Methanol und 130 ml Methylenchlorid gelöst und mit 30 ml 3n Salzsäure 1 h zum Sieden erhitzt. Nach Einengen der Lösung wird in Methylenchlorid aufgenommen, mit Natriumbicarbonatlösung und Wasser neutral gewaschen, getrocknet und eingedampft. Man erhält 7,1 g 17a-(3-Hydroxy propyl)-17ss-hydroxy-4-östren-3-on vom Schmelzpunkt 166167 C (lsopropyläther/Methylenchlorid).
UV: E241 = 17 100 (Methanol).
Eine Lösung von 4,0 g 17a-(3-Hydroxypropyl)-17ss-hydroy-4- östren-3-on, 3.3 g Chloranil und 0,05 g p-Toluolsulfonsäure in 400 ml Xylol wird 1 h zum Sieden erhitzt. Danach wird im Vakuum eingedampft und der Rückstand durch Gradientenchromatographie über Silicagel gereinigt. Man erhält 350 mg 17ss- Hydroxy-17a-(3-hydroxypropyl)-4,6-östradien-3-on vom Schmelzpunkt 193 - 194,50C (Aceton/Hexan).
UV: E2R4 = 26 900 (Methanol).
Die pharmakologisch wirksamen, erfindungsgemäss erhaltenen Verbindungen der allgemeinen Formeln können nach an sich bekannten Methoden der Galenik zu Arzneimitteln, insbesondere zur oralen Applikation, verwendet werden.
Die Dosierung der genannten neuen Verbindung liegt beim Menschen gewöhnlich bei 20-500 mg/Tag.
Die nachfolgenden Beispiele sollen das erfindungsgemässe Verfahren erläutern.
Beispiel 1
1,8 g Trimethylsulfoxoniumjodid und 2,8 g Natriumhydroxid werden in 32 ml Dimethylsulfoxid 15 min unter Argonbegasung gerührt. Anschliessend wird 1 g 17ss-Hydroxy-17a-(3-hydroxypro pyl)-4,6-androstadien-3-on eingetragen und 24 h bei 35 C weiter unter Argon gerührt . Man giesst in essigsaures Eiswasser ein, saugt das gefällte Produkt ab und löst in Methylenchlorid. Der Methylenchlorid-Extrakt wird mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum eingedampft. Der Rückstand wird durch Schichtchramatographie im System Chloroform-Methanol (9:1) gereinigt. Nach Umkristallisation aus Aceton erhält man 530 mg 17-Hydroxy-17a-(3-hydroxypropyl)- 6ss .7ss-methylen-4-androsten-3-on .
UV263m = 18 300, F. 170 bis 172"C.
Beispiel 2
Zu einer Lösung von 3 g (13,8 mMol) Trimethylsulfoxoniumjodid in 60 ml absolutem Dimethylsulfoxid gibt man 330 mg (13,8 mMol) Natriumhydrid als 50%ige Mineralölsuspension. Man rührt die Lösung bei Raumtemperatur unter Stickstoff, bis die Wasserstoffentwicklung beendet ist. Danach werden 3 g (0,87 mMol) 17ss-Hydroxy-17a-(3-hydroxypropyl)-18-methyl-4,6-östrE dien-3-on zugegeben und solange bei Raumtemperatur unter Stickstoff gerührt, bis das Ausgangsmaterial durch Dünnschichtchromatographie nicht mehr nachweisbar ist. Zur Aufarbeitung gibt man das Reaktionsgemisch in essigsaures Eiswasser und extrahiert mit Essigester. Nach Abdampfen des Lösungsmittels wird der Rückstand durch Gradientenchromatographie an Silicagel gereinigt.
Man eluiert als erstes 790 mg 17P-Hydroxy-17a-(3- hydroxyprnpyl)-l8.methyl-6a.7 -methylen-4.östren-3-on und danach 180 mg 17ss-Hydroxy-17α-(3-hydroxypropyl)-18-methyl- 63.7ss-methylen-4-östren-3-on.
Beispiel 3
Zu einer Lösung von 800 mg 17ss-Hydroxy-17a-(3-hydroxypro- pyl)-18-methyl-6ss.7ss-methylen-4-östren-3-on in 5 ml Pyridin gibt man 400 mg Acetoxyessigsäurechlorid und lässt 20 h bei Raumtemperatur stehen. Dann gibt man das Reaktionsgemisch in Eiswasser und filtriert den Niederschlag ab. Der Rückstand wird in Methylenchlorid gelöst, gewaschen, getrocknet und eingedampft. Man erhält 730 mg 17ss-Hydroxy-17a-63-acetoxyacetyl- oxypropyl)-18-methyl-6ss.7ss-methylen-4-östren-3-on.
Beispiel 4
Man löst 25 mg 17ss-Hydroxy-17a-(3-hydroxypropyl)-18- methyl-6(3.7(3-methylen-4-östren-3-onin2 ml Pyridin, gibt 1 ml Essigsäureanhydrid zu und lässt 20 h bei Raumtemperatur stehen. Danach wird derAnsatz in Eiswasser gefällt, die Substanz mit Methylenchlorid aufgenommen, die Lösung gewaschen, getrocknet und eingedampft. Man erhält 17ss-Hydroxy-17a-(3- acetoxypropyl)-18-methyl-6ss .7ss-methylen-4-östren-3-on.
Beispiel 5
Zu einer Lösung von 2 g (9,2 mMol) Trimethylsulfoxoniumjodid in 40 ml absolutem Dimethylsulfoxid gibt man 220 mg (9,2 mMol) Natriumhydrid als 50%ige Mineralölsuspension. Man rührt die Lösung bei Raumtemperatur unter Stickstoff, bis die Wasserstoffentwicklung beendet ist. Danach gibt man 2 g (7 mMol) 17ss-Hydroxy-17a-(3-hydroxypropyl)-4,6-östradien-3-on zu und rührt bei Raumtemperatur unter Stickstoff, bis das Ausgangsmaterial durch Dünnschichtchromatographie nicht mehr nachweisbar ist. Zur Aufarbeitung wird der Ansatz in essigsaures Eiswasser gegeben und mit Essigester extrahiert.
Nach Abdampfen des Lösungsmittels wird der Rückstand durch Gradientenchromatographie an Silicagel gereinigt. Man isoliert zunächst 850 mg 17ss-Hydroxy-17a-(3-hydroypropyl)-6a.7a-methy- len-4-östren-3-on, danach das gewünschte 17ss-Hydroxy-17(x-(3- hydroxy-propyl)-6(3.7(3-methylen-4-östren-3-on (190 mg).
Beispiel 6
Zu einer Lösung von 850 mg 17ss-Hydroxy-17a-(3-hydroxypro- pyl)-6ss.7ss-methylen-4-östren-3-on in 5 ml Pyridin gibt man 450 mg Acetoxyessigsäurechlorid und lässt 20 h bei Raumtemperatur stehen. Dann gibt man das Reaktionsgemisch in Eiswasser und filtriert den Niederschlag ab. Der Niederschlag wird in Methylenchlorid gelöst, gewaschen, getrocknet und eingedampft. Man erhält 530 mg 17ss-Hydroxy-17a-(3-acetoxyacetyloxypropyl)-6ss.7ss- methylen-4-östren-3-on.
Beispiel 7
Man löst 500 mg 17ss-Hydroxy-17a-(3-hydroxypropyl)-6ss.7ss- methylen-4-östren-3-on in 2 ml Pyridin, gibt 1 ml Essigsäureanhydrid zu und lässt 20 h bei Raumtemperatur stehen. Danach wird das Reaktionsgemisch in Eiswasser gefällt, die Substanz mit Methylenchlorid aufgenommen, die Lösung gewaschen, getrocknet und eingedampft. Man erhält 17ss-Hydroxy-17a-(3-acetoxy- propyl)-6ss.7ss-methylen-4-östren-3-on.
Beispiel 8
150 mg 17,8-Hydroxy-17a-(3-hydroxypropyl)-6,B.7,8-methylen-4- androsten-3-on werden in 2 mol Pyridin und 1 ml Acetanhydrid unter Argonbegasung 8 h unter Rückfluss erhitzt. Es wird mit Eiswasser versetzt, mit Methylenchlorid extrahiert, die Lösung mit 1 n Schwefelsäure und Wasser gewaschen und im Vakuum eingedampft. Nach Reinigung des Rückstandes durch Schichtchromatographie erhält man 17ss-Acetoxy-17a-(3-acetoxypropyl' 6B.7B-methylen-4-androsten-3-on.

27 g Trimethylsulfoxoniumjodid und 4,6 g 55%ige Natriumhydrid-Dispersion in Öl werden in 500 ml Dimethylsulfoxid 1,5 h unter Argonbegasung gerührt. Danach werden 55 g 17ss-Hydroxy 17α-(3-triphenylmethyloxypropyl)-4,6-androstadien-3-on eingetragen und 24 h bei Raumtemperatur gerührt. Anschliessend wird mit Wasser versetzt, mit Essigester extrahiert und die abgetrennte Essigesterlösung mit Wasser gewaschen und eingedampft. Der Rückstand wird an Silicagel chromatographiert.
Durch Elution mit Hexan-Essigester werden 26,5 g 6,7-Methylen-Addukt erhalten, das sich aus 70 % ss-Methylen- und 30 % cr- Methylenverbindung zusammensetzt.
Zur Ätherspaltung werden 26,5 g der 6,7-Methylenverbindung in 170 ml 80%iger Essigsäure gelöst und 15 min auf dem Dampfbad erhitzt. Die Lösung wird darauf in Eiswasser gegossen, das ausgefallene Produkt abgesaugt, getrocknet und aus Aceton/Hexan umkristallisiert. Man erhält 10 g 17ss-Hydroxy -17a (3-hydroxypropyl)-6ss.7ss-methylen-4-androsten-3-on vom F. 182 bis 1840C.
