Description
The present invention relates to a process for the preparation of cellulosic fibers incorporating enzymes.
It is known that many reactions, which otherwise could not be carried out or would require more drastic operating conditions, can instead be carried out when they are catalyzed by enzymes.
It is also known the use of enzymes incorporated in fibers which perform their catalytic activity without dispersing in the reaction mass.
The incorporation from the enzyme is achieved by using a fiber of artificial or synthetic polymeric material.
The enzyme incorporating fibers can be prepared from polymer solutions capable of giving fibers in which enzymatic preparations are dispersed in the form of minute droplets of the order of magnitude of the emulsions.
The emulsion thus obtained can be spun wet or dry to give a fiber which has very small cavities inside which the enzymes are disposed which are separated from the environment by a very thin membrane. The enzyme incorporating fibers hitherto used, and which form the subject of Italian patent no. 836 462 in the name of the same applicant, are obtained from polymers capable of giving fibers such as cellulose derivatives, but it is not known that they can be prepared directly from unregenerated cellulose due to the difficulties that the latter presents when subjected to subsequent spinning processes.
The use of cellulosic fibers incorporating enzymes has the advantage over other types of fibers of greater activity than enzymatic reactions, due to the increase in permeability of the fiber itself. The catalytic action of the enzymes contained in the fibers is in fact conditioned by the diffusion processes whereby the enzymatic activity is a function of permeability.
Enzyme incorporating fibers with high permeability are not always obtained by simply operating on the fiber preparation conditions, since it has been noted that, when the concentration of the enzyme is high, permeability is still a limiting factor.
It has now been found, what constitutes an object of the present invention, that it is possible to prepare cellulosic fibers incorporating enzymes starting from cellulosic polymers containing substituents which are suitably emulsified with the enzymatic solutions, then spun and finally the fibers thus obtained are subjected to reactions which cause the removal of the substituent, thus generating an unsubstituted cellulosic fiber incorporating the enzyme which has permeability far superior to that of the substituted cellulose fiber itself.
The process for the preparation of cellulosic fibers incorporating enzymes is defined in claim 1.
The process of the present invention is therefore essentially based on a chemical modification of the starting fibrogen material due to the action of reducing agents and operating conditions such as not to damage the enzyme.
The process of the present invention allows to overcome all the aforementioned drawbacks and the enzyme incorporating fibers, obtained after the reduction process and which in turn constitute a second object of the invention defined in claim 7, all have a high degree of permeability.
The starting material used consists essentially of high molecular weight nitrocellulose and a nitrogen content such as to make the material substantially soluble in organic solvents and preferably higher than 5%.
It is dissolved in a solvent which is not soluble in water selected from normalbutylacetate, dibutylphthalate, methylamylketone, ethylamylketone and others, which can be used as such or suitably diluted with aliphatic hydrocarbons such as pentane, hexane, heptane, octane, aromatic hydrocarbons such as toluene. and xylene, or mixtures of hydrocarbons such as petroleum ether. To the nitrocellulose solution is added the aqueous solution containing the enzyme which can be chosen by a wide range of names. For example urease, invertase, lactase, acylase, transaminase, glucose oxidase, catalase, papain, penicillin, acylase and others can be used. The emulsion is then spun according to known techniques using known coagulants such as, for example, those reported in the aforementioned Italian patent. In addition to the emulsion dispersion process of the enzyme in the polymer solution, other processes may be provided for preparing fibers containing the enzyme incorporated. For example said enzyme can be dispersed as a powder in the polymer which will then be spun according to known methods. The fiber thus obtained is reacted with a reducing agent which eliminates the nitrogroup thus giving rise to the cellulosic fiber incorporating the enzyme object of the present invention.
Come agenti riducente vengono impiegate soluzioni di solfidrato ammonico contenenti o no lievi eccessi di ammoniaca o idrogeno solforato. As reducing agents, ammonium sulfhydrate solutions are used, containing or not slight excesses of ammonia or hydrogen sulphide.
Further operational details will be more evident from the examination of the examples given below.

attraverso fori del diametro di 125 (a, utilizzando come coagulante una miscela di idrocarburi saturi aventi punti di ebollizione compresi tra 40 e 70° C. through holes with a diameter of 125 (a, using as a coagulant a mixture of saturated hydrocarbons having boiling points between 40 and 70 ° C.
About 2,600 g of fiber are obtained having the enzyme incorporated.
1,000 g of this fiber immersed in a 20% sucrose solution at pH 4.5 invert 40 g of sugar per minute.
After 6 hours of reaction this second fiber is washed and when it is immersed in the 20% sucrose solution at pH 4.5 it inverts 490 g of sugar per minute.
Esempio 2 Example 2
1,000 g of nitrocellulose are dissolved in a mixture consisting of 7,000 g of normal-butyl acetate and 3,750 g of toluene at room temperature. Once the solution is obtained, 2,000 g of an aqueous solution of the enzyme invented are added.
By stirring an emulsion is obtained which is spun at 1 ° C through holes with a diameter of 125 [a and coagulated at 20 ° C in a mixture of saturated hydrocarbons having the boiling points between 60 and 80 ° C. Approximately 3,000 g are obtained of fiber.
1,000 g of the latter fiber immersed in a 20% sucrose solution at pH 4.5 convert 505 g of sugar per minute.
Another 1,000 g of fiber are treated for 6 hours with an ammonia solution saturated with hydrogen sulfide as described in Example 1. After the treatment, this second portion of fiber, immersed in a sucrose solution equal to that of the first portion, convert 1,890 g. of sugar per minute.
Esempio 3 Example 3
1,000 g of nitrocellulose are dissolved in a mixture consisting of 7,000 g of normal-butyl acetate and 3,750 g of toluene.
2,000 g of an aqueous solution of the penicillin-amidase enzyme are then added and the mixture is stirred to obtain an emulsion.
The emulsion is spun as described in Example 1 to obtain about 3,000 g of fiber.
1.500 g di fibra vengono immersi in 37,5 litri di una soluzione di fosfato di potassio 10-i molare a pH 6.0 contenente 1.500 g di penicillina G potassica a 37° C. 1,500 g of fiber is immersed in 37.5 liters of a 10-i molar potassium phosphate solution at pH 6.0 containing 1,500 g of potassium penicillin G at 37 ° C.
The enzyme contained in the fiber catalyses hydrolysis (iella penicillin with consequent formation of 6-aminopene-cillanic acid and phenylacetic acid.
During the reaction the pH is kept constant with the addition of soda.
In this way, 90% of penicillin is converted after 227 minutes.
2.000 g di una soluzione acquosa dell'enzima penicillina-amidasi vengono inglobati in 1.000 g di nitrocellulosa analogamente a quanto descritto sopra e quindi trattati nella soluzione ammoniacale saturata con idrogeno solforato come descritto nell'esempio 1. 2,000 g of an aqueous solution of the penicillin-amidase enzyme are incorporated in 1,000 g of nitrocellulose in the same way as described above and then treated in the ammonia solution saturated with hydrogen sulphide as described in example 1.
An identical penicillin-amidase solution is dispersed in a solution obtained by dissolving 1,000 g of cellulose triacetate in 13,300 g of methylene chloride.
This emulsion is spun as described in Example 1 using toluene as a coagulant.
1,000 g of the first fiber, immersed at 37 ° C in 25 liters of a pH 8.0 molar potassium IO-1 phosphate solution containing 10% potassium penicillin G, hydrolyze 90% of the penicillin in 219 minutes.
The second fiber under the same conditions hydrolyzes 90% of penicillin in 298 minutes.
Another 1,500 g of fiber are treated with the hydrogen sulfide-saturated ammonia solution as described in Example 1. Then under the same conditions as the untreated fiber it causes the conversion of 90% of penicillin in 56 minutes.
It is obvious that what has been said for fibers could be extended to films, microcapsules or similar products containing enzymes.
Only the great interest of the applicant in the specific field of fibers has pushed the same not to extend beyond the exemplification, but it will be simple for the expert of the art to apply the principles of the invention to other materials without however leaving the scope of the same .
Similarly, it will be easy for anyone familiar with the field to go on to treat starting polymeric materials other than nitrocellulose, to which we have particularly referred, in order to better exemplify the invention itself.
