Die Chlorierungs- bzw. Bromierungsreaktion wird vorzugsweise in einem Lösungsmittel, zum Beispiel Tetrachlorkohlenstoff, Chloroform, Methylenchlorid, Dioxan, Tetrahydrofuran, Dimethylformamid oder einem geeigneten Kohlenwasserstoff, gegebenenfalls unter Zusatz einer tertiären organischen Base, wie Pyridin, oder aber mit Hilfe von Chlor- oder Bromsuccinimid durchgeführt. Die Reaktionstemperatur liegt bei dieser Umsetzung je nach dem eingesetzten Ausgangsstoff und der angewandten Methode zwischen Raumtemperatur und der Rückflusstemperatur des Reaktionsgemisches.
Soll die so erhaltene Chlor- bzw. Bromverbindung in die entsprechende Alkoxyverbindung übergeführt werden, so löst man sie zweckmässigerweise in einem Alkalialkoholat und erhitzt die Lösung unter Rückfluss.
Zur Überführung in die Alkylmercaptoverbindung wird die Chlor- bzw. Bromverbindung zweckmässigerweise in einem aprotischen Lösungsmittel, wie Dioxan, Tetrahydrofuran, Dimethylformamid oder Hexamethylphosphortriamid (HMPT), oder in einem Alkohol gelöst und mit einem Alkalimercaptid
umgesetzt. Das Alkalimercaptid kann bei dieser Umsetzung auch in situ erzeugt werden, beispielsweise durch Einleiten eines Mercaptans in die ein Alkalialkoholat enthaltende Lösung der Chlor- bzw. Bromverbindung.
Die Verbindungen der Formel I können in üblicher Weise in ihre physiologisch unbedenklichen Säureadditionssalze überführt werden. Zur Salzbildung geeignete Säuren sind beispielsweise Halogenwasserstoffsäuren, Schwefelsäure, Phosphorsäure, Salpetersäure, Cyclohexylsulfaminsäure, Zitronensäure, Weinsäure, Ascorbinsäure, Maleinsäure, Ameisensäure, Salicylsäure oder Methan- oder Toluolsulfonsäure.
Die Ausgangsverbindungen der Formel VI können nach dem in der DE-AS 2 410 030 beschriebenen Verfahren hergestellt werden. Dabei wird eine Verbindung der Formel II

worin X eine NH2-NHGruppe zu Gruppe bedeutet, mit einem Ortho- ameisensäureester umgesetzt.
Die Verbindungen der Formel I bzw. deren Säureadditionssalze weisen wertvolle therapeutische Eigenschaften auf. Sie haben sich bei der Anwendung verschiedener pharmakologischer Testmethoden als anxiolytisch, spannungslösend und muskelrelaxierend wirksam erwiesen und überdies eine intensive antikonvulsive Wirkung gezeigt. Sie haben auch die Eigenschaft, die Futteraufnahme bei Säugetieren beträchtlich zu verstärken. Bemerkenswert ist auch ihre ausserordentlich geringe Toxizität.
Sie sind bekannten Thieno-1,4-diazepinen ohne zusätzlichen Triazolring, wie sie beispielsweise aus den DE-OS 2 155 403 und 2 221 623 bekannt sind, insbesondere durch ihre stark ausgeprägte, im sogenannten Pentetrazol-Test nachweisbare antikonvulsive Wirkung überlegen, während sie den aus der DE-OS 2 229 845 bekannten 8-Alkyl-6-aryl-thieno [2,3e]-4H-s-triazolo-[3,4c]-1,4-diazepinen zwar im Wirkungsbild ähneln, ihnen aber hinsichtlich der Wirkungsstärke um mehr als eine Zehnerpotenz überlegen sind.
Als besonders wertvoll haben sich dabei solche Verbindungen bzw. deren Säureadditionssalze erwiesen, bei denen R1 ein Bromatom, R2 ein Chlor- oder Bromatom und R3 ein Bromatom oder eine Methoxygruppe bedeutet, insbesondere das 1,8-Dibrom-6-o chlorphenyl-4H-s-triazolo-[3,4c]-thieno-[2,3e]- 1 P-diazepin (R1 = Br,R2 = Cl,R3 = Br)unddas8-Brom-6-o-chlorphenyl-1- methoxy-4H-s-triazolo-[3 ,4c]-thieno-[2,3 e]- 1 ,4-diazepin (R, = Br, R2 = Cl, R3 = OCH3) und seine Säureadditionssalze.
Die Einzeldosis der erfindungsgemäss erhältlichen Verbindungen liegt bei 0,05 bis 50, vorzugsweise 0,1 bis 25 mg (oral) und 5 bis 150 mg als Tagesdosis.
Die erfindungsgemäss erhältlichen Verbindungen können allein oder in Kombination mit anderen erfindungsgemäss erhältlichen Wirkstoffen, gegebenenfalls auch in Kombination mit weiteren pharmakologisch aktiven Wirkstoffen wie Spasmolytika oder ss-Rezeptorenblocker zur Anwendung gelangen.
Geeignete Anwendungsformen sind beispielsweise Tabletten, Kapseln, Zäpfchen, Lösungen, Säfte, Emulsionen oder dispersible Pulver. Entsprechende Tabletten können beispielsweise durch Mischen des oder der Wirkstoffe mit bekannten Hilfsstoffen, beispielsweise inerten Verdünnungsmitteln, wie Calciumcarbonat, Calciumphosphat oder Milchzucker, Sprengmitteln, wie Maisstärke oder Alginsäure, Bindemitteln, wie Stärke oder Gelatine, Schmiermitteln, wie Magnesiumstearat oder Talk, und/oder Mitteln zur Erzielung des Depoteffektes, wie Carboxypolymethylen, Carboxymethylcellulose, Celluloseacetatphthalat, oder Polyvinylacetat erhalten werden. Die Tabletten können auch aus mehreren Schichten bestehen.
Entsprechend können Dragees durch Überziehen von analog den Tabletten hergestellten Kernen mit üblicherweise in Dragéeüberzügen verwendeten Mitteln, beispielsweise Kollidon oder Schellack, Gummi arabicum, Talk, Titandioxid oder Zukker, hergestellt werden. Zur Erzielung eines Depoteffektes oder zur Vermeidung von Inkompatibilitäten kann der Kern auch aus mehreren Schichten bestehen. Desgleichen kann auch die Dragéehülle zur Erzielung eines Depoteffektes aus mehreren Schichten bestehen, wobei die oben bei den Tabletten erwähnten Hilfsstoffe verwendet werden können.
Aus den erfindungsgemäss erhältlichen Wirkstoffen bzw.
Wirkstoffkombinationen hergestellte Säfte können zusätzlich noch ein Süssungsmittel, wie Saccharin, Cyclamat, Glycerin oder Zucker sowie ein geschmacksverbesserndes Mittel, z. B.
Aromastoffe, wie Vanillin oder Orangenextrakt, enthalten. Sie können ausserdem Suspendierhilfsstoffe oder Dickungsmittel, wie Natriumcarboxymethylcellulose, Netzmittel, beispielsweise Kondensationsprodukte von Fettalkoholen mit Äthylenoxid, oder Schutzstoffe, wie p-Hydroxybenzoate, enthalten.
Injektionslösungen werden in üblicher Weise, z. B. unter Zusatz von Konservierungsmitteln, wie p-Hydroxybenzoaten, oder Stabilisatoren, wie Alkalisalzen oder Äthylendiamintetraessigsäure hergestellt und in Injektionsflaschen oder Ampullen abgefüllt.
Die eine oder mehrere Wirkstoffe bzw. Wirkstoffkombinationen enthaltenden Kapseln können beispielsweise hergestellt werden, indem man die Wirkstoffe mit inerten Trägern, wie Milchzucker oder Sorbit, mischt und in Gelatinekapseln einkapselt.
Geeignete Zäpfchen lassen sich beispielsweise durch Vermischen mit dafür vorgesehenen Trägermitteln, wie Neutralfetten oder Polyäthylenglykol bzw. dessen Derivaten, herstellen.
Beispiel 1 1,8-Dibrom-6-(o-chlorphenyl)-4H-s-triazolo-[3,4c]- thieno-[2,3e]-1,4-diazepin a) 10 g 8-Brom-6-(o-chlorphenyl)-4H-s-triazolo-[3,4c]- thieno-[2,3e]-1,4-diazepin werden in einer Mischung aus 20 ml Pyridin und 100 ml Methylenchlorid gelöst und 7 Stunden erhitzt. Darauf werden innerhalb von 5 Minuten 6,3 g Brom, gelöst in 25 ml Methylenchlorid, zugegeben und weitere 3 Stunden unter Rückfluss erhitzt. Anschliessend wird das Reaktionsgemisch abgekühlt, mit Methylenchlorid verdünnt und die Lösung zweimal mit 1n Salzsäure und einmal mit Wasser ausgeschüttelt. Nach dem Trocknen wird die Methylenchloridlösung eingedampft und der Rückstand aus Äthanol umkristallisiert.
Man erhält 7,0 g = 60% der Titelverbindung vom Fp. 210 bis 211"C.
b) Die Ausgangsverbindung wurde auf folgendem Weg erhalten: 27 g 7-Brom-5-(o-chlorphenyl)-2-hydrazino-3H-[2,3 ei- thieno-1,4-diazepin vom Fp. - 300 C (Zers.) werden in eine Mischung von 23 ml Orthoameisensäureester und 300 ml Äthanol 30 Minuten unter Rückfluss gekocht. Man verdampft das Lösungsmittel und verreibt den Rückstand mit Äther.
Ausbeute: 26 g vom Fp. 214 bis 216 C.
Beispiel 2
8-Brom-6-(o-chlorphenyl)-1-methoxy-4H-s-triazolo [3,4c]-thieno-[2,3e]-1,4-diazepin
12 g 1,8-Dibrom-6-(o-chlorphenyl)-4H-s-triazolo-[3,4c]- thieno-[2,3e]-1,4-diazepin werden in einer Lösung von 0,6 g Natrium in 70 ml Methanol 2 Stunden unter Rückiluss erhitzt.
Anschliessend wird das Reaktionsgemisch eingedampft, mit Wasser verdünnt und wiederholt mit Methylenchlorid ausgeschüttelt. Die Methylenchloridphase wird getrocknet und das Methylenchlorid verdampft. Aus dem Rückstand erhält man nach Umkristallisation mit Äthanol 7 g = 76% d. Th. der Titelverbindung vom Fp. 198 bis 200 C.
Beispiel 3 8-Brom-6-(o-chlorphenyl)-1-methylmercapto-4H-s-triazolo- [3,4c]-thieno-[2,3e]-1,4-diazepin
0,15 g Natrium werden in 40 ml absolutem Methanol gelöst und in die Lösung 340 mg Methylmercaptan eingeleitet.
Danach wird die Lösung mit 2,5 g 1,8-Dibrom-6-(o-chlor phenyl)-4H-s-triazolo-[3,4cj-thieno-[2,3e]-1,4-diazepin versetzt und auf eine Temperatur zwischen 40 und 50 C erwärmt. Nach ca. 30 Minuten kann dünnschichtchromatographisch keine Dibromverbindung mehr nachgewiesen werden.
Die Lösung wird sodann eingedampft, der Rückstand in Methylenchlorid aufgenommen, das Methylenchlorid mehrmals mit Wasser ausgeschüttelt, die organische Base getrocknet, das Lösungsmittel verdampft und der Rückstand über eine SiO2-Säule chromatographiert.
Man erhält 920 mg = 40% derTitelverbindung vom Fp. 158 bis 160 C.
