La présente invention se rapporte au domaine des arômes et, plus précisément, elle a pour objet une composition aromatisante destinée à améliorer ou modifier le goût des aliments en leur conférant l'arôme du fromage. Ladite composition aromatisante est caractérisée par le fait qu'elle contient, en tant qu'ingrédient actif, un dipeptide cyclique de formule:

ce composé étant également défini comme la cyclo-(Asn-Phe).
L'industrie des arômes s'est attachée depuis fort longtemps à reproduire les notes gustatives caractéristiques de l'arôme de fromage. C'est ainsi qu'on a mis en lumière l'effet particulier exercé par certains composés, tels les acides gras inférieurs, les lactones et les phénols.
Bon nombre de compositions aromatisantes destinées à développer un arôme de fromage dans les aliments dans lesquels elles sont incorporées sont actuellement commercialisées; aucune de ces compositions ne permet cependant une reconstitution tout à fait satisfaisante de l'arôme naturel recherché. Un des problèmes particuliers que l'aromaticien doit résoudre dans une telle reconstitution consiste en la reproduction fidèle de ce qui est mieux connu dans l'art par le terme de mouth-feel. Ce caractère gustatif est provoqué par l'ingestion de certains aliments et traduit la sensation ressentie à ce moment par l'homme dans sa bouche et déterminée par le contact de l'aliment sur la muqueuse, notamment de la langue et du palais. Un tel caractère est déterminant pour le goût de certains aliments, le fromage notamment.
La présente invention apporte une solution originale au problème mentionné ci-dessus. Nous avons en effet découvert que, par l'utilisation du composé de formule I, il était désormais possible de produire une composition aromatisante de type fromage pouvant conférer aux aliments dans lesquels elle était incorporée un caractère gustatif très naturel. Ladite composition permettait notamment de reproduire d'une manière fidèle l'effet de mouth-feel propre aux aliments à base de fromage.
Cette découverte est fort surprenante. En effet, des études récentes ont montré que certains dipeptides, entre autres un dipeptide cyclique de structure

étaient caractérisés par une note gustative amère [voir à cet effet
J. Biochem. , 72, 841 (1972) et Tetrahedron Letters , 509 (1974)].
Or, un tel caractère, généralement provoqué dans les produits dérivés de la fermentation du lait par l'action de certaines bacté
ries, est reconnu comme étant désagréable dans l'arôme de fromage [voir par exemple: Z. Lebensm.-Unters. Forsch. , 137, 370
(1968)].
L'invention concerne en outre l'utilisation d'une composition aromatisante contenant comme ingrédient actif le composé de formule I pour conférer, développer ou améliorer le caractère
gustatif de l'arôme du type fromage dans les aliments.
Les proportions selon lesquelles on peut utiliser le composé de l'invention afin d'obtenir l'effet aromatisant recherché peuvent varier de façon relativement étendue. On utilise cependant de préférence des quantités de l'ordre de 10 à 200 ppm (parties par million) du poids total de l'aliment aromatisé, quoique des quantités plus élevées ou légèrement inférieures puissent également être utilisées. Lorsque le composé de l'invention est utilisé pour la préparation d'arômes artificiels par exemple, par mélange avec d'autres ingrédients, on peut l'utiliser à raison de 5 à environ 50% du poids de la composition considérée. Parmi les co-ingrédients préférentiels dans une composition aromatisante de type fromage, il convient de citer les acides lactique, butyrique et isovalérianique ainsi que la méthylpentylcétone et les lactones dérivées d'acides hydroxycarboxyliques inférieurs.
Ces compositions aromatisantes peuvent convenablement être ajoutées aux aliments sous forme de solution, dispersion ou émulsion dans l'eau, dans de l'alcool ou dans un liquide oléagineux comestible.
Le composé de l'invention est un composé de structure connue et sa synthèse a été décrite pour la première fois dans Hoppe
Seyler's Z. physiol. Chem. , 174, 76 (1928).
Etant donné la présence, au sein de la molécule, d'atomes de carbone asymétriques, le composé de formule I peut se présenter sous la forme de plusieurs isomères de configuration. C'est ainsi que le terme cyclo-(Asn-Phe) sert à désigner indifféremment l'un ou plusieurs des isomères que voici:
cyclo-(D-Asn-L-Phe),
cyclo-(D-Asn-D-Phe),
cyclo-(L-Asn-D-Phe), et
cyclo-(L-Asn-L-Phe).
En considération de la similitude relative de leurs propriétés organoleptiques, tout au moins dans le domaine défini par l'invention, chacun des isomères ci-dessus peut être remplacé par un autre, ou tout mélange desdits isomères, sans pour autant que l'on puisse observer des modifications sensibles dans l'arôme développé.
A titre indicatif, nous donnons ci-après la méthode suivie pour la préparation du composé de l'invention: 5 g d'aspartame (Ajinomoto Co., Inc., Tokyo) ont été chauffés, dans un tube scellé, pendant 20 h à 135 C en présence de 100 mg (2%) de terre d'infusoires acide dans 80 ml d'alcool méthylique. La solution a été ensuite refroidie à 60 C, puis filtrée. 5 g d'ammoniac ont été ajoutés au filtrat clair et le tout a été chauffé pendant 20 h à 40 C.
Après évaporation des parties volatiles sous pression réduite, on a
obtenu 4 g (90%) du produit désiré.
L'invention est décrite de manière plus détaillée, sans en être cependant limitée, par l'exemple suivant.
Exemple:
On a préparé deux compositions destinées à développer un arôme de type fromage en mélangeant les ingrédients suivants (parties en poids):

Les compositions ci-dessus ont été utilisées séparément pour l'aromatisation de 1000 ml d'une solution d'eau salée à 0,5% à raison de 0,1 g de composition.
Deux échantillons de 0,1 g de ces mêmes compositions ont été séparément mélangés avec 0,1 g de cyclo-(Asn-Phe), ce qui a permis d'obtenir deux nouvelles compositions aromatisantes A' et
B' définies comme compositions tests. Puis on a procédé à la détermination de leurs propriétés aromatisantes comme indiqué ci-dessus pour les compositions de contrôle.
Le groupe d'experts, appelé à se prononcer sur l'arôme obtenu, a déclaré de manière unanime que les compositions tests permettaient d'obtenir un effet très satisfaisant de mouth-feel. Cet effet n'a pas été observé lors de l'emploi des compositions de contrôle.

The present invention relates to the field of flavors and, more precisely, it relates to a flavoring composition intended to improve or modify the taste of foods by giving them the aroma of cheese. Said flavoring composition is characterized in that it contains, as active ingredient, a cyclic dipeptide of formula:

this compound being also defined as cyclo- (Asn-Phe).
The flavoring industry has long sought to reproduce the flavor notes characteristic of cheese aromas. This is how we brought to light the particular effect exerted by certain compounds, such as lower fatty acids, lactones and phenols.
A good number of flavoring compositions intended to develop a cheese flavor in the foods in which they are incorporated are currently marketed; however, none of these compositions allows a completely satisfactory reconstitution of the desired natural flavor. One of the particular problems which the aromatician must solve in such a reconstruction consists in the faithful reproduction of what is better known in the art by the term of mouth-feel. This taste character is caused by the ingestion of certain foods and reflects the sensation felt at that time by the man in his mouth and determined by the contact of the food on the mucous membrane, in particular of the tongue and the palate. Such a character is determining for the taste of certain foods, in particular cheese.
The present invention provides an original solution to the problem mentioned above. We have in fact discovered that, by using the compound of formula I, it was now possible to produce a cheese-type flavoring composition capable of giving the foods in which it was incorporated a very natural taste character. Said composition made it possible in particular to faithfully reproduce the mouth-feel effect specific to cheese-based foods.
This discovery is very surprising. Indeed, recent studies have shown that certain dipeptides, including a cyclic dipeptide of structure

were characterized by a bitter taste note [see to this effect

The proportions according to which the compound of the invention can be used in order to obtain the desired flavoring effect can vary relatively widely. Preferably, however, amounts on the order of 10 to 200 ppm (parts per million) of the total weight of the flavored food are used, although larger or slightly smaller amounts may also be used. When the compound of the invention is used for the preparation of artificial flavors, for example, by mixing with other ingredients, it can be used in an amount of 5 to about 50% of the weight of the composition considered. Among the preferred co-ingredients in a flavoring composition of cheese type, mention should be made of lactic, butyric and isovaleric acids as well as methylpentyl ketone and lactones derived from lower hydroxycarboxylic acids.
These flavoring compositions may suitably be added to foods as a solution, dispersion or emulsion in water, in alcohol or in an edible oleaginous liquid.
The compound of the invention is a compound of known structure and its synthesis was described for the first time in Hoppe
Seyler's Z. physiol. Chem. , 174, 76 (1928).
Due to the presence, within the molecule, of asymmetric carbon atoms, the compound of formula I can be in the form of several configurational isomers. Thus the term cyclo- (Asn-Phe) is used to designate indifferently one or more of the isomers:
cyclo- (D-Asn-L-Phe),
cyclo- (D-Asn-D-Phe),
cyclo- (L-Asn-D-Phe), and
cyclo- (L-Asn-L-Phe).
Considering the relative similarity of their organoleptic properties, at least in the field defined by the invention, each of the above isomers can be replaced by another, or any mixture of said isomers, without however being possible observe significant changes in the aroma developed.
As an indication, we give below the method followed for the preparation of the compound of the invention: 5 g of aspartame (Ajinomoto Co., Inc., Tokyo) were heated, in a sealed tube, for 20 h at 135 C in the presence of 100 mg (2%) of acidic diatomaceous earth in 80 ml of methyl alcohol. The solution was then cooled to 60 ° C., then filtered. 5g of ammonia was added to the clear filtrate and the whole was heated for 20h at 40C.
After evaporation of the volatile parts under reduced pressure, it was
obtained 4 g (90%) of the desired product.
The invention is described in more detail, without however being limited thereto, by the following example.
Example:
Two compositions for developing a cheese-like flavor were prepared by mixing the following ingredients (parts by weight):

The above compositions were used separately for flavoring 1000 ml of a 0.5% salt water solution at 0.1 g of composition.
Two 0.1 g samples of these same compositions were separately mixed with 0.1 g of cyclo- (Asn-Phe), which made it possible to obtain two new flavoring compositions A 'and
B 'defined as test compositions. Then, the determination of their flavoring properties was carried out as indicated above for the control compositions.
The group of experts, called on to comment on the aroma obtained, declared unanimously that the test compositions made it possible to obtain a very satisfactory mouth-feel effect. This effect was not observed when using the control compositions.
