Die Erdindung bezieht sich auf ein Verfahren zur Herstellung neuer Indolderivate der allgemeinen Formel:

worin R1, R2 und R3 die gleiche Bedeutung wie in Formel I haben, cyclisiert.
Die Cyclisierungsreaktion kann in Anwesenheit von organischen Säuren oder in einem inerten Lösungsmittel stattfinden.
Anstelle von organischen Säuren können auch sogenannte Lewis-Säuren, wie z. B. Bortriftuoridätherat, ZnCl2, AlC13 usw. verwendet werden.
Die Reaktion wird im allgemeinen bei erhöhter Temperatur, am besten bei Siedetemperatur des Reaktionsgemisches, durchgeführt. Die besten Ergebnisse werden im Temperaturintervall von 60 bis 1200 C erzielt.
Die erfindungsgemäss erhaltenen Verbindungen der Formel I weisen analgetische, entzündungswidrige und antirheumatische Eigenschaften auf. Sie können auf übliche Weise zu galenischen Formen, wie Kapseln, Tabletten, Suppositorien, usw. verarbeitet werden. Im allgemeinen werden sie in täglichen Dosen von 100 bis 300 mg verabreicht und weisen wesentlich weniger unerwünschte Nebenreaktionen (insbesondere weniger ulcerogene Wirkung) auf als bekannte Verbindungen ähnlicher Art, wie z. B. Indomethacin (1-(4-Chlor- benzoyl)-5 -methoxy-2-methylindol-3 -essigsäure) bei gleicher oder besserer Wirksamkeit, wie aus den folgenden Vergleichen ersichtlich ist. Diese Versuche wurden mit der gemäss dem folgenden Beispiel 16 erhaltenen Verbindung durchgeführt, welche hier als CRC-4011 bezeichnet ist.

Präparation 1 (Hydrazone)
In 50 ml 90 %igem Äthylalkohol werden 9,7 g (0,04 Mol) das Na-Salzes der p-Methoxyphenylhydrazinsulfonsäure suspendiert und bis zum Sieden erhitzt.
In das siedende Gemisch werden 3,3 ml konzentriertes HC1 zugegeben und 15 Minuten unter Rückfluss erhitzt, dann eine Lösung von 6,1 g (0,045 Mol) NaOAc 3 H2O in 10 ml Wasser zugegeben und im Eisbad gekühlt. Unter Mischen wird langsam 2,2 g (0,05 Mol) Acetaldehyd zugegeben und noch 30 Minuten gemischt. Das Gemisch wird in 200 ml kaltes Wasser gegossen und mit dreimal 50 ml Äther extrahiert. Die ätherischen Extrakte werden über Na2SO4 getrocknet, der Äther abgedampft und das verbliebene rote Öl unter hohem Vakuum abdestilliert. Man erhält 4,1 g (61% der Theorie) p-Methoxyphenylhydrazonacetaldehyd als hellgelbes Öl, Kp. 128 bis 134" C/1 mm Hg.
Präparation 2
Auf dieselbe Weise wie in Präparation 1 wurde aus dem Na-Salz der p-Chlorphenylhydrazinsulfonsäure und Acetaldehyd das p-Chlorphenylhydrazon des Acetaldehyds als farblose Kristalle vom Smp. 58 bis 60 C, Kp. 115 bis 1200 C/ 1 mm Hg, in einer Ausbeute von 80% der Theorie erhalten.
IR-Spektrum: 1345-1350 (Ar-NH), 1630-1640 (C=N), 3310-3350 (NH) cm-t. NMR - 1,88 ppm (d, 3 H), 6,85 ppm (quart, 1 H).
Präparation 3
Auf dieselbe Weise wie in Präparation 1 wird aus dem Na Salz der m-Chlorphenylhydrazinsulfonsäure und Acetaldehyd das m-Chlorphenylhydrazon- des Acetaldehyds als hellgelbes Öl, Kp. 110 bis 1150 C/1 mm Hg, in einer Ausbeute von 91% der Theorie hergestellt. IR- und NMR-Spektren bestätigen die Struktur.
Präparation 4
24,2 g (0,1 Mol) des Na-Salzes der p-Methoxyphenylhydrazinsulfonsäure werden in 160 ml 90 %igem Äthylalkohol suspendiert, bis zum Sieden erhitzt, 9 ml konzentriertes HCI zugegeben und 15 Minuten unter Rückfluss erwärmt. Zu der warmen Mischung werden eine Lösung von 19 g (0,14 Mol) NaOAc 3 H2O in 25 ml Wasser und 13,8 g (0,07 Mol) 2 Methyl-4-nitroimidazolylaceton zugesetzt und unter Rückfluss und Rühren so lange erwärmt, bis sich das Letztere gelöst hat.
Das Gemisch lässt man nun stehen, bis es sich abgekühlt hat, und giesst es in 500 ml Eis und Wasser ein. Es kristallisiert das p-Methoxyphenylhydrazon des (2'-Methyl-4'-nitroimidazolyl)-acetons als hellbraunes, kristallines Pulver, Smp.
140 bis 143" C (Zersetzung), in einer Ausbeute von 94% der Theorie aus.
Präparation 5 (benzoylierte Hydrazone)
5,0 g (0,0287 Mol) des p-Methoxyphenylhydrazons des Acetaldehyds (Präparation 1) werden in 20 ml Pyridin gelöst, und unter Mischen und Kühlen im Eisbad werden 4,7 ml (6,4 g, 0,0365 Mol) p-Chlorbenzoylchlorid tropfenweise zugesetzt. Das Gemisch wird noch 4 Stunden gemischt und anschliessend in 100 ml kaltes Wasser geschüttet. Das ausgeschiedene Öl wandelt sich durch Stehen in das hellbraune kristalline Pulver des N-p-Methoxyphenyl-N-p-chlorbenzylhydrazons des Acetaldehyds um, das abfiltriert, gut mit Wasser gewaschen und im Vakuum-Exsikkator über Schwefelsäure getrocknet wird. Smp. 92 bis 95" C in quantitativer Ausbeute.
Präparation 6
Auf dieselbe Weise wie in Präparation 5 wird aus dem p Methoxyphenylhydrazon des Acetaldehyds und aus 3,5-Dinitrobenzoylchlorid das N-p-Methoxyphenyl-N-3,5-dinitrobenzoylhydrazon des Acetaldehyds, ein sehr dickes hellbraunes Öl, gewonnen.
Präparation 7
Auf dieselbe Weise wie in Präparation 5 wird aus dem p Methoxyphenylhydrazon des Acetaldehyds und p-Methoxybenzoylchlorid das N-p-Methoxyphenyl-N-p-methoxybenzoyl- hydrazon des Acetaldehyds als farblose Kristalle vom Smp.
161 bis 163 C in quantitativer Ausbeute erhalten.
Präparation 8
Auf dieselbe Weise wie in Präparation 5 wird aus dem p Methoxyphenylhydrazon des Acetaldehyds und N-Phthalylglycylchlorid das N-p-Methoxyphenyl-N-phthalimidoglycylhydrazon des Acetaldehyds als hellorange Kristalle, Smp. 162 bis 165" C (Zersetzung), in einer Ausbeute von 96% der Theorie erhalten.
Präparation 9
Auf dieselbe Weise wie in Präparation 5 wird aus dem p Chlorphenylhydrazon des Acetaldehyds und p-Chlorbenzoylchlorid das N-p-Chlorphenyl-N-p-chlorbenzoylhydrazon des Acetaldehyds als farblose Kristalle, Smp. 145 bis 155" C, in einer Ausbeute von 74% der Theorie erhalten.
Präparation 10
Auf dieselbe Weise wie in Beispiel 5 wird aus dem m Chlorphenylhydrazon des Acetaldehyds (Präparation 2) und p-Chlorbenzoylchlorid das N-m-Chlorphenyl-N-p-chlorbenzoylhydrazon des Acetaldehyds als farbloses kristallines Pulver, Smp. 91 bis 93" C, in einer Ausbeute von 40% der Theorie hergestellt.
Präparation 11 (Benzolylierte Hydrazin-Hydrochloride)
9,1 g (0,003 Mol) des rohen N-ss-Methoxy-phenyl-N-pchlorbenzoylhydrazons des Acetaldehyds (Präparation 5) werden in einer Lösung von 120 mi trockenem Toluol (oder 150 ml trockenem Äther) und 5 ml absolutem Methanol suspendiert und unter Kühlung im Eisbad 3 bis 4 g trockenes HCl in das Gemisch im Laufe 1 Stunde eingeleitet. Nach 20 bis 30 Minuten Stehen wird das überschüssige HCl mit einer Vakuumpumpe aus dem Gemisch abgetrennt, das auskristallisierte N-p-Methoxyph enyl-N-p-chlorbenzoylhydrazin-Hydrochlorid abfiltriert und mit Benzol und Äther gewaschen. Man trocknet es im Vakuumtrockner über festem NaOH und Paraffin. Das Rohprodukt wird als hellbraune Kristalle, Smp. 164 bis 166 C (Zersetzung), in einer Ausbeute von 79% der Theorie erhalten.
Präparation 12
Auf gleiche Weise wie in Präparation 11 wird aus dem Np-Methoxyphenyl-N-(2,3-dinitrobenzoyl)-hydrazon des Acetaldehyds (Verbindung aus Präparation 6) das N-p Methoxyphenyl-N-(2,3 -dinitrobenzoyl)-hydrazin-Hydro- chlorid als hellbraune Kristalle, Smp. 182 bis 184" C (Zersetzung), in einer Ausbeute von 86% der Theorie erhalten.
NMR (CO30D) 3,78 ppm (s, 3 H), 3,80 ppm (s, 3 H), 6,52 ppm (dd, 4 H), 6,88 ppm (dd, 4 H).
Präparation 13
Auf gleiche Weise wie in Präparation 11 wird aus N-p Methoxyphenyl-N-p-methoxybenzoylhydrazon des Acetaldehyds (Verbindung aus Präparation 7) das N-p-Methoxyphenyl-N-p-methoxybenzoylhydrazin-Hydrochlorid als farblose nadelförmige Kristalle, Smp. 152 bis 1550 C (Zersetzung), in einer Ausbeute von 89% der Theorie erhalten.
Präparation 14
Auf gleiche Weise wie in Präparation 11 wird aus N-p Methoxyphenyl-N-phthalimidoglycylhydrazon des Acetaldehyds (Verbindung aus Präparation 8) das N-p-Methoxyphe nyl-N-phthalimidoglycylhydrazin-Hydrochlorid gewonnen, ein hellbraunes kristallines Pulver, Smp. 145 bis 1480 C (Zersetzung), Ausbeute 94% der Theorie.
Präparation 15
Auf gleiche Weise wie in Präparation 11 wird aus N-p Chlorphenyl-N-p-benzoylhydrazon des Acetaldehyds (Verbindung aus Präparation 9) das N-p-Chlorphenyl-N-p-chlorbenzoyl)-hydrazin-Hydrochlorid bereitet, das als farblose Kristalle, Smp. 170 bis 173" C (Zersetzung), in einer Ausbeute von 85% der Theorie anfällt.
Präparation 16
Auf gleiche Weise wie in Präparation 11 wurde aus dem N-m-Chlorphenyl-N-p-chlorbenzoylhydrazon des Acetaldehyds (Verbindung aus Präparation 10) das N-m-Chlorphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid als farbloses kristallines Pulver, Smp. 170 bis 172" C (Zersetzung), in einer Ausbeute von 90% der Theorie erhalten.
Beispiel 1 (Indole)
3,1 g (0,01 Mol) N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 11) und
1,6 g (0,01 Mol) Benzoylaceton werden in 30 ml Eisessig gelöst und 3 bis 4 Stunden auf dem Wasserbad bei 70 bis 80" C erwärmt. Das ausgeschiedene Ammoniumchlorid wird abfiltriert und die Essigsäure im Vakuum eingedampft. Der Rest wird zweimal aus Äthylalkohol umkristallisiert. Man erhält das 1 - (p-Chlorbenzoyl) -2-methyl-3 -benzoyl-5 -methoxy-indol in Form von hellgelben nadelförmigen Kristallen, Smp. 112 bis 114" C in einer Ausbeute von 31% der Theorie.

NMR (Me2CO-d) 2,32 ppm (s, 3 H), 3,64 ppm (s. 3 H), 6,60 bis 7,10 ppm (m, 3 H), 7,40 bis 7,95 ppm (m, 9 H).
Beispiel 2
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus dem N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 11) und p-Methylbenzoylaceton das 1 -(p-Chlorbenzoyl)-2-methyl-3 -(p-methyl- benzoyl)-5-methoxyindol als hellgelbe flockige Kristalle, Smp.
142 bis 144" C, in einer Ausbeute von 52% der Theorie gewonnen.

Auf die gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 11) und 3,4-Dimethylbenzoylaceton das 1-(p-Chlorbenzoyl)-2-methyl-3-(3,4-di- methylbenzoyl)-5-methoxyindol als hellgelbe Kristalle, Smp.
144 bis 146 C, in einer Ausbeute von 33% der Theorie erhalten.

Beispiel 4
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 11) und p-Methoxybenzoylaceton das 1-(p-Chlorbenzoyl)-2-methyl-3-(p-meth- oxybenzoyl)-5-methoxyindol als farblose flockige Kristalle aus Äthylacetat-Petroläther, Smp. 127 bis 129 C, in einer Ausbeute von 40 % der Theorie erhalten.
Analyse für: C2sH2oclNo4 berechnet: C 69,20 H 4,65 N 3,23% gefunden: C 69,60 H 5,04 N 3,08%
Beispiel 5
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 11) und 2,4-Dimethoxybenzoylaceton das 1-(p-Chlorbenzoyl)-2-methyl-3-(2,4-dimethoxybenzoyl)-5-methoxyindol als hellgelbe, flockige Kristalle, Smp. 110 bis 111" C, in einer Ausbeute von 58 % der Theorie erhalten.
Analyse für: C24H22ClNOs berechnet: C 67,31 H 4,78 N 3,02% gefunden: C 67,21 H 4,95 N 2,74%
Beispiel 6
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 11) und p-Chlorbenzoylaceton das 1,3 -Bis-(p-chlorbenzoyl)-2-methyl-5 -methoxy- indol als gelbe Kristalle, Smp. 181 bis 183 C (aus Äthylacetat), in einer Ausbeute von 60% der Theorie erhalten.
Analyse für: C24H17CliNO3 =438,298 berechnet: C 65,76 H 3,91 N 3,19% gefunden: C 65,53 H 4,25 N 3,05%

Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 11) und p-Brombenzoylaceton das 1-(p-Chlorbenzoyl)-2-methyl-3-(p-brombenzoyl)-5-methoxyindol als hellgelbe Kristalle, Smp. 185 bis 186 C, in einer Ausbeute von 50% der Theorie erhalten.
Analyse für: C24H17BrCINO3=482,757 berechnet: C 59,71 H 3,55 N 2,90% gefunden: C 59,63 H 3,73 N 2,95%
Beispiel 8
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydro- chlorid (Verbindung aus Präparation 11) und 2-Naphthoylaceton das 1 - (p-Chiorbenzoyl) -2-methyl-3 - (2' -naphthoyl)- 5-methoxyindol als farblose flockige Kristalle, Smp. 143 bis 145 C, in einer Ausbeute von 45% der Theorie erhalten.
Analyse für: C28H2oClNO3=453,905 berechnet: C 74,08 H 4,45 N 3,09% gefunden: C 73,79 H 4,67 N 3,17%
Beispiel 9
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-p-methoxybenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 13) und p-Chlorbenzoylaceton das 1 -(p-Methoxybenzoyl)-2-methyl-3 -(p- chlorbenzoyl)-5-methoxyindol als farblose, flockige Kristalle, Smp. 141 bis 143" C, in einer Ausbeute von 51 % der Theorie erhalten.

Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p -Methoxyphenyl-N-p-methoxybenzoylhydrazin-Hy- drochlorid (Verbindung aus Präparation 13) und2-Naph- thoylaceton das 1-(p-Methoxybenzoyl)-2-methyl-3-(2'- naphthoyl)-5-methoxyindol als hellgelbe Kristalle, Smp. 135 bis 137 C, in einer Ausbeute von 40% der Theorie erhalten.
Analyse für; C29H23NO4 = 449,482 berechnet: C 77,49 H 5,16 N 3,11% gefunden: C 77,42 H 5,33 N 3,27%
Beispiel 11
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-methoxybenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 13) und Levulinsäure die 1-(p-Methoxybenzoyl)-2-methyl-5-methoxyindol-3-essigsäure als farblose, nadelförmige Kristalle, Smp. 163 bis 165 C, in einer Ausbeute von 75% der Theorie erhalten.
Analyse für: C20H19N05 = 353,360 berechnet: C 67,98 H 5,42 N 3,98% gefunden: C 67,64 H 5,55 N 4,20%
Beispiel 12
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-(3 ,5-dinitrobenzoyl) -hydrazin- Hydrochlorid (Verbindung aus Präparation 12) und Levulinsäure die 1 -(3,5-Dinitrobenzoyl)-2-methyl-5-methoxy-3 - indolessigsäure als gelbes, kristallines Pulver, Smp. 219 bis 211" C. in einer Ausbeute von 70% der Theorie erhalten.
Analyse für: C19H15N3O8=413,331 berechnet: C 55,21 H 3,65% gefunden: C 55,47 H 3,41%
Beispiel 13
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Methoxyphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 11) und Äthylmethylketon das 1 -(p-Chlorbenzoyl)-2,3 -dimethyl-5 -methoxyindol als farblose Kristalle, Smp. 81 bis 83" C, in einer Ausbeute von 76% der Theorie erhalten.
Beispiel 14
Auf gleiche Weise wie in Beispiel 1 beschrieben, wurde aus N-p-Chlorphenyl-N-p-chlorbenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 15) und Äthylmethylketon das 1 -(p-Chlorbenzoyl)-2,3 -dimethyl-5-chlorindol als gelbe Kristalle, Smp. 128 bis 130"C, in einer Ausbeute von 80% der Theorie erhalten.
Analyse für: C17H13CiNO berechnet: C 64,16 H 4,11 N 4,40% gefunden: C 64,03 H 3,90 N 4,16%
Beispiel 15
2,15 g (0,007 Mol) N-p-Methoxyphenyl-N-p-methoxybenzoylhydrazin-Hydrochlorid (Verbindung aus Präparation 13) und 1,3 g (0,007 Mol) 2-Methyl-4-nitroimidazolylaceton in 15 ml Eisessig werden 8 Stunden unter Mischen auf einem Wasserbad von 85 bis 90" C erwärmt. Das ausgeschiedene Ammoniumchlorid wird von der heissen Lösung abfiltriert, und das Filtrat wird zum Abkühlen stehengelassen. Das ausgeschiedene Rohprodukt wird durch Filtration abgetrennt. Durch Umkristallisieren aus wässrigem Aceton erhält man 1-(p Methoxybenzoyl)-2-methyl-3-(2'-methyl-4'-nitroimidazolyl)-5-methoxyindol als farblose Kristalle, Smp. 227 bis 229 C, in einer Ausbeute von 38% der Theorie.
Analyse für: C22H20N4O5=420,412 berechnet: C 62,85 H 4,79% gefunden: C 62,91 H 4,84%
Beispiel 16
