Die Verbindung der Formel I, worin Rl Methyl und R2 Wasserstoff bedeuten, ist bekannt.
Der Ausdruck nieder Alkyl , wie er hier verwendet wird, bezieht sich allein oder in Kombination, wie beispielsweise Halogen-nieder-alkyl auf geradkettige und verzweigte Kohlenwasserstoffreste mit bis zu 7 Kohlenstoffatomen, wie Methyl, Äthyl, Propyl, Isopropyl, Butyl, tert.-Butyl, Pentyl und dgl.
Der Ausdruck Salze umfasst die in der Pharmazie infolge ihrer physiologischen Eigenschaften (geringe Toxizität, Resorption etc.) gebräuchlichen Salze.
Bevorzugte Verbindungen der Formel I sind: N-Isobutyl-3,5-dinitro-2-thiophenamin N-sec. -Butyl-3, 5 -dinitro -2-thiophenamin N-Isopropyl-3 ,5-dinitro-2-thiophenamin N-Äthyl-3,5-dinitro-2-thiophenamin N-Butyl-3,5-dinitro-2-thiophenamin N-Isobutyl-N-methyl-3 ,5-dinitro-2-thiophenamin N-Äthyl-N-methyl-3, 5 -dinitro -2-thiophenamin N,N-Dimethyl-3 ,5-dinitro-2-thiophenamin N-(2-Chloräthyl)-3,5-dinitro-2-thiophenamin
Eine ganz bevorzugte Verbindung der Formel list das 3- (3,5-Dinitro-2-thienyl)-thiazolidin.
Gemäss vorliegender Erfindung wird ein Ausgangsmaterial der allgemeinen Formel II mit einem Amin der Formel III umgesetzt. Diese Reaktion wird in einem inerten Lösungsmittel, wie beispielsweise in einem niederen wässrigen Alkanol, vorzugsweise Methanol oder Äthanol, in Dimethylformamid, in einem chlorierten Kohlenwasserstoff, vorzugsweise Methylenchlorid oder Chloroform, oder in Dimethylsulfoxyd durchgeführt. Die Reaktionstemperatur liegt vorteilhaft zwischen OoC und dem Siedepunkt des Reaktionsgemisches. Ein besonders bevorzugtes Temperaturintervall liegt zwischen 150 und 40ob. Der Druck ist nicht kritisch; die Reaktion wird vorzugsweise im offenen Gefäss durchgeführt.
Wird das Amin der Formel III in Form eines Salzes, wie beispielsweise des Hydrohalogenides, eingesetzt, so wird zweckmässigerweise dem Reaktionsgemisch eine schwache Base, wie beispielsweise Kaliumcarbonat, zugesetzt. Die Aufarbeitung des Reaktionsgemisches erfolgt in der üblichen Weise durch Entfernen des Lösungsmittels und Umkristallisieren des Rückstandes.
Die Verbindungen der Formel I sind teilweise basische Substanzen und die Herstellung ihrer Salze, insbesondere solcher, die pharmazeutisch verwendbar sind, wird ebenfalls von der vorliegenden Erfindung umfasst. Diese Salze können aus den entsprechenden Basen nach allgemein bekannten Methoden durch Umsetzung mit geeigneten anorganischen oder organischen Säuren hergestellt werden. Beispiele von organischen und anorganischen Säuren, welche zur Herstellung von pharmazeutisch verwendbaren Salzen Verwendung finden können, sind Chlor- oder Bromwasserstoff, Schwefelsäure, Essigsäure, Bernsteinsäure, Maleinsäure, Methan-, Benzoloder p-Toluolsulfonsäure usw.
Die pharmazeutisch nicht verwendbaren Säureadditionssalze der Verbindungen der Formel I eignen sich als Zwischenprodukte für die Herstellung von entsprechenden pharmazeutisch verwendbaren Säureadditionssalzen, welche beispielsweise durch Umsalzung oder durch Freisetzen der Base und anschliessende Salzbildung mit einer geeigneten Säure erfolgen kann. Diejenigen Verbindungen der Formel I, die amphoteren Charakter besitzen, können gegebenenfalls auch mit geeigneten Basen in Salze übergeführt werden.
Die Ausgangsverbindungen der Formel II sind teilweise ebenfalls neu. Sie werden hergestellt, indem man eine Verbindung der Formel

worin Hal die in Formel I angegebene Bedeutung besitzt und Y Wasserstoff oder eine Nitrogruppe bedeutet, wobei mindestens eines der Symbole Y Wasserstoff bedeutet, nitriert.
Für die Nitrierungsreaktion ist es unerheblich, ob Y in der Formel IV Wasserstoff oder eine Nitrogruppe bedeutet. In jedem Fall wird zweckmässigerweise die Verbindung der Formel IV zuerst in konzentrierter Schwefelsäure bei tiefer Tem peratur, vorzugsweise 0-200C, gelöst und zu diesem Gemisch eine beispielsweise äuf ebenfalls 0-200C vorgekühlte Lösung von konzentrierter Schwefelsäure und konzentrierter Salpetersäure unter Kühlung zugefügt. Die Art und Weise, in welcher die Salpetersäure dem Gemisch zugeführt wird, ist nicht kritisch. Beispielsweise kann zu Schwefelsäure auch Natriumnitrat zugegeben werden. Wichtig ist lediglich, dass die Salpetersäure etwa in stöchiometrischer Menge bzw. in kleinem Überschuss vorliegt und die Temperatur tief genug gehalten und kontrollierbar bleibt.
Letzteres wird durch vorsichtiges Zutropfen der Nitriersäure auf einfache Weise erreicht. Nachdem die Nitriersäure zugegeben ist, wird das Reaktionsgemisch auf Eis gegossen und mit beispielsweise Essigester extrahiert. Essigester-Extrakte werden mit Wasser mehrfach gewaschen, getrocknet und eingedampft. Der Rückstand wird in üblicher Weise gereinigt.
Die Verbindungen der Formel I finden Verwendung als Arzneimittel. Sie besitzen eine protocoide, bactericide, anthelmintische und insbesondere eine schistosomicide Wirkung. Sie können beispielsweise zur Therapie der Bilharziose verwendet werden. Der nachstehende Versuchsbericht demonstriert die schistosomicide Wirkung der Verbindungen der Formel I in beispielhafter Form.
Goldhamster werden subcutan mit 60 Cercarien von Schistosoma mansoni infiziert. Ca. 42 Tage nach der Infektion werden die Tiere an fünf aufeinanderfolgenden Tagen mit den zu prüfenden Präparaten behandelt. Pro Präparat und Dosierung (mg/kg) werden 5-10 Tiere eingesetzt. Als Kontrolle dienen 10 unbehandelte Tiere. Die Sektion erfolgt 6 Tage bzw.
2-3 Wochen nach Behandlungsabschluss. Wurmpaare in Mesenterialvenen, Pfortader und Leber werden herauspräpariert und gezählt. Die prozentuale Verteilung der Wurmpaare in Mesenterialvenen, Pfortader und Leber wird berechnet und der Zustand der Würmer (lebend, tot) registriert. Die Präparatwirkung zeigt sich in einem erhöhten Anteil der Würmer in den Gefässen der Leber und im Auftreten toter Würmer.
Zur Auswertung vergleicht man den prozentualen Anteil lebender und toter Wurmpaare in den Gefässen der Leber sowohl bei infizierten behandelten Tieren als auch bei infizierten aber unbehandelten Kontrolltieren. Die Bestimmung der SD9o (= Shift-Dosis 90%: Dosis, die bei einer Gruppe behandelter Tiere 90% der Wurmpaare in die Leber vertreibt) und der VDsn (= Vermizide Dosis 90%: Dosis, die 90% der Wurmpaare abtötet) erfolgt nach der Probitmethode.

Die Verfahrensprodukte der Formel I können als Heilmittel, z.B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial, wie Gelatine, Milchzucker, Stärke, Gummi arabicum, Magnesiumstearat, Talcum, pflanzliche Öle, Polyalkylenglykole, Vaseline u. dgl. mehr, enthalten.
Die Präparate können in fester Form, z.B. als Tabletten, Dragees, oder in flüssiger Form, z.B. als Lösungen,
Suspensionen oder Emulsionen, vorliegen. Sie können Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer enthalten. Auch andere therapeutisch wirksame Stoffe können beigemischt sein.
Zweckmässige pharmazeutische Dosierungsformen enthal ten 100-500 mg, vorzugsweise etwa 250 mg, einer Verbindung der Formel.
Die Dosierung wird entsprechend den individuellen Erfor dernissen gewählt. Zum Beispiel können diese Verbindungen in Dosierungen von etwa 10 mg/kg bis etwa 50 mg/kg pro Tag p. o., vorzugsweise 25 mg/kg pro Tag p. o. verabreicht wer den. Diese Menge kann in einer einzelnen Dosierung oder in mehreren unterteilten Dosierungen verabreicht werden, je nach Bedürfnis des Patienten und Vorschrift des Fachmannes.
Zweckmässigerweise wird diese Dosis in Anpassung an den
Zustand des Patienten an mehreren aufeinanderfolgenden
Tagen verabreicht, vorzugsweise an 5 bis 8 aufeinanderfolgen den Tagen.
Die nachstehenden Beispiele illustrieren die Erfindung. Die Temperaturen sind in oC angegeben.
Beispiel 1
5 g 2-Chlor-3,5-dinitrothiophen werden in 50 ml absolutem Methanol aufgeschlämmt und mit 5 g Chloräthylaminhydrochlorid und 5 g Kaliumcarbonat versetzt. Das Reaktionsgemisch wird auf dem Dampfbad 5 Minuten erwärmt, das Methanol abgedampft und der Rückstand mit Methylenchlorid/Wasser extrahiert.
Die Methylenchloridlösung wird dann über Magnesiumsulfat getrocknet, filtriert und eingeengt. Der Rückstand wird an einer Kieselgelkolonne (Laufmittel: Methylenchlorid) gereinigt. Aus Äthanol kristallisiert man N-(2-Chloräthyl)-3,5dinitro-2-thiophenamin vom Fp.: 137"C.
Beispiel 2
In Analogie zur Arbeitsvorschrift in Beispiel 1 erhält man aus 10 g 2-Chlor-3,5-dinitrothiophen, 8 g Isobutylamin und 120 ml Methanol das N-Isobutyl-3,5-dinitro-2-thiophenamin.

Beispiel 4
In Analogie zur Arbeitsvorschrift in Beispiel 1 erhält man aus 10 g 2-Chlor-3,5-dinitrothiophen, 120 ml Methanol und 6,5 g Isopropylamin das N-Isopropyl-3,5-dinitro-2-thiophenamin. Aus Methylenchlorid/Petroläther, Fp.: 140-142 C.
Beispiel 5
In Analogie zur Arbeitsvorschrift in Beispiel 1 erhält man aus 10 g 2-Chlor-3,5-dinitrothiophen, 100 ml Methanol und 5 g Äthylamin (letzteres gelöst in 20 ml Methanol) das N Äthyl-3,5-dinitro-2-thiophenamin. Aus Methylenchlorid/ Petroläther, Fp.: 141-143 C.
Beispiel 6
In Analogie zur Arbeitsvorschrift in Beispiel 1 erhält man aus 10 g 2-Chlor-3,5-dinitrothiophen, 100 ml Methanol und 8 g n-Butylamin das N-Butyl-3,5-dinitro-2-thiophenamin. Aus Äthanol, Fp.: 60-64 C.
Beispiel 7
In Analogie zur Arbeitsvorschrift in Beispiel 1 erhält man aus 2-Chlor-3,5-dinitrothiophen, 100 ml Methanol, 5 g Dimethylamin das N,N-Dimethyl-3 ,5-dinitro-2-thiophenamin, welches aus Äthanol umkristallisiert wird, Fp.: 1550C.
Beispiel 8
5 g 2-Chlor-3,5-dinitrothiophen werden in 70 ml Äthanol mit 4,5 g Thiazolidin versetzt und bei Raumtemperatur gerührt. Das Reaktionsprodukt fällt aus und wird abgesaugt.
Das ausgefallene Produkt wird aufgearbeitet, indem das Äthanol abdestilliert und das Produkt direkt an einer Kieselgelkolonne gereinigt wird. Aus Methylenchlorid/Petroläther kristal lisiert 3-(3,5-Dinitro-2-thienyl)-thiazolidin. Fp.: 126-128"C.
Beispiel 9

The precipitated product is worked up by distilling off the ethanol and purifying the product directly on a silica gel column. 3- (3,5-Dinitro-2-thienyl) thiazolidine crystallizes from methylene chloride / petroleum ether. M.p .: 126-128 "C.
