Die vorliegende Erfindung betrifft ein Verfahren zur
Herstellung von neuen und wertvollen Cephalosporin-Verbin dungen, insbesondere von 7-acylierten, 3-substituierten
Cephalosporin-Verbindungen.
Es ist bekannt, dass Cephalosporine auf verschiedene Arten hergestellt werden können; so kann nach einem bekannten
Verfahren durch Behandlung von heterocyclisch acylierten
Cephlosporin-Derivaten mit heterocyclischen Thiolen die Acet oxy-Gruppe in 3-Stellung des Cephalosprin-Kernes verdrängt werden, wie von T. Takano et al. in der USA-Patentschrift Nr.
3 516 997 beschrieben wird. Es ist ferner durch R.J. Stedman aus der USA-Patentschrift Nr. 3 124 576 bekannt geworden, dass 3-desacetoxylierte Cephalosporine durch katalytische
Hydrierung der Acetoxymethyl-Gruppe der Cephalosporine über Palladium auf einem inertern Träger hergestellt werden können. Durch T. Takano et al. wird in der erwähnten USA
Patentschrift ferner gezeigt, dass 3-heterocyclisch thiolierte 7
Aminocephalosporan-Säuren auf herkömmliche Art acyliert werden können. Die britische Patentschrift Nr. 1 073 530 lehrt, dass Cephalosporine in wesentlich höheren ausbeuten aus 7
Aminocephalosporan-Säure durch Silylierung und anschlies sende Acylierung als durch blosse Acylierung allein erhalten werden können. Verschiedene andere Silylierungsmittel, die für die Silylierung von 7-Aminocephalosporansäure verwendet werden können, werden von B.G.
Jackson in der belgischen
Patentschrift Nr. 737 761 beschrieben. Es wurden schon verschiedene Cephalosporin-Derivate nach den verschiedenen, oben kurz erwähnten Methoden hergestellt und für den therapeutischen Gebrauch zur Behandlung bakterieller Infektionen getestet. Bei der therapeutischen Verwendung der Cephalosporin-Antibiotika, besonders bei oraler Verabreichung, werden diese nicht zu einer genügenden Menge konzentriert bzw. nicht in genügender Menge absorbiert, um eine zufriedenstellende Hemmung des Wachstums der infektiösen Bakterien zu bewirken. Selbst wenn solche Cephalosporin Derivate in recht hoher Konzentration absorbiert werden, sind diese häufig gegen eine Reihe von Organismen nicht genügel.d aktiv.
Es wurde nun gefunden, dass einige neue Cephalosporin Verbindungen bei oraler Verabreichung in genügender Menge absorbiert werden, um das Wachstum infektiöser Organismen zu hemmen und sich so wertvoll erwiesen in der oralen Behandlung von bakteriellen Infektionen, wie sie durch Staphylokokken und andere infektiöse Mikroorganismen verursacht werden.
Es ist daher Ziel der vorliegenden Erfindung, ein Verfahren zur Herstellung neuer und wertvoller 7-acylierter, 3-substituierter Cephalosporin-Verbindungen zu schaffen, die nützlich sind als antibakterielle Mittel bei der Behandlung von Krankheiten, wie sie von verschiedenen Mikroorganismen, einschliesslich gram-positiver und gram-negativer Bakterien, verursacht werden, und die sich besonders darin auszeichnen, dass sie auch bei oraler Verabreichung in genügend hoher Konzentration absorbiert werden, um das Wachstum der Mikroorganismen zu verhindern.
Die neuen 7-acylierten, 3-substituierten Cephalosporin Verbindungen können durch die folgende allgemeine Formel

Der Ausdruck Heterocyclyl umfasst insbesondere N-haltige Ringe, wie Thiazolyl-, Thiadiazolyl-, Oxadiazolyl- oder Tetrazolyl-Ringe. Die N-haltigen Ringe sowie die Thienyl und Furylreste können gegebenenfalls substituiert sein durch Halogenatome, Amino-, Merkapto-, Niederalkyl- oder Nieder alkylthio-Gruppen.
Der Acylrest R2 mit mehr als einem Kohlenstoffatom entspricht der allgemeinen Formel R-CO-, worin R eine Niederalkyl-, Aryl-, Aryl-niederalkyl- oder Thienylgruppe sein kann, wobei besagte Niederalkyl-, Aryl- oder Aralkyl-Gruppen durch Sauerstoff unterbrochen sein können.
Der Ausdruck Niederalkyl bedeutet gegebenenfalls substituierte geradkettige oder verzweigte gesättigte aliphatische Kohlenwasserstoffreste mit einem bis vier Kohlenstoffatomen, die beispielsweise mit Halogenatomen, Niederalkyloxy oder Aryloxyresten substituiert sein können, z. B.
Alkylreste wie Methyl, Äthyl, Propyl, i-Propyl, Butyl, i-Butyl und tert.Butyl, die gegebenenfalls durch Chlor, Methoxy, Äthoxy, Propoxy, Chlormethoxy, Chloräthoxy, Dichlormethoxy, Phenoxy, Chlorphenoxy u. dgl. substituiert sein können.
Der Arylrest ist ein aromatischer Kohlenwasserstoffrest, welcher gegebenenfalls durch Halogen, Hydroxyl, Nitro, Amino, Niederalkyl, Niederalkoxy, Niederalkylcarbonyloxy oder Niederalkylthio substituiert sein kann. Als Beispiele für solche Arylreste seien erwähnt: Phenyl, Naphthyl, Chlorphenyl, Hydroxyphenyl, Nitrophenyl, Aminophenyl, Tolyl, Methoxyphenyl, Acetoxyphenyl, Methylthiophenyl u. dgl.
Der Ausdruck Halogen umfasst Brom, Chlor u. dgl.
Die gewünschten Ausgangsprodukte können z. B. hergestellt werden unter Verwendung eines Ausgangsmaterials, welches gewöhnlich als Cephalosporin C bekannt ist, und welches durch Aufzucht eines Cephalosporium-Stammes in einem passenden Nährmedium nach bekannten Methoden gewonnen werden kann.
7-Acylierte Aminocephalosporan-Säuren, wie sie im vorliegenden Prozess verwendet werden, können durch Hydrolyse des Cephalosporin-C-Antibiotikums und anschliessende Acylierung der erhaltenen 7-Aminocephalosporan Säuren nach bekannten Methoden erhalten werden.
Wenn die betreffenden Cephalosporin-Derivate in Form ihrer freien Säuren in die Reaktion eingesetzt werden, so kann diese in Gegenwart einer Base, wie Alkalihydroxyd, Alkalibicarbonat, oder einer organischen Base, wie Trialkylamin, oder einer Pufferlösung wie Phosphatpuffer, durchgeführt werden.
Bei Abwesenheit einer solchen Base oder einer solchen Pufferlösung können die Salze der Säuren verwendet werden.
Das erfindungsgemässe Verfahren besteht nun darin, dass man eine Verbindung der allgemeinen Formel:

in welcher Rl, R2 und M obige Bedeutung haben, mit einer nukleophilen Verbindung der allgemeinen Formel:
R3-H (V) in welcher R3 obige Bedeutung hat, oder deren Salze umsetzt.
Die Reaktion kann in einem Lösungsmittel, wie Wasser, wässerigem Aceton oder Alkoholen, durchgeführt werden.
Auch eine Pufferlösung kann verwendet werden. Bevorzugte
Puffer sind Phosphat-Puffer. Die Reaktion kann zwischen
Raumtemperatur und etwa 80" C ausgeführt werden, bevor zugt zwischen 55 und 65" C bei einem pH-Wert von etwa 3,5 bis 8,0, vorzugsweise zwischen 4,0 und 7,0, wobei die Bedin gungen abhängig von der Art der eingesetzten Reaktionsmittel variieren können. Weiters kann die Reaktion, wenn erwünscht oder notwendig, unter Überdruck z.B. in einer Stickstoffatmo sphäre, durchgeführt werden. Die Salze der angeführten Thiole, die ebenfalls verwendet werden können, sind z. B. Alkalimetall salze, Erdalkalimetallsalze, Ammonium- oder Aminsalze.
Der a-Hydroxysäure-Teil der erwähnten Verbindungen kann ebenfalls ein asymmetrisches Kohlenstoffatom enthalten und daher in zwei optisch-aktiven Formen, der D- und L-Form, vorkommen. Es versteht sich, dass die vorliegende Erfindung sowohl die D- und L-Form als auch die DL-Gemische mit umfasst.
Alle Reaktionskomponenten, die im Verfahren gemäss vorliegender Erfindung verwendet werden, sind entweder im
Handel erhältlich oder können nach bekannten Methoden oder durch eine Reihe von analogen Methoden, wie sie zur Herstellung solcher Reaktionsmittel eben geeignet sind, hergestellt werden.
Gegebenenfalls während der Reaktion ausfallende Niederschläge werden vom Reaktionsgemisch durch übliche Methoden abgetrennt und die erhaltenen Reaktionsprodukte werden routinemässigen Reinigungsvorgängen unterworfen, wie z. B.
Umkristallisation aus einem geeigneten Lösungsmittel oder Lösungsmittelgemisch.
Die erfindungsgemäss hergestellten neuen Verbindungen können als Säuren nach herkömmlichen Methoden in die pharmazeutisch anwendbaren, im wesentlichen nicht giftigen Salze umgewandelt werden, z.B. durch Reaktion mit Alkalihydroxyd, Alkalibicarbonat, Alkalicarboxylat oder mit einer organischen Base. Bevorzugt werden Natriumsalze. Die bevorzugte Methode besteht darin, die Säure in einem Lösungsmittel aufzulösen, in welchem das Salz unlöslich ist, und zu der Lösung die salzbildende Verbindung oder Base hinzuzufügen, wobei das Salz aus dem Reaktionsgemisch ausfällt.
Die erfindungsgemäss erhaltenen Verbindungen zeigen eine hohe antibakterielle Wirksamkeit und hemmen das Wachstum einer Anzahl von Mikroorganismen, einschliesslich grampositiver und gram-negativer Bakterien. Wie bereits erwähnt, besitzen die erfindungsgemäss hergestellten Verbindungen den Vorteil, selbst bei oraler Verabreichung in der Galle und im Harn in so hoher Konzentration ausgeschieden zu werden, dass das Wachstum der krankheitserregenden Mikroorganismen gehemmt wird. Für die therapeutische Anwendung werden die erfindungsgemäss erhaltenen Cephalosporin-Verbindungen in Form pharmazeutischer Zubereitungen verabreicht, in welchen die betreffenden Verbindungen in Gemisch mit pharmazeutisch geeigneten organischen oder anorganischen, festen oder flüssigen Streckmitteln vorliegen, so dass sie für die orale oder parenterale Verabreichung geeignet sind.
Die pharmazeutischen Zubereitungen können in fester Form als Kapseln, Tabletten oder Dragees vorliegen, oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen. Wenn erwünscht, können den obigen Zubereitungen auch Hilfsstoffe, Stabilisierungsmittel, Netzmittel oder Emulgatoren, Puffer und andere für gewöhnlich verwendete Additive beigemischt werden.
Die Dosierung der erfindungsgemäss erhaltenen Verbindungen wird im allgemeinen vom Alter und vom Zustand des Patienten abhängig sein, doch haben sich durchschnittliche Einzeldosen von etwa 100 mg, 250 mg und 500 mg als wirkungsvoll in der Behandlung von bakteriellen Infektionskrankheiten erwiesen. Im allgemeinen können Mengen zwischen 10 und etwa 1000 mg und sogar noch mehr verabreicht werden.
Beispiel 1
7-[DL-2-Acetoxy-2-(2-thienyl)-acetamido]- 3-(5-methyl
1,3,4-oxadiazol-2-ylthiomethyl)- 3-cephem-4-carbonsäure.
Eine Lösung von 1,0 g 7-[DL-2-Acetoxy-2- (2-thienyl)acetamido]- cephalosporansäure, 0,22 g Natriumbicarbonat und 0,32 g 5-Methyl-1,3,4-oxadiazol-2-thiol in 20 ml Phosphat Puffer (pH-Wert 6,4) wurde 3 Stunden bei 70" C gerührt. Das Reaktionsgemisch wurde mit 200 ml Äthylacetat gewaschen, die abgetrennte wässerige Schicht mit Salzsäure auf einen pH
Wert 1 angesäuert und sodann mit 400 ml Äthylacetat extra hiert. Der Extrakt wurde zuerst mit Wasser, dann mit Na triumchloridlösung gewaschen, getrocknet und auf kleineres
Volumen eingeengt. Der Rückstand wurde mit Äther angerie ben und über Nacht stehen gelassen, wobei 0,4 g der obigen
Verbindung in Form eines schwach braun gefärbten Pulvers erhalten wurden. Fp. 125 bis 1300 C (Zersetzung).
Bei Verwendung anderer entsprechender Ausgangs produkte oder anderer nukleophiler Verbindungen oder deren
Natriumsalze wurden bei Einsatz äquimolekularer Mengen nach analoger Arbeitsweise die folgenden 7-acylierten 3 substituierten Cephalosporin-Verbindungen erhalten:
2) 7-(DL-2-Acetoxy-2-phenylacetamido)- 3-(5-methyl 1, 3,4-thiadiazol-2-ylthiomethyl)- 3 -cephem-4-carbonsäure,
Rohprodukt, Fp. 115 bis 125 C (Zers.);
3) 7-(D-2-Acetoxy-2-phenylacetamido)- 3-(5-methyl 1 ,3,4-thiadiazol-2-ylthiomethyl)- 3 -cephem-4-carbonsäure,
Rohprodukt, Fp. 91 bis 980 C (Zers.).
Beispiel 4
Natrium-7-(DL-2-Acetoxy-2-phenylacetamido)- 3-(5-me thyl- 1 ,3,4-thiadiazol-2-ylthiomethyl)- 3-cephem-4-carboxylat.
Zu einer Lösung von 2 g des nach Beispiel 2 erhaltenen
Rohproduktes in 15 ml Aceton wurde eine Lösung von 750 mg Natrium-a-äthylhexanoat in 3 ml Aceton hinzugefügt. Die
Lösung wurde zur Trockene eingedampft und der erhaltene
Rückstand mit Aether angerieben, wobei 1,9 g des obigen
Natriumsalzes erhalten wurden, Fp. 165 bis 168 C (Zerset zung).
Bei Verwendung anderer entsprechender Ausgangsprodukte oder des Natriumsalzes derselben und der disubstituierten Essigsäuren oder ihrer Säurehalogenide, wurde bei Einsatz äquimolekularer Mengen nach den gleichen Verfahren wie in Beispiel 1 und 4 beschrieben, die folgenden 7-acylierten, 3-substituierten Cephalosporansäuren erhalten.

Die UV-Spektren wurden in Phosphat-Puffer (pH-Wert
6,4) beobachtet, ausser bei den mit ' bezeichneten Proben, die in Wasser beobachtet wurden.
Die erfindungsgemäss erhaltenen, neuen Verbindungen zeigen eine gewisse Wirksamkeit gegen eine Anzahl von Mikroorganismen. Die Wirksamkeit der Verbindungen wird in der minimalen Hemmkonzentration (MHK) angegeben, welche nach der üblichen Serien-Agar-Verdünnungsmethode bestimmt wird. Die Bestimmungen wurden mit Hilfe eines Glukose Bouillon-Nährbodens durchgeführt. Dieser wurde für 24 bis 72 Stunden bei 30 C inkubiert und die MHK der Verbindungen, ausgedrückt in meg/ml Substanz, die das Wachstum der Organismen verhinderte, bestimmt.

Wie schon erwähnt, zeigen einige der erfindungsgemäss erhaltenen Verbindungen hervorragende Vorteile beim
Gebrauch als orales Antibiotikum. Die Natriumsalzverbindun gen der Beispiele 3 und 15 und ein bestimmtes im Handel erhältliches Cephalosporin wurden getestet. 3 mg der betreffen den Verbindungen in 0,5 ml destilliertem Wasser wurden oral an wahllos zusammengestellte Gruppen von 8 Mäusen verab reicht, die vorher mit von Kranken isolierten Staphylokokken infiziert worden waren. Als Kontrolle wurde ebenfalls 8 infizierten Mäusen nur destilliertes Wasser verabreicht.
Zehn
Tage nach der Verabreichung wurde an der Kontrollgruppe eine 100%ige Mortalität beobachtet, während bei den mit den
Natriumsalzen der Beispiele 15 und 3 nur eine 12,5%ige bzw.
25%ige Mortalität festgestellt wurde. Beim Handelsprodukt wurde dagegen eine 37,5 %ige Sterberate beobachtet.

U.S. Patent No. 737,761. Various cephalosporin derivatives have been prepared by the various methods briefly mentioned above and tested for therapeutic use in the treatment of bacterial infections. When the cephalosporin antibiotics are used therapeutically, especially when administered orally, they are not concentrated in sufficient quantity or not absorbed in sufficient quantity to bring about a satisfactory inhibition of the growth of the infectious bacteria. Even if such cephalosporin derivatives are absorbed in very high concentrations, they are often insufficiently active against a number of organisms.
It has now been found that some new cephalosporin compounds, when administered orally, are absorbed in sufficient quantities to inhibit the growth of infectious organisms and are found to be of value in the oral treatment of bacterial infections such as those caused by staphylococci and other infectious microorganisms.
It is therefore an object of the present invention to provide a process for the preparation of new and useful 7-acylated, 3-substituted cephalosporin compounds which are useful as antibacterial agents in the treatment of diseases such as those caused by various microorganisms, including gram positive and gram-negative bacteria, and which are particularly characterized in that, even when administered orally, they are absorbed in a concentration high enough to prevent the growth of the microorganisms.

The new compounds prepared according to the invention can be converted as acids into the pharmaceutically acceptable, essentially non-toxic salts by conventional methods, e.g. by reaction with alkali hydroxide, alkali bicarbonate, alkali carboxylate or with an organic base. The UV spectra were recorded in phosphate buffer (pH

The new compounds obtained according to the invention show a certain effectiveness against a number of microorganisms.
