Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Benzophenonderivaten.

Diese Dibenzazepinderivate lassen sich aus den Benzophenonderivaten der allgemeinen Formel I, z.B. nach dem Verfahren der schweizerischen Patentschrift Nr. 570 403, herstellen.
Diese Dibenzazepinderivate besitzen wertvolle pharmakologische Eigenschaften, die in der genannten Patentschrift näher beschrieben sind.
In den Verbindungen der allgemeinen Formel list Rl als Alkylgruppe z.B. die Methyl-, Äthyl- oder Propylgruppe.
Unter einer Alkylengruppe A wird ein beliebiger zweiwertiger, gesättigter aliphatischer Kohlenwasserstoffrest mit 1 bis 3 Kohlenstoffatomen, wie die Methylen-, Äthylen-, 1-Methyl äthyliden-, Äthylen-, Propylen- oder Trimethylengruppe verstanden; von besonderer Bedeutung ist unter diesen Gruppen die Methylengruppe.
R2 ist als Alkoxygruppe z.B. die Propoxy-, Isopropoxy-, Butoxy-, Isobutoxy-, Pentyloxy-, Isopentyloxy-, Hexyloxyoder Isohexyloxygruppe, vorzugsweise jedoch die Methoxyoder Athoxygruppe, als Monoarylmethoxygruppe z.B. die Benzyloxy-, o-, m- oder p-Chlor-benzyloxy-, o-, m- oder p Methyl-benzoxy-, o-, m- oder p-Methoxybenzyloxy- oder 3,4,5-Trimethoxy-benzyloxygruppe, und als Diarylmethoxygruppe insbesondere die Diphenylmethoxygruppe.
Die Alkylgruppen der Dialkylamino- und N-Alkyl-aralkylaminogruppen R2 sind beispielsweise Propyl-, Isopropyl-, Butyl-, Isobutyl-, Pentyl-, Isopentyl- oder Hexylgruppen und vorzugsweise Methyl- oder Äthylgruppen, und Aralkylgruppen in Diaralkylamino- oder N-Alkyl-aralkylaminogruppen R2 z.B.
Phenäthyl-, z-, o-, m- oder p-Methylbenzyl-, 3-Phenyl-propyla-Methylphenäthyl- und insbesondere Benzylgruppen. Als Polymethyleniminorest ist R2 ein 1-Pyrrolidinyl-, Piperidinooder Hexahydro-lH-azepin-1-yl-rest. Diese Reste können ebenso wie der Morpholinorest durch Äthyl-, Propyl- oder insbesondere Methylgruppen ein- oder mehrfach substituiert sein, sofern die maximale Zahl von 10 Kohlenstoffatomen nicht überschritten wird.
Als Dialkoxymethylgruppe ist R2-A z.B. die Dimethoxymethyl- oder Diäthoxymethylgruppe.
Halogenatome als Substituenten der Ringe B und C sind Fluor-, Chlor- oder Bromatome, während als Alkylgruppen bzw. Alkoxygruppen mit 1 bis 6 Kohlenstoffatomen z.B.
Methyl-, Athyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, Tert.butyl-, Pentyl-, Isopentyl-, 2,2-Dimethyl-propyl-, Hexyl- oder Isohexylgruppen bzw. Methoxy-, Äthoxy-, Propoxy-, Isopropoxy-, Butoxy-, Isobutoxy-, Pentyloxy-, Isopentyloxy-, 2,2 Dimethylpropoxy-, Hexyloxy- oder Isohexyloxygruppen in Betracht kommen. Ein Substituent des Ringes B befindet sich insbesondere in 8-Stellung und ist vorzugsweise Fluor, Brom, die Trifluormethylgruppe und vor allem Chlor. Der Ring C ist vorzugsweise unsubstituiert oder durch Fluor, Chlor oder Brom in beliebiger Stellung, insbesondere jedoch durch Fluor oder Chlor in o-Stellung, substituiert.
Eine bevorzugte Gruppe von Verbindungen der allgemeinen Formel I, die in Dibenzazepinderivate mit wertvollen pharmakologischen Eigenschaften übergeführt werden können, sind solche, die Wasserstoff als Rl, eine niedere Alkoxygruppe als R2 und eine Methylen- oder Äthylen- gruppe als A enthalten, wie das 2-[5-(Methoxymethyl)-, das 2-[5-(Äthoxymethyl)- und das 2-[5-(2-Äthoxyäthyl) s-triazol-4-yl]-5-chlorobenzophenon sowie solche mit einer Benzyloxygruppe R2 und einer Äthylengruppe A, wie das 2-[5-(2-Benzyloxyäthyl)-s-triazol-4-yl]-5-chloro- benzophenon.
In solchen Verbindungen ist vorzugsweise der Ring B in 5 Stellung durch Chlor substituiert und der Ring C unsubstituiert oder in o-Stellung durch Fluor oder Chlor substituiert.
Für die Acylierung der Verbindungen der allgemeinen Formel II kann man als reaktionsfähige funktionelle Derivate von Carbonsäuren der allgemeinen Formel III beispielsweise deren Halogenide, insbesondere Chloride oder Bromide, deren Anhydride oder gemischte Anhydride, beispielsweise mit Kohlensäure-monomethylester oder -monoäthylester, und als Kondensationsmittel z.B. eine tertiäre organische Base, wie Triäthylamin, Äthyldiisopropylamin, Pyridin und insbesondere 2-Methyl-imidazol verwenden und die Acylierung in einem inerten organischen Lösungsmittel, z.B.
in einem Kohlenwasserstoff, wie Benzol oder Toluol, in einem niederen Polyhalogenalkan, wie Methylenchlorid oder Chloroform, in einer ätherartigen Flüssigkeit, wie Dioxan oder Tetrahydrofuran, in einem Amid, wie Dimethylformamid oder N,N,N',N',N",N"- Hexamethyl-phosphorsäuretriamid, oder in Dimethylsulfoxid durchführen.
Von den 2-Amino-benzophenonen der allgemeinen Formel III ist eine grössere Anzahl in der Literatur beschrieben, vgl.
z.B. F. D. Chattaway, J. Chem. Soc. 85, 344 (1904), L. H. Sternbach et al., J. Org. Chem. 26, 4488 (1961) und 27, 3781-3788(1962).
Besonders vorteilhaft ist die Acylierung mit niederen Alkylestern, wie den Äthyl- oder Methylestern, von Carbonsäuren der allgemeinen Formel III, die in Gegenwart eines alkalischen Kondensationsmittels, wie Natriumamid, Lithiumamid und insbesondere Natriumhydrid, in einem inerten organischen Lösungsmittel, z.B. in Dimethylformamid oder vorzugsweise in N,N,N',N',N",N"-Hexamethylphosphorsäure-triamid, beispielsweise bei Temperaturen zwischen 0O und 80OC, vorzugsweise zwischen 200 und 50OC, durchgeführt wird. Die niederen Alkylester werden hierbei in zu den o-Aminobenzophenonen der allgemeinen Formel II äquimolarer Menge oder in geringem Überschuss von 5 bis 20% eingesetzt.
Die Reaktion von Verbindungen der allgemeinen Formel IV mit Hydrazin wird vorzugsweise im Temperaturbereich von 00 bis 1000C, insbesondere bei Raumtemperatur bis mässig erhöhter Temperatur, d.h. zwischen ca. 200 und 600C, durchgeführt. Das Hydrazin kann als solches oder auch in Form des Hydrats in zur Verbindung der allgemeinen Formel IV äquimolarer Menge oder vorzugsweise in grösserem Überschuss eingesetzt werden. Im Überschuss angewendet kann es als alleiniges Reaktionsmedium dienen, doch kann man die Reaktion auch in einem geeigneten organischen Lösungsmittel, z.B.
einem niederen Alkanol, wie Methanol, Äthanol, Propanol oder Isopropanol, durchführen. Gegebenenfalls kann die Reaktion durch Zufügen eines sauren Kondensationsmittels, z.B. einer anorganischen Säure, wie Chlorwasserstoff, Bromwasserstoff, Schwefelsäure oder Phosphorsäure, einer Carbonsäure, wie Essigsäure oder Propionsäure, oder einer Sulfonsäure, wie Benzolsulfonsäure oder p-Toluolsulfonsäure, beschleunigt werden, wobei die benötigte Säure auch in Form der Säurekomponente des entsprechenden Hydrazinsalzes zugefügt werden kann.
Die Umsetzung von Verbindungen der allgemeinen Formel V mit den Halogeniden, insbesondere Chloriden oder Bromiden, oder den Anhydriden von 2-Chlor- oder 2-Brom-alkansäuren, die entsprechend der Definition des in ihnen enthaltenen Rl 2 bis 5 Kohlenstoffatome aufweisen, wie z.B. mit
Chloracetylchlorid oder Chloressigsäureanhydrid, erfolgt beispielsweise im Temperaturbereich von 0O bis 800C, vorzugsweise bei Raumtemperatur oder schwach erhöhter Temperatur, d.h. zwischen ca. 200 und ca. 400C. Als säurebindende Mittel werden insbesondere tertiäre organische Basen, wie Pyridin, 2-Methyl-imidazol, Äthyldiisopropylamin oder Tri äthylamin, ferner anorganische säurebindende Stoffe, wie Natrium- oder Kaliumcarbonat oder -bicarbonat, venvendet.
Die Umsetzung erfolgt in einem grösseren Überschuss einer als säurebindendes Mittel dienenden organischen Base oder in einem inerten organischen Lösungsmittel, z.B. in einem Kohlenwasserstoff, wie Benzol oder Toluol, in einem niederen Polyhalogenalkan, wie Chloroform oder Methylenchlorid, in einer ätherartigen Flüssigkeit, wie Diäthyläther oder Tetrahydrofuran oder in Dimethylformamid. Wenn die Acylierung bei Raumtemperatur durchgeführt wird, erhält man auch bei Venvendung eines grösseren Überschusses an Säurechlorid oder Anhydrid ausschliesslich oder vonviegend das Monoacylierungsprodukt der allgemeinen Formel VII, während unter energischeren Bedingungen gegebenenfalls auch ein oder ein Gemisch von mehreren Diacylierungsprodukten mit an ein Ringstickstoffatom und/oder an das Sauerstoffatom der Hydroxylgruppe gebundenem zweitem Acylrest entstehen kann.
Ferner ist auch Triacylierung möglich, doch lassen sich sowohl Di- wie Triacylierungsprodukte gleich wie die Monoacylierungsprodukte der allgemeinen Formel VII in die entsprechenden gewünschten Verbindungen der allgemeinen Formel I umwandeln, so dass der Acylierungsgrad für die Durchführbarkeit der Reaktionsfolge nicht von ausschlaggebender Bedeutung ist.
Als schwach-saures Medium zur Umwandlung der Acylierungsprodukte von Verbindungen der allgemeinen Formel V, z.B. der Monoacylverbindungen der allgemeinen Formel VII und deren Gemischen mit Di- und Triacylverbindungen, in Verbindungen der allgemeinen Formel I eignet sich insbesondere eine Carbonsäure, beispielsweise eine niedere, gegebenenfalls halogensubstituierte Alkansäure, wie Ameisensäure, Essigsäure, Chloressigsäure oder Dichloressigsäure, eine aromatische Carbonsäure, wie Benzoesäure oder Salicylsäure, oder eine araliphatische Carbonsäure, wie Phenylessigsäure oder Hydrozimtsäure. Niedrige Alkansäuren wie Ameisensäure oder Essigsäure, dienen vorzugsweise zugleich als Reaktionsmedium, doch kann man auch diesen und insbesondere höheren Carbonsäuren ein geeignetes inertes organisches Lösungsmittel, wie z.B.
Benzol, Toluol, Xylole oder Xylolgemische, Chlorbenzol, Chloroform, Tetrachloräthan oder Diäthylenglykoldimethyläther beifügen. Die Reaktion wird z.B. im Temperaturbereich von ca. 20-1600C, vorzugsweise zwischen 800 und 1200C, bzw. Siedetemperatur des Reaktionsmediums, falls diese niedriger ist, durchgeführt und das Reaktionsprodukt der allgemeinen Formel I beispielsweise durch Eindampfen des Reaktionsgemisches im Vakuum gewonnen.
Gewünschtenfalls wird das Chlor- oder Bromatom X der Verbindungen der allgemeinen Formel I durch Jod ersetzt, beispielsweise durch Umsetzung der betreffenden Verbindung mit Natrium- oder Kaliumjodid in einem organischen Lösung mittel, wie Methanol, Äthanol oder Aceton. Die Einführung des Jodatoms kann aber auch in situ unmittelbar vor der weiteren Umsetzung der Verbindung der allgemeinen Formel I mit Ammoniak oder Hexamethylentetramin erfolgen, so wie das in der oben erwähnten Patentschrift beschrieben ist.
Die nachfolgenden Beispiele erläutern die Durchführung des erfindungsgemässen Verfahrens zur Herstellung der Verbindungen der allgemeinen Formel I, sollen jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in Celsiusgraden angegeben. Als Petrol äther wird stets solcher vom Siedebereich 40 bis 65o venven det.
Beispiel 1 a) Eine Lösung von 92,7 g (0,4 Mol) 2-Amino-5-chlor benzophenon in 900 ml abs. Hexamethylphosphorsäuretriamid wird innerhalb 23/4 Stunden portionenweise unter Rühren und Stickstoffatmosphäre mit 63,4 g einer 55%igen Dispersion von Natriumhydrid in Mineralöl und 81,6 g (0,42 Mol) Benzyloxyessigsäure-äthylester versetzt, wobei durch gelegentliches
Kühlen mit einem Eisbad die Temperatur zwischen 25-390 gehalten wird. Hierauf rührt man das Reaktionsgemisch 15 Stunden bei 250. Dann neutralisiert man es unter Eiskühlung durch Zugabe von Eis und wässriger Salzsäure. Die erhaltene Lösung wird mit Äther extrahiert, die Ätherphase mit Wasser und mit gesättigter Natriumchloridlösung gewaschen, über Natriumsulfat getrocknet und eingedampft.
Die Kristallisation des Rückstandes aus Äther-Petroläther ergibt das 2-(2-Benzyloxyacetamido)-5-chlorbenzophenon vom Smp. 82-830.
b) 9,50 g (0,025 Mol) 2-(Benzyloxyacetamido)-5-chlor- benzophenon werden mit 100 ml wasserfreiem Hydrazin 21/2
Stunden bei 250 gerührt. Unter Eiskühlung fügt man 100 ml Wasser zu, filtriert den Niederschlag ab und wäscht ihn mit
150 ml Wasser und 50 ml Äther. Durch Umkristallisation des Filtergutes aus Methylenchlorid-Petroläther erhält man das
3-Amino-2-(benzyloxymethyl)-6-chlor
3 ,4-dihydro-4-phenyl-chinazolin-4-ol von Smp. 176-1810.
c) 0,985 g (0,0025 Mol) 3-Amino-2-(benzyloxymethyl) 6-chlor-3,4-dihydro-4-phenyl-chinazolin-4-ol, 4,92 g (0,06 Mol) 2-Methyl-imidazol und 50 ml abs. Benzol wird unter Rühren bei 0O innerhalb 30 Minuten tropfenweise mit 5 ml einer abs. Benzollösung versetzt, die 2,35 ml (3,34 g, ca. 0,03 Mol) Chloracetylchlorid enthält. Dann wird das Gemisch 11/2 Stunden bei 25o weitergerührt, anschliessend mit 50 ml Wasser versetzt, noch weitere 45 Minuten gerührt und dann 16 Stunden bei 25 stehengelassen. Hierauf trennt man die Benzolschicht ab, wäscht sie mit gesättigter Natriumchloridlösung und dampft sie im Vakuum ein.
Der Rückstand wird an 60 g Kieselgel Merck (Korngrösse 0,063-0,2 mm) mit Benzol Äthylacetat (3 1) als Eluiermittel chromatographiert. Die Fraktionen, die das gewünschte Produkt [Rf-Wert im System Benzol-Chloroform-Athylacetat (4 :4 :2) = 0,24] enthalten, wurden vereinigt, eingedampft und der Rückstand aus Benzol Äther-Petroläther kristallisiert, wobei man das 2-(B enzyloxymethyl)-6-chlor-3- (chloracetamido)- 3,4-dihydro-4-phenyl-chinazolin-4-ol vom Smp. 102-1070 in Form von gelben Kristallen erhält.
d) 142 mg (0,0003 Mol) 2-(Benzyloxymethyl)-6-chlor 3-(chloracetamido)-3,4-dihydro-4-phenyl-chinazolin-4-ol werden mit 2,5 ml Ameisensäure 21/2 Stunden unter Rückfluss gekocht. Dann dampft man die Lösung im Vakuum ein. Der Rückstand wird in Athylacetat gelöst, mit Natriumbicarbonatlösung und gesättigter Natriumchloridlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Das Rohprodukt chromatographiert man an 15 g Kieselgel Merck (Korngrösse 0,063-0,2 mm) mit Äthylacetat-Benzol (1 :2) als Eluiermittel, wobei man in Fraktionen à ca. 10 ml unterteilt.
Die Fraktionen 6-10 wurden vereinigt, eingedampft und der Rückstand [Rf-Wert im System Benzol-Isopropanol (9 :1) = 0,43J aus Äther-Petroläther kristallisiert, wobei man das 5-Chlor-2-[5-(benzyloxymethyl)-3-chlor methyl-5-triazol-4-yl]-benzophenon vom Smp. 1631660 erhält.
Beispiel 2 a) Analog Beispiel la) erhält man unter Verwendung von 94,1 g (0,42 Mol) (p-Methoxybenzyloxy)-essigsäure-äthylester das 2-[2-(p-Methoxybenzyloxy)-acetamido] -5 -chlorbenzophenon, unter Verwendung von 49,6 g (0,42 Mol) Methoxyessigsäure-äthylester das 2-(2-Methoxyacetamido)5-chlorbenzophenon, unter Verwendung von 55,4 g (9,42 Mol) Äthoxyessigsäure-äthylester das 2-(2-Athoxyacetamido)5-chlorbenzophenon, und unter Verwendung von 61,3 g (0,42 Mol) 3-Äthoxypropionsäure-äthylester das 2-(3-Äthoxypropionamido)5-chlorbenzophenon.
b) Analog Beispiel lb) erhält man ausgehend von 10,25 g (0,025 Mol) 2-[2-(p-Methoxybenzyloxy)-acetamido]-5-chlorbenzophenon das 2-[(p-Methoxybenzyloxy)-methyl]-3-amido-3,4-dihydro- 4-phenyl-6-chlor-chinazolin-4-ol; ausgehend von 7,60 g (0,025 Mol) 2-(2-Methoxyacetamido)-5-chlorbenzophenon das 2-(Methoxymethyl)-3 amino-3,4-dihydro-4-phenyl-6-chlor-chlnazolin-4-ol; ausgehend von 7,95 g (0,025 Mol) 2-(2-Äthoxyacetamido)-5-chlorbenzophenon das 2-(Äthoxymethyl)-3-amino- 3 ,4-dihydro-4-phenyl-6-chlor-chinazolin-4-ol; und ausgehend von 8,30 g (0,025 Mol) 2-(3-Äthoxypropionamido)-5-chlorbenzophenon das 2-(2-Äthoxyäthyl)-3 amino-3,4-dihydro-4-phenyl-6-chlor-chinazolin-4-ol.
c) Analog Beispiel 1c) erhält man durch Umsetzung von 2,35 ml (ca. 0,03 Mol) Chloracetylchlorid mit 1,060 g (0,0025 Mol) 2-[(p-Methoxybenzyloxy) methyl]-3-amino-3,4-dihydro-4-phenyl-6-chlor-chinazolin- 4-ol das 2-[(p-Methoxybenzyloxy)-methyl]-3-(2-chloracet- amido)-3,4-dihydro-4-phenyl-6-chlor-chinazolin-4-ol; mit 0,795 g (0,0025 Mol) 2-(Methoxymethyl-3-amino3,4-dihydro-4-phenyl-6-chlor-chinazolin-4-ol das 2-(Methoxymethyl) -3- (2-chloracetamido)-3 ,4-dihydro-4- phenyl-6-chlor-chinazolin-4-ol;
mit 0,830 g (0,0025 Mol) 2-(Äthoxymethyl)-3-amino 3 ,4-dihydro-4-phenyl-6-chlor-chinazolin-4-ol das 2-(Äthoxymethyl)-3-(2-chloracetamido)-3,4-dihydro-4- phenyl-6-chlor-chinazolin-4-ol; und mit 0,865 g (0,0025 Mol) 2-(2-Äthoxyäthyl)-3-amino 3 ,4-dihydro-4-phenyl-6-chlor-chinazolin-4-ol das 2-(2-Äthoxyäthyl)-3-amino-3,4-dihydro-4-phenyl-6chinazolin-4-ol.

b) Analog Beispiel lb) erhält man ausgehend von 10,70 g (0,025 Mol) 2-[2-(p-Methoxy benzyloxy)-acetamido]-5-chlor-2'-fluorbenzophenon das 2-[(p-Methoxybenzyloxy)-methyl] -3 -amino-3 ,4-dihydro- 4-(o-fluorphenyl)-6-chlor-chinazolin-4-ol; und ausgehend von 11,12 g (0,025 Mol) 2 [2-(p-Methoxy- benzyloxy)-acetamido]-5 ,2'-dichlorbenzophenon das 2-[(p-Methoxybenzyloxy)-methyl] - -amino-3 ,4-dihydro- 4-(o-chlorphenyl) -6-chlor-chinazolin-4-ol.

The acylation with lower alkyl esters, such as the ethyl or methyl esters, of carboxylic acids of the general formula III, which are formed in the presence of an alkaline condensing agent such as sodium amide, lithium amide and especially sodium hydride, in an inert organic solvent, e.g. in dimethylformamide or preferably in N, N, N ', N', N ", N" -hexamethylphosphoric acid triamide, for example at temperatures between 0O and 80OC, preferably between 200 and 50OC. The lower alkyl esters are used in an amount equimolar to the o-aminobenzophenones of the general formula II or in a slight excess of 5 to 20%.
The reaction of compounds of the general formula IV with hydrazine is preferably carried out in the temperature range from 00 to 1000C, in particular at room temperature to moderately elevated temperature, i.e. between approx. 200 and 600C. The hydrazine can be used as such or in the form of the hydrate in an amount equimolar to the compound of the general formula IV or preferably in a larger excess. Used in excess it can serve as the sole reaction medium, but the reaction can also be carried out in a suitable organic solvent, e.g.
a lower alkanol such as methanol, ethanol, propanol or isopropanol. If the acylation is carried out at room temperature, the monoacylation product of the general formula VII is obtained exclusively or predominantly even if a larger excess of acid chloride or anhydride is used, while under more energetic conditions, optionally also one or a mixture of several diacylation products with a ring nitrogen atom and / or a second acyl radical bonded to the oxygen atom of the hydroxyl group can arise.
Triacylation is also possible, but both di- and triacylation products, like the monoacylation products of the general formula VII, can be converted into the corresponding desired compounds of the general formula I, so that the degree of acylation is not of decisive importance for the feasibility of the reaction sequence.
As a weakly acidic medium for converting the acylation products of compounds of general formula V, e.g. of the monoacyl compounds of the general formula VII and their mixtures with di- and triacyl compounds, in compounds of the general formula I, a carboxylic acid is particularly suitable, for example a lower, optionally halogen-substituted alkanoic acid, such as formic acid, acetic acid, chloroacetic acid or dichloroacetic acid, an aromatic carboxylic acid such as benzoic acid or salicylic acid, or an araliphatic carboxylic acid such as phenylacetic acid or hydrocinnamic acid. Lower alkanoic acids, such as formic acid or acetic acid, preferably also serve as the reaction medium, but these and especially higher carboxylic acids can also be a suitable inert organic solvent, e.g.
Add benzene, toluene, xylenes or xylene mixtures, chlorobenzene, chloroform, tetrachloroethane or diethylene glycol dimethyl ether. The temperatures are given in degrees Celsius. Petroleum ether with a boiling point of 40 to 65o is always used.
Example 1 a) A solution of 92.7 g (0.4 mol) of 2-amino-5-chloro benzophenone in 900 ml of abs. Hexamethylphosphorsäuretriamid is added in portions over 23/4 hours with stirring and under a nitrogen atmosphere with 63.4 g of a 55% dispersion of sodium hydride in mineral oil and 81.6 g (0.42 mol) of ethyl benzyloxyacetate, with occasional

c) 0.985 g (0.0025 mole) 3-amino-2- (benzyloxymethyl) 6-chloro-3,4-dihydro-4-phenyl-quinazolin-4-ol, 4.92 g (0.06 mole) 2 -Methyl-imidazole and 50 ml abs. Benzene is added dropwise with 5 ml of an abs within 30 minutes while stirring at 0O. Added benzene solution containing 2.35 ml (3.34 g, about 0.03 mol) of chloroacetyl chloride. The mixture is then stirred for a further 11/2 hours at 25 °, then 50 ml of water are added, the mixture is stirred for a further 45 minutes and then left to stand at 25 for 16 hours. The benzene layer is then separated off, washed with saturated sodium chloride solution and evaporated in vacuo.
The residue is chromatographed on 60 g of Merck silica gel (particle size 0.063-0.2 mm) with benzene / ethyl acetate (3 l) as the eluent. The fractions containing the desired product [Rf value in the system benzene-chloroform-ethyl acetate (4: 4: 2) = 0.24] were combined and evaporated, and the residue was crystallized from benzene ether-petroleum ether, the 2 - (B enzyloxymethyl) -6-chloro-3- (chloroacetamido) -3,4-dihydro-4-phenyl-quinazolin-4-ol of melting point 102-1070 is obtained in the form of yellow crystals.

b) Analogously to Example lb), starting from 10.70 g (0.025 mol) of 2- [2- (p-methoxybenzyloxy) acetamido] -5-chloro-2'-fluorobenzophenone, 2 - [(p-methoxybenzyloxy) methyl] -3-amino-3, 4-dihydro-4- (o-fluorophenyl) -6-chloro-quinazolin-4-ol; and starting from 11.12 g (0.025 mol) of 2 [2- (p-methoxybenzyloxy) -acetamido] -5, 2'-dichlorobenzophenone the 2 - [(p-methoxybenzyloxy) -methyl] - -amino-3, 4-dihydro-4- (o-chlorophenyl) -6-chloro-quinazolin-4-ol.
