Spezielle Beispiele für niedere Alkylreste sind die Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl- oder tert. Butylgruppe. Spezielle Beispiele für niedere Alkoxyreste sind die Methoxy-, Äthoxy-, n-Propoxy-, Isopropoxy-, n-Butoxy-, Isobutoxy- und tert.-Butoxygruppe. Spezielle Beispiele für niedere Cycloalkylreste sind die Cyclopropyl-, Cyclobutyl-, Cyclopentyl-, Cyclohexyl-, Methylcyclopropyl- und Dimethylcyclopropylgruppe.
Spezielle Beispiele für niedere Alkanoyloxyreste sind die Acetoxy- und Propionyloxygruppe. Spezielle Beispiele für die Trihalogenmethylgruppe sind die Trifluormethyl-, Trichlormethly- und Chlor-difluormethylgruppe. Als Halogenatome kommen Fluor-, Chlor-, Brom- und Jodatome in Frage. Beispiele für den Alkylenrest CnH2n sind die Methylen-, Äthylen-, 1 -Methyläthylen-, 2-Methyläthylen- und Trimethylengruppe.
Chinazolinone der allgemeinen Formel I werden gemäss der Erfindung dadurch hergestellt, dass man eine Verbindung der allgemeinen Formel:

in der R1 R2, R, R und n die vorstehende Bedeutung haben, erhitzt.
Die Umsetzung der Verbindung der allgemeinen Formel II wird vorzugsweise in Gegenwart einer Säure durchgeführt.
Als Säuren können Mineralsäuren, wie Schwefelsäure und Salzsäure, oder organische Säuren, wie Essigsäure, verwendet werden. Die Umsetzung wird vorzugsweise bei Temperaturen von 90 bis 120 C durchgeführt.
Die Chinazolinone der allgemeinen Formel I sind aufgrund ihrer ausgezeichneten antiphlogistischen und analgetischen Eigenschaften und ihrer geringen Toxizität wertvolle Arzneistoffe sowie wertvolle Zwischenprodukte zur Herstellung anderer Arzneistoffe. Die antiphlogistische Wirkung dieser Verbindungen ist höher als die des 1,2-Diphenyl-3,5-dioxo4-n-butyl-pyrazolidins (Phenylbutazon), während ihre akute, subakute und chronische Toxizität wesentlich niederiger ist.
Die folgenden Beispiele erläutern die vorliegende Erfindung.
Beispiel I
Eine Lösung von 2,9 g 2-(2'-Äthoxyäthylamino)-5-chlor -benzhydrol in 30 ml Essigsäure wird mit 0,9 g Kaliumcyanat versetzt. Das Gemisch wird gerührt und unter Rückfluss erhitzt. Nach dem Abkühlen wird das Reaktionsgemisch in Wasser gegossen. Das erhaltene Gemisch wird mit Chloroform extrahiert. Der Chloroformextrakt wird mit verdünnter Natronlauge und anschliessend mit Wasser gewaschen und dann über wasserfreiem Natriumsulfat getrocknet. Nach Abdestillieren des Lösungsmittels unter vermindertem Druck wird der Rückstand aus Athanol umkristallisiert, wobei 1 -(2'-Äthoxy- äthyl)-4-phenyl-6-chlor-3,4-dihydro-2(1H)-chinazolinon vom Smp. = 127,0 bis 128,5"C erhalten wird.
Beispiel 2
Durch einstündiges Erhitzen eines Gemisches aus 1,58 g 2-(2',2',2'-Trifluoräthylamino)-5-chlor-benzhydrol, 3,0 g Carbaminsäureäthylester und 0,2 g Zinkchlorid auf 180"C wird 2-N-Carbamoyl-(2',2',2'-trifluoräthyl)-amino] -5-chlor- benzhydrol hergestellt, worauf das erhaltene Gemisch noch während weiteren 2 Stunden erhitzt wird. Das Reaktionsgemisch wird nach dem Abkühlen mit Chloroform versetzt, worauf die erhaltene Lösung mit Wasser gewaschen und über wasserfreiem Natriumsulfat getrocknet wird.
Durch Verdampfen des Lösungsmittels unter vermindertem Druck wird ein Rückstand erhalten, der beim Umkristallisieren aus ittha- nol 1-(2',2',2'-Trifluoräthyl)4-phenyl-6-chlor-3,4-dihydro- -2(1H)ihinazolinon, Smp. = 169,5 bis 170,50C, liefert.
Beispiel 3
Nach der in Beispiel 2 angegebenen Arbeitsweise lassen sich auch die folgenden Chinazolinone herstellen.

Specific examples of lower alkyl radicals are methyl, ethyl, n-propyl, isopropyl, n-butyl, isobutyl or tert. Butyl group. Specific examples of lower alkoxy radicals are the methoxy, ethoxy, n-propoxy, isopropoxy, n-butoxy, isobutoxy and tert-butoxy groups. Specific examples of lower cycloalkyl radicals are the cyclopropyl, cyclobutyl, cyclopentyl, cyclohexyl, methylcyclopropyl and dimethylcyclopropyl groups.
Specific examples of lower alkanoyloxy groups are the acetoxy and propionyloxy groups. Specific examples of the trihalomethyl group are trifluoromethyl, trichloromethyl and chlorodifluoromethyl groups. The halogen atoms are fluorine, chlorine, bromine and iodine atoms. Examples of the alkylene radical CnH2n are the methylene, ethylene, 1-methylethylene, 2-methylethylene and trimethylene groups.
Quinazolinones of the general formula I are prepared according to the invention by a compound of the general formula:

The reaction of the compound of general formula II is preferably carried out in the presence of an acid.
The acids used can be mineral acids such as sulfuric acid and hydrochloric acid, or organic acids such as acetic acid. The reaction is preferably carried out at temperatures from 90 to 120.degree.
The quinazolinones of the general formula I are valuable drugs and valuable intermediates for the production of other drugs because of their excellent anti-inflammatory and analgesic properties and their low toxicity. The anti-inflammatory effect of these compounds is higher than that of 1,2-diphenyl-3,5-dioxo4-n-butyl-pyrazolidine (phenylbutazone), while their acute, subacute and chronic toxicity is significantly lower.
The following examples illustrate the present invention.
Example I.
A solution of 2.9 g of 2- (2'-ethoxyethylamino) -5-chloro-benzhydrol in 30 ml of acetic acid is treated with 0.9 g of potassium cyanate. The mixture is stirred and refluxed. After cooling, the reaction mixture is poured into water. The resulting mixture is extracted with chloroform. The chloroform extract is washed with dilute sodium hydroxide solution and then with water and then dried over anhydrous sodium sulfate. After the solvent has been distilled off under reduced pressure, the residue is recrystallized from ethanol, with 1 - (2'-ethoxy-ethyl) -4-phenyl-6-chloro-3,4-dihydro-2 (1H) -quinazolinone having a melting point = 127.0 to 128.5 "C is obtained.
Example 2
By heating a mixture of 1.58 g of 2- (2 ', 2', 2'-trifluoroethylamino) -5-chlorobenzhydrol, 3.0 g of carbamic acid ethyl ester and 0.2 g of zinc chloride to 180 "C for one hour, 2-N -Carbamoyl- (2 ', 2', 2'-trifluoroethyl) -amino] -5-chlorobenzhydrol, whereupon the resulting mixture is heated for a further 2 hours. After cooling, the reaction mixture is treated with chloroform, whereupon the obtained solution is washed with water and dried over anhydrous sodium sulfate.
Evaporation of the solvent under reduced pressure gives a residue which, when recrystallized from itthanol 1- (2 ', 2', 2'-trifluoroethyl) 4-phenyl-6-chloro-3,4-dihydro- -2 ( 1H) ihinazolinone, m.p. = 169.5-170.50C, provides.
Example 3
The following quinazolinones can also be prepared using the procedure given in Example 2.
