Die Erfindung betrifft ein Verfahren zur Herstellung neuer lH-Imidazo[4,5-b]- bzw. .[4,5-cjpyridin-Derivate der Formel I (siehe Formelblatt), worin R1 für Wasserstoff, Chlor, Brom, Fluor oder eine niedere Alkylgruppe, R2 für Wasserstoff oder eine niedere Alkylgruppe, R3 für eine niedere Al Alkylgruppe stehen, X Stickstoff und Y eine Methylidyngruppe oder umgekehrt X eine Methylidyngruppe und Y Stickstoff bedeuten, wobei, wenn X für Stickstoff steht, R2 stets Wasserstoff bedeutet, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I, indem man Verbindungen der Formel II, worin R1, R2, X und Y obige Bedeutung besitzen, mit einer den R,,CO- Rest abgebenden Verbindung umsetzt.
Beispielsweise kann das erfindungsgemässe Verfahren ausgeführt werden, indem man eine Verbindung der Formel II in einem unter den Reaktionsbedingungen inerten Lösungsmittel z.B. in Pyridin löst und mit einem Acylierungsmittel, z.B. mit Acetanhydrid versetzt. Nach Beendigung der Reaktion kann das Reaktionsprodukt aus dem Reaktionsgemisch nach an sich bekannten Methoden isoliert und gegebenenfalls gereinigt werden.
Die Verbindungen der Formel I können in ihre Säureadditionssalze überführt werden und umgekehrt.
Die durch Rl, R2 und R3 symbolisierten niederen Alkylgruppen besitzen 1 bis 4 Kohlenstoffatome und bedeuten insbesondere die Methylgruppe.
Die als Ausgangsprodukte benötigten Verbindungen der Formel II können erhalten werden, indem man Verbindungen der Formel III, worin Rl, R2, X und Y obige Bedeutung besitzen, reduziert.
Beispielsweise kann das Verfahren ausgeführt werden, indem man eine Verbindung der Formel III in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. in einem niederen Alkohol wie Isopropanol aufschlämmt und dann ein Reduktionsmittel, z.B. Natriumborhydrid zusetzt. Nach beendeter Reaktion kann das Reaktionsprodukt aus dem Reaktionsgemisch nach an sich bekannten Methoden isoliert und gegebenenfalls gereinigt werden.
Die als Ausgangsprodukte benötigten Verbindungen der Formel III können erhalten werden, indem man Verbindungen der Formel IV, worin X, Y, R1 und R2 obige Bedeutung besitzen, oxidiert. Beispielsweise kann das Verfahren durchgeführt werden, indem man eine Benzylverbindung der Formel IV in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. in einem cyclischen Äther wie Dioxan löst, die Lösung mit einem Oxidationsmittel, z.B. mit Mangandioxid oder Selendioxid versetzt und bei verschiedenen Temperaturen, deren Bereich sich vom Raumtemperatur bis zur Siedetemperatur des Reaktionsgemisches erstreckt, reagieren lässt. Das Reaktionsprodukt kann nach an sich bekannten Methoden isoliert und gegebenenfalls gereinigt werden.
Die als Ausgangsprodukte benötigten Verbindungen der Formel IV können erhalten werden, indem man eine Verbindung der Formel V, worin Rl, R2, X und Y obige Bedeutung besitzen, mit einem Säureadditionssalz einer Verbindung der Formel VI, worin R4 für eine niedere Alkylgruppe steht, umsetzt.
Beispielsweise kann das Verfahren durchgeführt werden, indem man eine Verbindung der Formel V in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. in einem niederen Alkohol wie Äthanol löst und mit einem Säureadditionssalz einer Verbindung der Formel VI, z.B. mit einem Hydrochlorid versetzt. Die Reaktion kann bei erhöhter Temperatur durchgeführt werden, vorteilhafterweise bei Siedetemperatur des Reaktionsgemisches. Nach Beendigung der Reaktion, z.B. nach ca. 8 Stunden, kann das Reaktionsprodukt nach an sich bekannten Methoden isoliert und gegebenenfalls gereinigt werden.
Soweit die Herstellung der Ausgangsprodukte nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze besitzen bei geringer Toxizität wertvolle pharmakodynamische Eigenschaften und können daher als Heilmittel verwendet werden. Sie entfalten eine antivirale Wirksamkeit, wie sich durch Untersuchungen in Plaque-Reduktionstests unter Verwendung verschiedener Viren zeigen lässt. Diese antivirale Wirksamkeit zeigt sich durch eine Hemmung des Wachstums, die bei einer Konzentration von ca. 20 bis 100 ylml festgestellt wurde. Daher können die erfindungsgemässen Substanzen als Mittel gegen Virusinfektionen verwendet werden.
Als Heilmittel können die Verbindungen der Formel I und gegebenenfalls ihre wasserlöslichen, physiologisch verträglichen Salze allein oder in geeigneten Arzneiformen gemeinsam mit anorganischen oder organischen, pharmakologisch indifferenten Hilfsstoffen verabreicht werden. Beispielsweise werden sie als Bestandteil von Kapseln eingesetzt, die eine Suspension mit einem Gehalt von ca. 5 bis 30% an Wirkstoff enthalten.
In dem nachfolgenden Beispiel, das die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 2-,-A cetoxybenzylimidazo[4,5-b] pyridin
0,5 g 2-a-Hydroxybenzylimidazo[4,5-b]pyridin wurden in einem Gemisch aus 2 ml Acetanhydrid und 2 ml Pyridin gelöst und 24 Stunden bei 25 gehalten. Das Reaktionsgemisch wird im Vakuum zur Trockne eingedampft, in Chloroform aufgenommen und durch eine kurze Säule filtriert (Aluminiumoxid Merck, neutral, Aktivität Stufe III). Nach Umkristallisation aus Aceton/Hexan erhält man die Titelverbindung vom Schmp. 182 - 1840.
Das benötigte Ausgangsprodukt kann folgendermassen erhalten werden: a) 2-Benzylimidazo[4,5-b]pyridin:
5 g 2,3-Diaminopyridin wurden in 100 ml Äthanol gelöst, mit 5,5 g Phenylacetimidoäthylesterhydrochlorid versetzt und die Lösung 8 Stunden unter Rückfluss gekocht. Das Reaktionsgemisch wurde im Vakuum eingedampft, mit Wasser digeriert und der kristalline Rückstand isoliert. Durch Umkristallisation aus Äthanol erhält man die Titelverbindung vom Schmp. 192 - 1940.
b) 2-Benzoylimidazo[4,5-bipyridin:
1 g 2-Benzylimidazo[4,5-b]pyridin wurde in 30 ml Dioxan gelöst, mit 5 g Mangandioxid versetzt und bei 25 60 Stunden gerührt. Zur Vervollständigung der Reaktion wurde anschliessend noch 3 Stunden auf dem Wasserbad bei 50 erwärmt. Das Reaktionsgemisch wurde über Celite filtriert und das Filtrat zur Trockne im Vakuum eingedampft. Nach Umkristallisation aus Propanol erhält man die Titelverbindung vom Schmp. 262 - 2640 (Umwandlungspunkt bei 246 - 2480).
c) 2-a-Hydroxybenzylimidazo[4,5-b]pyridin:
550 mg 2-Benzoylimidazo[4,5-b]pyridin wurden in 50 ml Isopropanol aufgeschlämmt, mit 500 mg Natriumborhydrid versetzt und bei 20 15 Minuten lang gerührt. Das Reaktionsgemisch wurde mit Natriumchlorid-gesättigtem Wasser versetzt, wiederholt mit Essigester ausgeschüttelt, die vereinigten Extrakte mit Natriumsulfat getrocknet und die Essigesterlösung im Vakuum eingedampft. Nach Umkristallisieren aus Aceton/Hexan erhält man die Titelverbindung vom Schmp.

The compounds of the formula IV required as starting materials can be obtained by reacting a compound of the formula V, in which Rl, R2, X and Y have the above meanings, with an acid addition salt of a compound of the formula VI in which R4 is a lower alkyl group .
For example, the process can be carried out by treating a compound of formula V in a solvent which is inert under the reaction conditions, e.g. dissolved in a lower alcohol such as ethanol and treated with an acid addition salt of a compound of formula VI, e.g. mixed with a hydrochloride. The reaction can be carried out at an elevated temperature, advantageously at the boiling point of the reaction mixture. After completion of the reaction, e.g. after about 8 hours, the reaction product can be isolated and, if necessary, purified by methods known per se.
If the production of the starting products is not described, these are known or can be produced by processes known per se or analogously to those described here or analogously to processes known per se.
The compounds of the formula I and their pharmacologically acceptable acid addition salts have valuable pharmacodynamic properties with low toxicity and can therefore be used as medicaments. They develop an antiviral activity, as can be shown by studies in plaque reduction tests using various viruses. This antiviral effectiveness is shown by an inhibition of growth, which was found at a concentration of about 20 to 100 μl. Therefore, the substances of the present invention can be used as agents against viral infections.
The compounds of the formula I and, if appropriate, their water-soluble, physiologically tolerated salts, can be administered as medicaments on their own or in suitable medicinal forms together with inorganic or organic, pharmacologically inert adjuvants. For example, they are used as a component of capsules which contain a suspension with an active ingredient content of approx. 5 to 30%.
In the following example, which explains the invention in more detail but is not intended to limit its scope in any way, all temperatures are given in degrees Celsius.
Example 2 -, - Acetoxybenzylimidazo [4,5-b] pyridine
0.5 g of 2-a-hydroxybenzylimidazo [4,5-b] pyridine was dissolved in a mixture of 2 ml of acetic anhydride and 2 ml of pyridine and kept at 25 for 24 hours. The reaction mixture is evaporated to dryness in vacuo, taken up in chloroform and filtered through a short column (aluminum oxide Merck, neutral, activity level III). After recrystallization from acetone / hexane, the title compound is obtained with a melting point of 182-1840.
The required starting material can be obtained as follows: a) 2-Benzylimidazo [4,5-b] pyridine:
5 g of 2,3-diaminopyridine were dissolved in 100 ml of ethanol, 5.5 g of phenylacetimidoethyl ester hydrochloride were added and the solution was refluxed for 8 hours. The reaction mixture was evaporated in vacuo, digested with water and the crystalline residue was isolated. The title compound of melting point 192-1940 is obtained by recrystallization from ethanol.
b) 2-Benzoylimidazo [4,5-bipyridine:
1 g of 2-benzylimidazo [4,5-b] pyridine was dissolved in 30 ml of dioxane, 5 g of manganese dioxide were added and the mixture was stirred at 25 for 60 hours. To complete the reaction, the mixture was then heated on the water bath at 50 for a further 3 hours. The reaction mixture was filtered through Celite and the filtrate was evaporated to dryness in vacuo. After recrystallization from propanol, the title compound is obtained with a melting point of 262-2640 (conversion point at 246-2480).
c) 2-a-Hydroxybenzylimidazo [4,5-b] pyridine:
550 mg of 2-benzoylimidazo [4,5-b] pyridine were slurried in 50 ml of isopropanol, added with 500 mg of sodium borohydride and stirred at 20 for 15 minutes. Sodium chloride-saturated water was added to the reaction mixture, the mixture was shaken out repeatedly with ethyl acetate, the combined extracts were dried with sodium sulfate and the ethyl acetate solution was evaporated in vacuo. After recrystallization from acetone / hexane, the title compound is obtained with a mp.
