Gegenstand der vorliegenden, Erfindung ist ein Verfahren zur Herstellung des 2-Chlordexamethasons der Formel

Dieses Derivat des Dexamethasons, das d 1,4-2-Chlor- 9a -fluor-16a -methyl-1 1ss, 17a,21 -trihydroxy-3,20-dioxo- pregnadien, besitzt wertvolle pharmakologische Eigenschaften. So weist es eine antiinflammatorische Wirkung im Fremdkörpergranulomtest an der Ratte ab Dosen von 0,03 mg/kg bei peroraler Verabreichung auf, was etwa einem Drittel der entsprechenden Wirkungsintensität beim Dexamethason entspricht. Zur Erzielung einer signifikanten Nebennieren-Hemmung werden jedoch im Vergleich zu Dexamethason etwa 30mal höhere Dosen benötigt. Durch die Einführung von Chlor in 2-Stellung des Dexamethasons wird somit die unerwünschte Nebennieren-Hemmung sehr stark zurückgedrängt.
Die neue Verbindung kann jedoch auch als Zwischenprodukt zur Herstellung anderer pharmakologisch wirksamer Präparate verwendet werden.
Die genannte neue Verbindung kann auch als Tierfutterzusatzmittel verwendet werden.
Die neue Verbindung der vorliegenden Erfindung kann nach an sich bekannten Methoden gewonnen werden. Das Verfahren des vorliegenden Patentes ist dadurch gekennzeichnet, dass man in einer Verbindung der Formel

in welcher R, und R2 ist eine Hydroxygruppe bedeuten, von denen mindestens eine in geschützter Form vorliegt, die vorhandenen geschützten Hydroxygruppen in freie Hydroxygruppen überführt.
Eine geschützte Hydroxygruppe R, und R2 ist insbesondere eine veresterte oder verätherte Hydroxygruppe. Veresterte Hydroxygruppen sind insbesondere solche, die sich von einfachen aliphatischen Carbonsäure wie Essig-, Propionoder Buttersäure ableiten, vorzugsweise auch halogenierten Derivaten derselben, in erster Linie Trifluoressigsäure.
Die verfahrensgemässe Entfernung von Schutzgruppen in 11- und/oder 21-Stellung geschieht in an sich bekannter Weise. So werden veresterte oder verätherte Hydroxygruppen durch basische oder saure Hydrolyse in freie Hydroxygruppen überführt.
Die Ausgangsstoffe sind bekannt oder können in an sich bekannter Weise hergestellt werden. So werden z. B. die obiben Verbindungen der Formel II dadurch hergestellt, dass man ein Derivat des Dexamethasons, worin mindestens die 21-Hydroxygruppe verestert ist, mit Chlor und anschliessend mit einem dehydrohalogenierenden Mittel behandelt, und, gegebenenfalls, in an sich bekannter Weise eine veresterte
Hydroxygruppe in 11ss- oder 21-Stellung selektiv durch alka lische Behandlung verseift.
Die gemäss dem Verfahren der vorliegenden Erfindung herstellbare neue Verbindung kann zur Herstellung von phar mazeutischen Präparaten für die Human- oder Veterinär medizin verwendet werden. Als pharmazeutisches Träger material verwendet man organische oder anorganische Stoffe, die für die enterale, z. B. orale, parenterale oder topische
Gabe geeignet sind. Für die Bildung derselben kommen sol che Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole,
Gummi, Polyalkylenglykole, Vaseline, Cholesterin und andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können in fester Form, z. B. als Tabletten, Dragees oder Kapseln, oder in flüssiger oder halbflüssiger Form als Lösungen, Suspensionen, Emulsionen, Salben oder Cremen vorlie gen.
Gegebenenfalls sind diese pharmazeutischen Präparate sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Die erfindungsgemäss erhältliche neue Verbindung kann auch als Ausgangsprodukt für die Herstellung anderer wertvoller Verbindungen dienen.
In dem folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
47,5 g Dexamethason-1 1 -trifluoracetat-2 1 -acetat werden in 4,5 Liter Dioxan gelöst. Dazu gibt man eine Lösung von 72 g Chlor in einen Liter Propionsäure und lässt bei 10 C stehen. Nach 10 Tagen wird das Reaktionsgemisch auf 15 Liter Wasser und 2 kg Eis ausgetragen, 15 Minuten gerührt, abgenutscht und mit 10 Liter Wasser gewaschen. Das erhaltene farblose Kristallisat wird im Hochvakuum bei 50 C getrocknet. Es stellt das 1,2-Dichlor-1,2-dihydro-dexametha- son-ll-trifluoracetat-21-acetat dar. Durch Auswaschen der Nutsche und des Filters mit Essigsäureäthylester werden weitere 7,5 g desselben Dichlorids erhalten.
57 g rohes 1,2-Dichlor-1,2-dihydro-dexamethason-11- trifluoracetat-21-acetat werden in einem Liter Pyridin gelöst und über Nacht bei Raumtemperatur stehengelassen. Zur Aufarbeitung wird die Reaktionslösung auf 5 kg Eis und 1 Liter Wasser gegossen, 4mal mit 500 ml Methylenchlorid extrahiert, der Extrakt 2mal mit 250 ml 1n Salzsäure und 2mal mit 500 ml Wasser gewaschen. Nach dem Trocknen der Methylenchlorid-Lösung und Eindampfen im Vakuum erhält man rohes 2-Chlordexamethason-ll-trifluoracetat-21-acetat.
Zur Hydrolyse des 1 1-Trifluoracetyl-Restes werden 54,5 g 2-Chlor-dexamethason-ll-trifluoracetat-21-acetat in 4 Liter Methanol gelöst und unter Rühren mit 500 g Natriumazid versetzt. Nach einer Stunde Rühren bei Raumtemperatur wird das Reaktionsgemisch auf 7,5 Liter Wasser gegossen, 4mal mit 500 ml Methylenchlorid extrahiert und der Extrakt 4mal mit Wasser gewaschen. Die über Natriumsulfat getrocknete Methylenchloridlösung hinterlässt nach Verdampfen des Lösungsmittels rohes 2-Chlor-dexamethason-21-acetat. Nach Chromatographie an Alex und Kristallisation aus Methanol schmilzt die reine Verbindung bei 247248 OC unter Zersetzung.
Eine Lösung von 14,1 g 2-Chlor-dexamethason-21 acetat in einem Liter Methanol wird unter Stickstoff bei 0 bis 5" C innerhalb 15 Minuten unter Rühren mit einer Lösung von 4,2 g Pottasche in 50 ml Wasser versetzt. Anschliessend wird mit 50 ml 2n Essigsäure schwach sauer gestellt, im Vakuum bei 40 C bis zur Kristallisation eingeengt und 3mal mit Essigsäureäthylester extrahiert. Nach Waschen des Extraktes mit Wasser, Trocknen über Natriumsulfat und Eindampfen erhält man rohes 2-Chlor-dexamethason, das nach Kristallisieren aus Essigsäureäthylester bei 251-252 C schmilzt, Cr]D20 + 440 (+ 10) in Dioxan.
Das als Ausgangsstoff verwendete Dexamethason-ll-trifluoracetat-21-acetat lässt sich durch Trifluoracetylierung von Dexamethason mittels Trifluoracetanhydrid in Pyridin zum 11,21-Bistrifluoracetat, Spaltung zum ll-Monotrifluoracetat mittels p-Toluolsulfonsäure in Methanol und Acetylierung in 21-Stellung gewinnen.
